간편한 고등식물 RNA 분이 방법 (A Simple Procedure for RNA Isolation from Plants and Preservation of Plant Material for RNA Analysis)

  • Total RNA was isolated from two months old wheat, rice, tobacco and sweet potato. The procedure used was simple and provided pure RNA preparation. Lysis of plant tissue in a buffer with guanidine thiocyanate and CsCl density gradient centrifugation separated RNA from the rest of the cellular components. Subsequent cholroform/1-butanol extraction and ethanol precipitation were necessary to ensure contaminant-free RNA preparation. Storage of the lysed plant tissue in the buffer with guanidine thiocyanate preserved the sample for two months without noticeable RNA degradation.

Oxyfluorfen에 대한 담배의 엽령별 생리적 반응 (Physiological Responses of Tobacco(Nicotiana tabacum L.) Leaves of Different Age to Oxyfluorfen)

  • Diphenyl ether계 제초제인 oxyfluorfen에 의해 나타나는 세포내 구성물질의 누출, 엽록소 파괴, 지질과산화작용 등 전형적인 생리적 반응을 재배종 담배를 대상으로 하여 엽령별로 조사하였다. Oxyfluorfen을 담배의 엽절편에 처리하고 암조건에서 12시간 동안 배양한 후 광조건하에서 시간별로 전해물질의 누출을 측정한 결과, 암조건하에서는 전해물질의 누출은 얼어나지 않았으나 광조건하에서 일정 기간의 lag period를 거친 후 배양시간이 경과함에 따라 크게 증가하였다. Oyfluorfen에 의한 전해물질의 누출은 처리 농도의 증가에 따라 증대되었으며 담배 잎에 대한 oxyfluorfen의 효과는 엽령에 따라 크게 다르게 나다나 어린 잎일수록 전해물질의 누출이 뚜렷하였으며 반응의 lag period도 단축되었다. Oxyfluorfen에 의한 담배 잎의 엽록소 함량 감소와 malondialdehyde의 생성도 엽령에 따라 크게 좌우되어 엽령별로 어린 잎일수록 크게 나타났다. 이상의 결과에서와 같이 oxyfluorfen에 의해 담배 잎에 나타나는 생리적 반응이 잎의 엽령에 따라 크게 달라져 담배는 diphenyl ether계 제초제에 대한 내성기작연구에 유용한 식물 재료가 될 것으로 생각된다.

옥수수 calreticulin 과발현 토마토에서 tobamovirus의 상엽 이동 억제 (Suppression of tobamovirus movement toward upper leaves in the tomato plant over-expressing a maize calreticulin)

  • To ascertain the effect of over-expressed maize calreticulin in tomato plant on tobamovirus movement in addition to validating potentiality of the gene (ZmCRT) as a means for the virus-resistance resource, four ZmCRT-expressing homozygous lines were generated from the T0 plants as using an Agrobacterium-mediated transformation, nucleic acid analyses, and a conventional breeding method. Of them, a line was subjected to the bioassay for tolerances to tobacco mosaic virus-U1 (TMV-U1) and tomato mosaic virus (ToMV) followed by RT-PCR and a chlorophyll fluorescence quenching analyses. Both transgenic plants transcribing ZmCRT and wild-type plants showed no symptom by 20 days after viruses inoculation, however the photosystem II quantum yield parameter measured from the upper leaves of ToMV-inoculated plants revealed that ZmCRT transgenic plants have higher photosynthetic ability than wild-type ones at that time, which indirectly implies that over-expressed ZmCRT product acts as a barrier to the cell-to-cell and/or systemic movement of ToMV. Moreover, ZmCRT transgenic plants showed remarkably longer shoot length than wild-type ones in 40 days after TMV-U1 or ToMV inoculation each, which might be resulted from higher photosynthetic ability during the phase not yet showing any external symptoms. Collectively, over-expressed ZmCRT protein in tomato plants is able to interrupt the systemic movement of infected TMV-U1 and ToMV even though not perfect.

Expression of Chitinase Gene in Solanum tuberosum L.

  • In order to protect fungal diseases, leaf disc explants of Solanum tuberosum cultivar, Belchip, was infected with an Agrobacterium MP90 strain containing chimeric gene construct, consisting of antibiotic resistance and chitinase gene driven by the CaMV 35S promoter, for transformation. Regenerated multiple shoots were selected on a medium containing kanamycin and carbenicillin after exposure to Agrobacterium. The presence and integration of the npt II and chitinase gene were confirmed by polymerase chain reaction(PCR). Northern blot analysis indicated that the genes coding for the enzyme could be expressed in potato plants. The chitinase activity of transgenic potato plants was higher than the control potato.

Use of Serological-Based Assay for the Detection of Pepper yellow leaf curl Indonesia virus

  • Diseases caused by Pepper yellow leaf curl virus infection is considered to be emerging plant diseases in Indonesia in the last five years. One key factor for disease management is the availability of accurate detection of the virus in plants. Polyclonal antibody for Pepper yellow leaf curl Indonesia virus-Bogor (PYLCIV-Bgr) was produced for detection of the virus using I-ELISA and DIBA methods. The antibody was able to detect PYLCIV-Bgr from infected plants up to dilution 1/16,384 and cross reaction was not observed with Cucumber mosaic virus (CMV), Tobacco mosaic virus (TMV), and Chilli veinal mottle virus (ChiVMV). Positive reaction was readily detected in membrane containing Begomovirus samples from Yogyakarta (Kaliurang and Kulonprogo) and West Java (Bogor and Segunung). Infection of PYLCIV-Bgr in chillipepper, tomato, and Ageratum conyzoides was also confirmed using polyclonal antibody for PYLCIV-Bgr in DIBA. Polyclonal antibody for PYLCIV-Bgr is suggested to be included in disease management approach due to its good detection level.

Ethyl Acetate Extract of Bacillus pumilus SH122 Induces Resistance Against Phytophthora Blight in Pepper Plant

  • In order to obtain bacterial metabolites inducing disease resistance in pepper plant, two hundred bacterial isolates were isolated from the rhizosphere soil of tobacco, cucumber, and pepper plant. Ethyl acetate extract of each bacterial culture was used to screening for induction of resistance against phytophthora blight of pepper plant. Application of ethyl acetate extract of an isolate SH122 culture to pepper plant conferred resistance against phytophthora blight consistently and significantly. According to cellular fatty acid analysis and other characteristics, the SH122 culture were significantly lower than those on control plants treated with ethyl acetate extract of nutrient broth. The B. pumilus SH122 itself of ethyl acetate extract of its culture did not show antifungal activity against phytophthora blight in pepper plants.

아황산가스(${SO_2}$)와 오존(${O_3}$)에 대한 지표식물의 가시적 피해 발현시간 조사 (Evaluation of ${SO_2}$ or ${O_3}$ Exposure Durations Requiring Foliar Damage Development by Using Bioindicating Plants)

  • 들깨 '수원25호', 담배 'L8', 'KF109'와 'NC82', 시금치 '입추'와 나팔꽃을 생장혼경조절이 가능한 대기오염원 노출상에서 SO? 140ppb/시간 또는 O? 150ppb/시간의 농도로 하루 24시간 10일 동안 노출시킨 결과 '수원25호' 들깨와 담배 'L8'은 SO?에 민감한 반면, 담배 'L8'과 나팔꽃은 오존에 아주 민감한 것으로 판명되었다. 위의 농도에서 가시적 피해가 조사식물의 50%에서 발생하는 병반발생빈도 50%(SF?)를 기준으로 설정한 한계노출시간 조사에서 오존에 대한 한계 노출시간은 담배 'L8'의 경우 24시간, 나팔꽃의 경우 72시간, 담배 'NC82'와 'KF109'의 경우 144시간 내외, 시금치의 경우 216시간으로 추정되었으며, SO?에 대한 한계노출시간은 담배 'L8'의 240시간을 제외하고는 모든 식물에서 240시간 이상으로 추정되었다.

  • We have tested 101 species traditional medicinal plants on Acetylcholinester ase inhibition. Among them. Caesalpina sappan. Cnidium offcinale, Coptis japonica, Ephedra sinica, Eribotrya japonica, Gardenia jasminoides, Liriope platyphylla, Lonicera japonica, Paeonia suffruticosa, Phyllostachys nigra, Polygonum multiflorum, Prunella vulgaris, Rheum undulatum, Salvia miltiorrhiza and Uncaria rhynchophylla showed strong inhibition potency over 88%. In addition, Aconitum carmichaeli, Caesalphina sappan, Cinnamomun japonica, Citrus unshiu, Hordeum vulgare, Glycine max and Rehmannia glutinosa showed relatively good inhibition potency over 80%.
