Objective: This study aimed to compare the effects of diets with and without supplements of methionine/lysine (met/lys) and methionine/α-tocopherol (met/α-tocopherol) on the performance, carcass traits and meat quality of Hanwoo steers. Methods: A total of 21 Hanwoo steers were divided into three different groups. Each group consisted of 7 animals that received different dietary treatments for 120 days as follows: a control (C) diet composed of a basal diet with 74% total digestible nutrient and 12% crude protein; treatment 1 (T1) composed of a basal diet enriched with methionine 69%+lysine 31%; and treatment (T2) composed of a basal diet enriched with methionine 84.65%+α-tocopherol 15.35%. Daily supplementation was given using the top-dressing method (20 g/animal). Results: The met/lys groups yielded a longissimus lumborum with increased water holding capacity (p<0.01) and lower shear force value (p<0.05). Dietary met/lys did not have an adverse effect on the animal performance and carcass traits. Instead, these supplements contributed significantly to the higher protein content compared to the control diet (p<0.05). Myristic acid (C14:0) was the only fatty acid affected by the supplementation. While the met/α-tocopherol group led to significantly higher protein and oxymyoglobin contents during storage (p<0.05). It also contributed significantly to the lower shear force value and 2-thiobarbituric acid reactive substances score during storage (p<0.05) compared to the control and treatment 1 since the initial storage day. The met/α-tocopherol diet also yielded meat with a redder color (p<0.05) after 3 days of storage. However, it did not significantly contribute to the fatty acid profiles of Hanwoo steers. Conclusion: Met/lys supplementation resulted in higher protein scores, water holding capacity and lower shear force scores. While met/α-tocopherol supplementation contributes to the production of redder meat, reduces lipid oxidation, production of more tender meat and increases the content of protein and oxymyoglobin percentage.
This study was conducted to investigate the effects of $\alpha$-, ${\gamma}$-, $\delta$-Tocopherols on oxidative stability of lipid in hamburger patties during frozen storage. Hamburger patty samples were prepared with the addition of tocopherol by fat basis: control, treat 1 (control+0.02% $\alpha$-tocopherol), treat 2 (control+ 0.02% ${\gamma}$-tocopherol were cooked at 85$^{\circ}C$ for 3 minutes and the stored at -1$0^{\circ}C$ or 3$0^{\circ}C$). The fatty acids composition of the hamburger samples were analyzed during the storage periods (1, 20, 40, 60 and 80 days). there was no difference in the fatty acids composition among control and treatment groups in early storage time; however, content of unsaturated fatty acid was decreased in control group and slightly decreased or almost not changed in treatment groups as the storage period passed. No difference in the ratio of saturated fatty acid : unsaturated fatty acid (SFA : USFA) and saturated fatty acid : monounsaturated fatty acid : unsaturated fatty acid (SFA : MUFA : PUFA) between control and treatment groups at early storage time. However, SFA : USFA and SFA : MUFA : PUFA was decreased in control and slightly decreased or not changed in all treatment groups as the storage period passed.
This study examined the effects of vacuum packaging and modified atmosphere packaging (MAP) on the meat quality of ${\alpha}$-tocopherol supplemented broiler chicks after prolonged refrigeration. The supplemented ${\alpha}$-tocopherol levels had no effect on the body weight and feed efficiency. The dietary treatments and packaging methods had no significant effects on the level of microbial growth, lightness, and the metmyoglobin content. The vacuum packaging showed the highest level of purge loss. The broilers supplemented with either 200 or 400 IU of ${\alpha}$-tocopherol had a lower level of malondialdehyde than the control group. The vacuum packaged meats from chicks fed the control diet showed the highest levels of lipid oxidation. The lipid oxidative stability was most improved in modified atmosphere (20% $CO_2$ + 80% $N_2$) packaged breast meat from the broilers supplemented with either 200 or 400 IU of ${\alpha}$-tocopheryl acetate.
The longitudinal change of vitamin E($\alpha$-Tocopherol) content and secretion in transitional milk of 32 Korean lactating women and vitamin E intake of infants was studied 7, 10 and 15 days postpartum, Vitamin E content of the milk decreased at 7, 10 and 15 days postpartum with the respective amount of 784, 633 and 539$\mu\textrm{g}$/100ml. The average vitaimin E secretion in the milk of mothers appeared 350$\mu\textrm{g}$/days. The average vitamin E intake in the milk was 3033 and 2424$\mu\textrm{g}$/day in boys and girls respectively and was 174.7% for infants of the recommended lower limit for vitamin E in infant formular of 0.5mg/100㎉. Vitamin E intake per body weight of infants in the milk were 929.731 and 676$\mu\textrm{g}$/kg/day respectively.
Twenty three healty women on self - selected diet were given 800IU of tocopherol daily for 4 weeks. The levels of tocopherol in plasma and red blood cells( RBC ) and total choles terol, TG, HDL- chol, HDL subfractions and lipoprotein pattern in serum were determined pre-and postregimen at 2 and 4 weeks. No significant change was noted in VLDL, LDL, HDL fraction and LDL/HDL ratio separated by electrophoresis, even though HDL fraction was decreased at 2 wk but slightly increased at 4 wk. There were also no significant changes in the relative amount of HDL-chol and VLDL-chol when cholesterol content of each lipoprotein fraction was assayed. A transient increase in LDL-chol was observed at 2 wk but returned to the pretreatment level. Plasma and RBC tocopherol levels were significantly ( p ^lt;0.05 ) increased and decreased respectively, at both 2 and 4 wk, and LDL-chol was positively correlated to plasma tocopherol level ( p<0.05 ) but not to RBC tocopherol. However HDL-chol fractionated by heparin-Mn was increased at 2 and 4 wk by a significant increase in $HDL_{2}$-chol but no change in $HDL_{3}$-chol, which resulted in a significant reduction of $HDL_{3}$/HDL ratio and increase of $HDL_{2}/HDL$ and $HDL_{2}/HDL_{3}$ ratios. HDL- chol was negatively correlated to the levels of LDL-chol ( p<0.05), VLDL-chol (p<0.01), and T-chol/HDL ratio ( p<0.01 ). Serum TG was significantly decreased ( p <0.05 ) but total cholesterol was decreased only at 4 weeks.

The effect of ${\alpha}$-tocopherol and ${\beta}$-carotene supplementation on reducing the oxidative damag in the liver of rats were studied. Forth-five male Sprague Dawley aged 4 weeks were randomly assigned to 9 groups of five for the 12 weeks of the study. Nine groups, sardine oil, sardine oil+Vt E, sardine oil+${\beta}$-carotene, soybean oil, soybean oil+Vt E, soybean oil+${\beta}$-carotene, lard, lard+Vt E, lard+${\beta}$-carotene group, were prepared. Sardine oil, soybean oil, or lard was used for dietary fat and 200% of ${\alpha}$ -tocopherol or 150% of ${\beta}$-carotene was supplemented to each diet. Each diet supplied 65% of total energy as carbohydrate, 15% as protein, and 20% as lipid. The MDA value and protein carbonyl contents of sardine oil group were significantly different(p<0.05) to those of other fat groups indicating that the most severe lipid oxidation occurred in the group fed diet containing highly polyunsaturated fatty acid. When ${\alpha}$-tocopherol or ${\beta}$ -carotene was supplemented to the sardine oil diet, MDA value(-35%, -15%, respectively) and protein carbonyl content(-44%, -32%, respectively) decreased significantly(p<0.05). Cu, Zn-superoxide dismutase(SOD) and catalase activities of three different sardine oil groups with or without antioxidants were lower than those of soybean oil or lard group. The reducing effect of ${\alpha}$-tocopherol on oxidative damage in sardine oil group supplemented with ${\alpha}$-tocopherol was noticeable(p<0.05). However the adverse effect of ${\beta}$-carotene was observed. SOD and catalase activities of ${\beta}$-carotene supplemented groups were that the lowest among the same fat groups, but the differences were not statistically significant. The possible cause of decreased enzyme activity seemed to be related to the vitamin A(Vt A) toxicity in the liver where retinol converted from dietary ${\beta}$-carotene in the intestinal mucosa was stored.
There are two groups of significant functional constituents in sesame seeds on the whole; one is the vegetable oils and another is the anti-oxidative compounds. However, although high amounts of major fatty acids are synthesized in sesame seeds, their composition is unfavorable because the contents of alpha- and gamma-linolenic acid, the essential fatty acids, are very low or do not produced in sesame seeds. So, to increase these fatty acids in sesame seeds, one strategy is to overexpress their genes, ${\omega}$-3 fatty acid desaturase for alpha-linolenic acid and delta-6 fatty acid desaturase for gamma-linolenid acid, in them. Another molecular target is to enhance alpha-tocopherol, vitamin E, because its content is very low in sesame seeds. The enzyme, gamma-tocopherol methyltransferase, catalyzes the conversion of gamma-tocophero to alpha-tocopherol. Overexpression of this enzyme in sesame seeds could be also a good molecular breeding target. Reduction of phytic acid is also another molecular target in sesame seeds because phosphorus pollution may be caused by its high content in sesame seeds. Accordingly, to do so, one of target enzymes could be myo-inositol 1-phosphate synthase which is a key regulatory enzyme in the pathway of phytic aicd biosyntheses. In this lecture, a molecular strategy for development of value-added sesame crop is described in association with some results of our experiments involved in the molecular characterizations of the genes mentioned above.

Legumes are one of the largest families of crop plants and are widely consumed and produced for their nutritional and commercial benefits. Mung bean (Vigna radiata L.) is a legume crop that contains various functional compounds ; moreover, it has strong antioxidant properties and is becoming an increasingly important food crop. However, most previous studies on mung beans have focused on their primary metabolites. In this study, we investigated the composition and contents of phenolic compounds, fatty acids, soyasapogenol and tocopherol in mung beans cultivated in different regions and cultivated at different seeding dates. Material analysis was conducted using the following methods: LC-MS/MS, GC-FID and HPLC-ELSD. In total, 57 different samples were analyzed. Thirteen phenolic compounds were detected in mung beans. Of these, vitexin and isovitexin were the most abundant compounds, accounting for approximately 99% of phenolic compounds. The difference in phenol compounds according to the seeding dates of mung bean was not statistically significant. The total fatty acid content in beans was the highest in Pyeongchang. Significant differences in total fatty acid content were found according to the cultivation regions. Crops grown in Sohyeon and Dahyeon showed the highest soyasapogenol B content in the Suwon region, and these were the lowest in Jeonju. The total tocopherol content of beans cultivated in Dahyeon and Sohyeon was the lowest and highest in Pyeongchang. Soyasapogenol B and total tocopherol content were not significantly different according to seeding dates. This study was conducted to obtain basic data for the cultivation of mung beans with a high content of various functional materials in terms of regional specialization and optimal seeding time.
An investigation of various olive oils available in Korea was carried out to assess their quality properties such as color, oxidative stability, fatty acid composition, tocopherol content, sterol content and benzo(a)pyrene content. In color measurement, by using a Lovibond color scale and Hunter color difference meter, both a and b values of extra virgin olive oil were higher than those of pure olive oil by Tintometer (Lovibond PFX995). However, extra virgin olive oil showed higher a value and lower L value than pure olive oil by the Hunter color difference meter. In the rancimat test, the induction period of extra virgin olive oil $(38.03{\sim}8.47hr)$ was longer than that of pure olive oil $(32.40{\sim}9.94hr)$. In fatty acid composition, C18:1 $(72.01{\sim}78.53wt%)$ was present in the greatest amount, with lesser amounts of C18:2 $(4.88{\sim}10.36wt%)$ and C18:3 $(0.56{\sim}1.09wt%)$. The tocopherol content ranged from ${\alpha}-Toc\;4.09{\sim}13.89mg/100g$, ${\beta}-Toc\;0.57{\sim}1.34mg/100g$, and ${\gamma}-Toc$$3.41{\sim}8.03mg/100g$, and ${\alpha}-tocopherol$ was found to be the main isomer in all oil samples. Therefore, there was little difference in the fatty acid composition and tocopherol content among the different types of olive oils. In sterol content, ${\beta}-sitosterol$$(124.52{\sim}19.33mg/100g)$ and campesterol $(1.10{\sim}0.62mg/100g)$ of extra virgin olive oil were higher than that of pure olive oil $({\beta}-sitosterol\;92.68{\sim}17.44mg/100g,\;campesterol\;0.59{\sim}0.35mg/100g)$. Benzo(a)pyrene was found in almost all samples, with $0.287{\sim}0.106{\mu}g/kg$ in extra virgin olive oil and $1.204{\sim}2.130{\mu}g/kg$ in pure olive oil.

This experiment was conducted to study the effects of Korean ginseng and tocopherol on serum testosterone level, protein metabolism and swimming endurance in rats. Powdered ginseng (2.5 g/kg) and ${\alpha}-tocopherol$ (250 mg/kg) were added directly into the basal diet of rats feeding for 6 weeks. The serum testosterone level of ginseng-treated rats had no difference from that of control rats, but the level was slightly increased in tocopherol-treated rats. In the case of ginseng-treated rats, there were no significant difference in the contents of albumin, ${\alpha}_1-globulin$, ${\alpha}_2-globulin$, ${\gamma}-globulin$ and A/G ratio compared with those of the control rats, however, the levels of total protein and ${\beta}-globulin$ were increased significantly(p<0.005). Total protein value of tocopherol-treated rats was higher than that of the control rats(p<0.005). In contrast, albumin content was slightly decreased. The contents of ${\alpha}_1-globulin$, ${\alpha}_2-globulin$, ${\beta}-globuiln$ and ${\gamma}-globulin$ were higher in the rats fed tocopherol diet that those obtained from the control rats. Accordingly A/G value was markedly decreased (p<0.001). In swimming endurance test, both Korean ginseng and ${\alpha}-tocopherol$ did not appear to have any effect on rats' stamina.
