Chitosan, a deacetylated polymer of chitin, was applied and characterized as an adsorbent for a dye Toluidine Blue O. The adsorption of Toluidine Blue O by chitosan was affected by the particle sire and mass of chitosan, initial dye concentration, reaction time, and solution pH. More dye was adsorbed the smaller the chitosan particles. When initial ratio of dye to chitosan was over 1 : 500, the adsorption of dye was rapidly declined. Adsorption rate of dye to chitosan showed 2 phases, rapidly occurred one at the beginning within 3 min and thereafter very slowly occurred one. The amount of dye adsorbed was increased with increase in pH.
A simple and rapid method was developed for monitoring of Gram-positive bacteria in the wastewater treatment system. Culture suspensions of 4 Gram-positive and 4 Gram-negative strains were filtrated and stained with a polyethersulfone membrane filter and Toluidine Blue-O. To establish quantitative color image analysis, the intensity value of RGB (red-green-blue) color of a scanned filter image was analyzed with a photographic program. Red and green color values of Gram-positive bacteria were higher than those of Gram-negative bacteria. This method was applied to the activated sludge mixed with the Gram-positive bacteria. Although evaluation was difficult due to the irregular size and shape of flocs, the population of Gram-positive bacteria in the activated sludge could be monitored with floc dispersion technique. The more amounts of Gram-positive bacteria in the activated sludge led to the increase of red and green color values. This method provides a rapid and quantitative measurement of Gram-positive bacteria within the wastewater treatment systems.
A 2-year old female African hedgehog (Atelerix albiventris) weighing 645 grams was presented with a well circumscribed mass in the subcutaneous of the left shoulder. It measured $2{\times}2cm$ in size and had a hard texture. There were no clinical signs. In a fine-needle aspiration (FNA) cytological evaluation, mast cells were identified with Wright-Giemsa stain. Mast cell tumor was confirmed and characterized by surgical sampling and histopathological evaluation. Four stains (hematoxylin-eosin, toluidine blue, periodic acid-Schiff, and Safranin O) were examined in order to find an applicable stain for a mast cell tumor histopathological evaluation in hedgehog. This case showed that periodic acid-Schiff and toluidine blue staining were the most useful for assessment of a mast cell tumor in a hedgehog.
The author observed histochemically the nature of the calcified tissue in the rabbit dental pulp, induced by pulpal injection of potassium permanganate. The pulp of rabbit mandibular incisors were exposed and enlarged by a dental hand reamer. The exposed pulps were injected with 0.05ml of 20mM solution of potassium permanganate dissolved in Ringer's solution in experimental tooth. Also the control tooth received a pulpal injection of 0.05ml of Ringer's solution. After pulpal injection, the tooth was plugged with a gutta-percha root canal point. The staining techniques were hematoxylin-eosin stain, van Gieson stain, PAS reaction, toluidine blue stain, alcian blue-hematoxylin stain and colloidal iron-picric acid stain. The results were as follows: 1. The pulp on experimental tooth showed osteodentin-like calcified tissue. Also, in some areas, false denticle-like substance were observed. 2. The central portion of the calcified matrix showed metachromasia in toluidine blue stain had strong staining capacity in alcian blue stain. 3. The peripheral portion of the calcified tissue revealed marked van Gieson positive reaction for collagen. But their staining ability in alcian blue was slight and metachromasia was not appeared.
The purpose of this study was to determine the effect of electrical stimulation on the number of MCs and percent of degranulated MCs in rat skin. Twelve male Sprague-Dawley rats were divided into two group; electrical stimulation group (n=6) and control group (n=6). Each animals hair on the back was removed. The electrical stimulation group received an positive rectangular pulsed electrical stimulation, while the control group was given the same treatment without electricity. The biopsy specimens were fixed in formalin, embedded in paraffin and stained with toluidine blue-nuclear fast red and alcian blue-safranin O. respectively. The MCs were counted using a light microscope and computerized image analysis system and calculated as the density and the percent. A t-test showed a significantly higher density of MCs in the electrical stimulated rats than control rats(p<0.05), and the percent of degranulated MCs in the electrical stimulated rats was higher than in the control rats (p<0.05) in toluidine blue stained sections. The density of MCs was significantly higher in the electrical stimulated rats than the control rats in alcian blue-safranin O Stained sections (P<0.01). An analysis of variance showed that the densities of CTMCs was significantly lower than the densities of MMCs and mixed MCs in electrical stimulated rat in alcian blue-safranin O Stained sections (p<0.05). These results suggest that the electrical stimulation may have potential for increasing the number of MCs and lead to degranulate the MCs in rat skin.

We prepared hydrophilic porous supporters for the reinforced composite fuel cell membrane by radiation grafting of acrylonitrile (AN) and hydrophilic sodium allylsulfonate (SAS) into a porous polytetrafluoroethylene (PTFE) supporter. The physicochemical properties of the supporters prepared under various reaction conditions such as molar ratio of SAS/AN, monomer concentration, and irradiation dose were evaluated. FTIR was utilized to confirm the successful introduction of SAS/AN copolymer chains into the porous PTFE. The pores of the porous PTFE film were found to be decreased with an increase in the degree of grafting by using FE-SEM and gurley number. Furthermore, by analyzing the degree of grafting, contact angle, and TBO (toluidine blue O) uptake, the hydrophilicity of the prepared supporters was found to increase with an increase in the degree of grafting.
APUD cells in the oviduct of the sheep at standing estrus were localized as paraneurons in the lamina propria sandwiched between this structure and the tunica muscularis by the method of masked metachromasia to toluidine blue after hot mineral acid hydrolysis. These were also confirmed by lead haematoxylin stain and argyrophilia. The oviduct was serialized into 66 zones. Cells were absent in the first and last 2 zones, and most parts of the isthmus. There was however abundant number of APUD cells in the ampulla which were fusiform shaped and were about $5{\mu}m$ width and also in the juncture, where the cells were of a smaller width ($3{\mu}m$) and were quite numerous reaching 180-200 in some zones. It is concluded that peptide secreting cells are numerous in the oviduct and that this may qualify the oviduct as an endocrine organ.
This study was investigated the changes of root length, tissue structure of root tip, and dry matter production of a Dongjinbyeo (DJ) cultivar subjected to 0.4 % agar medium with various concentration of NaCl (salt stress)- and polyethylene glycol 6000 (PEG, drought stress). Root length and dry weight of DJ plant were declined along the high concentration of PEG and NaCl in rice plants. To elucidate the changes of tissue structure in root tip to PEG- and NaCl-treatments, we examined the microscopic observation of root tip in NaCl- and PEG-treated rice plants using Toluidine blue O. By Toluidine blue O staining, methyl-lignin accumulation was found in the epidermis and outer cortex of the elongation zone at an early stage of PEG treatment, whereas was found only the outer cortex of the elongation zone of NaCl-treated root tip. The epidermis of NaCl-treated root tip became soften instead of methyl-lignin accumulation. TR ratio was increased along the high concentration in PEG- and NaCl-treated rice plant as a result of inhibited root elongation under PEG- and NaCl-treatment. From these morphological changes in root stimulated by drought and salt stress, we suggest that agar medium is useful to identify tolerant variety in germination stage under stressful environments.
Various long-term studies have shown that titanium implants as abutments for different types of prostheses have become a predictable adjunct in the treatment of partially or fully edentulous patients. The continuous exposure of dental implants to the oral cavity with all its possible contaminants creates a problem. A lack of attachment, together with or caused by bacterial insult, may lead to peri-implantitis and eventual implant failure. Removal of plaque and calculus deposits from dental titanium implants with procedures and instruments originally made for cleaning natural teeth or roots may cause major alterations of the delicate titanium oxide layer. Therefore, the ultimate goal of a cleaning procedure should be to remove the contaminants and restore the elemental composition of the surface oxide without changing the surface topography and harming the surrounding tissues. Among many chemical and mechanical procedure, air-powder abrasive have been known to be most effective for cleaning and detoxification of implant surface. Most of published studies show that the dental laser may be useful in the treatment of pen-implantitis. $CO_2$ laser and Soft Diode laser were reported to kill bacteria of implant surface. The purpose of this study was to obtain clinical guide by application these laser to implant surface by means of Non-contact Surface profilometer and X-ray photoelectron spectroscopy(XPS) with respect to surface roughness and atomic composition. Experimental rough pure titanium cylinder models were fabricated. All of them was air-powder abraded for 1 minute and they were named control group. And then, the $CO_2$ laser treatment under dry, hydrogen peroxide and wet condition or the Soft Diode laser treatment under Toluidine blue O solution condition was performed on the each of the control models. The results were as follows: 1. Mean Surface roughness(Ra) of all experimental group was decreased than that of control group. But it wasn't statistically significant. 2. XPS analysis showed that in the all experimental group, titanium level were decreased, when compared with control group. 3. XPS analysis showed that the level of oxygen in the experimental group 1, 3($CO_2$ laser treatment under dry and wet condition) and 4(Soft Diode laser was used under toluidine blue O solution) were decreased, when compared with control group. 4. XPS analysis showed that the atomic composition of experimental group 2($CO_2$ laser treatment under hydrogen peroxide) was to be closest to that of control group than the other experimental group. From the result of this study, this may be concluded. Following air-powder abrasive treatment, the $CO_2$ laser in safe d-pulse mode and the Soft Diode laser used with photosensitizer would not change rough titanium surface roughness. Especially, $CO_2$ laser treatment under hydrogen peroxide gave the best results from elemental points of view, and can be used safely to treat peri-implantitis.
Following the extensive use of implant, the incidence of peri-implantitis increases. Guided bone regeneration has been used for the optimal treatment of this disease. Because implant surface was contaminated with plaque and calculus, cleaning and detoxification were needed for the reosseointegration when guided bone regeneration was performed. Various mechanical and chemical methods have been used for cleaning and detoxification of implant surface, air-powder abrasive and oversaturated citrate were known to be most effective among these methods. However, these methods were incomplete because these could not thoroughly remove bacteria of implant surface, moreover deformed implant surface. Recent studies for detoxification of the implant surface using laser were going on, $CO_2$ laser and Soft Diode laser were known to be effective among these methods. The purpose of this study was to obtain clinical guide by application these laser to implant surface. 15 experimental machined pure titanium cylinder models were fabricated. The $CO_2$ laser treatment under dry, wet and hydrogen peroxide condition or the Soft Diode laser treatment under Toluidine blue O solution condition was performed on the each of models. Each groups were examined with SPM and SEM to know whether their surface was changed. The results were as follows : 1. Surface roughness and surface form weren't changed when $CO_2$ laser was usedunder dry condition(P>0.05). 2. Surface roughness and surface form weren't changed when $CO_2$ laser was used under wet condition(P>0.05). 3. Surface roughness and surface form weren't changed when $CO_2$ laser was used under hydrogen peroxide condition(P>0.05). 4. Surface roughness and surface form weren't changed when Soft Diode laser was used under toluidine blue O solution condition(P>0.05). From the result of this study, it may be concluded that the $CO_2$ laser having relatively safe pulse mode and the Soft Diode laser used with photosensitizer can be used safely to treat peri-implantitis.
