Effects of Spirulina Added Salad Dressing on the Antioxidant Index and DNA Protection in Mice (스피루리나 첨가 샐러드 드레싱 보충 식이가 마우스 혈장 항산화 지표 및 DNA보호에 미치는 영양)

  • The effects of spirulina-added salad dressing on lipid profiles and antioxidant biomarkers such as total glutathionine, TBARS value, carbonyl value, GPx, GR, SOD and paraoxonase activity in plasma or liver of mice were evaluated Sixteen male ICR mice weighing 20$\pm$2 g were divided into two groups and fed low fat ($5\%$ fat) diet (low fat control: LFC) and low fat control plus dressing diet (LFD) for eight weeks. Body weight, tissue weights of liver, heart and kidney, and the distribution of body fat deposition were not significantly different between two groups. Also, the profile of TG, TC, LDL and HDL cholesterol were similar between two groups. The DNA damage was determined using the comet assay (single cell gel assay) with alkaline electrophoresis and quantified by measuring tail length (TL). Spirulina salad dressing consumption resulted in significant decrease in lymphocyte DNA damage expressed by TL (LFC: $28.8{\mu}m$, LFD: $20.3{\mu}m$). Additionally, salad dressing consumption for 8 wks decreased the lipid peroxidation assayed by TBARS to $12.6\%$ compared with the control. The levels of antioxidant vitamins such as $\beta$-carotene were significantly higher in plasma of LFD group than those in LFC group based on HPLC method This study shows that spirulina-added salad dressing exerts degenerative disease-protective effects on oxidative DNA damage and lipid peroxidation possibly via a free radical levels.

Protective effects of dandelion extract against liver damage by repeated administration of thioacetamide in rats (랫드에서 thioacetamide의 반복 투여로 유도된 간 손상에 대한 민들레 추출물의 효과)

  • This study was conducted to investigate of hepatoprotective effect of dandelion water extract (DWE) according to repeated administration of thioacetamide (TAA) induced hepatotoxicity in Spraque-Dawley rats. Thirty rats were randomly assigned to 5 groups; normal control, DWE-control, TAA-control (TAA injection during the feeding of normal diet), TAA&DWE600 (TAA repeated injection during the feeding of DWE 600 mg/kg BW), TAA&DWE1200 (TAA repeated injection during the feeding of DWE 1,200 mg/kg BW). Rats in DWE-control and TAA&DWE groups were treated with DWE (600 or 1,200 mg/kg BW daily) by gavage for 20 days (twice a day). All the rats in the TAA-control and TAA&DWE groups were repeated injection of TAA (100 mg/kg BW) into the abdominal cavity 3 days interval and 12 hrs later, all rats were sacrificed. At the same time, normal control and DWE-control groups were injected normal saline. In TAA&DWE groups, serum alanine and aspartate aminotransferase (ALT, AST) were significantly decreased and triglyceride (TG) synthesis was significantly increased compared to TAA group. As well as total billilubin and GGT were slightly decreased by the treatment of DWE. Lipid peroxidation (MDA) concentration was significantly decreased and hepatic GSH content was slightly or significantly increased in the TAA&DWE groups compared to TAA group. Hepatic anti-oxidative enzyme activities, such as GSH, GST, SOD and catalase were slightly or significantly elevated by the treatment of DWE. According to these results, When dandelion extract was long term supplied, it could be used as a potential protective material for a longer time liver damage by repeated adminstration of the TAA.

Search of Experimental Studies(35) on Anti aging and Anti oxidant in Korea (국내문헌(國內文獻)(35)에 나타난 항노화(抗老化) 및 항산화(抗酸化)의 실험적(實驗的) 연구(硏究)에 대(對)한 검색(檢索))

  • 35 theses conducted in Korea on the topic of senescence or anti-oxidant were classified on the basis of research methods, animals used for the experiments, and research items. Evaluating these research works with respect to the Free Radical Theory, the following conclusions were reached. 1. Of the 17 theses written in the Oriental medicine aspect, three theses used a single herb, nine theses used a complex prescription, four theses concentrated on the usage of the medicinal acupuncture, and one research paper focused on using scientific components. Common objection of these papers were on the verification of the efficacy of herbs. 2. Of the 18 these written in the Western medicine aspect, five theses used a single drug, seven theses conducted a research on the changes due to senescence, and 6 papers were on variety of topics. The main focus of these works were on the mechanism and pathology related to the senescence rather than on the suppression of senescence. 3. Among the theses written in the Oriental medicine perspective, a total of 48 herbs were utilized. 26 of these herbs has a tonification function on the Kidney. Six out of nine complex prescription mentioned above has a function of tonifying the Kidney. 4. With respect to the research subjects used on the experiments, 8 theses have used Senescence-Accelerated mice, 13 theses have used Sprague-Dawley mice, and remaining 7 papers have used human or other animals. 5. These are the categorization of the research items used: the weight (11) and weight changes of the visceral organs (9), the measurement of the content of peroxide-disease (9), the measurement of enzyme vitality (21), the blood and urine test (10), the experiment concerning immune system (3), the influence on the hepatic capability of metabolizing foreign substance (3), the effect on hepatic cell protection (3), the measurement of both the suppression of Free Radical and ability to create Free Radical (2), the measurement of effect of suppresing MDA(malondialdehyde) (4), the effect of eliminating DPPH Radical (2), and experiements about the functions (2). 6. The rate of vitalization of well known anti-oxidants such as Superoxide dismutase (SOD), Protein-bound SH, Nonprotein-bound SH, Glutathione(GSH), Catalase, and etc. were tested in 17 theses. Considering the conclusions mentioned above, the theses related to the senescence published in Korea elected different animals used for experiments, research items and the methods of research, the end result seems to be a lack of objectivity. Thus, I would argue that research methods to overcome such a deficiency need to be developed systematically.

Characteristics of Barley Polyphenol Extract (BPE) Separated from Pearling By-products (보리 도정부산물로부터 분리한 폴리페놀 추출물의 특성)

  • Barley brans, divided into fractions I $(pearlers\;1{\sim}4)$, II $(pearlers\;5{\sim}20)$, and III $(pearlers \;21{\sim}24)$, germ, and broken kernels were collected as pearling by-products produced by an industrial process consisting of 24 consecutive barley pearlers. The pearling by-products were extracted with 75% ethanol, and polyphenol extracts were separated using Sepabeads SP-850 resin. Total polyphenol content was the highest (43.68%) in the polyphenol extract separated from fraction I. Polyphenol compounds analyzed by HPLC were tentatively identified as proanthocyanins and prodelphinidin polymers, which were known to have antioxidative and antimutagenic activities. Antioxidative abilities were estimated to be in the following order: fraction I>germ>fraction II>fraction III. At 0.01% polyphenol extract, angiotensin I-converting enzyme inhibitory activity was the highest in the germ fraction (40.04%). Nitrite-scavenging activity was over 70% in all fractions. Superoxide dismutase-like activity was in the range of $64{\sim}78%$ in fractions I and II.

Effects of Medicinal Plants on the Quality and Physiological Functionalities of Traditional Ginseng Wine (각종 약용 식물 첨가가 전통 인삼주의 품질 특성과 생리기능성에 미치는 영향)

  • The goal of this study was to develop a highly valuable Korean traditional ginseng wine containing various bioactive compounds with good acceptability. The effect of some medicinal plants on the quality and physiological functionality of Korean traditional ginseng wine were investigated. Advanced traditional ginseng wine (AG wine) prepared by addition of 0.5% each Pleuropterus multflorus and Pueraria lobata into the rice mash containing 1% ginseng, 0.4% Fermivin(commercial alcohol fermentation yeast) and Koji (2:1 mixture of nuruk and amylase containing 36 Saccharogenic power per g) showed the highest acceptability and ethanol content (16.8%). Changes of functionalities of the AG wine during fermentation at $25^{\circ}C$ for 30 days were investigated. The highest antihypertensive angiotensin I-converting enzyme inhibitory activity (78.9%) and total acceptability of the AG wine were shown after fermentation at $25^{\circ}C$ for 20 days. However, antioxidant activity, SOD-like activity and fibrinolytic activity of the AG wines were not detected or very low. HMG-CoA reductase inhibitory activity of the AG wine was also shown to be 8.2% and 9.2% after fermentation for 15 days and 20 days, respectively.

Effects of Hijikia fusiforme Extracts on Lipid Metabolism and Liver Antioxidative Enzyme Activities in Triton-Induced Hyperlipidemic Rats (톳(Hijikia fusiforme) 추출물이 고지혈증 횐쥐의 지질대사 및 간 효소활성에 미치는 영향)

  • This study was conducted to investigate the effect of Hijikia fusiforme extracts on serum lipid and liver antioxidative enzyme activities in triton-induced hyperlipidemic rats. Sprague-Dawley male rats divided into 7 groups : We injected saline to a normal group (N), saline and tween 80 to control groups (CS, CT) and tot extracts to experimental groups (CSA, CTEtOH, CTE, CTH) for 7 days and then injected triton at the last day. Serum and liver free cholesterol contents were significantly lower in hexane-treated group (CTH) than control group (CT) whereas serum HDL-cholesterol content was higher in aqueous extract group (CSA) than control group (CS). Total cholesterol and phospholipid contents in serum and liver were lower in aqueous extract group (CSA) than control group (CS). Serum and liver triglyceride contents were significantly lower in ethanol (CTEtOH) and hexane treated group (CTH) than control group (CT). Thiobarbituric acid reactive substances of liver were lower in tot extract groups (CSA, CTEtOH, CTE, CTH) than control groups (CS, CT). Superoxide dismutase activities in liver were significantly lower in aqueous extracts group (CSA) and hexane treated group (CTH) than control groups (CS, CT). Liver catalase activity was the lowest in ethylacetate extract group. These results showed that some Hijikia fusiforme extracts have reduction effect of lipid and antioxidative effect in triton-induced hyperlipidemic rats.

Effects of Schizandra chinensis BAILL on Lipid Lowering and Antioxidant in Hyperlipidemic Rat

  • The present study examined the effects of Schizandra chinensis extract on the serum lipid composition and the antioxidant of rats in which obesity was induced through high fat diet. Fifty male Sprague-Dawely rats weighing 163.91$\pm$4.17g on the average were adjusted to basic diet and laboratory environment and were fed with high fat diet freely for 6 weeks to induce obesity. Forty rats, the final weight of which was 400g, were selected and were divided into a control group(C), treated groups(T I ; body weight of 100mg/kg, TII ; 150mg/kg and TIII ; 200mg/kg), 10 heads of similar weight for each, and test breeding was performed for 4 weeks. During the test breeding, all treated groups were fed with basic diet and difference in intake among the treated groups were maintained to be less than 5%. According to the result, the quantity of Triglyceride in serum was lower in all of the groups treated with Schizandra chinensis than the control group, but the difference was not significant except the treated group of 200mg (P>0.05). The quantity of Total cholesterol in serum was significantly lower in all the groups treated with Schizandra chinensis than in the control group (P<0.05) but differences according to the quantity of Schizandra chinensis applied were not observed. The quantity of HDL-cholesterol was not significantly different among all the groups including the control group (P<0.05) and no regular tendency of change in the quantity was observed according to the quantity of Schizandra chinensis applied. The quantity of LDL-cholesterol was lower in all the groups treated with Schizandra chinensis, but the treated group of 100mg was not significantly different from the control group. The quantity of TBARS in serum was lower in all the groups treated with Schizandra chinensis than in the control group (P<0.05), but no regular tendency of change in the quantity was observed according to the quantity of Schizandra chinensis applied. The quantity of liver TBARS was not significantly different among all the treated groups (P>0.05). The levels of glutathione peroxidase activity (GSH-Px), superoxide dismutase activity (SOD) and catalase activity were higher in all the groups treated with Schizandra chinensis treated group than in the control group (P<0.05), and the treated group of 200mg showed the highest activity among the treated groups.

RAPD Analysis and Antimicrobial Susceptibility of Streptococcus equi subsp. zooepidemicus Isolated from Thoroughbred Horses (더러브렛 말에서 분리한 Streptococcus equi subsp. zooepidemicus의 RAPD 분석 및 약제 감수성)

  • A total of 68 samples were collected including vaginal mucosa (n=66) from Jangsu stud farm, an equine aborted fetus (n=1), and uterine contents (n=1) from Jeju island. Seventeen Streptococcus equi subsp. zooepidemicus (S. zooepidemicus) strains isolated from horses in Korea were identified as S. zooepidemicus by biochemical tests and sodA.seeI specific multiplex PCR. All isolated strains were divided into 4 clusters: group 1 (No. 2, 3, 5, 6, 7, 11, 12, 13, 14, 15), group 2 (No. 4, 9), group 3 (No. 10, 16, 17), and group 4 (No. 1, 8) by RAPD typing. In group 3, No. 10 isolate that was isolated from vaginal mucosa was indistinguishable from No. 16 and 17 isolates, which were isolated from the equine uterine contents and the equine aborted fetus, respectively. The results of this study suggest that a limited epidemiological relationship exists between the strains from Jangsu (No. 10) and Jeju (No. 16 and No. 17). All isolates showed a high susceptibility to ampicillin, cefoxitin, ceftiofur, cephalothin, florofenicol, gentamicin, nalidixic acid, oxacillin, penicillin, tiamulin, tylosin and vancomycin in antimicrobial susceptibility tests. These results may provide the basic information needed to establish strategies for the treatment and prevention of reproductive diseases in mares in Korea.

A Survey of Weed Occurrence and Management on Apple Orchard Fields in Chungnam Province in Korea (충남지역 사과원의 잡초관리방법과 잡초발생특성)

  • This survey was conducted to identify weed occurrence and to investigate weed management methods in the apple orchard fields in Chungnam province including Daejeon and Sejong from April to June and from September to October, 2015. In this survey, 64 weed species in 27 families including 39 annuals and 25 perennials were identified. Based on the importance values, the most dominant weed species in the first survey was Poa annua (6.51), followed by Veronica didyma (5.36), Plantago asiatica (5.36). In the second survey, Stellaria media (5.73), Digitaria ciliaris (5.36), and Rumex crispus (5.18) were dominant. When the 64 weed species were classified by family, the most abundant weed species belong to Compositae (12 species), followed by Poaceae (7 species), Polygonaceae (6 species), and Cruciferae (6 species). These 31 weed species in the most occurred four families accounted for 48% of total weed occurrence. Based on the questionnaire survey in which weed management methods in the apple orchard were asked, applied mowing + herbicide, mowing + tillage, and mowing + sod-culture was methods commonly conducted in apple orchard fields.

Bioaccumulation Patterns and Responses of Fleece-flower; Persicaria thunbergii to Cadmium and Lead

  • Application of phytoremediation in the polluted area to remove undesirable materials is a complex and difficult subject without detailed investigation and experimentation. We investigated the accumulation patterns of cadmium and lead in plants naturally grown, the bioavailability of plants to accumulate these toxic metals and the responses of P. thunbergii to cadmium and lead. The soil samples contained detectable lead (<$17.5_\mu$g/g), whereas cadmium was not detected in the soils of study area. The whole body of Persicaria thunbergii contained detectable lead (<320.$8_\mu$g/g/g) but cadmium was detected only in the stem (<7.$4_\mu$g/g/g) and root (<10.$4_\mu$g/g/g) of P. thunbergii. Cadmium was not detected in Trapa japonica and Nymphoides peltata, whereas lead was detected in T. japonica (<323.$7_\mu$g/g/g) and N. peltata (<177.$5_\mu$g/g/g). Correlation coefficient between lead content in soil and in these plant samples represented positive correlation. The total content of lead in each plant sample increased in the order of N. peltata$\leq$P. thunbergii
