Occurrence and Biovar Classification of Bacterial Wilt Caused by Ralstonia solanacearum in Eggplant (Solanum melongena) (가지의 Ralstonia solanacearum에 의한 풋마름병 발생과 생리형의 분화)

  • Batcterial wilt caused by Ralstonia solanacearum is one of important and widespread diseases worldwide as well as in Korea. Bacterial wilt disease caused by R. solanacearum has been reported mainly in solanaceous crops including eggplant (Solanum melongena), tomato (Solanum lycopersicum), potato (S. tuberosum), and pepper (Capsicum annuum). A total of 48 strains of R. solanacearum from eggplant were collected during 2005 and 2006. They were confirmed as R. solanacearum by PCR amplification with primer pair flipcF/flipcR resulting in production of 470-bp DNA fragment. The 15 isolates exhibited pathogenicity on eggplant and tomato, but less virulent on pepper than other species. The biovar of collected isolates, which have been reported of five types worldwide, were classified as biovars 3 and 4 by physiological test. Biovar 4 was the dormant type without pathogenicity on eggplant rootstock, whereas biovar 3 had pathogenicity on eggplant rootstocks that is resistant to R. solanacearum, indicating necessity of breeding new rootstock with resistance to R. solanacearum biovar 3

Screening of Tomato Cultivars Resistant to Bacterial Canker by Seedling Test (유묘검정법을 이용한 궤양병 저항성 토마토품종 선발)

  • Bacterial canker, caused by Clavibacter michiganensis subsp. michiganensis, is a very damaging disease to tomato (Lycopersicon esculentum) farm in Korea. It infects tomato, spreads through the xylem and causes bacterial wilt and canker. Selection of resistant cultivar is the best way to prevent or reduce the occurrence of the disease. Thirty-nine tomato cultivars, twenty-one cherry tomato cultivars and thirteen rootstock tomato cultivars were inoculated with Clavibacter michiganensis subsp. michiganensis, to evaluate tomato cultivarspecific resistance against bacterial canker. In the evaluation of 73 major commercial cultivars, 'Sunmyung', 'Sweet', 'Akiko', 'Dadaki', 'Match', 'Magnet', 'Friend', and 'Greenpower' were found to have a high level of resistance to bacterial canker of tomatoes.

Isolation, Identification and Biological Control Activity of SKU-78 Strain against Ralstonia solanacearum (풋마름병균, Ralstonia solanacearum의 길항세균 SKU-78 균주의 분리 동정 및 특성)

  • Six stains of plant growth promoting rhizobacteria were selected through germinating seed assay and root colonization assay. Among them, SKU-78 strain induced significant suppression of bacterial wilt disease in tomato and pepper plants. Seed treatment followed by soil drench application with this strain resulted in over 60% reduction of bacterial wilt disease compared with the control. It was suggested that SKU-78 strain activated the host defense systems in plants, based on lack of direct antibiosis against pathogen. According to Bergey's Manual of Systemic Bacteriology and 16S rDNA sequence data, SKU-78 stain was identified as Bacillus sp. SKU-78.

Tomato spotted wilt virus Isolates Giving Different Infection in Commercial Capsicum annuum Cultivars

  • $Tomato$ $spotted$ $wilt$ $virus$ (TSWV)-infected $Capsicum$ $annuum$ plants were collected from open fields during June to July in 2010. The TSWV isolates were designated as Gneung, Ghang-Kjj, Gchang-Njc, Ghae, and Pap. The nucleotide sequence of the nucleocapsid protein (N) and movement protein (NSm) of the five isolates was determined. The pathogenicity of the five isolates was determined on 14 $C.$ $annuum$ cultivars two times by using mechanical inoculation. The five isolates induced different response: Both Gneung and Gchang-Kjj did not infect any of the cultivars in the 2nd trial, while Gchang-Njc, Ghae and Pap infected 11, 6 and 13 of 14 cultivars, respectively. The five isolates also were tested on $Solanum$ $lycopersicum$ breeding line TGC09-71 and three $Nicotiana$ species. $S.$ $lycopersicum$ showed a similar response to the five isolates as did $C.$ $annuum$. Both Gchang-Njc and Ghae infected systemically all three $Nicotiana$ species tested. While both Pap and Gneung did not infect any of the $Nicotiana$ species tested. In conclusion, five TSWV isolates induced different infection spectra in $C.$ $annuum$ cultivars, $Nicotiana$ species and an $S.$ $lycopersicum$ breeding line. Amino acid sequence analysis of the NSm gene could not support or explain the different infection spectra of the five isolates. This study indicated that various isolates must be used as virus inocula for evaluation of $C.$ $annuum$ and $S.$ $lycopersicum$ cultivars in breeding programs for TSWV resistance.

An Unusual Potyvirus from Pepper in Taiwan (대만에서 고추에 발생한 미보고 Potyvirus에 관한 연구)

  • A virus which induced yellowing, vein banding and ruffling on pepper in the field was investigated. The virus reacted strongly with PVY - antiserum in ELISA, but not with antisera of cucumber mosaic virus, tobacco mosaic virus, tomato black ring virus, alfalfa mosaic virus, tomato spotted wilt virus, tobacco etch virus, pepper mottle virus, and tobacco ringspot virus. Electron micrographs revealed that the virus was a flexuous rod of 750-760nm in length. The virus was transmitted mechanically and by Myzus persicae in a nonpersistent manner. The host range was similar to that of PVY, except that Chenopodium amaranticolor and C. quinoa were infected systemiclly.

Viruses and Symptoms on Peppers, and Their Infection Types in Korea (우리나라 고추 바이러스 종류, 병징 및 발생 형태)

  • The incidence of virus disease on peppers was investigated at the 52 areas in the whole country in 2002, 2004 to 2006. Among the six viruses, Cucumber mosaic virus (CMV), Pepper mottle virus (PepMoV), Pepper mild mottle virus (PMMoV), Broad bean wilt virus 2 (BBWV2), Tobacco mild green mosaic virus (TMGMV) and Tomato spotted wilt virus (TSWV), infecting peppers in Korea, the incidence of CMV, PepMoV, PMMoV and BBWV2 was 29.4%, 13.6%, 14.3%, 25.6%, orderly. TMGMV and TSWV had the same low infection rate of 1.0%. The infection rate of CMV was higher as 53.3% and 34.2% in 2002 and 2004, but it was decreased to 18.2% and 11.9% in 2005 and 2006, respectively. The infection rate of BBWV2 was lower as 1.3% in 2002 and 1.8% in 2004, but it was increased abruptly to 41.3% in 2005 and 58.2% in 2006. For the types of mixed infections of pepper viruses, the incidence of CMV+PepMoV was 62.6% in 2002 and 50.0% in 2004, and that of CMV+BBWV2 was increased suddenly from 33.3% in 2005 to 83.2% in 2006. The triplex infection of CMV+BBWV2+PepMoV was 6.4% in average. CMV caused severe mosaic and BBWV2 induced ring spots, and the two mixed virions caused chlorosis on the leaves of red peppers. TSWV induced the typical symptoms of multiple ring spots on the leaves and fruits of red peppers.
