Infection Route of Bacterial Wilt of Tomato Caused by Ralstonia. solanacearum in Hydroponic Culture (수경재배에서 토마토풋마름병의 전염경로)

  • Hydroponic culture has been developed to control soilborn diseases, to increase yield and to enhance the quality of vegetable, The pathogen could be detected from infected plant materials, hydroponic tanks, culture solution and solid media of the severely infected greenhouse, The density of pathogen population was coincided with the severity of disease incidence, For example, 1,900cfu m$L^{-1}$ pf pathogens were counted from tomato plants sampled in a 20% diseased greenhouse. The pathogens may be introduced in the greenhouse through the contaminated soil surrounding the house and/or through the infected young seedlings grown on the nursery soil. Also, not detected to Ralstonia solanacearum from tomato seeds (House Momotaro, Bbaebbae, Ggoggo, and Minicarol cultivar) selling at a market.

Contamination Level of Ralstonia solanacearum in Soil of Greenhouses Cultivating tomato Plants in Chungbuk Province and Characteristics of the Isolates (충북지역 토마토 시설재배지의 풋마름병균(Ralstonia solanacearum) 오염도 및 분리균주의 특성)

  • Contamination level and characteristics of Ralstonia solanacearum in soil of greenhouses cultivating tomato plants in Chungbuk province was determined. R. solanacearum was isolated with the semiselective media in 27 greenhouse soil samples out of 133 greenhouses soil investigated, which indicates 20.3 % of tomato cultivating greenhouses in Chungbuk contaminated with the bacterial wilt pathogen. Density of R. solanacearum was estimated to 10$^{2.4}$ cfu/g in the contaminated soil. All 71 isolates of R. solanacearum which containing 12 isolates from the diseased tomato plants were race 1, ann 35 isolates of them were biovar 3 and 36 isolates were biovar 4. More than 88% of 71 isolates were inhibited growth on nutrient agar containing oxolinic acid 0.5 $\mu\textrm{g}$/ml, streptomycin 25 $\mu\textrm{g}$/ml, tetracycline 5 $\mu\textrm{g}$/ml and cupric sulfate 375 $\mu\textrm{g}$/ml (1.5 mM). The 11.3%, 4.2% and 5.6 % of the isolates can grow on nutrient agar containing 10 times more oxolinic acid, streptomycin, tetracycline than minimal inhibitory concentration of the sensitive strains. Five isolates were resistant to 2 bactericides and one isolates was resistant to all 3 bactericides.

  • Fourteen isolates of Pseudomonas solanarearum virulent to tobacco cultivars were selected among isolates obtained from tobacco plants shelving wilt symptoms throughout the country during $1980\~1981$. The fourteen isolates were classified into two races based on reactions in eggplant, tomato, red pepper, potato and tobacco, and also classified into biochemical type 1 and type 4 according to physiological characteristics.

An Antifungal Agent Produced by Bacillus thuringiensis BK4, an Antagonistic Bacterium against Fusarium Wilt Disease of Tomato (항진균성 항생물질을 생산하는 Bacillus thuringiensis BK4의 항생물질 정제와 토마토 시들음병의 효과적인 방제)

  • The optimum production condition for the antibiotic from Bacillus thuringiensis BK4 was determined, and the suppression rate of Fusarium-wilt by the butanol-extracted antibiotic was verified by employing tomatoes in vitro and in vivo pot tests. Cell growth and antifungal activity were the best when 0.5% xylose and 0.2% peptone No.3 were given as carbon and nitrogen sources, respectively, in the presence of 5mM $CaCl_2$. The partially purified antibiotic successfully prevented Fusarium oxysporum pathogen in pot experiments. When the pots were treated with both live cells and the partially purified antibiotic, an additive-effect was seen in the suppression of Fusarium-wilt, but synergistic effect was not detected. The antibiotic, denoted BK4, purified by Sephadex LH-20 column chromatography was eluted with a single peak at a retention time of 38 min. on prep-HPLC; Minimum inhibition concentration of the homogenous antibiotic was determined to be 50${\mu}$g/ml.

  • The kinds of crop requested from agricultural actual places of famers, Agricultural extension services and so forth was 8 including red pepper for vegetables, 4 including apple for fruit trees and 6 including chrysanthemum for flowers in 2010. The important vegetables in clinical diagnosis of viral diseases were tomato, watermelon and red pepper having the requested rate of 31.8%, 21.4% and 19.5%, respectively. On fruit trees, grape and apple were most common with the requested rate of 63.6% and 33.0%, orderly. On floral crops, tulip and cactus were damaged by viral diseases with the requested rate of 60.0% and 20.0%, orderly. On peppers and tomatoes, six viruses including Cucumber mosaic virus (CMV) and Broad bean wilt virus 2 (BBWV2) infected. Five viruses including Melon necrotic spot virus (MNSV) and CMV were identified from watermelons. On grapes, six viruses including Grapevine fleck virus (GFkV) infected. CMV was identified from six vegetables including pepper out of 8 kinds of vegetables and tulip plant. Total agents of virus and viroid species were 32 and 4 species, respectively, in 2010. Tomato yellow leaf curl disease by Tomato yellow leaf curl virus and Tobacco yellow leaf curl virus was occurred newly at 18 Si/Gun areas including Buan, Jeonbuk province in 2010 and the total areas were increased up to 58 Si/Gun from the first incidence in 2008. Tomato spotted wilt virus (TSWV) occurred newly at two areas of Jinan, Jeonbuk and Jeju in Jeju province in 2010, and the incidence areas were expanded to 25 Si/Gun areas from severe occurrence at Anyang area in 2004. No incidence of TSWV was recorded only in Gyeongbuk and Chungbuk province. Tomato bushy stunt virus occurred newly at Jinju, Gyeongnam, and it had the total incidence areas of 5 Si/Gun after first observation at Sacheon, Gyeongnam in 2004.

Application of Multiplex RT-PCR for Simultaneous Identification of Tomato Spotted Wilt Virus and Thrips Species in an Individual Thrips on Chrysanthemum (시설재배 국화에서 총채벌레의 종 동정 및 보독 바이러스 동시 검출을 위한 다중 진단법 적용)

  • We have developed a simultaneous diagnostic method that can identify both the species of thrips and tomato spotted wilt virus (TSWV) that are problematic in chrysanthemum plants. This is a method of amplifying DNA by performing reverse transcription-polymerase chain reaction by simultaneously adding primers specific to TSWV coat protein (N) gene and primers specific to the internal transcribed spacer 2 region of Frankliniella occidentalis and F. intonsa using total nucleic acid extracted from one thrips. The sizes of DNA fragments for TSWV, F. occidentalis, and F. intonsa were 777, 287, and 367 bp, respectively. These results showed species identification of thrips and whether thrips carrying TSWV can be simultaneously confirmed. Further usefulness of the simultaneous diagnostic method was made from greenhouse survey at chrysanthemum greenhouses in Taean (Chungcheongnam-do) and Changwon (Gyeongsangnam-do) to investigate the identification of thrips species and the rate of thrips carrying TSWV. Of thrips collected from the greenhouses, 83.7% thrips was F. occidentalis and 72.9% F. occidentalis carried TSWV in Taean. Similarly, the diagnostic method showed that 92.2% thrips was F. occidentalis and 84.0% F. occidentalis carried TSWV in Changwon. These results confirm that F. occidentalis is a dominant thrips species and the thrips species plays a crucial role in the transmission of TSWV in chrysanthemum plants in the greenhouses. Taken together, this study showed a simple diagnostic method for thrips identification and epidemiological studies of the timing and spread of TSWV through thrips in chrysanthemum greenhouses in South Korea.

Antibacterial Activity of Pharbitin, Isolated from the Seeds of Pharbitis nil, against Various Plant Pathogenic Bacteria

  • This study aimed to isolate and characterize antibacterial metabolites from Pharbitis nil seeds and investigate their antibacterial activity against various plant pathogenic bacteria. The methanol extract of P. nil seeds showed the strongest activity against Xanthomonas arboricola pv. pruni (Xap) with a minimum inhibition concentration (MIC) value of $250{\mu}g/ml$. Among the three solvent layers obtained from the methanol extract of P. nil seeds, only the butanol layer displayed the activity with an MIC value of $125{\mu}g/ml$ against Xap. An antibacterial fraction was obtained from P. nil seeds by repeated column chromatography and identified as pharbitin, a crude resin glycoside, by instrumental analysis. The antibacterial activity of pharbitin was tested in vitro against 14 phytopathogenic bacteria, and it was found to inhibit Ralstonia solanacearum and four Xanthomonas species. The minimum inhibitory concentration values against the five bacteria were $125-500{\mu}g/ml$ for the n-butanol layer and $31.25-125{\mu}g/ml$ for pharbitin. In a detached peach leaf assay, it effectively suppressed the development of bacterial leaf spot, with a control value of 87.5% at $500{\mu}g/ml$. In addition, pharbitin strongly reduced the development of bacterial wilt on tomato seedlings by 97.4% at $250{\mu}g/ml$, 7 days after inoculation. These findings suggest that the crude extract of P. nil seeds can be used as an alternative biopesticide for the control of plant diseases caused by R. solanacearum and Xanthomonas spp. This is the first report on the antibacterial activity of pharbitin against phytopathogenic bacteria.

Thrips and TSWV Occurrence in Geographically Different Open Fields Cultivating Hot Peppers (상이한 지역별 노지 고추재배지의 총채벌레 연중 발생 및 토마토반점위조바이러스 발병)

  • Thrips infest hot peppers (Capsicum annuum) cultivating in open fields and give serious economic damages. This study reports their yearly occurrence from transplanting to harvest at three different places in Andong, an intense hot pepper-cultivating area. Two main occurrence peaks were detected in June and September. Two dominant thrips were the flower flowers, Frankliniella occidentalis and F. intonsa, which comprised of over 87% of the total occurring thrips. Other thrips did not follow the occurrence peaks of the two dominant species. Tomato spotted wilt virus (TSWV) was detected from the dominant two species, in which higher viruliferous rate was recorded in F. intonsa. Although the hot peppers were the resistant varieties against TSWV, some of them cultivating in the fields exhibited a characteristic disease symptom infected by the virus over the growing seasons. TSWV was isolated from the viruliferous thrips and assessed in NSs sequences encoded in S RNA segment of the virus. Compared to the known resistance breaking (RB) strains, the TSWV isolated from the viruliferous thrips in Andong did not show any RB mutations.

Physiological, Biochemical and Genetic Characteristics of Ralstonia solanacearum Strains Isolated from Pepper Plants in Korea (고추에서 분리된 Ralstonia solanacearum 계통의 생리, 생화학 및 유전적 특성)

  • Totally sixty three bacteria were isolated from lower stems showing symptoms of bacterial wilt on pepper plants in 14 counties of 7 provinces, Korea. The isolates showed strong pathogenicity on red pepper (cv. Daewang) and tomato (cv. Seogwang) seedlings. All virulent bacteria were identified as Ralstonia solanacearum based on colony types, physiological and biochemical tests and polymerase chain reaction (PCR). All R. solanacearum isolates from peppers were race 1. The bacterial isolates consisted of biovar 3 (27%) and biovar 4 (73%). Based on polymorphic PCR bands generated by repetitive sequence (rep-PCR), the 63 R. solanacearum isolates were divided into 12 groups at 70% similarity level. These results will be used as basic materials for resistant breeding program and efficient control against bacterial wilt disease of pepper.
