Statistics and Probability Distribution of Total Coliforms in Wastewater (하수에서의 대장균수 확률분포 특성 분석)

  • Probability distribution of microbes in wastewater is a crucial factor to be determined for microbial risk assessment associated with its reuse. The objective of this study was to investigate probability distribution of an indicator microorganism in wastewater. Daily total coliform counts measured from nationwide wastewater treatment plants in 2010 by the Ministry of Environment were used for statistical analysis. Basic statistics and probability distributions were estimated in the three different spatial scales using the MS Excel software and FARD2006 model. Overall, wastewater from manure and livestock treatment plants demonstrated greater median coliform counts than from sewage and village treatment plants. Generalized logistic (GLO) and 2-parameter Weibull (WBU2) appeared to be the two probability distributions that fitted best for total coliform numbers in wastewater. The study results of microbial statistics and probability distributions would provide useful data for quantitative assessment of microbial risk from agricultural wastewater reuse.

The Conductance Determination of Total, Coliform and Psychrotrophic bacteria Counts in Raw Milk by Using Malthus (Malthus를 이용한 원유(原乳)내의 총균수, 대장균군수, 저온성균수 측정)

  • This study was performed to obtain fast, consistant and reliable estimation system of bacterial counts of raw milk, which effectively related to the quality of sanitaion and the condition of production at the farm. This study compared regression equation and correlation coefficient relationship between standard plate counts and data of Malthus conductance method for the detection time of total, psychrotrophs, coliform bacterial counts in raw milk. Regression equation (RE) between conductance detection time (Y) and total bacterial log counts (X) was Y=18.27651 - 2.07550X, with correlation coefficient -0.95(n=201). In coliform, RE was Y=9.320848 - 1.15598X with correlation coefficient -0.90 (n=207). Psychrotrophs had the RE of Y=29.96008-3.02487 with correlation coeffecient -0.9 (n=201). This conductance method gave results more quickly and was less labor-intensive than traditional standard plate count method.

Survey of bacterial contamination of chicken meat (도계육에 대한 미생물 오염실태 조사)

  • In order to monitoring the chicken meat hygiene, 165 chicken samples were tested for contamination agents such as the total bacterial counts, coliform bacterial counts and falling bacterial counts. The prevalence of level on number of standard plate count(SPC), total bacterial counts in less than 1033cfu/cm2, 103∼104cfu/cm2 and 104∼10\^5/cfu/cm2 were 30 samples (18.2%), 98 samples(59.4%) and 37 samples(22.4%), respectively. The coliform bacterial counts in less than 102cfu/cm2, 102∼103cfu/cm2 and 103∼104cfu/cm2 were 7 samples(4.2%), 66 samples (40.0%), 92 samples(55.8%), respectively. The falling bacterial counts in plants were higher 9.1, in summer 8.4, in fall 7.2 in spring and 6.7 in winter decreased.

Effect of Microbial Control on Alfalfa Sprout Vegetable by Depending on Sanitization Method in Kindergarten Foodservice (유치원 급식으로 이용되는 알팔파 새싹채소의 소독방법에 따른 미생물제어 효과)

  • Purpose : This study was designed to provide basic data of effective sanitization method of sprout vegetable for microbiological safety. Methods : Sanitization treatments were performed by dipping the sprout into chlorine and vinegar water. Microbial analysis were composed of the total plate count(TPC) and coliform group count(CGC). Result : Among chlorine water sanitization, the microbial reduction was largest in 100ppm chlorine water, and its TPC and coliform group count decreased to 6.01 log CFU/g and 5.06 log CFU/g. The effective dipping time in 100ppm chlorine water treatment was 5.97 log CFU/g and 5.91 log CFU/g for 30min and 60min, in which TPC were below the microbiological safety limits of 6.00 log CFU/g. Coliform group counts were decreased to 5.44, 5.46, 5.42 log CFU/g in the dipping the spouts for 30min, 60min and 90min. As a result of sanitizing alfalfa spout by vinegar water, a microbial counts tended to decrease with increasing concentration and the dipping time. The effective concentration of vinegar water was 2% for TPC(6.00 log CFU/g) and 1% for coliform group count(5.20 log CFU/g). With 1% vinegar water treatment, TPC became below the microbiological safety limits in all samples and in particular, the sample treated for 60min and 90min(4.93, 4.54 log CFU/g). While coliform group counts were decreased to 3.91 log CFU/g in the dipping the sprouts for 90min, those were still beyond the permitted limit. Conclusion : To secure the food safety of food service facilities including kindergarten feeding, it is considered that along with the systematic study of effective disinfection method for microbiological control at the preconditioning level of spout vegetable within the range of secured maleficence to human body, and the study regarding the measures to lower the initial microbiological pollution of spout vegetable.

A Study on the Characteristics of Water Quality for Groundwater and Purified Water of Several Schools in Gyeongsangbukdo Province (경상북도지역 학교지하수 및 정수기통과수의 수질특성에 관한 연구)

  • This study characterized water quality of groundwater and purified water used for drinking water in 30 schools in Gyeongsangbukdo Province. The results of the study showed that 43% of groundwater and 45% of purified water were not suitable for drinking. Among them, microbial contaminations were the most serious problem. In raw groundwater, the exceeding rates of total colony counts was the highest in August (53.6%). Purified water samples showed higher rate of exceeding drinking water standard for total colony counts while showing lower rate of exceeding drinking water standard for total coliforms and fecal coliforms in March and August. Overall, proper managements for microbial contaminations are required for both groundwater and purified water. Furthermore, special attention should toward students not to drink water when groundwater or purified water exceeds the drinking water quality standard.

The Changes on Total Bacterial Counts and Coliform Counts of Human Milk vs Formula Milk Stored under Cold and Room Temperature Conditions (냉장 및 실온 보존에 따른 모유와 환원조제분유 중의 총균수와 대장균수의 변화)

  • Human milk is normally contaminated with various microorganisms, which seem to produce no ill effects. A contamination of hand-expressed and pump-expressed human milk is a major concern in the collection of storage milk. In this study we compared milks collected by two methods, hand expression and suction breast pump, to quantify differences in the degree of bacterial contamination. Thirty-one samples had been manually expressed. The mean of total bacterial counts was 10,600 CFU/mL (range: 360 ∼59,200 CFU/mL) and coliform counts was 43 CFU/mL (range: 20 ∼ 1,060 CFU/mL) in these samples. Whereas in the 118 breast pump-expressed samples, the mean of total bacterial counts was 20,200 CFU/mL (range: 240 ∼ 492,000 CFU/mL) and coliform counts was 158 CFU/mL (range: 4∼10,600 CFU/mL). There was no bacterial growth when the samples were incubated for 10 days at 4\^C. We also compared total bacterial growth in colostrum and in matured human milk for 24 hr at 20\^C and 30\^C. Although bacterial growth had not shown for 24 hr at 20\^C, but shown slight growth in colostrum and rapidly increase in matured human milk for 24 hr at 30\^C. The coliform bacteria in all samples, particulary in formula milk, had grown at 20\^C and 30\^C.

A Study on the storage of Ginseng Powder by r-Irradiation (방사선조사에 의한 인삼분말의 저장성에 관한 연구)

  • To determine the effect of r-irradiation on the microorganisms destruction and the changes of general constituents of Ginseng Powder, three samples were analyzed at 1st and 3rd month after r-irradiation. The results were as follows: 1. Total bacterial counts were decreased during storage as the irradiation dose increased. It seems that about 300 Krad was satisfactory to the regulation of ginseng powder. (5.0×104/g) 2. Coliform group was also decreased during storage as the irradiation dose increased. Coliform group was not detected at the irradiation dose higher than 500 Krad. 3. Total bacterial counts and coilform group were in proportion to the content of moisture in ginseng powder. 4. The contents of moisture, ash, crude protein, crude lipid and total sugar in ginseng powder during storage had nearly no changes. 5. The content of reducing sugar in ginseng powder during storage had the increasing tendencies as the irradiation dose increased. 6. The content of amino-nitrogen in ginseng powder during storage had the decreasing tendencies as the irradiation does increased.

The Effect of Jujubi, Ginseng and Garlic on the TBA value and microbial count of Samgaetang during Refrigerated Storage (대추, 마늘, 수삼이 냉장 저장한 삼계탕의 산패와 미생물증식에 미치는 영향)

  • The objective of this study was to evaluate the effects of ingredients on the change of Samgyetang quality during refrigerated storage. The Samgyetang was prepared with five treatments. The five treatments were chicken cooked alone (Tl), cooked with jujubi, ginseng and garlic (T2), cooked with jujubi (T3), cooked with ginseng (T4) and cooked with garlic (T5). The TBA values of the Samgyetang over 4 days of refrigerated storage were T1(0.89) > T3(0.74) T5(0.74) > T4(0.57) > T2(0.42). The total plate counts of the Samgyetang in the T2 and T3 treatments were lower than with the other treatments. The coliform counts of the Samgyetang in the T2 and T5 treatments were lower than with the other treatments. The results from this study showed that ginseng had an antioxidant activity, jujubi lowered the total plate count and garlic lowered the coliform count in refrigerated Samgyetang. Therefore, the addition of these ingredients maintains the quality of Samgyetang during refrigerated storage.

A Study on the Deterioration of Raw Milk Quality by the Growth of Psychrotrophic Bacteria (원유의 저온성 세균의 증식에 의한 유질변화에 관한연구)

  • A trial was carried out to check the seasonal variation in total bacterial counts (T.B.C.) and psychrotrophic bacterial counts (P.B.C.) and to investigate the quality deterioration of raw milk collected from dairy farms in Kyunggi area, Korea. T.B.C. of raw milk exceeded 107/ml in summer and 106/ml in winter 1980, but they have gradually decreased from 1983. In 1985, the counts showed less than 2×106/ml through the whole year. The same tendency of improvement in coliform counts was shown. The counts were higher than 106/ml in summer and 105/ml in winter 1982, but they were kept lower than 1×105/ml in 1985 through the whole year. Free fatty acids and free amino acids were increased in raw milk stored at 5Cand10C, by the growth of psychrotrophic bacteria.
