Inactivation of Microorganisms and Enzymes in Foxtail Millet Takju by High Hydrostatic Pressure Treatment (초고압 처리에 의한 좁쌀탁주의 미생물 살균 및 효소 불활성화)

  • High hydrostatic pressure was applied to Foxtail Millet Takju to investigate the effects of high pressure on inactivation of microorganisms and enzymes. Total bacteria, lactic acid bacteria and yeast in untreated Takju were $6.8{\times}10^7,\;1.3{\times}10^8\;and\;8.4{\times}10^7\;CFU/mL$, respectively. Total bacterial count in Takju reduced to $2.2{\times}10^5\;CFU/mL$, while lactic acid bacteria and yeast were sterilized completely when heated at $65^{\circ}C$ for 30 min. Lactic acid bacteria and yeast decreased with the increase of treatment pressure, and pressurization of 400 MPa for 10 min at room temperature sterilized completely the lactic acid bacteria and yeast in Takju. Total bacteria were not sterilized with pressurization of even 600 MPa at room temperature. Total bacteria were completely sterilized at $66^{\circ}C/400\;MPa/60\;min\;and\;66^{\circ}C/600\;MPa/10\;min$. Pressurization of Takju caused a partial inactivation of ${\alpha}-amylase$, and after pressurization at 600 MPa for 10 min at room temperature, 73.2% of the original activity remained. The activity of glucoamylase increased with the increase of treatment pressure. Treatment at $66^{\circ}C/400\;MPa/10\;min$ reduced the activity of ${\alpha}-amylase$ by 59.7% and glucoamylase by 20.5%. ${\alpha}-Amylase$ was inactivated to less than 1.2% of the original activity at $66^{\circ}C/600\;MPa/30\;min$.

  • An experiment with alfalfa (Medicago sativa L.) plants was designed to investigate the changes in non-structural carbohydrate (NSC) contents and the activities of amylolytic enzymes during a regrowth period following defoliation. Sampling from hydroponic grown-plants were carried out at intervals during 24 days of regrowth. Shoot regrowth was very slow during the first 10 days and root growth was depressed after defoliation. Defoliation induced a great decrease in both total sugar and starch contents in taproots during the first 10∼14 days. A major recovery of NSC occurred from day 15. Averaged over sampling dates, the activity of exo-amylase was about 400-fold higher than that of endo-amylase. Exo-amylase activity in defoliate plants slightly increased until day 6 (maximum level) and then decreased. Endo-amylase rapidly increased for the first 4 days after defoliation and slightly increased afterwards to a maximum on day 24. These results showed that increase in amylolytic enzyme activity in taproots coincided with the time of starch utilization during regrowth and that indicated it plays an important role in starch degradation.

Effects of Chitosan and Lactic Acid on Enzymatic Activities and Bioactive Compounds during Germination of Black Rice

  • The effect of chitosan on enzymatic activities and on bioactive compounds was characterized during germination at $25^{\circ}C$ for 7 days to search for a method to produce a germinated black rice. The germination rate was reduced by the addition of lactate and chitosan. The rotting rate was greatly decreased by chitosan, suggesting that the addition of chitosan into a germination solution might be an effective method for controlling fungal contamination during the germination of cereals. The addition of 100 and 200 ppm chitosan increased $\alpha$-amylase activity after 7 days by up to 152 % and 197 %, respectively. The activities of $\beta$-amylase and $\beta$-glucosidase were lower with 200 ppm chitosan than in distilled water and 100 ppm lactate. The amount of total soluble phenolics and total flavonoids decreased rapidly for four days and thereafter remained constant until the seventh day. The antioxidant activity of germinated black rice, in terms of hydrogen-donating activity, increased slowly and did not correspond to the changes of total soluble phenolics and total flavonoids. The amount of phytic acid was reduced by the addition of 200 ppm chitosan compared to distilled water, indicating that chitosan could be used as an elicitor for the increase of phytase activity during the germination of black rice.

  • Changes of Enzymatic activities and chemical components during Koji preparation of soy sauce with various ratio of soy bean and wheat were studied as first step for checking the current ratio of raw materials for improved soy sauce and determination of proper ratio of them in the respect to its quality and taste, and following results could be obtained. 1. The Protease in the dryed Koji were mainly conmposed of a part which active at the neutral (about pH 6.0) range, while parts which active at acid and alkaline side were inferior. The more amount of wheat increases as raw materials of Koji, the stronger Protease activities of acid and neural side were, while the weaker alkaline side were. 2. Activity of Enzymes were increased rapidly in earlier stage then gradually in later stage or Protease and ${\beta}-Amylase$ rapidly throughout except drying of ${\beta}-Amylase$ during the course of Koji preparation. The more amount of wheat as raw material increases, the stronger Protease and ${\beta}-Amylase$ activity except. ${\beta}-Amylase$ were. 3. Reducing sugar, amino nitrogen, total nitrogen were increased, while total sugar were decreased during the course of Koji preparation. 4. The more amount of wheat increases as raw materials, the more increase reducing sugar, total sugar were, while the total nitrogen were decreased, no noticeable differences were observed in the amino nitrogen among the dryed Kojies.

Antioxidant Activity and Inhibition Activity against α-Amylase and α-Glucosidase of Viola mandshurica Extracts (제비꽃 추출물의 항산화 활성 및 α-Amylase와 α-Glucosidase에 대한 저해 활성)

  • This study was performed to investigate the physiological activities of Viola mandshurica. Antioxidant activity was evaluated by measuring total phenolic contents, reducing power, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2,2'-azino-di-2-ethyl-benzthiazoline sulphonate (ABTS) radical scavenging activity while anti-diabetic activity was measured by inhibition activities on ${\alpha}$-amylase and ${\alpha}$-glucosidase. V. mandshurica extracts were prepared by extracting with four different solvents (methanol, ethanol, acetone, and water). The methanol extract showed the highest total phenol content (34.49 mg/g gallic acid equivalents) among the extracts. The water extract showed the highest reducing power (0.454) at the concentration of $1,000{\mu}g$/mL. The acetone extract showed the most potent radical scavenging activity. DPPH and ABTS radical scavenging activity of the acetone extract at the concentration of $1,000{\mu}g$/mL were 21.13% and 43.53%, respectively. The inhibitory activity of acetone extracts against ${\alpha}$-amylase and ${\alpha}$-glucosidase showed more than 100% at the concentration of $1,000{\mu}g$/mL. The results indicate that V. mandshurica might have potential antioxidant and anti-diabetic activities.

A Study on Contents of Sugar and the Activities of Amylase in Enzyme Foods and Enzyme-shaped Foods (효소식품과 효소표방식품 중 아밀라아제 활성과 당 함량 조사연구)

  • The purpose of this study was to investigate the contents of sugars and ${\alpha}$-amylase and ${\beta}$-amylase activities in 98 specimen with enzyme foods and enzyme-shaped foods (the other processed foods, beverage bases, fermented drinks, liquid teas). The ${\alpha}$-amylase activity in enzyme foods and the other processed foods were ranged 4.9~53,854.6 U/g and 2.9~1,182.7 U/g, respectively, there was a big difference in the same type. The ${\alpha}$-amylase activity of the fermented products (beverage bases, fermented drinks, liquid teas) were ranged 0.1~1.7 U/g. The average of ${\beta}$-amylase activity in enzyme foods, the other processed foods, the fermented products were found 126.0 U/g, 5.6 U/g and 10.5 U/g, respectively, enzyme-shaped foods were a lot lower than enzyme foods. Total contents of sugars were average 22.4 g/100 g in enzyme foods, 14.8 g/100 g in the other processed foods, 46.9 g/100 g in beverage bases, 41.1 g/100 g in fermented drinks, 39.5 g/100 g in liquid teas, total contents of sugars appeared high amount in the fermented products. Correlations between ${\alpha}$-amylase activity and lactose content was statistically significant in enzyme foods (r = 0.644) and it was strong in the other processed foods (r = 0.903). Correlations between ${\beta}$-amylase activity and lactose content was statistically significant in enzyme foods (r = 0.648) and it was strong in the other processed foods (r = 0.757). There was a significant relationship between ${\alpha}$-amylase and ${\beta}$-amylase activities in enzyme foods and the other processed foods (r = 0.869, r = 0.760). That is, it was found that also the proportional relationship established among the ${\alpha}$-amylase activity, ${\beta}$-amylase activity.

Antioxidant and anti-diabetic effects of Ixeris strigosa extract (선씀바귀 추출물의 항산화 및 항당뇨 효과)

  • Purpose: Ixeris strigosa (IS) is a perennial plant that commonly grows in meadows. The leaves and roots of IS have been used in medicine as a sedative. This study evaluated the antioxidant and carbohydrate-digestive-enzyme inhibitory effects of IS to determine its potential as an essential antioxidant and glycemic inhibitor for type 2 diabetics. Methods: The antioxidative and α-amylase and α-glucosidase inhibitory activities were examined using the water extracts (ISW), ethanol extracts (ISE), and solvent fractions from IS. The antioxidative activities were measured using in vitro methods by measuring the 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity. Results: Investigations of the total polyphenol, flavonoid content, in vitro antioxidant activity, and α-amylase and α-glucosidase inhibitory activities of the IS extract showed that the ISE had higher total phenolic and flavonoid contents than the ISW, as well as high antioxidant activity. The ethanolic extracts of IS (70%) had an α-amylase inhibitory activity of 78.55%. The ethyl acetate fraction (90.56%) showed higher α-glucosidase inhibitory activity than the positive control, acarbose (83.01%). Conclusion: Among the ISE fractions, the ethyl acetate and butanol fractions showed the best digestive enzyme inhibitory activity. Moreover, the antioxidant activity of the extract and the carbohydrate, α-amylase, and α-glucosidase inhibitory effects showed a stronger correlation with the total phenol and flavonoid contents compared to the ISW. As a result, the antioxidant and digestive enzyme inhibitory activities of high ISE are due to the phenolic compounds, particularly the flavonoid compounds. Therefore, ethyl acetate and butanol fractions of the 70% ethanol extract are excellent anti-diabetic functional materials.

Morphological Changes of Cooked Rice Kernel During Saccharification for Sikhe (식혜제조과정 중 밥알의 형태 변화)

  • Growth of acrospire length from germinated covered barley with 1.5∼2.0 times length of buds had the highest amylase activity for 9 days at 15$^{\circ}C$. When the extraction of malt was carried out at 50$^{\circ}C$ for 3.5 hr., total sugar, reducing sugar, sweetness determined by refractometer and amylase activity were the highest, and 2.33%, 1.61%, 3.4 brix(%), 28,332 units, respectively. The sikhe saccharificated at 60$^{\circ}C$ for 8 hr. showed total sugar content increased to 3.90∼9.27% in nonwaxy rice, 4.19∼11.91% in waxy rice, and reducing sugar-content increased 3.30∼7.61% in nonwaxy rice, 3.31∼9.11% in waxy rice. Also, brix was increased to 3.6∼10.8 brix (%) in nonwaxy rice, 3.6∼12.8 brix(%) in waxy .ice, as saccharification time increased. The amylase activity was decreased as saccharification time was increased. And pH was gradually decreased according to time increase, however, it changed little after 4 hr. Morphology of cooked rice kernel during saccharification for sikhe gradually enlarged the oval for hydrolyzed starch granule by increasing saccharification time.

Antioxidant and antidiabetic Activity of Jehotang according to Different Cooking Methods (제조방법에 따른 제호탕의 항산화 및 항당뇨 활성)

  • Jehotang is a cold traditional Korean drink made with honey and several ingredients used in traditional Korean medicine. The ingredients include Fructus mume, Fructus amomi, Fructus tsaoko, Santalum album and honey. In this study, Jehotang and its ingredients were determined through the analysis of antioxidant activity, total phenolic content, ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity. In addition, quality characteristics of Jehotang made by a traditional recipe(Yeonmil) and a modern recipe(double boiling and boiling) were also compared in terms of pH, color and sugar content. Total phenolic content of extract from Fructus ammomi was found to be 120.45 mg, and Yeonmil recipe was discovered 152.66 mg equivalent of gallic acid per g of extract. DPPH free radical scavenging activity were Feuctus amomi(93.13%) and Yeonmil recipe(56.44%). The Fructus amomi extract showed the highest ${\alpha}$-glucosidase inhibitory activity(89.51%) at the concentration of $100{\mu}g/mL$. ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity of boiled Jehotang were 52.38% and 72.52%, respectively. These results suggest that extract of Fructus amomi has an antioxidant activity and antidiabetic effects. Yeonmil recipe is useful for antioxidant effects more than the others. Also, the double boiling recipe has an excellent antidiabetic effect.
