Varietal difference in the occurrence of grey leaves in upland diverted from paddy field were investigated with several flue-cured tobacco varieties in Korea. Grey leaves caused by Fe, Mn and Cl toxicity were occurred in all varieties used in this experiment. The variety, NC82 and KF113, however, were more sensitive than other varieties to the toxicity. It's revealed that grey leaves had little effects on the contents of total sugar, nicotine and total nitrogen. NC82 and KF113 were increased in the ratio of occurrences in the paddy soils which had a tendency to concrete with the passage of days after transplanting. NC82 showed high occurrences of grey leaves in each of variety used and it was estimated that this variety had a traits of easy occurrences of grey leaves, and it might be presumed that there would be a close connection between the ratio of occurrence and amounts of absorbed causal materials accumulated in plants and tobacco varieties on the occurrence of grey leaf tobacco.
The coat protein (CP) gene was cloned from RNA genome of the Cucumber Mosaic Virus strain ABI (CMV-ABI) isolated in Korea. The comparisons of the nucleotide sequence of the cloned CP gene and its deduced amino acid sequences with other CP genes revealed that the CMV-ABI belongs to subgroup I (type I), CMV-ABI developed the typical mosaic symptom in infected plants. Tobacco plants (Samsun and NC82) were transformed by leaf-disc transformation via Agrobacterium, temefaciens LB4404 harboring pVCP, witch CMV-ABI CP gene was inserted into the pBI121, and a number of mature transgenic tobacco plants were developed. Southern and PCR analysis of genomic DNA from the transgenic plants showed that the CP gene was integrated into the genomes of the most of the transgenic plant. Result of the segregation patterns of resistance in T1 seedlings of the plants to kanamycin showed that the transgenic plants containing l,2 and 3 copies of CP gene were50%, 39% and 11% of the total transgenic plants, respectively.
In order to establish the integrated management of white grubs in ginseng field damages of ginseng plants (Panax ginseng C.A. Meyer) by the larvae of the larger black chamfer (Holotrichia morose Waterhouse) and the Korean black chamfer (Holotrichia diomphalia Bates) were investigated. Most of ginseng damages due to H, morose usually occurred in September through October, mainly on 2nd year plants. However, the damage by H. diomphalia occurred in September through October (usually in odd number of years) on 2nd year plants and in the following May and June on 3rd year plants. Therefore, in the fall of every odd number of years, synchronized occurrence of the two species caused severe damage on ginseng plants. Both of the 3rd instar larvae of the forementioned two species damaged ginseng roots regardless of the age of the plants when ginseng plants were artificially infested with them. Ginseng fields located at the slopes with good drainage are most likely damaged by them.
This study was carried out to screen herbs among herbal plants showing antimicrobial activity against some food spoilage and pathogenic microorganisms. One hundred fifteen kinds of herbal plants were extracted by 70% ethanol, and then they have been screened for antimicroorganisms. Six herbal plants such as Salviae radix, Dryopteris rhizoma, Terminaliae fructus, Araliae radix, Psoraleae fructus and Schisandrae fructus showed strong antimicrobial activities against Bacillus subtilis. Antimicrobial activities were showed in Anemarrhena radix and Dryopteris rhizoma on Candida albicans, and in Anemarrhenae radix, Dryopteris rhizoma and Polygalae radix on Schizosaccharomyces sp. It was revealed that eight herbal plants such as Dryopteris rhizoma, Salviae radix, Sappan ligunum, Sinomeniae radix, Schisandrae fructus, Rhui fructus, Sophorae radix and Inulae radix also showed antimicrobial activities on Streptococcus mutans. In addition, Anemarrhena radix, Curcuma tuber, Inulae radix, Polygonum radix, Sanguisorbae radix, Scutellariae radix and Terminaliae fructus and showed antimicrobial activities on Trichophyton mentagrophytes. Four kinds of herbal plants such as Dropteris rhizoma, Salviae radix, Terminaliae fructus and Scutellariae radix which showed broad antimicrobial spectrums were mixed by 1 : 1 ratio with the other herbal paints showing relatively strong microbial activities such as Terminaliae fructus, Sinomeniae radix and Scutellariae radix etc. The extracts of mixed herbal paints showed higher antimicrobial activities than those of single herbal plant.
A program was initiated to transfer potato virus Y vein-necrosis strain resistance from N. africana to N. tabacum The Fl plants between the above species were self-sterile, but all amphidiploid plants from the Fl plants and backcrossed flowers, that is, the N. tabacum flowers crossed with amphidiploid were self-fertile. The parent, amphidiploid plants of Fl, F2 population of the amphidiploid and the backcrossed generation were screened for a resistance of potato virus Y vein-necrosis strain isolated in Korea. The Chi-square values for the F2 population of the amphidiploid and the backcrossed generation fitted 35: 1 and 5 : 1 ratios of resistant to susceptible for the potato virus Y vein-necrosis strain, respectively. Therefore, it was found that the resistance of N. tabacum for the potato virus Y vein-necrosis strain was controlled by a single dominant gene.
Interleukin-11 (IL-11) is a cytokine that plays a key regulatory role in the immune system. Recombinant human IL-11 (rhIL-11) exerts a preventative effect against apoptotic cell death and inhibits preadipocyte differentiation. IL-11 also is used to stimulate the bone marrow to produce platelets in order to prevent low platelets that may be caused by chemotherapy. Unfortunately, the high production cost of IL-11 associated. In this study, we investigated the feasibility of transgenic plants for the cost-effective production of rhIL-11. Production of rhIL-11 proteins in whole-plant expression system will be more economical when compared to the current E. coli based expression system. The human rhIL-11 gene was codon optimized to maximize plant host system expression. IL-11 expression vector under the control of a constitutive cauliflower mosaic virus 35S (CaMV 35S) promoter was introduced into tobacco by Agrobacterium-mediated transformation. The 5'-leader sequence (called ${\Omega}$) of tobacco mosaic virus (TMV) as a translational enhancer was added to construct. Transgenic tobacco plants expressing various levels of rhIL-11 protein were generated. Western blotting of the stably transformed lines demonstrated accumulation of the appropriately sized rhIL-11 protein in leaves. This research demonstrated the efficacy of using tobacco as an expression system for the production of rhIL-11.
Cabbage plants were transformed with the potato proteinase inhibitor II (PINII) gene, bar gene, and hpt gene using Agrobacterium. The expression of the PINII gene was driven by its own promoter which was wound-inducible. Ten transgenic plants were obtained from medium containing hygromycin as a selection antibiotic. The integration and expression of PINII and bar genes were confirmed by Southern and Northern hybridization. Growth and development of diamondback moths (Plutella xylostella) and tobacco cutworm (Spodoptera litura) larvae were examined on $T_1$ plants. The weight of the larvae and pupae of these two insects grown on transgenic plants was not different compared to those grown on wild type plants. However, the pupation and emergence rate of diamondback moths and tobacco cutworms fed on some transgenic plants was lower than on wild type plants. These results suggest that the PINII transgene under the control of a wound-induced promoter may be used for control of insects in transgenic cabbage through reduction of insect progeny number.
Effects of neutron beam irradiation on seed germination, growth and RAPD pattern of tobacco (Nicotiana tabacum L. cv.; N. plumbaginifolia) and rice (Orya sativa L. cv.) plants were estimated. Seed germination rate was not significantly changed by the neutron beam treatment in both tobacco and rice seeds. And there was no significant differance in growth of the plants by the neutron beam treatment. Interestingly, however, some of morphological changes, including leaf shape (about $36\%$), stem color and leaf color were observed in neutron beam treated tobacco plants. In addition, abnormal flower in petal was observed in the neutron beam treated plant. This results indicate that neutron beam is able to use as an effective mutagen in plant mutations. Scorable products from 20 primers were obtained by RAPD analysis in the leaves of the beam irradiated tobacco plants and most of the plants showed the similar band patterns.
This experiment was conducted to investigate the amount of natural crossing at varying isolation distance and various transplanting time in tobacco. Normal variety and male sterile line showed an average of 1. 6 and 29% natural crossing in the adjacent plantings, respectively. The natural crossing was highest in the adjacent planting, as expected, and less at the farther distance from pollinator plants. Plants transplanted in April 20th were higher in natural crossing than those of other transplanting time.
Effect of liming material application on the prevention or cure of Mn toxicity symptoms including marginal leaf chlorosis in 3-year-old ginseng plants grown in acidic soils were investigated. It was found that the ratio of Fe to Mn was above about 0.5 and the ratio of Mn to Fe was below about 2.00 in 4-year old ginseng leaves when liming materials were applied in field experiments. It was apparent that the occurrence of marginal leaf chlorosis was decreasing affected by application of Ca, Mg and Fe. The appearance of marginal leaf chlorosis was decreased to about 78 percent in 4-year-old ginseng and to about 69 percent in 5-year-old ginseng, and then the fresh root weight was increased up to 66 percent in 4-year ginseng plants.
