Gray Mold Rot of Eggplant Caused by Botrytis cineraea in Greenhouse (시설재배에서 Botrytis cinerea에 의한 가지 잿빛곰팡이병)

  • Botrytis cinerea E12 was isolated from the leaves, flowers and fruits of eggplant in the greenhouse in Halrim, Kimhae and Dejeo, Pusan. The leaves infected with the pathogen were appeared initially brown-color, small gray spots at the edge, and finally fall down. The fruit was showed the symptoms of circular or irregular shapes, followed by sunken. When the symptoms were developed, the conidia formed on the surface with gray color. To determine the pathogenicity of B. cinerea E12 against the eggplants, the conidia were suspended with 30% tomato juice, PDB and sterile water, respectively. The result showed that the conidial suspension with 30% tomato juice was highly effective on the pathogenicity as more than 90%. Moreover, the symptoms caused by inoculum were the same as those of wild-type pathogen.

  • In an attempt to find a satisfactory medium which would facilitate abundant conidial production of Piricularia oryzae agar and liquid media containing various nutrients were tested as to the efficiency of conidial production. Among the media tested most number of conidia occurred on V-8 juice broth and tomato juice agar and the least on rice straw medium. Thus in conidial productivity, V-8 juice broth and tomato juice agar far exceeded the rice polish and yeast starch agar which were known to be best by previous workers.

Studies on the Juice Manufacturing Properties of Various Tomato Varieties (토마토의 품종별 쥬스제조 적성에 관한 연구)

  • For the elucidation of some informations on juice manufacturing properties of tomato, physical, chemical and sensory characteristic were analyzed for two groups of cultivation pattern-nonproped cultivation (5 varieties) and proped cultivation (4 varieties). The proped cultivation showed higher sugar content of juice than the nonproped on. Among the proped Jeokpung and Master 2 showed high value of sugar content. The lowest value of sugar content was observed in 79078$\times$ARC of the nonproped. There was no difference in acidity between cultivation groups but high value was observed in TM103, Jinhong from the nonproped and Jeokpung from the proped. There was no difference in pH between tomoto juice. High content of vitamin C was observed in 79078$\times$CL1561 from the nonproped and 76Moll-3-2-2 from the proped. High viscosity was observed in 79078$\times$CL1561 from the nonproped and Horgju from the proped. The nonproped showed greater value in dominant wavelength than the proped. Good Hope showed the greatest value in dominant wavelength, color score of panel test and yield. It was found that Good Hope from the nonproped and Jeokpung from the proped were suitable for juice processing. But better quality of juice can be possibly made by using various varieties rather than using single variety because each variety has each goodness of juice preparation separately.

Protective Effects of Sweet Orange, Unshiu Mikan, and Mini Tomato Juice Powders on t-BHP-Induced Oxidative Stress in HepG2 Cells

  • The aim of this study was to investigate the protective effects of juice powders from sweet orange [Citrus sinensis (L.) Osbeck], unshiu mikan (Citrus unshiu Marcow), and mini tomato (Solanum lycopersicum L.), and their major flavonoids, hesperidin, narirutin, and rutin in tert-butyl hydroperoxide (t-BHP)-induced oxidative stress in HepG2 cells. The increased reactive oxygen species and decreased glutathione levels observed in t-BHP-treated HepG2 cells were ameliorated by pretreatment with juice powders, indicating that the hepatoprotective effects of juice powders and their major flavonoids are mediated by induction of cellular defense against oxidative stress. Moreover, pretreatment with juice powders up-regulated phase-II genes such as heme oxygenase-1 (HO-1), thereby preventing cellular damage and the resultant increase in HO-1 expression. The high-performance liquid chromatography profiles of the juice powders confirmed that hesperidin, narirutin, and rutin were the key flavonoids present. Our results suggest that these fruit juice powders and their major flavonoids provide a significant cytoprotective effect against oxidative stress, which is most likely due to the flavonoid-related bioactive compounds present, leading to the normal redox status of cells. Therefore, these fruit juice powders could be advantageous as bioactive sources for the prevention of oxidative injury in hepatoma cells.

Comparison of Physicochemical Properties and Physiological Activities of Commercial Fruit Juices (시판 과일주스의 이화학적 특성 및 기능성 비교)

  • In this study, the physicochemical and physiological activities of 10 different commercially available juices (apple juice-A: cloud type; apple juice-B: clear type; blueberry juice; grape juice-A; grape juice-B; orange juice-A; orange juice-B; pineapple juice; pomegranate juice; and tomato juice) were investigated. The average pH, titratable acidity, Brix, and reducing sugar were 3.57, 0.57%, $13.1^{\circ}Brix$, and 57.58%, respectively. The total polyphenol and flavonoid contents were within the ranges of 98.29-878.12 ug/mL and 23.82-156.63 ug/mL, respectively, and pomegranate juice showed the highest content, followed by blueberry juice and apple juice-A. In the DPPH radical scavenging assay, apple juice-A, grape juice-A, and pomegranate juice exhibited the highest activity (greater than 90%) while orange juice-B, pineapple juice and tomato juice showed the weakest. The tyrosinase inhibitory effect was highest in apple juice-A, followed by orange juice-A and blueberry juice. The reducing power was highest in pomegranate juice and lowest in pineapple juice and grape juice-A. The antioxidant activity by reducing power was highly correlated with the total polyphenol content.

Leaf Blight of Perilla Caused by Alternaria alternata (Alternaria alternata에 의한 들깨 잎마름병)

  • A new leaf blight was found on the perilla leaves at the major perilla-cultivating areas such as Kangdong in Busan and Miryang in Kyungnam province. Symptoms of the disease initially appeared on the edge of perilla leaves showing black necrosis and drying, and the infected leaves were finally fell down. The SD1 isolate showing strong pathogenicity and forming abundant conidial spores on the diseased lesions was isolated. Among the tested media, mycelial growth was abundant on PDA (Potato Dextrose Agar) medium at $25^{\circ}C$ under dark condition, but conidial formation was greater on V8A (V-8 juice A8ar) medium than that on PDA medium. Optimal temperatures for mycelial growth and conidial formation on PDA medium were respectively $25^{\circ}C$ and 3$0^{\circ}C$. The rate of conidial germination and the elongation of germ tube were more effective in 10% tomato juice than those in PDB (Potato Dextrose Broth) and sterile water. In 10% tomato juice, the rate of conidial germination and the length of germ tube were 100% after incubation for 24k and 535.2${\mu}{\textrm}{m}$ after incubation for 36hr, respectively. According to the result of pathogenicity, it revealed that conidial suspention with 10% tomato juice was the most effective for pathogenicity test showing as 100% of disease incidence, and the symptoms caused by artificial inoculum were same as those of naturally infected perilla. In this study, the SD1 isolate according to the results of morphological characteristics, the incubation characteristics and pathogenicity was firstly identified A. alternata, and named as leaf blight of perilla.

Feasibility of RF Sensor Application for On-line Monitoring of Tomato Paste Processing (토마토 패이스트 가공공정의 온라인 모니터링용 RF 센서의 활용 가능성)

  • On-line monitoring of fresh tomato paste processing was done using two radio frequency (RF) sensors resonant at 85 and 110 MHz. Fresh tomato juice with soluble solid content of about $5^{\circ}Brix$ was evaporated up to $23^{\circ}Brix$ and diluted down to about $5^{\circ}Brix$ again with a pilot scale evaporator. The RF sensors were installed in a processing pipe and monitored. The pastes at a specific $^{\circ}Brix$ level were sampled and analyzed for physical properties such as soluble solid content and viscosity. The relationships between sensor outputs and measured physical properties were analyzed. Analysis results showed RF sensor is feasible to apply on-line monitoring of tomato paste processing.

  • A Corynespora species was isolated from brown target spots developed on leaves of tomato. The fungus was identified as Corynespora cassiicola and proved to be pathogenic to tomato plants. Growth of the fungus in vitro was optimum at $27^{\circ}C$, and poor at 19 and $35^{\circ}C$. Sporulation was abundant on potato dextrose malt agar and moderate on V-8 juice agar, potato dextrose agar and Czapek agar.

Utilization of Domestic Vegetables Juices as a Medium for Growth and Reproduction of Phytophthora species (국내산 야채쥬스의 역병균 영양생장 및 생식생장용 배양기 이용)

  • V8 juice agar (V8A) has been the most popular and commonly used medium for growth and reproduction of Phytophthora. However, frequently V8 vegetable juice is not available or difficult to obtain in Korea. We therefore developed widely accessible medium to substitute for V8A using domestically available five juices; two carrot (KCJA), two tomato (KTJA) and a vegetable-mix (KVMA). To prepare 10% juice medium, each vegetable juice 100 ml, DW 900 ml, agar 17 g and Ca CO3 0.5∼1.0g were supplemented to adjust pH ca. 6.0 Mycelial growth of P. cactorum and P. capsici on KTJA and KVMA was equally effective as V8A for the growth of P. cactorum, P. capsici, P. drechsleri and P. nicotianae under light. Sporangial production of P cactorum, P. capsici and P. nicotianae on KTJA and KVMA was as good as V8A and slightly better than CKJA, but the difference was insignificant by P. cactorum and P. nicotianae. The four fungi successfully formed oospores on all the media although the numbers were varied among species and media. While KTJA was the best for P. cactorum and P. capsici, V8A was the best for P. capsici and P. drechsleri. However, KCJA stimulated highest number of oopspores of P. nicotianae. Overall results showed that domestically available vegetable juices were highly effective on growth and reproduction of Phytophthora and comparable to V8 juice. Therefore, the domestic juice medium can be successfully replaced V8A in Phytophthora study.
