Fermentation properties of fermented milk with added cacao nibs (Theobroma cacao L.)

  • Cacao, a major ingredient for making chocolate, has been shown to have potent antioxidant effects. The consumption of raw cacao has beneficial effects on health. This study determined the fermentation properties of yoghurt that was manufactured with cacao nibs (Theobroma cacao L.). The pH of the yogurt containing the cacao nibs was lower than that of the control, and this reduction was both concentration and fermentation time-dependent. Furthermore, the number of lactic acid bacteria in the yogurt made with cacao and its viscosity increased in a cacao nibs dependent manner. In particular, the yoghurt with 5% cacao nibs added had the highest number of lactic acid bacteria at 48 hours at the end point of the fermentation. Whey protein degradation in yogurt containing cacao nibs increased with the fermentation time, and the organic acid production, especially lactic acid, in the yogurt containing cacao nibs increased significantly after 16 hours of fermentation. In addition, the total polyphenol content of the yogurt containing cacao nibs increased, which was proportional to the amount added. Sensory evaluations of the yogurts indicated that overall taste was less desirable as cacao nib loadings increased from 1 to 5% compared with controls. This result is due to the excessive acidity, which was strongly perceived in the absence of added sugar. The development of yogurts with cacao nibs is expected to contribute much to the appeal of fermented milk.

  • 식염농도 3로$15^{\circ}C$에서 발효시킨 물김치에서 젖산균과 효모의 균수변화를 측정한 결과 숙성기까지 효모의 수가 증가한 후 후기에 감소하는 것으로 나타났으며 숙성적기에 효모를 분리동정한 결과 Saccharomyuces sp.가 24주로 대부분을 차지하였으나 Kluyveromyces fragilis 3주, Torulopsis candida 2주도 발견되었다. 이 중에서 S.saitoanus Y17, S.capensis Y29, S.chevalieri Y13, K.fragilis Y2, T.candida Y9를 같은 김치에서 분리한 젖산균 Lactobaccillus plan tarum Lp2, Pediococcus pentosaceus P1, Leuconostoc mesentroides Lu5와 각각 혼합배양하여 효모와 생육에 미치는 영향을 조사한 결과 모두 생육이 억제되었으며, 이는 lacti acid나 H2O2에 의한 억제가 아닌 다른 물질에 의한 것으로 밝혀졌다. 이 세 종류의 젖산균에 의해 영향을 받는 정도는 효모의 종류에 따라 매우 큰 차이를 나타내었다.

배추의 환원당 함량이 김치 발효에 미치는 영향 (Effect of Reducing Sugar Content in Chines Cabbage on Kimchi Fermentation)

  • This study was conducted to investigate and make comparison between treatment which was reduced the reducing sugar content from Chinese cabbage using salting and desalting processes prior to Kimchi fermentation , and control for the effect of reducing sugar content on Kimchi fermentation at $25^{\circ}C$. In the early stage of Kimchi fermentation , the amount of reducing sugar (5.7mg/ml) in treatment was much smaller than that (15.1mg/ml) in control. Reducing sugar content of treatment decreased drastically during the first two days and then levelled off . Whereas, that of control dropped significantly up to the first four days of fermentation. pHs of treatment and control decreased significantly during the first two days and then showed gentle slopes. Acidities of treatment and control were increased continuously during the entire range of fermentation . The acidity of control reached to 0.75% in 3 days of fermentation, while that of treatment was shown after 6 days. Hardnesses of treatment and control using a puncture test were almost constant and the hardness value of treatment was higher than that of control during whole fermentation period. The total bacteria and lactic acid bacteria counts increased drastically during the first day of fermentation and the increase of total bacteria counts was mainly caused by that of lactic acid bacteria counts.

김치숙성 중에 미치는 단삼과 감초의 혼합효과 (The Mixed Effect of Salvia miltiorrhiza and Glycyrrhiza uralensis on the Shelf-life of Kimchi)

  • The Mixed effect of Salvia miltiorrhiza and Glycyrrhiza uralensis(SG) on kimchi fermentation was investigated by measuring changes of physicochemical, icrobiological and sensory qualities of kimchi during fermentation. The pH of SG-added kimchi was a little higher than that of control during the fermentation. Titratable acidity, viable cell of total bacteria and lactic acid bacteria in mixed medicinal herbs(SG) added kimchi were changed more slowly than those in control. The inhibitory effect of the mixture on kimchi fermentation was increased as the concentration of the mixture was increased from 1% to 5%. Total bacteria and lactic acid bacteria of 3% and 5% SG-added kimchi reduced to 1.3~2.9 and 1.2~4.0 log10 cycle after 15 days fermentatin compared to control. The changes in texture of SG-added kimchi was a higher and sour taste of SG-containing kimchi excepts of 1% SG-added kimchi was more weak than that of control. Sensory score of flavor and overall acceptability did not show any significant difference between SG-added kimchi and control during fermentation. But SG-added kimchi decreased its sensory quality by 5% the other kimch.

이온음료 제조를 위한 동치미의 최적 담금 조건에 관한 연구 (Studies on the Optimum Fermenting Conditions of Dongchimi for Production of Ion Beverage)

  • 본 연구의 목적은 이온음료 제조를 위한 동치미 최적 담금 조건을 설정하고자 각 담금 조건 소금농도 2.0, 2.4%, 숙성온도 0.5, $10^{\circ}C$에 따른 동치미액의 성분 분석을 행하였다. pH 감소속도 및 총산도는 동치미의 숙성온도가 높을수록 빠른 증가경향을 나타내었으며 유기산은 lactic acid, citric acid, malic acid가 검출되었고, 그 중 lactic acid 생성량이 가장 많았으며 succinic acid는 $0^{\circ}C$ 숙성 말기에 미량 검출되었다. 젖산균은 염도에 관계없이 숙성초기에 증가하다가 감소하는 경향을 나타내었고 젖산균이 증가하는 기간에 lactic acid 함량도 증가하는 경향을 나타내어 젖산균수와 lactic acid 함량 변화는 일치하는 경향을 보여주고 있다. 휘발성 성분은 3-methyl pentane, ethyl thioethene, 2,3-diaza-indolizine, dimethyl disulfide 등으로 추정되었다. 이온 음료 제조를 위한 최적 담금 조건은 비휘발성 유기산이 최대로 검출되는 최적 숙기인 pH 4.2~3.9, 산도 0.1~0.4에 이르는 동치미로 $0^{\circ}C$에서 24~29일, $5^{\circ}C$에서 9~12일, $10^{\circ}C$에서 16~22 숙성시킨 동치미를 최적 담금조건으로 설정하고자 한다.

Lactobacillus plantarum GBL17 균주를 이용한 복분자 유산발효 특성 (Characteristics of Lactic Acid Fermentation of Black Raspberry Juice Using the Lactobacillus plantarum GBL17 Strain)

  • Lactic acid fermentation of black raspberry (Rubus occidentalis) juice was carried out by using the Lactobacillus plantarum GBL17 strain. The sterilized black raspberry juice was fermented using the L. plantarum GBL17 strain at $30^{\circ}C$ for 72 hours after which the total acidity increased and the pH value decreased. In addition, the highest total acidity content (2.38%) was reached, the lowest pH value (3.22) was observed, and the sugar content decreased by $9.8^{\circ}Brix$ after the 72 hour fermentation. The number of viable cells rapidly increased up until 24 hours, after which it gradually decreased. HPLC analysis of the organic acids showed 14.51 mg/g of lactic acid content in the fermented black raspberry juice, which was not detected in the non-fermented black raspberry juice (control). The content of fructose and glucose slightly decreased after fermentation. The total polyphenol and flavonoid contents of the fermented black raspberry juice increased significantly after fermentation. The DPPH radical scavenging activity of fermented black raspberry juice (70.92%) was higher than that of the control (62.96%). After lactic acid fermentation, there was no significant increase in ABTS radical scavenging activity. These results confirm that lactic acid bacteria, such as L. plantarum GBL17, showed generally higher activities with a potential as a functional beverage.

젖산균과 온도가 김치 발효에 미치는 영향(II) (Effect of Lactic Acid Bacteria and Temperature on Kimchi Fermentation (II))

  • 김치발효 중에 주로 나타나는 Lac. plantarum, Leu. mesenteoides, Ped. acidilactici, Lac. brevis 등의 젖산균들이 김치발효에 미치는 영향을 규명하고자, 이 젖산균들을 순수 배양하여 염도 2.31%의 김치즙에 접종한 후 $30^{\circ}C$, $21^{\circ}C$, $7^{\circ}C$에서 발효시키면서 알코올 당의 변화 및 생균수의 변화를 관찰하고 관능검사를 하였다. Ethyl alcohol은 control group과 Leu. mesenteoides를 접종한 시료, 4종의 젖산균을 혼합 접종한 시료에서 검출되었으며 특히 $14^{\circ}C$에서 더 많은 양이 검출되었다. 당은 발효가 진행됨에 따라 감소하였고, 감소 정도는 pH 및 총산도의 변화와 잘 일치하였다. 총 생균수는 발효초기에는 증가하다가 서서히 감소하는 경향을 보였으며 control group과 Leu. mesenteroides에 있어서 김치가 적당히 익은 정도의 pH및 산도를 보이는 시기와 최고의 균수에 도달하는 시기가 일치하였다. 관능검사 결과 모든 온도에서 Control group이 냄새와 맛이 가장 좋았고 그 다음 4종의 젖산균을 혼합접종한 시료 Leu. mesentenoides를 접종한 시료의 순으로 나타났으며, 온도에 있어서는 $14^{\circ}C$에서 맛과 냄새가 좋았다.

Production of lactic acid by Lactobacillus paracasei isolated from button mushroom bed

  • A galactose fermentation bacterium producing lactose from red seaweed, which was known well to compromise the galactose as main reducing sugar, was isolated from button mushroom bed in Buyeo-Gun, Chungchugnamdo province. The lactic acid bacteria MONGB-2 was identified as Lactobacillus paracasei subsp. tolerans by analysis of 16S rRNA gene sequence. When the production of lactic acid and acetic acid by L. paracasei MONGB-2 was investigated by HPLC analysis with various carbohydrates, the strain MONGB-2 efficiently convert the glucose and galactose to lactic acid with the yield of 18.86 g/L and 18.23 g/L, respectively and the ratio of lactic acid to total organic acids was 1.0 and 0.91 g/g for both substrates. However, in the case of acetic acid fermentation, other carbohydrates besides galactose and red seaweed hydrolysate could not be totally utilized as carbon sources for acetic acid production by the strain. The lactic acid production from glucose and galactose in the fermentation time courses was gradually enhanced upto 60 h fermentation and the maximal concentration reached to be 16-18 g/L from both substrates after 48 h of fermentation. The initial concentration of glucose and galactose were completely consumed within 36 h of fermentation, of which the growth of cell also was maximum level. In addition, the bioconversion of lactic acid from the red seaweed hydrolysate by L. paracasei MONGB-2 appeared to be about 20% levels of the initial substrates concentration and this results were entirely lower than those of galactose and glucose showed about 60% of conversion. The apparent results showed that L. paracasei MONGB-2 could produce the lactic acid with glucose as well as galactose by the homofermentation through EMP pathway.

당의 종류와 호소처리가 분리대두단백으로 제조한 대두요구르트의 품질특성에 미치는 영향 (Effects of Sweeteners and Enzyme Treatments on the Quality Attributes of Soy Yogurt Containing Soy Protein Isolate)

  • This study was carried out to investigate the effects of enzyme treatments on the functional properties of soy protein isolate (SPI) and to examine the quality attributes of soy yogurt prepared by different enzyme treatments, sweeteners and starter cultures. Enzyme treatment increased the solubility and emulsifying capacity of soy proteins, but decreased the emulsifying stability; the enzymatic activity of ${\alpha}$-chymotrypsin was higher than that of trypsin. Enzyme treatments decreased the pH of soy yogurts prepared by both culture methods, the culture of L. bulgaricus and S. thermophilus and the culture of L. bulgaricus and K. fragilis, but increased the titratable acidity, total numbers of lactic acid bacteria and yeast. Trypsin was more effective than ${\alpha}$-chymotrypsin in decreasing pH and increasing titratable acidity and total numbers of lactic acid bacteria and yeast. Fructose decreased the pH of soy yogurts more than sucrose in the culture of L. bulgaricus and S. thermophilus, and vice versa in the culture of L. bulgaricus and K. fragilis. Fructooligosaccharides were more effective in the culture of L. bulgaricus and K. fragilis than in the culture of L. bulgaricus and S. thermophilus in increasing the titratable acidity, total count of lactic acid bacteria and yeast. In sensory evaluation, soy yogurts containing trypsin treated SPI, fructose and fructooligosaccharides (75%:25%) were more acceptable than those containing untreated or trypsin treated SPI and fructose. This was because of more smooth and less sour, in which the values of pH, titratable acidity, microbial growth, and viscosity were in the range of commercial yogurts. Soy yogurts fermented by L. bulgaricus and K. fragilis showed more smooth mouthfeel than those fermented by L. bulgaricus and S. thermophilus.

Quality and Probiotic Lactic Acid Bacteria Diversity of Rabbit Meat Bekasam-Fermented Meat

  • Rabbit meat bekasam is a traditional fermentation product from Indonesia. This study aimed to determine the chemical and microbiological characteristics of rabbit meat bekasam during the fermentation process in order to isolate, characterize (in vitro and in vivo), and identify lactic acid bacteria (LAB) as the probiotic candidate. The chemical contents of bekasam on 7-day fermentation were investigated in explorative and experimental methods in a completely randomized design. A proximate analysis reported a decrease in the moisture content, fat and carbohydrate content, and an increase in protein content. Also, lactic acid content was increased from 0.48% to 1.12%, and pH was decreased from 5.3 to 4.3. Other properties indicated different values, such as bacteria (2.75×106 to 4.45×107 CFU/g), total LAB (3.82×106 to 4.67×108 CFU/g), total yeast (9.89×106 to 3.82×108 CFU/g) and total mould (4.34×101 to 4.86×103 CFU/g). The experiment produced nine LAB isolates, including two probiotics subjected to further 16S rRNA gene analysis, which indicated that Lactobacillus buchneri was the potential probiotic isolate. After being tested on BALB/c mice, L. buchneri could improve the immune system by inhibiting the growth of Coliform and Salmonella.
