Characteristics of the Inhibitory Action of Protease Inhibitors on the Glucose-6-phosphate Transporter

  • The present paper reports characteristics and specificity of the inhibitory action of $N^{\alpha}-tosyl-L-lysine-chloromethyl\;ketone$ (TLCK) and $N^{\alpha}-tosyl-L-phenylalanine-chloromethyl\;ketone$ (TPCK) on the glucose6-phosphate transporter of rat liver microsomes. The TLCK-induced inhibition was pH dependent. The inhibition constants for TPCK were determined by following pseudo-Lst order reaction mechanism. The inhibition was protected by preincubation with excess amount of glucose-6-phosphate. The results proved that (a) TLCK inactivates the microsomal glucose-6-phosphate transporter, (b) the inhibition results from the modification of sulfhydryl groups of the transporter.

  • For the development of new COX-2 inhibitor, novel compound were synthesized through induction an arylsulfonyl group to 1-position, an arylcarboxamidyl group to 2-position and an alkyl group to 4-position of L-proline. We started from 4-hydroxy-L-proline, the 4-methylphenylsulfonyl of 1-position was introduced through N-tosylation and the carboxylic acid group was protected by esterification. We sucessfully converted to a various derivatives 4b-d for O-alkyl-(or aralkyl)ation of 4-position using silver oxide as catalysis. The 4-alkyloxy-1-tosyl L-prolines 5b-d were synthesized through base-hydrolysis for the deprotection of carboxylic acid. Final compound 1,2,4-substituted pyrrolidines, 4-alkyloxy-2-phenylcarboxamidyl-1-tosyl pyrrolidines 6a-d were synthesized through the condensation of arylamine with 3 and 5b-d using DCC.

Induction and Inhibition of Amphibian(Rana dybowskii) Oocyte Maturation by Proteolytic Enzymes In vitro. (단백질분해효소들의 양서류 난자에 대한 성숙유도와 억제작용에 관하여)

  • Fully grown amphibian oocytes undergo their maturation (germinal vesicle breakdown, GVBD) during in vitro follicle culture when they are stimulated with frog pituitary homogenate (FPH) or progesterone. Present experiments were designed to determine whether proteolytic enzymes are involved in the regulation of the matunation process. Treatment of a $\alpha$ -chymoiyypsin inhibitor, N a -tosyl-L-phenylalanine-chloromethyl-ketone(TP) to the oocytes exhibited a biphasic phenomenon, the induction of the maturation without added hormone at relatively low doses (0.001-1 $\mu$M) and inhibition of the hormone induced oocyte maturation at a high dose (100 $\mu$M). Treatment of a trypsin inhibitor, N a -tosyl-L-lysine-chloromethyl ketone(TLCK) to the oocytes did not induce the maturation, but rather suppressed the hormone induced oocyte maturation in a high dose(100 $\mu$ M). Treatment of exogenous iyypsin to the oocyte induced their maturation without added hormone in a dose dependent manner (0.001-1 $\mu$ M). The data presented here indicate that some proteolytic enzymes play a role in the regulation of the maturation(meiotic arrest or reinitiation) in amphibians.

Stereospecfic Synthesis of Cis-Alkenyl Thioethers of Mercaptolycerol (Mercaptoglycerol 중 cis-alkenyl thioethers의 입체 특이적 합성)

  • Methods were developed to synthesize optically active mercaptoglycerol from optically active isopropylidene glycerols. 1,2-Isopropylideneglycerol was tosylated and the tosyl group displaced with thiolacetate. Base hydrolysis and oxidation gave 1,1'-dithiobis-2,3-isopropylidene-2,3-propanediol. This compound could be used as a source of mercaptoglycerol, or reacted with 1-decenyl lithium to form cis-1-S-dec-1'enyl-2,3-isopropylidene-1-mercapto-2,3-propanediol. The latter is a stereospecific synthetic route to cis-alkenyl thioethers of protected mercaptoglycerol, and it may be useful for the preparation of a thioplasmalogen substrate for plasmalogenase.

  • The chemical engrafting of polymers on particle surface, plays an important role on selective partitioning of micro/nano-particles in the separated layers of liquid media, such as aqueous two phase systems (ATPSs). Three polymers, dextran, poly (ethylene glycol) and albumin were chosen and chemically conjugated to the polystyrene (PS) magnetic microparticle surface. The attachment of long-chained polymer chains which may switch the partition behavior, can be simply performed by SN2 substitution of various polymers having primary amine functional groups, with p-toluenesulfonyl (tosyl)-activated polystyrene magnetic micro-particles. The surface modification of microparticle was probed by infrared microscopy. The distinctive peak represents N-H stretching vibration mode for microparticles after the reaction and it is common for all three polymers examined. The locations of main peaks are similar for all micro-particles but different and distinguishable in fingerprint region.

  • N,N'-Dicyclohexylcarbodiimide (DCC) known as powerful dehydrating reagent in amide or ester synthesis is converted into N,N'-dicyclohexylurea (DCU) during the reaction. In the paper, DCU was recovered from the reaction for the synthesis of the hydrophilic derivative of ${\beta}$-sitosterol, and the purification of the recovered DCU and the dehydration of DCU into DCC were investigated. In the presence of tosyl chloride, (TsCl) and triethylamine (TEA), DCU was converted into DCC, and the optimum molar ratio of [DCU] : [TsCl] : [TEA] was found to be 1.0 : 1.5 : 3.0. Pure DCC was obtained with a 46% yield by the sublimation after the purification process, and characterized by GC/MS, FT-IR and $^{13}C-NMR$.
