Differential expression of rice calmodulin promoters in response to stimuli and developmental tissue in transgenic tobacco plants

  • The promoters of OsCaM1 and OsCaM3 were characterized after sequencing and fused to the reporter gene, GUS. The constructs were then transformed into the tobacco plant. Histochemical analysis of GUS showed different expression patterns in pOsCaM1::GUS and pOsCaM3:: GUS transgenic plants. The expression of pOsCaM1::GUS in 4- to 15-day-old seedlings in particular was observed only in the root, while the expression of pOsCaM3::GUS was detected in both the cotyledons and root. Also, pRCaM1::GUS was detected in all the tissues surrounding the root system, while the presence of pOsCaM3::GUS was observed in the root, except in the root meristem. However, in mature transgenic plants, the expression of pOsCaM1::GUS and OsRCaM3::GUS was scarcely detected. Under wounding stress, the GUS activity of pOsCaM1 and pOsCaM3 was strongly induced, and the activity of pOsCaM3 especially, was retained for long periods. In the phloem, pOsCaM3 activity induced by hormone treatments and abiotic stresses was also identified.

Characteristics of Tobacco Mosaic Virus Isolated from Wasabi (Eutrema wasabi) in Korea

  • Wasabies showing mosaic symptoms were collected and extracted for virus purification. Tobacco mosaic virus (TMV) was identified as causal agent by electron microscopy and nucleic acid and coat protein analyses. TMV strains were determined by enzyme-linked immunosorbent assay (ELISA). TMV was identified as W and C strain in wasabi. The results of host reaction indicated that this virus induced local lesions on Nicotiana tabacum cv. Bright Yellow and N. glutinosa, leaf spots on Chenopodium amaranticolor and mosaic symptoms on wasabi. Rot shape virus particles were observed and was about 300 nm in length. About 6.5 kb single RNA molecule was observed from extracted viral RNA sample and 26 KDa coat protein was detected in denatured acrylamide gel. Infection ratio of TMV was 8% for the first cultivation year, but was 22% for the second year when TMV-W antiserum was used. The results of this experiment showed that infection ratios of both TMV-W and TMV-C strains were higher compared to that of TMV-P strain.

Accelerating Effect of 2-Chloroethyl Phosphonic Acid Foliar Applications on Leaf Tobacco Maturity (2-Chloroethyl Phosphonic Acid가 잎담배 조열에 미치는 영향)

  • Cultivar Yeollow Special A, the most leading Korean economic leaf tobacco in Korea, were field-cultivated in four different localities of Korea for the period of two years (1970 and 1971) and sprayed at varied levels of 2-chloroethyl phosphonic acid (CEPA) for foliar application few days after topping. While no striking difference in leaf yield by weight was obtained among the treatments when compared with control, leaf quality as expressed for shipment price in won tended when compared with control, leaf quality as expressed for shipment price in won tended to improve. The treated leaves with 300 to 900 ppm of CEPA (approximately 140 1/acre of 500 ppm) not only showed yellowing and accelerated maturity to pick 4 to 5 days with practicable optimal level earlier than control, but also speeded up to take nearly with practicable optimal level earlier than control, but also speeded up to take nearly last half of the total time required for the five stages of flue-curing. It is therefore considered that CEPA is as effective maturity-accelerating agent and useful as known for other solanaceous plants showing climacteric stage respiration, and discussions were made about physiological actions of ethylene gas released from CEPA at plant tissues sprayed.

  • Interspecific cross between Nicotiana trigonophylla and N. tabacum cv. BY4 is highly sterile because of abnormal ovule and embryo development. In vitro culture of excised N. trigonophylla ovules after polination by N. tabacum allows significant numbers of hybrid embryos to develop into mature plants. Total yield of seedlings and number of normal seedlings were produced following in vitro culture of individual fertilized ovules of N. trigonophylla X N. tabacum at four days post-pollination on B5 medium containing 6% sucrose. Hybrids were uniform in morphology and peroxidase isozyme composition and the majority were cytologically stable: flower characteristics were generally intermediate between those of the parents. All hybrids evaluated were self-sterile.

The Viruses in Gladiolus hybridus cultivated in Korea 2. Broad Bean Wilt Virus, Cucumber Mosaic Virus and Tobacco Rattle Virus (한국산 글라디올러스에 발생하는 바이러스)

  • Gladioli (Gladiolus hybridus) showing flower colour breaking leaf mosaics, notched leaf, and dwarfing or lack of visible symptoms were collected from gladioli growing areas in Taegu and Kyungpook province, Korea. The three viruses isolated from the naturally infected gladioli were identified as broad bean wilt virus (BBWV), cucumber mosaic virus (CMV), and tobacco rattle virus (TRV) by their host range, immunosorbent electron microscopy (ISEM), enzyme-linked immunosorbent assay (ELISA), direct tissue blotting immunoassay (DTBIA), and intracellural symptoms. By DTBIA and ISEM, TRV was detected in gladiolus showing notched leaf, while CMV was frequently detected in gladioli with dwarfing, color breaking and malformation of flowers. BBWV was also often detected in many symptomless gladiolus plants, but TRV was detected in notched-leaf of gladiolus. Electron microcopic examination of negatively stained preparations showed that BBWV and CMV are spherical particles of 28 nm and 30 nm in diameter, and TRV is rigid rod-shaped particles of 40∼200 nm in length. The rigid rodshaped virus particles reacted positively with TRV antiserum in ISEM and DTBIA, respectively.

  • A lot of individual ginseng plants were selected in the farmer's fields to develop new ginseng varieties with desirable traits. Among them, a promising line has been developed through comparative cultivation of several lines selected with pure line separation of local raced. It was then designated as "KC (denotes Korean Ginseng)" and tested in the regional yield and adaptation trials for 10 years. KG lines grew vigorously after 4 years of age. Especially, KG 102 line arson운 them showed traits of multiple and short stems. As for the root characters, the length of taproot of KG 101 line was longer than that of local race, Jakyung-jong, or other lines ailed the root weight of KG 102 line waIn 15 % higher than that of local race. In general, KG 101, KG 103, KG 104, and Hwangsauk-jong had good root shape. Total amount of ginsenosides of ginseng taproot was the highest in KG 103 line than in local race and other lines. In these studies, we, elected three superior lines, KG 101, KG 102, and KG 103, having characteristic of good root shape, high yield, and large amount of ginsenoside, respectively.

Selection of Glyphosate-Resistant Clones form MNNG-treated Mesophyll Protoplasts of Haploid Tobacco Plants (반수체 담배의 엽육 원형질체로부터 MNNG 처리에 의한 Glyphosate 저항성 클론의 선별)

  • Selection of glyphosate-resistant clones from MNNG-treated mesophyll protoplasts of haploid tobacco and their differentiation were studied. The protoplasts were treated with 0.1 to 100 $\mu\textrm{g}$/mL N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) for 30 min when they expanded to oval shapes. After the treatment, the protoplasts in 4-16 cell stages were transferred to the selective medium containing 1 mM glyphosate for the selection of the glyphosate-resistant colonies. The efficiency of the cell division of the protoplasts in the selective medium decreased as the MNNG concentrations in creased. Optimal MNNG concentration for induction of the glyphosate-resistant clones was 10$\mu\textrm{g}$/mL and mutation frequency was 2.66$\times$10-6. The stability of the glypohsate-resistance of the clones was examined by prolonged subculture in the medium with 1 mM glyphosate, and the resistant clones were survived more than 10 months. Among them one clone has been proliferating and greening and the others were proliferating without greening or greening with slower proliferating.

  • BcGRl gene encoding cytosolic glutathione reductase of Chinese cabbage (Brassica campestris var. Pekinensis cv. Seoul) was placed under the control of the CaMV 35S promoter and introduced into tobacco (Nicotiana tabacum L. cv. Samsun) via Agrobacterium-mediated transformation. T$_{0}$ 32 independent plants transformed with BcGRl gene were selected with kanamycin and they were confirmed by polymerase chain reaction (PCR) and Southern blot analysis. Northern blot analysis revealed that the constitutive expression of BcGRl gene and there was no relationship between the copy number of introduced gene and the levels of BcGRl transcripts.

  • Leaf mesophyll protoplasts from five cultivars of tobacco (N. tabacum L.) were cultured. The protoplasts did not survive in culture medium containing Murashige and Skoog inorganic salts for over 6 days. NH4NO3 and FeSO4.Na2EDTA concentration of Murashige and Skoog medium were toxic in tobacco leaf mesophyll protoplast culture. Therefore we investigated optimum condition in Murashige and Skoog medium. High plating efficiency was obtained by reducing the concentrations of NH4NO3 and FeSO4.Na2EDTA to 1/3 and 1/10, respectively, on the supplemented with 5$\mu$M IAA, 0.5 $\mu$M 2.4-D 5 $\mu$M BAP. Plants were regenerated from protoplast-derived calluses.

Development of Environmental Stress-Tolerant Plants by Gene Manipulation of Antioxidant Enzymes

  • Oxidative stress is one of the major limiting factor in plant productivity. Reactive oxygens species (ROS) generated during metabolic processes damage cellular functions and consequently lead to disease, senescence and cell death. Plants have evolved an efficient defense system by which the ROS is scavenged by antioxidant enzymes such as superoxide dismutase (SOD) and ascorbate peroxidase (APX). Attempts to reduce oxidative damages under the stress conditions have included the manipulation of 갠 scavenging enzymes by gene transfer technology. Increased SOD activities of transgenic plants lead to increased resistance against oxidative stresses derived from methyl viologen (MV), and from photooxidative damage caused by high light and low temperature. Transgenic tobacco plants overexpressing APX showed reduced damage following either MV treatment of photooxidative treatment. Overexpression of glutathion reductase (GR) leads to increase in pool of ascorbate and GSH, known as small antioxidant molecules. These results indicate through overexpression of enzymes involved in ROS-scavenging could maintain or improve the plant productivities under environment stress condition. In this study, the rational approaches to develop stress-tolerant plants by gene manipulation of antioxidant enzymes will be introduced to provide solutions for the global food and environmental problems in the $21^\textrm{st}$ century.
