We examined the distribution of uterine mast cells in cycling mice of different ages between 7 weeks and 38 weeks by toluidine blue staining. Mast cell density increased gradually depending on the age and culminated at 30 weeks after birth. Of the estrous stages, uteruses at metestrus showed markedly higher density of mast cells than those at the other stages in cycling mice of all ages analysed in this study and the majority of the mast cells were found in myometrium. We also discriminated types of the uterine mast cells in cycling mice at 10 weeks old by Alcian blue-safranin double staining. Mucosal type was most abundant among three types, including connective-tissue types and mixed types, through the entire estrous cycles. The portion of mixed types increased slightly after estrous. The abundance of uterine collagen protein determined by Masson trichrome staining was exactly consistent with the density distribution of the uterine mast cells during estrous cycles. Taken together, these results may imply that uterine mucosal mast cells, together with collagen proteins, are heavily involved in tissue remodeling of cycling mouse uterus.

The morphological findings of the parotid, mandibualr and sublingual salivary glands of the Korean native goat have been investigated by the histological and histochemical observation using the light microscope. Tissues were fixed with 10% neutral buffered formalin and Bouin's solution, and embedded in paraffin. The tissue sections were stained with hematoxylin and eosin, Heidenhain's azocarmine-aniline blue, alcian blue, toluidine blue, periodic acid Schiff, aldehyde fuchsin, alcian blue-periodic acid Schiff and aldehyde fuchsin-alcian blue. Some sections were stained with the alcian blue after each teatment of diastase digestion, methylation, methylation-saponification, and neuraminidase digestion. The results were as follows ; 1. The major salivary glands were compound. tubuloacinar glands, and the parenchyma was composed of acini, intercalated ducts, striated ducts and excretory ducts. 2. The acini were composed of serous cells in the parotid gland, and mucous cells, serous cells and seous demilunes in the mandibular gland. The acini of the sublingual glands were composed of mucous cells and serous demilunes. 3. In histochemistry, the serous cells of the parotid gland contained neutral mucin and enzyme-liable silaic acid. 4. The serous cells and demilunes of the mandibular gland contained neutral mucin and enzyme-liable sialic acid, and the mueous cells contained sulfated mucin, enzyme liable sialic acid and neutral mucin 5. In the sublingual gland, the mucous cells contained sulfated mucin, enzyme-resistant sialic acid and neutral mucin, and the serous demilunes contained neutral mucin and enzyme-resistant sialic acid.
Outdoor sculptures of modern art works are being damaged and deteriorated as they are exposed to the outdoor environment due to the nature of exhibition in the outdoor environment, but secure of basic data through the measures for conservation and advanced researches still remain in the early stage. The surface of "Conversion" which is exhibited in the Busan Cheonmasan Sculpture Park has been exfoliated and deteriorated due to outdoor exhibition for a long time, so systematic conservation and management of works are considered necessary. Prior to the conservation and management, this study conducted observation of cross section, analysis of inorganic components, FT-IR, Raman and Py-GC/Mass analysis to examine the nature and type of paints used for the work through a scientific analysis. As a result of analysis, paints used for the "Conversion" include paint mixed with silvery aluminium powder and white pigment, reddish paint mixed with toluidine red, bluish paint that mixed prussian blue and titanium white and mixture of phthalocyanine blue and titanium white. The result is expected to be used as basic data for selecting materials necessary for conservative treatment of and establishing a plan for conservative treatment of the "Conversion".
For the purpose of evaluating the response of the periodontal tissue in relation to the experimental tooth movement, orthodontic force was applied to 15 female rats. The animals were investigated histopathologically and histochemically by several staining methods. Findings were as follows: 1. The application of 60gm on the rat's molar caused mesial tipping movement and undermining resorption. 2. One day after the insertion of the appliance, the osteoblast was increased in activity but not in number. 3. The osteoclastic activity was decreased as soon as removing the appliance. 4. With the P.A.S. staining, osteoclasts Stained mildly in the initial stage and stained more intensely in the last stage. Osteoclasts revealed negative reaction in the initial stage and stained mildly in the last stage. 5. Slides stained with toluidine blue revealed no conclusive variations between osteoblasts and osteoclasts.
APUD cells in the oviduct of the sheep at standing estrus were localized as paraneurons in the lamina propria sandwiched between this structure and the tunica muscularis by the method of masked metachromasia to toluidine blue after hot mineral acid hydrolysis. These were also confirmed by lead haematoxylin stain and argyrophilia. The oviduct was serialized into 66 zones. Cells were absent in the first and last 2 zones, and most parts of the isthmus. There was however abundant number of APUD cells in the ampulla which were fusiform shaped and were about $5{\mu}m$ width and also in the juncture, where the cells were of a smaller width ($3{\mu}m$) and were quite numerous reaching 180-200 in some zones. It is concluded that peptide secreting cells are numerous in the oviduct and that this may qualify the oviduct as an endocrine organ.

A rat (Rottus norvegicus) infected with C. hepatica was trapped incidentally. At necropsy, grossly yellowish-white nodules were scattered on the liver surface. Microscopically, granulomatous and fibrotic nodules containing eggs and/or adult worms of C. hepatica were detected in the liver. Septal fibrosis forming pseudolobules was observed as a diffuse change throughout the liver. In double staining with immunostaining of -SMA and toluidine blue, myofibroblasts and mast cells were generally observed within the fibrous septa with mast cells being along the myofibroblasts. In this case, we hypothesized that myofibroblast and mast cell might playa role in septal fibrosis of rat liver induced by C. hepatica infection.
Early esophageal carcinoma is defined as a lesion wherein invasion is confined to the mucosa and submucosa without metastasis to lymph node or other organs. Postoperative 5-year survival rate for early esophageal carcinoma is much superior than advanced carcinoma. Unfortunately, because of the anatomic characteristic of esophagus and absence of specific early symptoms, detection is frequently belated, and advanced disease is present at the time of the initial diagnosis. We experienced 2 cases of early esophageal carcinoma. They complained no specific symptoms. The diagnosis was made by barium esophagogram, esophagofiberscopy with dye staining and endoscopic biopsy. We performed esophagectomy with esophagogastrostomy. All had good postoperative course without any complication. We concluded that the combined use of double contrast radiography, esopagofiberscopy aided by intraluminal staining with Toluidine blue or Lugol`s solution, and endoscopic biopsy is very important in the diagnosis of early esophageal carcinoma in high risk patient group.
Objectives: This study aimed to investigate the anti-atopic properties of Glechoma hederacea var. longituba extracts (GHE) in murine atopic dermatitis model. Methods: BALB/c mouse ear stimulated with oxazolone (OX) for 4 weeks, then 1% GHE was topically applied every two days for 3 weeks to mouse. Ear thickness was measured by a digital thickness gauge. The ears tissues were collected and subjected to hematoxylin-eosin (H&E) and toluidine blue (TB) staining. Results: Treatment with GHE successfully alleviated the symptoms of atopic dermatitis, such as erythema, horny substance, and swelling. The infiltration of lymphocytes and mast cells were significantly reduced following GHE treatment. Conclusion: Taken together, our results showed that GHE possessed the potential to be a novel immunomodulatory drug against atopic dermatitis.
This study was investigated the changes of root length, tissue structure of root tip, and dry matter production of a Dongjinbyeo (DJ) cultivar subjected to 0.4 % agar medium with various concentration of NaCl (salt stress)- and polyethylene glycol 6000 (PEG, drought stress). Root length and dry weight of DJ plant were declined along the high concentration of PEG and NaCl in rice plants. To elucidate the changes of tissue structure in root tip to PEG- and NaCl-treatments, we examined the microscopic observation of root tip in NaCl- and PEG-treated rice plants using Toluidine blue O. By Toluidine blue O staining, methyl-lignin accumulation was found in the epidermis and outer cortex of the elongation zone at an early stage of PEG treatment, whereas was found only the outer cortex of the elongation zone of NaCl-treated root tip. The epidermis of NaCl-treated root tip became soften instead of methyl-lignin accumulation. TR ratio was increased along the high concentration in PEG- and NaCl-treated rice plant as a result of inhibited root elongation under PEG- and NaCl-treatment. From these morphological changes in root stimulated by drought and salt stress, we suggest that agar medium is useful to identify tolerant variety in germination stage under stressful environments.
Five dogs were used to determine whether 0.1% tacrolimus ointment application for one day would inhibit IgE-mediated late-phase reactions (LPRs). It was consisted of three periods: one period without therapeutic administration (control) and two periods of treatment with either the tacrolimus ointment or vehicle. Induction of IgE-mediated LPRs was induced by intradermal injections of 0.05 ml (0.14 mg/ml) of solution of goat anti-canine IgE polyclonal antibodies. Each section for mast cells (MCs) and eosinophils (EPs) was stained with acidified toluidine blue, and Luna's stain, respectively. Assessment of anti-inflammatory effect of tacrolimus ointment composed of cell counts of MC and EP from lesions of induced LPR. In normal canine biopsies, the number of dermal MCs and EPs were $12.3{\pm}1.4cells/mm^2$ and $3.1{\pm}1.3cells/mm^2$, respectively. MC counts dramatically decreased at time dependent manner after anti-IgE administration. However, the number of MCs on 6 hours after challenge was significantly less decreased in the groups treated with the tacrolimus, as compared with control and vehicle group. The number of EPs on 24 hours after challenge was significantly lower in the group treated with the tacrolimus than in the control and vehicle groups. In conclusion, this study revealed that 0.1% tacrolimus ointment in dogs may exert a potent anti-inflammatory effect on inhibition of MC degranulation and also secondary prevention of EP infiltration during LPR.
