버어리종 역병저항성 계통 KB 104와 KB 106의 육성경과 및 특성 (Development of Black Shank Resistant Burley Tobacco Germplasm, KB 104 and KB 106, and Their Characteristics)

  • Black shank(Phytophthora parasitira roar. nicotianae) resistant burley tobacco (Nicotiana tabacum L.) germplasms, KB 104 and KB 106, were developed by Korea Ginseng & Tobacco Research Institute. KB 104 was developed from the single cross of Burley 21$\times$Newton 77, using a modified pedigree method. KB 104 was highly resistant to black shank, and its agronomic characteristics and chemical contents were comparable to those of Burley 21, and value per 10a was slightly higher than Burley 21, KB 106 is a maternally derived doubled haploid made by N. africana method from the single cross of Burley 21$\times$ Va 509. KB 106 was also highly resistant to black shank, had two more harvestable leaves per plant and flowered three days later than Burley 21 did. Total alkaloid and nicotine contents of KB 106 were significantly lower than those of Burley 21. But its nornicotine content was higher than Burley 21 5.   • This study was conducted to identify useful germ plasm sources of resistance to black shank, Phytophthora. parasitica, in Nicotiana tabacum and to characterize the interaction between host plant resistance to black shank and pathogen variability. The six internationally represen-tative set of black shank resistant and susceptible tobacco genotypes and four Korean lines were tested for black shank on the field infested with P. parasitica at Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute during the 1992~1997 growing season. Almost 100% of the plants of the most susceptible genotype, Ky 14, did not survive. Complete survival of (MS Ky 14 x L8)F$_1$, which is resistant to reee 0 and ausceptible to reace 1, suggests that Race 0 is the only race of the P. parasitica in this bield. Coker 371 Gold and Beinhart 1000-1 having excellent resistance to black shank could be recommended as the parental cultivars for improving black shank resistance in Korea. Data of the diseased plants showed that KB 101 and KB 110 were medium high resistant and KB 103 and KB 108 being low resistant to black shank.

감자바이러스 Y 및 역병 저항성 연초 버어리종 신품종 KB 110의 육성 및 농경적 특성 (Breeding of Burley Tobacco KB 110 Resistant to PVY and Black Shank and its Agromomic Characteristics)

  • The vein-necrosis strain or potato virus Y (PVY-Vff) and black shank (Phytophlhora parasitica roar. nicotianae) causes severe damage on burley tobacco(Wicotiana tabacum L.) in Korea, A new burley tobacco resistance to PVY and black shank, KB 110, was developed by Korea Ginseng and Tobacco Research Institute. It was developed from the cross of Burley 21 with TC 591 in 1990, and was backrossed to Burley 21 in the following season. TC 591 has resistance to PVY and moderate resistance to race 0 of black shank, but it is susceptible to tobacco mosaic vim (TMV). KB 110 was evaluated for its resistance to PVY, TMV and black shank in the greenhouse and at fields for preliminary and performance trials. KB 110 which has secreting glandular trichomes was resistant to PVY-VN, TW and black shank. It had an erect growth habit and two more leaves per plant than that of Burley 21, and matures two to three days later. It yielded approximately 3 percent more cured leaf than the standard cultivar Burley 21, but other plant characteristics were very similar to those of Burley 21. It had acceptable standards for chemical and physical characteristics of lured leaf on regional farm test in 1995-1997. KB 110 produced average yields of good quality tobaccos and was appeared to be resistant to PVY inwhere occurrence of the virus are severe chronic at burley growing area.

연초 버어리종 웅성불임 일대잡종 KB 111의 육성경과 및 농경적 특성

  • The vein necrotic strain of Potato Virus Y (PVY - VN) and black shank (Phyto-phthora parasitica var. nicotianae) are the two major diseases causing severe damages especially in burley tobacco (N. tabacum L.) area in Korea. A new tobacco variety, KB 111, resistant to PVY and black shank disease, was developed by Korea Ginseng & Tobacco Research Institute in 1997. It is a male sterile(MS) F$_1$ hybrid of the cross between MS TC 613 and KB 108. KB 111 was compared to Burley 21 on the agronomic characteristics and disease resistances in performance tests: It possessed upright growth habit and flowered two days later than Burley 21. It was resistant to both PVY and black shank and yielded about 3% more cured leaf than Burley 21, but other characteristics are very simiar to those of Burley 21. The chemical composition and physical properties of the cured leaf of KB 111 were as much acceptable as those of Burley 21 while it produced average yield of good quality leaf and appeared to resistant to PVY and black shank disease on regional farm test in 1998.

In Vitro Sensitivity to Metalaxyl of Phytophthora parasitica var. nicotianae Isolates from Burley Tobacco in Korea

  • In disease surveys from 1986 ti 1998, disease incidence of tobacco black shank was gradually increased in burley tobacco from 1996. To study the causes of the disease occurrence, one hundred and fourteen isolates of Phytophthora parasitica var. nicotianae (Ppn) were collected from burley tobacco-growing areas in the southern part of Korea during 1996-1997, and tested in vitro for meatlaxyl sensitivity which was determined by measuring the mycelial growth on corn meal agar (CMA) amended with metalaxyl. Of the tested isolates, 78.1% showed sensitive to metalaxyl, having $\textrm{ED}_{50}$ values less than 1.0 $\mu\textrm{g}/$\textrm{ml}, while 1.7% was resistant weth $\textrm{ED}_{50}$ greater than 100 $\mu\textrm{g}/$\textrm{ml}. Ppn isolates from three provinces, Chungnam, Chonbuk and Chonnam showed similar distributions of metalaxyl sensitivity. Metalaxyl-resistant isolates were not significantly different from metalaxyl-sensitive ones in mycelial growth rate, chlamydospore formation capacity and size of the spore, and pathogenicity on tobacco plant (cv. Burley 21). These results suggest that the metalaxyl-resistant Ppn in burley tobacco may be one of the major factors to cause the higher occurrence of the tobacco black shank in the burley tobacco-growing area.

내병성 버어리종 신품종 "KB 101"의 육성경과 및 특성 (Performance of "KB 101" a New Burley Tobacco Resistant to Bacterial Wilt and Black Shank)

  • KB 101 is a bacterial wilt(Pseudomonas solanaceamm E.F. Smith) and black shank (Phytophthora nicotianae Breda de Haan Var. nicotianae Waterhouse) resistant cultivar of burley tobacco (Nicotiana tabacum L.) KB 101 was developed by the Korea Ginseng&Tobacco Research Institute, and released in 1987. KB 101 was developed from a single plant selection in the F2 generation derived from the double cross, [(Burley 21X Burley 37) X (Burley 64X Ky 16)]. Burley 37 and Burley 64 were the source of resistance to bacterial wilt and black shank. Yield trials were conducted in the Fs through F6 generations at the four Exp. Stn. of Korea Ginseng &Tobacco Research Institute as JB 7705-1. On-farm yield trials were conducted in the F7 through F9 generations at the 45 locations of burley tobacco growing area from 1984 to 1986 as KB 101. KB 101 has an erect growth habit similar to that of Burley 21: plant size is larger and has more leaves than those of Burley 21. It is late maturing cultivar that flowers approximately 3 days later than Burley 21. The physical characteristics and chemical composition of KB 101 were similar to those of Burley 21.

Metalaxyl 저항성 담배역병균(Phytophthora nicotianae) 방제를 위한 약제 선발 (Chemical screening for control of Metalaxyl-resistant tobacco black shank disease)

  • 우리 나라 농가에서 담배역병 방제약제로 사용되고 있는 Metalaxyl의 약제 저항성균 발생억제 및 방제를 위한 대체 약제를 선발하기 위하여 시중에서 판매되고 있는 역병방제용 약제 중 Dimethomorph를 비롯한 5종의 약제를 구입하여 시험하였다. 실내 시험에서는 Propamocarb hydrochloride는 Metalaxyl과 유사한 반응을 보여 서로 교차저항성이 있는 것으로 조사되었으며 Oxadixyl + Mancozeb, Dimethomorph + Mancozeb 등이 실내시험 및 포장시험 결과 약효가 우수하여 Metalaxyl 대체약제로 선발되었다.

  • 각 품종간 역병 저항성 정도는 접종 4일째 부터 심하게 발생되었으며 특히 황색종에서는 접종 4-12일 사이에 심하게 이병증상을 나타냈고 재래종은 6-20일 사이, 일본종은 4-13일, 중국종은 6-14일, 터어키종은 8-17일, 음건종은 5-14일 사이에 발병이 심하였으나 발병이 늦은 품종을 저항성으로 본다면 1. 황색종 : Cash, Oxford-1 N.C-75 N.C-95 Goldencure, Delcrest, Oxford-3. 2, 재래종 : Sinzangtchio Muktchio, Sosa-3, Mokgitchio. 3. 일본종 : Nambu, Ibusukey, Maruha, Enshu. 4, 중국종 : Gkzuoe, Yungzung Tongzuoe. 5. 터키종 : Samsun, Baffra, Xanthi. 6. 음건종 : B-Hw, Connecticut-Broad leaf, C. Havana, Tennessee Black 등이 저항성 품종으로 나타났고 특히 재래종 계통이 내병성이 강한 품종으로 육종모본선발에 가치가 크다고 하겠다.

  • Burley tobacco (Nicotiana tabacum L.) KB 108 was developed from a single cross between KB 104 and TC 591 which was developed from a cross between Burley 49 and Tobacco Introduction 1406. It was tested for its resistance to black shank, potato virus Y(PVY), TMV and agronomic characteristics under field conditions. KB 108 has resistance to tobacco mosaic virus(TMV) and necrotic strain of potato virus Y(PVY-VN) with secreting glandular trichomes. It has also moderate resistance to black shank caused by phytophthora parasitica val. Nicotianae. KB 108 has an up-right plant growth habit similar to Burley 21. It flowers about 1-2 days later than Burley 21. The leaf width and length of KB 108 are approximately 3 cm wider and longer than those of Burley 21. The yield of KB 108 was higher 4%, nearly equal in value per kg compared to Burley 21.

Stable Expression of TMV Resistance and Responses to Major Tobacco Diseases in the Fifth Generation of TMV CP Transgenic Tobacco

  • TMV resistant lines (TRLs) originated from the Blo plant of Nicotiana tabacum cv. NC82 transformed with TMV coat protein cDNA which initially showed delayed disease symptom were selected for increased resistance in each subsequent generation. The result of field experiment of the transgenic tobacco lines in the fifth generation for TMV resistance and their response to other tobacco diseases (black shank, bacterial wilt, and powdery mildew) is described in this report. When fifteen TRLs of the fifth generation were tested for TMV resistance by mechanically inoculating the individual plants, over 95 percent of the plants of 6 lines showed complete resistance even 8 weeks after the inoculation. Average frequency of the resistant plants in TRLs of the fifth generation 8 weeks after the inoculation was 87%. Stable insertion and expression of TMV coat protein cDNA in the fifth generation of the transgenic tobacco plant were confirmed by PCR and immunoblot hybridization, respectively. All TRLs were resistant to the black shank but were susceptible to the bacterial wilt disease and the powdery mildew to the same degree as non-transgenic NC82 was. Therefore, it was indicated that the phenotypes related at least to disease resistance were not changed in the transgenic tobacco.
