EFFECTS OF THE CULTURE FILTRATE OF PSEUDOMONAS SOLANACEARUM ON THE CALLUS AND CUTTING OF TOBACCO PLANT (담배 Callus 및 삽수에 대한 세균성마름병균 (Pseudomonas solanacearum) 배양여액의 처리효과)

  • The typical dark brown stripe symptom of bacterial wilt disease was observed in the cuttings of tobacco stem treated with the culture filtrate of virulent Pseudomonas solanaceamm. And the tobacco callus create.4 with that culture filtrate showed deterioration of the callus 2 days after the treatment. On the contrary, the cuttings and the callus treated with the culture filtrate of the avirulent bacteria expressed no typical symptom and vigorous growth respectively. Therefore it was suggested that certain toxin which might be produced by the virulent bacteria could break down tobacco cells.

Effects of Ammonium, and Nitrate on Callus Growth of Tobacco and Soybean and Activities of Nitrogen Metabolizing Enzymes (암모늄태와 질산태 질소가 담배와 콩의 조직배양시 생육 및 질소대사 효소의 활성에 미치는 영향)

  • Soybean and tobacco tissue cultured with modified MS media containing 4 different ratio(as N) of nitrate to ammonium combination which were 3:0, 2:1, 1:2 and 0:3. The highest callus growth in soybean were observed in the 2:1 medium. The medium containing nitrate only was detrimental to soybean callus growth. Tobacco callus grown with nitrate-only grew as well as those in the 2:1 and very slowly with ammonium-only. In tobacco callus, the total nitrogen in the callus increased with the increase of nitrogen concentration in the medium, but in soybean callus, the opposite result was noted. Nitrate reductase activity in tobacco callus was high when grown with nitrate-only but low with ammonium-only. In case of soybean callus, nitrate reductase activity was high in the 2:1 and remarkably low in nitrate-only medium. Both in soybean and tobacco callus, the activity of glutamine synthetase was high with nitrate-only, but low with ammonium.

Regeneration of Tobacco Tissue Introduced with the Maize Transposable Element Activator (옥수수 전이인자 Ac가 도입된 연초조직의 재분화)

  • To explore the possibility of introducing Zea mays transposable element Ac(activator) which can be used as a mutagen and gene tag in tobacco plants other than maitre, we tried to introduce a cloned Ac element into tobacco cells by an Agrobacterium tumefaciens binary vector system. Transformation of N. babacum cv. Burley 21 tissues and regeneration to whole plant were carried out. The frequency of the transformed callus induced in shoot induction media was higher than that of transformed callus induced in callus induction media. However, the calli were not grown in the second selection media, and became yellow senescent calli. Regenerated tobacco plantlets with foreign gene were also obtained in shoot induction media containing 100 $\mu\textrm{g}$/ml kanamycin and 100$\mu\textrm{g}$/ml carbenicillin. The leaf tissues of transformant was also resistant to 1000 $\mu\textrm{g}$/ml kanamycin. The chromosomal DNAs of transformant and normal plant of N. tabacum were digested by EcoR I and Hind III but not by Pst I.

  • This study was conducted to obtain basic information about the transformation of ginseng tissue, identification of opine compound and protein, and saponin production from ginseng callus transformed with Ti-plasmic of AW$.$obacterium tumefaiens C58. Ginseng crown gall callus induced by pTiC58 could be continuously cultured on the Phytohormone-free medium. The transformation was reconfirmed by the detection and identification of opine compound, from the gall callus. The transformed ginseng callus contained higher amounts of protein than normal callus and the protein pattern of transformed callus was quite different from that of normal callus. The xylose which is not detected in the normal callus and ginseng root was identified in gall callus. The saponin contents of gall callus of ginseng were three times higher than that of normal callus, and ginsenoside composition of the transformed callus was similar to that of the cultivated ginseng root, but quite different from that of normal callus.

Effect of NaCl on Salt-tolerant Callus in Tobacco (내염성 담배 캘러스에 대한 NaCl의 효과)

  • Effects of various NaCl concentrations on salt-tolerant callus of tobacco were investigated. Selection of NaCl-tolerant (S) callus was conducted by subculturing Nicotiana tabacum cv. BY 4 callus in 200 mM NaCl-containing MS medium for more than 18 months. In spite of the long subculture period, characteristics of salt tolerance were maintained very stably. Significant differences were found in ion contents of each callus which was subcultured with treatment of various NaCl concentrations: Na+ and Cl- became higher but Mg2+, Ca2+ and K+ became lower with the increasing external salt contents. Therefore, the ratios of Na+/Ca2+ and Na+/K+ also increased resulting close to those of halophytic property. The contents of chlorophylls and carotenoids in S callus were estimated to 3.1 and 2.9 times more, respectively. than those of non-selected (NS) callus (control). The higher content of external NaCl tended to increase the amount of water soluble proteins and to decrease the amounts of the total sugars, reducing sugars and free amino acids. The activity of peroxidase was increased with higher contents of external NaCl in S callus, but it was maintained at a higher level than S callus at lower NaCl, followed by a subsequent decrease above 80 mM NaCl in NS callus. These results suggest that S callus may have a biological system converting energy source to efficient growth leading to reduction of the growth inhibition under stress environment.

  • The present study was conducted to obtain some basic information on the shoot formation from crown gall callus and the characteristics of teratoma shoot derived from crown gall induced by inoculation of Agrobacterium tumefaciens C58. Crown gall callus could be continuously cultured on the phyohormone free basic medium. The growth of crown gall callus was inhibited when BA were added to the cultural media. Shoot formation from crown gall callus fail to be initiated except teratoma shoot which induced on the phytohormone free medium after several subculture on rare occasions. Teratoma shoot could not form root and grow as normal shoot. Addition of BA to cultural media was not effective for shoot elongation, reduction in multiple shoot formation, but IBA was somewhat effective for shoot elongation of teratoma shoot, never for root formation.

  • Structures of the adventitious root meristem induced from callus culture of tobaco (Nicotiana tabacum cv. NC 82) leaves were investigated by light and transmission electron microscopy. Structural differences between in vitro root and callus cells were also examined by the microscopy. The submicroscopic features of the in vitro root cells were as follows. Intercellular spaces were not developed and nuclei with two nucleoli were observed occasionally. Plasmodesmate were found in groups or sing1y on transverse and longitudinal walls. Amyloplast solely filled with starch grains, with one to five electron - dense bands, was surrounded by single membrane. in the callus cells, vacuolization of central part in the cytoplasm having mitochondria with swollen cristae and starch grains like those of in vitro root cells was a distinct feature. Vesicles which were found between cell wall and plasma membrane may be arisen by a process of protoplasmic invagination. By comparing of ultrastructures between the cells of callus and in vitro roots we found that the distinct differences lied on thickened cell walls and hypertrophed vacuoles in the former, and less thickened cell walls and several small vacuoles in the later.

Varietal Response of Tobacco Plants Through Tissue Culture to Butachlor and Bialaphos Herbicides (조직배양(組織培養)에 의한 제초제(除草劑) Butachlor와 Bialaphos에 대(對)한 담배의 품종간반응(品種間反應))

  • This study was carried out to determine effect of butachlor [N.-(buthoxymethyl)-2-chloro-N-(2,6-diethylphenyl) acetamide] and bialaphos [2-amino-4(hydroxy)(methyl) phosphionyl] butyryl-alanylalanine sodium salt on the germination of tobacco seed, induction and growth of callus from tobacco. Further, fatty acids and ammonia content of tobacco calli were determined. Bialaphos had no effect on tobacco seed germination, but the growth of seedling was markedly affected by an application of 10 ppm bialaphos. However, regardless of varieties tested, tobacco seed germination was completely inhibited by $5{\times}10^{-5}M$ of butachlor. At an application of $5{\times}10^{-5}M$ butachlor, tobacco seeds were to some extent germinated and showed further growth. Hyangcho among varieties tested, showed the most tolerant response to butachlor. In induction of callus from various tobacco varieties and their growth, aromatic type of tobacco varieties exhibited the most tolerance against bialaphos. However, no distinct varietal differences were determined in the treatment of butachlor. The major fatty acids identified in tobacco calli were palmitic, oleic and linoleic acid. No marked difference in terms of fatty acids was observed among tobacco varieties used, but it was observed that there was the higher ratio of quantity in unsaturated fatty acids over saturated one, bialaphos treatment accumulated about 9 times higher ammonia content than that of the untreated control, giving an evidence that bialaphos might inhibit glutamine synthetase activity.

연초(Nicotiana tabacum cv. Xanthi) 배양세포로부터 Ubiquinone-10 생산을 위한 현탁배양

  • The effect of phytohormones, light and phosphate on in vitro production of ubiquinone 10 from the suspension cultures of Nicotiana tabacum cv. Xanthi callus was investigated. The inoculum size and cultured time in the suspension culture had to be at least over 2 % of medium volume at 15 days for the excellent growth of Xanthi callus. The growth of Xanthi callus in the suspension culture was improved by addition of NAA and 2,4-D, especially NAA 1.0mg/1 alone, at the light condition. The optimal concentration of phytohormone was 0.1 mg/l 2.4-D and 1.0 mg/l NAA for productivity of ubiquinone 10 in the suspenseion of Xanthi callus. Addition of 3mM KH$_2$PO$_4$ to the medium was more effective in promoting ubiquinone-l0 formation than other concentration in the light condition. Content and production of ubiquinone-l0 in the suspension cultures of Xanthi callus were the highest at the MS media containing 0.5mg/L kinetin, 0.5mg/L 2,4-D, 1.0mg/1 NAA, 3mM phosphate and 2 % inoculum in the light.

Protoplast Isolation and Fusion of Nicotiana glauca and Solanum tuberose Transformed by Selectable Marker Genes (표지유전자로 형질전환된 연초와 감자로부터 원형질제의 유리 및 융합)

  • Protoplasts were isolated from mesophyll of tobacco(Nicotiana glauca) transformed with kanamycin-resistant gene (NPT II gene) and potato hairy root callus containing Ri plasmid of Agrobacterium rhiEogenes, and protoplasm fusion was made between the isolated protoplasts. The transgenic tobacco leaf tissue could grow on the media containing high concentrations of kanamycin, but not on the phytohormone-free media. On the other hand, the potato hairy root calli could be cultured on the phytohormone-free media but not on media containing more than 40 ㎍/ml kanamycin. In these conditions, the viability of both protoplasts were above 90%, These selection markers were used for the selection of protoplasts fused between the two, i.e. protoplast fusion was detected using selection media containing 100㎍/ml kanamycin and with no phytohormone. The mixture of 1.0% cellulase, 0.3% macerozyme, and 0.7M mannitol was best for the maximum protoplast production for tobacco, and that of 2.0% cellulase, 2.0% macerozyme, 1.0% dricelase, and 0.5M mannitol for potato. Both tobacco mesophyll and potato callus protoplasts were fused by using PEG solution on the selectable medium. Cell walls were regenerated after 5 days in this medium, and colonies were alive until 4 weeks after cultural, but died after 6 weeks.
