Bacillus spp., as a type of plant growth-promoting rhizobacteria (PGPR), were studied with regards promoting plant growth and inducing plant systemic resistance. The results of greenhouse experiments with tobacco plants demonstrated that treatment with the Bacillus spp. significantly enhanced the plant height and fresh weight, while clearly lowering the disease severity rating of the tobacco mosaic virus (TMV) at 28 days post-inoculation (dpi). The TMV accumulation in the young non-inoculated leaves was remarkably lower for all the plants treated with the Bacillus spp. An RT-PCR analysis of the signaling regulatory genes Coil and NPR1, and defense genes PR-1a and PR-1b, in the tobacco treated with the Bacillus spp. revealed an association with enhancing the systemic resistance of tobacco to TMV. A further analysis of two expansin genes that regulate plant cell growth, NtEXP2 and NtEXP6, also verified a concomitant growth promotion in the roots and leaves of the tobacco responding to the Bacillus spp.
Effect of the amount of fertilizer, plant density, and harvesting time on the production of tobacco leaf protein and fresh biomass was investigated. Flue-cured tobacco(M tabacum, L., cv. NC 82) seedlings were transplanted in the field dressed 200kg N per ha at 1$\times$105, 3$\times$105, 5$\times$105, 7$\times$105 plants per ha, and were harvested at the time when 6 and 10 weeks after transplanting, respectively. Harvest at 10 weeks after transplanting increased greatly number of leaves per plant and fresh weight of a plant, Precentage of senescent leaf weight, but significantly decreased fresh weight of a leaf and total protein contents g-1 fresh weight of leaf and stalk over the amount obtained from the harvest at 6 weeks after transplanting. Also, fresh leaf numbers of a plant, fresh weight of a leaf and of a plant, and total protein contents g-1 fresh weight of biomass were more decreased, but percentage of senescent leaf weight were remarkably increased under higher plant density. Therefore, it was seemed that harvesting at 6 weeks after transplanting under 1$\times$105 plant density per ha is more effective for producing higher yield of biomass and protein per plant than 10 weeks harvesting with 7$\times$105 population per ha. A trend was observed that biomass and protein yields per ha are positively correlated with plant population. Biomass yield per ha was the greatest at 7$\times$105 density(80.5t), but the peak of protein yield was at the near of 5$\times$105 population(2454kg as total protein) per ha on the regression curve. It was assumed that if tobacco plants are transplanted under 5$\times$105 plant density at the mid of May, and thereafter harvest at 6 weeks repeatedly during the growing season, it is possible to harvest 2~3 times per year, and to yield more 6.024kg of protein and over 140me1ric tons of fresh biomass ha 1 year 1 statistically in the korea tobacco growing regions.
This experiment was conducted to study the effect of soil water potenial on the growth and internal changes of stressed plants. The experimental imposition of soil water potential ( $\Psi$soil) were -0.1 to -0.2, -0.2 to -0.5, -0.5 to -3.0, -3.0 to -10.0 bar respectively. During water stress all growth rates were depressed, and the most sensitive period to water stress was found to be 10 to 25 days after transplanting. The water potential of leaf was declined rapidly within 12 hours after with holding of water. Nitrate reductase activity was decreased progressively as water deficit was built up in tobacco leaves, but the activity of alpha- amylase and super contents were increased. There were differences in peroxidase isozyme patterns between tile control and water stressed plant. New isozymes started to appear as tobacco leaf water potential decreased.
To explore the possibility of introducing Zea mays transposable element Ac(activator) which can be used as a mutagen and gene tag in tobacco plants other than maitre, we tried to introduce a cloned Ac element into tobacco cells by an Agrobacterium tumefaciens binary vector system. Transformation of N. babacum cv. Burley 21 tissues and regeneration to whole plant were carried out. The frequency of the transformed callus induced in shoot induction media was higher than that of transformed callus induced in callus induction media. However, the calli were not grown in the second selection media, and became yellow senescent calli. Regenerated tobacco plantlets with foreign gene were also obtained in shoot induction media containing 100 $\mu\textrm{g}$/ml kanamycin and 100$\mu\textrm{g}$/ml carbenicillin. The leaf tissues of transformant was also resistant to 1000 $\mu\textrm{g}$/ml kanamycin. The chromosomal DNAs of transformant and normal plant of N. tabacum were digested by EcoR I and Hind III but not by Pst I.
A double - stranded cDNA fragment (436bp) encoding coat protein of tobacco mosaic virus(TMV) was derived from the total 480nucleotides gene after reverse transcription of TMV RNA, and subclorled into a plant expression vector pBl 121, resulting in pBL 430. The plasmid DNA containing this chimeric gene was moved from E. cofi to Agrobacterium tumefaciens strain A28l, and was introduced in시 the tobacco plant by the Agrobocterium Ti - mediated transformtion system. The transformants were selected on a selection media containing kanamycin. The shoots add roots could be differentiated from the explants and whole plants were obtained. From Southern blot hybridization analysis, DNA extracted from transformants, it could be conformed that the chimeric gene fragment was inserted into the genomic DNA of tobacco plant.
This study was conducted to find out the effects of chlorine on the yield, chemical constituents and physical properties of Burley leaf tobacco. The results obtained are as follows. 1. There was no significant differences in growth and yield among chlorine application levels. 2. When chlorine being applied 4kg/10a or more, the cured leaf showed higher chlorine content and lower leaf quality remarkably. It was estimated that the critical application of chlorine was 3kg/10a. 3. Total nitrogen and alkaloid contents of cured leaf were increased by chlorine application, while the phosphorus, calcium and magnesium were not affected. 4. The reflectance color of red and yellow were decreased by cvhlorine application in cured leaf of field grown tobacco.
In vitro-tested ribozyme against synthesized cucumber mosaic virus (CMV) RNA (Agric. Chem. & Biotech. 37:56-63(1994)) was expressed in tobacco plant to develop virus resistant plants. The ribozyme sequence was linked to cauliflower mosaic virus 35S promoter and nopaline synthase(nos) terminator and this chimeric 35S-ribozyme-nos gene was sequenced. The sequenced chimeric gene was transferred to Agrobacterium tumefaciens LBA4404 using tri-parental mating system. The E. coli HB101 containing chimeric gene was incubated with E. coli HB101(pRK2073) as a helper and Agrobacterium tumefaciens LBA4404. Then Agrobacterium cells containing the ribozyme construct was cocultivated with tobacco leaf pieces. Ten different plants were regenerated from kanamycin containing MS medium. The presence of the ribozyme construct in the transgenic tobacco plants was confirmed by polymerase chain reaction (PCR). Seven different transgenic plants in ten different kanamycin resistant plants showed the expected size (570 base pairs) of 35S-ribozyme-nos gene fragment. Total RNAs were isolated from four different transgenic plants and separated on a 1% agarose gel containing formamide. Northern hybridization with 35S-ribozyme-nos gene fragment as a probe indicated that ribozyme transcripts may be degraded tv nuclease. Therefore, nuclease-resistant ribozymes are needed for the development of virus-resistant transgenic plants using ribozymes.
This study was conducted to evaluate the affect of soil conditioners and their application rates on the growth, Yield and quality of Flue-cured tobacco. Soil conditioners for this study were Montmorillonite arid Zeolite as clay mineral source and Rice hull and Sawdust as organic source. Their application rates were 500, 1000, 2000kg/10a. The growth of tobacco plants was promoted, compared with that of Non- treated plot (control) by Zeolite, Montmorillonite and Rice hull application but decreased by Sawdust application. During tobacco growing season soil moisture content, compared with control, was increased by Zeolite and Montmorillonite application but decreased by Rice hull and Sawdust application. In contrast to soil moisture content, soil temperature was higher in the Rice hull and Sawdust plot than in the plot of Zeolite and Montmorillonite. The treatment of Sawdust, compared with control, obviously accelerated the maturity of tobacco, increased reducing Sugar but decreased total alkaloids, nicotine and nitrogen content of cured leaves. The Sawdust Plot decreased the Yield (kg/10a) with significant difference by 1 %level but significantly increased leaf quality (Won/kg) and value (Won/10a) by 1%and 5 %level, respectively. The present application , ate of Nitrogen in Korea (12.5kg/10a) seems to be excessive for Flue-cured tobacco production.
To assess resistance of transgenic tobacco plants which overexpress superoxide dismutase (SOD) and ascorbate peroxidase (APX) in chloroplasts to water stress, changes in leaf water potential, turgor potential, stomatal conductance and transpiration rate were measured. Leaf water potential in all plants remained high up to day 4 after withholding water but thereafter decreased markedly. In spite of a remarkable decrease in leaf water potential, some of transgenic plants maintained higher turgor potential compared with control plant on day 12. In particular, the transgenic plant expressing MnSOD showed an outstanding maintenance in turgor pressure by osmotic adjustment throughout the experiment, resulting in high stomatal conductance and transpiration rate. However, among transgenic plants, osmotic potential was reduced more effectively in multiple transformants such as the double transformant expressing both MnSOD and APX, and the triple transformant expressing CuznSOD, MnSOD and APX than single transformants. Consequently, further research is needed to get general agreement on the tolerance of transgenic plants to water stress at different growth stages for each transgenic plant.
This study was conducted to identify useful germ plasm sources of resistance to black shank, Phytophthora. parasitica, in Nicotiana tabacum and to characterize the interaction between host plant resistance to black shank and pathogen variability. The six internationally represen-tative set of black shank resistant and susceptible tobacco genotypes and four Korean lines were tested for black shank on the field infested with P. parasitica at Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute during the 1992~1997 growing season. Almost 100% of the plants of the most susceptible genotype, Ky 14, did not survive. Complete survival of (MS Ky 14 x L8)F$_1$, which is resistant to reee 0 and ausceptible to reace 1, suggests that Race 0 is the only race of the P. parasitica in this bield. Coker 371 Gold and Beinhart 1000-1 having excellent resistance to black shank could be recommended as the parental cultivars for improving black shank resistance in Korea. Data of the diseased plants showed that KB 101 and KB 110 were medium high resistant and KB 103 and KB 108 being low resistant to black shank.
