Seasonal Changes of Chemical Compositions in Leaves, Shoot and Berries of ‘Delaware’Seedless Grapes induced by Gibberellic Acid (Delaware 포도의 무핵재배에서 엽${\cdot}$신초${\cdot}$과립의 생장 및 성숙에 따른 화학성분의 변화)

  • This study was carried out to clarify seasonal changes of chemical compositions and their interrelation in leaves, shoots and berries treated with gibberellin($GA$_{3}$)for seedless grapes in ‘Delaware’grapevines. the clusters were dipped twice with 100ppm $GA_{3}$: 10 days befor and after the full bloom. The reaults obtained as follows; 1. Cumulative growth curve of berry fresh weight showed a double sigmoid curve and the characteristics of three distinctive growth stages(I, II, III)were weekened with $GA_{3}$ treatment. 2. while the contents of ash, total carbon, total nitrogen and total carbohydratd had little reation with edvelopment and ripeness of berries, those of total sugar and starch jad close relation, viz., they decreased with enlargement and maturity fo berries, but increased rapidly after harvest in leaves and shoots. Especially, total sugars in leaves and shoots decreased coincidently with starch-increasing in shoots at November. 3. The contents of total soluble solid and reducing sugar in berries increased rapidly at growth stage III, but those of total titratable acidity and organic acid decreased coincidently with sugar-increasing. 4. The berry-hardness increased until growth stage I, and then stagnated until gtowth stage II, and then increased rapidly at growth stage III. pH of berry-juice decreased until growth stageII, afterwards increased at growth stage III. 5. By correlation and path coefficient analysis between qualitative characters and the ratio of total soluble solid to titratable acidity($^{\circ}$Brix/Acidity), total correlation coefficients were all highly significant. Of these characters, pH and viscosity of berry-juice were positive, but brightness and hardness of berry, negative. The direct effect of pH on $^{\circ}$Brix/Acidity ratio, p4y=0.9090, was large positively and those of berry-hardness and juice-viscosity, p1y=0, 5938, median and, p2y=0, 3550, small, respectively. Direct effect of brightness was negatively small.

Production and physicochemical properties of black bean yogurt made with lactic acid bacteria isolated from vinegar and kimchi (식초와 김치 유래 젖산균으로 제조한 서리태 발효우유 제조 및 물리화학적 품질 특성 연구)

  • This study evaluated the physicochemical properties of a novel black bean milk yogurt made by lactic acid producing bacteria isolated from vinegar (Bacillus acidiproducens, BA) and kimchi (Lactobacillus plantarum, LP). Commercially available Lactobacilli mixture for yogurt (YF) were regarded as control bacteria. To investigate optimal conditions to make black bean yogurt, different types of bacteria (BA and LP) and dilution ratios of black bean milk and water (1:1, 1:1.5, and 1:2) were used. Titratable acidity and pH were affected by these dilution ratios, but not by the different types of Lactobacilli used. Titratable acidity and pH were lowest in the 1:1 diluted sample. The visible cell count, sugar content, and viscosity were significantly higher in the sample containing the black bean milk: water ratio of 1:1 than the rest of the samples. In the sensory test, black bean yogurt made with BA showed a higher overall acceptability score, similar to yogurt made with YF. In conclusion, black bean yogurt made by fermentation with BA and with 1:1 black bean milk:water dilution ratio was better than ones made with LP and at other dilution ratios.

Measurement of Lactoferrin, IgA, IgG1, IgG2, Antibacterial Activity, and Lactic Acid Bacterial Growth in Holstein Colostrum (Holstein 초유 중 Lactoferrin, IgA, IgG1, IgG2 정량과 미생물의 성장에 미치는 영향)

  • This experiment was carried out to measure the content of lactoferrin, IgA, $IgG_1,\;IgG_2$, in Holstein colostrum, and to test the effect of it's colostrum on the antibacterial activity to pathogenic bacteria and the growth stimulation of lactic acid bacteria. Colostrum was collected at the first, second, and third day after parturition in summer and winter season. The levels of lactoferrin, IgA, $IgG_1,\;and\;IgG_2$ in Holstein cow colostrum were 0.30 mg/mL, 0.37 mg/mL, 4.00 mg/mL, 0.37 mg/mL, respectively, on the first day of the summer season whereas they were 1.16 mg/mL, 2.60 mg/mL, 13.35 mg/mL, 1.30 mg/mL on the first day of the winter season, postpartum. Heat treated ($65^{\circ}C$ for 30 min) or non-treated colostrum showed antibacterial activity toward Escherichia coli. The growth of commercial mixed strains (Bifidobacterium longum, Lactobacillus acidophilus, Streptococcus themophilus), L. acidophilus, L. casei, L. bulgaricus, and L. lactis subsp. cremoris were improved in first, second and third day colostrum compared to normal milk. Commercial miked strains (B. longum, L. acidophilus S. themophilus) lowered the pH to 4.97-5.22 and 4.89 while increasing the titratable acidity to 0.75-0.88% and 0.70% in colostrum and normal milk, respectively. However, L. casei, L. bulgaricus, L. lactis subsp. cremoris lowered the pH to 5.96-6.47 and 6.5-6.8 while increasing the titratable acidity to 0.29-0.48% and 0.20-0.25% in colostrum and normal milk, respectively.

Quality Characteristics of Brown Rice Vinegar by Ferment Ratio (발효제 비율에 따른 현미식초의 품질특성)

  • Brown rice vinegar was made from brown rice mash with different brewing starter addition rates in static culture, and its quality characteristics were investigated. As a result, the amounts of alcohol produced in the fermentation process were shown to be 9.1, 8.8, 8.6 and 8.5% in the Nuruk 75 : crude enzyme 25 (B), Nuruk 50 : crude enzyme 50 (C), Nuruk 25 : crude enzyme 75 (D) and Nuruk 0 : crude enzyme 100 (E), respectively. The higher the percentage of crude enzyme added was, the lower the alcohol content that was produced. Nuruk 100 : crude enzyme 0 (A) was appeared to contain the lowest alcohol content (7.7%). In addition, the titratable acidity in all the groups was about 0.7%. The final titratable acidity (BV) of brown rice vinegar made with static culture was the highest (approximately 5.2%). The initial pH appeared to be between 3.6~4.0 and steadily decreased as the fermentation progressed, and the pH was almost unchanged after 15 day fermentation. The examination of the changes in the organic acids showed that the acetic acid content was similar in all the groups, and that the single starter added (AV, EV) group had much more other organic acids than the mixed starters added (BV, CV, DV) group. From these results, the mixed starters (Nuruk and crude enzyme) added group appeared to be superior to the single starter added in terms of alcohol production ability and vinegar quality. As the future aging process, however, is expected to change the flavor components and sensory characteristics, studies on various quality factors of vinegar are needed.

Changes in Physicochemical Characteristics during Soaking of Persimmon Pickles Treated with Organic Acids and Sugars (단감 장아찌의 저염 침지 중 유기산과 당의 종류에 따른 이화학적 특성의 변화)

  • In order to develop a new persimmon pickle with a low salt concentration, changes in physicochemical characteristics during soaking of persimmon pickles treated with different organic acids and sugars were investigated at $15^{\circ}C$ for 70 days. The pH of persimmon pickles decreased from pH 6.64 at initial stage to pH 3.99 (control group), pH 2.76-3.33 (citric acid treatment, CT), and pH 3.08-3.34 (acetic acid treatment, AT) after 70-days of soaking. Titratable acidity of AT was higher (0.86-1.18%) than that of CT (0.61-0.78%). After 70-days of soaking, salinity of persimmon pickles treated with stevia leaf powder, sodium saccharin, sucrose, and isomalto-oligosaccharide were 4.94-4.96, 4.00-5.15, 2.99-3.31, and 3.03-3.30%, respectively, and 1.54-3.70% lower than that of the control group (6.69%). The hardness of persimmon pickles in all cases was increased up to 20-40-days of soaking and then decreased. L (lightness), a (redness), and b (yellowness) value of persimmon pickles treated with citric acid were higher than those of AT and the control group. Sensory evaluation of persimmon pickles after 70-days of soaking revealed that CT had the highest scores for color, texture, and overall acceptability. These results indicate that the salt concentration of persimmon pickles could be lowered by the addition of citric acid and various sugars during soaking.

Comparison of the organic acids, fusel oil contents and antioxidant activities of Yakju with the additions of various rice cultivars (쌀 품종별 약주의 유기산, fusel oil 함량 및 항산화활성 비교)

  • The organic acids, fuel oil contents and antioxidant activities of Yakju with the addition of rice varieties were investigated. The rice cultivars that were used in this study were Ilmi rice, Hopyung rice, Hopum rice, Hanarum rice, Anda rice and Indica rice. The study results are as follows: the pH values of Ilmi Yakju and Hanarum Yakju were 4.14 and 4.07, respectively, and those of the other Yakjus, 3.92~3.98. The total titratable acid content of the Yakju for which Indica rice was imported from Thailand was highest among the samples. The reducing sugars content of the Yakju for which Indica rice was used was higher than that of the other samples. The ethanol content of the Hanarum Yakju was higher than that of the other Yakjus, and the lowest ethanol content was found for the Indica Yakju. The major organic acid component of the Yakju was lactic acid. The total organic acid content was highest in the Indica rice Yakju. The amounts of the following main fusel oils were highest, in the following order: 2-butyl alcohol, 2-pentanol, N-propyl alcohol, acetaldehyde, ethyl acetate, methyl alcohol and 2-methyl-1-propyl alcohol. The DPPH free radical scavenging activity of the Indica rice Yakju was much higher than that of the Ilmi rice Yakju and the Hanarum rice Yakju. Also, the SOD activity of the Indica rice Yakju was strongest among all the samples. The total polyphenol content of the Indica rice Yakju was the highest among all the samples.

Quality Properties of Capsule Type Meju Prepared with Aspergillus oryzae (Aspergillus oryzae를 이용한 캡슐형 메주의 품질특성)

  • In order to improve some problems such as contamination of undesirable mold, mycotoxin production and excessive drying on the surface of traditional meju, we developed a capsule type-meju(CM) coated with steamed black bean and Aspergillus oryzae - rice koji(0.3%, w/w) mixture to surface of traditional meju and fermented at 25$^{\circ}C$ for 14 days under 80% relative humidity. Contamination of undesirable mold on the surface of CM was not detected within 2 weeks and some genus Penicillium molds on control meju without koji were found naturally after 12 days of fermentation. The moisture content of meju was showed to be in the range of 34.7 - 29.4% being 32.7%(w/w) of mean value. Titratable acidities in CMs prepared with black bean(BCM) and soybean(SCM) were much higher than that in control meju, and BCM was similar to SCM. Free sugar content in BCM(123.98 mg%) was 10 times and 2.1 times higher than that in control meju(15.02 mg%) and SCM(59.85 mg%), respectively. Amino type nitrogen content in control meju was 147.00 mg% and its content in BCM(255.50 mg%) was 1.37 times higher than that in SCM(187.25 mg%). Total organic acid content in BCM(95.98 mg%) and SCM(1l9.98 mg%) were much higher than that in control meju(26.44 mg%), and then lactic and malic acid contents were markedly changed according to capsulation of meju. Lightness value(L) of Hunter color index was much higher in BCM than in SCM. Fatty acid composition of CM was not different as compared to control meju. Total free amino acid content in BCM(1039.70 mg%) was 4.4 times and 2.4 times higher than that in control meju(236.45 mg%) and SCM(556.07 mg%), respectively.

Effect of Chemical and Anti-fogging Agent Treated MA Packaging Film on Freshness Extension of 'Fuji' Apples (산, 염기 처리된 제올라이트와 결로 방지제를 함유한 기능성 MA포장재가 '후지' 사과의 선도유지에 미치는 영향)

  • To investigate the effects of functional MA films (FMA) masterbatched by zeolite powder treated with 1 N HCl, 0.5 N NaCl solution and anti-fogging agent (NA) compared with control on the freshness extension of 'Fuji' apples during storage at $15^{\circ}C$. Preference, weight loss, total ascorbic acid, sugar content, acidity, change of gas composition in package were measured. After 150 days of storage, the weight loss of control (L) apples was 1%, that of apple in FMA film (NA) was 1.5% after 150 days. Total ascorbic acid content of apples in control (L) after 150 days was 2.09 mg%, those of apple in FMA film (NA) was 2.72 mg%. The titratable acidity of apple in FMA film was higher than that in control, while soluble solids content of apples in FMA film was lower than that in control after 150 days. Ethylene gas adsorbability in control was 192.2 ppm and those in FMA film was 141.4 ppm after 150 days. Overall, apples in FMA film was better than that of control. It was verified that apples packed with LLDPE film (control) lasted about 130days, while those with FMA films lasted about 170 days. Also, FMA films treated with anti-fogging agent few different compare to nontreated film, but commerdity on the display in market was considered higher than that of non-treated anti-fogging agent.

Quality characteristics of Yakju at addition sprout and root of reed (갈대 뿌리와 갈대순 첨가에 따른 약주의 품질특성)

  • This study was performed to improve the availability of reed and quality of traditional Yakju by using roasted reed. The pH of sprout Yakju was 3.43, whereas the pHs of other Yakjus were 3.57. No remarkable differences were observed in contents of total titratable acids and reducing sugars. Color changes were measured by Hunter's color value; L value decreased, whereas redness (+a) and yellowness (+b) increased during fermentation. Ethanol content of Yakjus prepared with sprout and root were 15.30% and 15.28%, respectively, which were lower than that of control Yakju (15.31%). However, there were obvious differences of ethanol contents in three types of Yakjus. The major free sugar and organic acid were glucose and lactic acid, respectively. The total polyphenol contents of sprout Yakju (4.26 mg%) and root Yakju (4.21 mg%) were much higher than that of control Yakju (2.56 mg%). The DPPH (1,1-diphenyl-2-picryl-hydrazyl) free radical scavenging activities of sprout Yakju and root Yakju were higher than those of control Yakju. Yakju prepared from 0.3% addition of sprout showed the highest score in overall preference from the sensory test.

Effect of Lysozyme, Glycine and EDTA on the Kimchi Fermentation (Lysozyme, Glycine 및 EDTA의 첨가가 김치 발효에 미치는 영향)

  • Effects of addition of 120ppm lysozyme and 0.12% glycine into Kimchi liquid on the growth of lactic acid bacteria (LAB) were investigated. The results showed that the growth of LAB was significantly reduced as much as 10$^1$~10$^4$cfu/$m\ell$ for the liquid having pH 5.5 during storage at 32$^{\circ}C$ while the liquid of pH 4.4 showed a reduction of LAB growth by 10$^1$~10$^2$cfu/$m\ell$. Combined addition of lysozyme and glycine was found to be more effective than those of individual additions. The decrease in pH of Kimrhi added with 100, 200 and 1, 000ppm lysozyme was slower than that of control, but there was no significant pH difference at late stage of fermentation. However Kimchi added with 100ppm lysozyme and 1% glycine decreased in the rate of pH change. Change in titratable acidity was also slowed up for Kimchi added lysozyme and glycine. The growth of LAB could be inhibited as much as 10$^1$to 10$^2$cfu/$m\ell$ in the Kimrhi sample added with 120ppm lysozyme and 2mM EDTA compared with other treatments including control.
