  • It is our common understanding that biological materials like microorganisms, tissue and cell cultures, seeds, plants and animals are inevitable resources for the development of science and technology. Culture collections which are reservior of biological materials now occupy a central position in life sciences and biotechnology. The World Data Center of Microorganisms (WDC) is a infrastructure of culture collections in the world realizing quick and smooth exchanges of information and microorganisms to support research and development in those fields. The WDC was relocated from University of Queensland in Australia to out institute RIKEN in 1986. This article introduces a function for WDC in RIKEN (RIKAGAKU-KENKYUUSHO, the Institute of Physical and Chemical Research)

  • Ultrastructural observations of mycelial and tissue phase with dimorphic fungal pathogen Coccidioides immitis were studied by electron microscopy of thin sections. 1. In mycelial phase of C.immitis contains normal cell components such as nucleus, mitochondria, endoplamsic reticulum, intracytoplasmic membrane system, cell wall and cell membrane as observed in the other encaryotic cells. 2. In tissue phase of C. immitis was larger than mycelial phase in cell size and observed much more vacuoles than mycelial phase. 3. In the contrast of mycelial phase of C. immitis, the tissue phase of cells were observed fibril form of capsular layer.

  • The comparative study on the predominant cultivable periodontopathic bacteria were done 2 weeks after the application of the e-PTFE membrane and collagen membrane in the controlled tissue regeneration procedures. The purpose of the present study also included the antibiotic susceptibility test (ciprofloxacin, tetracycline, clindamycin) of these cultured organisms. 0.1% chlorhexidine mouthwash (10ml twice/day for 6 weeks) and systemic doxycycline (200mg/day for 2 weeks) were administered for supragingival and subgingival plaque control respectively. Four clinical isolates of A.a. from 2 patients were found to be resistant to tetracycline which were susceptible to clindamycin and ciprofloxacin. One isolate of W.r. and two unidcntified microorganisms were resistant only to clindamycin and one isolate of NID BPB and E.c. and two isolates of unidentified microorganisms were resistant only to ciprofloxacin. Overall susceptibility of tested microorganisms to ciprofloxacin, tetracycline and clindamycin were 85%, 77% and 89% respectively. The results indicated no significant differences in the percentage of cultivable periodontopathic bacteria between the two membranes, and also the microorganisms resistant to tetracycline after systemic administration of doxycycline turned out to be susceptible to either ciprofloxacin or clindamycin.

  • The meaning of obturating root canal is to substitute an inert filling materials in the prepared canal space in order to eliminate all avenues of leakage from the oral cavity or periradicular tissue into root canal system. Inadequate obturation induce the infiltration of periapical tissue fluids, which provide materials for growth of microorganisms or localization of bacteria, into dead space of loosely filled canal. Most parts of endodontic failure is attributed to inadequate obturation of root canal system.(omitted)

Subcutaneous Caseous Necrotic Mass in a Pet Iguana (Iguana iguana) (애완 이구아나의 피하에 발생한 건락괴사성 종류)

  • A subcutaneous mass foud in a nine-year-old male Iguana (Iguana iguana) was pathologically and microbiologically examined. Grossly, this mass, measuring $2 cm{\times}1.5 cm{\times}1 cm$ in size, contained white to yellow caseous-crumbly material which was surrounded by the thin connective tissue. Histopathologically, a mass consisted of central caseous necrotic debri in which heterophils and macrophages were scattered, and peripheral fibrous connective tissue in which scattered heterophils, macrophages and lymphoid cell aggregates were present. This mass composed of central necrotic core and peripheal connective tissue did not have any detectable microorganisms including acid-fast bacteria and fungi. Histopathologically, severe atrophy of seminiferous tubules in both testicles was observed. In addition, peripheri of ductus deferens was thickened by fibrous connective tissue. This is the first report of subcutaneous caseous necrotic mass of an Iguana in Korea. In addition, the bilateral severe atrophy of seminiferous tubules may be due to fibrosis of ductus deferens.

Study on the Discoloration Identified from the Column of Wooden house, Hyunchungsa(Shrine) - Focused on Influence of Microorganisms and Correlation with Strength - (현충사 옛집의 기둥 하부 변색에 관한 연구 - 미생물에 의한 영향 및 강도와의 상관관계를 중심으로 -)

  • In general, it is thought discoloration on wood is frequently found in decorative wood products. So this study was conducted focusing on white rot found lower parts of columns and baseboards of a wooden house, Hyunchungsa (shrine) to know whether microorganisms have any influence on discoloration or there is correlation with strength by investigating resistograph, occurrence of microorganisms and microscopy for analysis(SEM, tissue analysis etc.). The results obtained were as follows: (1) The result of measurement of resistograph showed there are little correlation between discoloration and strength though there was a spot indicating low resistance. (2) The moisture content of discolored part was relatively higher than that of normal parts, but occurrence of microorganisms was less in discolored parts while more kinds of microorganisms were identified in normal parts with high CFU(Colony Forming Unit). (3) The result of SEM (with a magnification of 500 times) on the surface of discolored parts, it was found out there are many kinds of particles in different sizes on the surface and those were composed of elements such as C, O, Si, Ca, and a small amount of Na and Cl (weight %) were detected in part. (4) The result of tissue analysis showed discoloration occurs limitedly to the outer surface of column. As these results, it is concluded that discoloration has nothing to do with strength, damage by microorganisms and salt.

Isolation of Antibiotic-producing Microorganisms Antagonistic to Soilborne Pathogenic Fungi of Bentgrass and Their Antifungal Activity (잔디 토양전염성병원진균에 대한 길항미생물의 분리 및 길항효과)

  • Recently, the importance of management and cultivation of grasses has been increased in Korea. Among these cultural practices, the appropriate control of diseases is considered more important than other cultivation techniques such as fertilization and irrigation. The damages of brown patch and large patch caused by Rhizoctonia spp. and Pythium blight caused by Pythium spp. are serious in the major cultivation area of turfgrass in Korea. Since these diseases are difficult to control by agrochemicals, the damages are very serious if these are occured. The periodic spray of agrochemicals, to protect and control these diseases could make some problems of toxicity and environmental pollution as well as rising of non-target diseases. Therefore, the biological methods to control diseases have been required to decrease problems resulted from overuse of agrochemicals, to conserve natural ecosystem, and to control effectively diseases of grasses in the long period. The number of studies about biological control using antagonistic microorganisms have been increased for last half century. However, the application of biological control method has been very limited. In this study, thirteen isolates of R. cerealis, 8 isolates of R. solani and 3 isolates of Phthyn spp. have been isolated from diseased turfgrass in golf course and grass-culture area that have patch and wilting symptoms of zoysia grass and creeping bentgrass. Isolation frequency of R. cerealis and R. solani was high in especially zoysiagrass, while Pythym spp. was isolated from bent grass at low frequency but showed high pathogenicity. Totally, 205 isolates of soil microorganisms were isolated in this study as primary antagonistic microorganism by Herr's triple agar layer plate and dual culture method using rhizosphere of grasses, soil of crop field as the source of antagonistic microorganisms. Among the 205 isolates, 23 isolates were actinomycetes and 182 isolates were bacteria. All of the actinomycetes were isolated by Herr's method. Antagonistic effect of primary isolated microorganisms was tested for in vitro mycelial growth inhibition against pathogenic fungi isolated from grasses and for inhibition of disease occurrence in 24 well tissue culture plate and pot experiment. Then, four isolated of bacteria which are BG23, BG74, BG136 and BG171 were selected as antagonistic microorganisms against soil-born pathogenic fungi of bentgrass.

The Microbiome-Immune Axis Therapeutic Effects in Cancer Treatments

  • During the last decades, research and therapeutic methods in cancer treatment have been evolving. As the results, nowadays, cancer patients are receiving several types of treatments, ranging from chemotherapy and radiation therapy to surgery and immunotherapy. In fact, most cancer patients take a combination of current anti-cancer therapies to improve the efficacy of treatment. However, current strategies still cause some side effects to patients, such as pain and depression. Therefore, there is the need to discover better ways to eradicate cancer whilst minimizing side effects. Recently, immunotherapy, particularly immune checkpoint blockade, is rising as an effective anti-cancer treatment. Unlike chemotherapy or radiation therapy, immunotherapy has few side effects and a higher tumor cell removal efficacy depend on cellular immunological mechanisms. Moreover, recent studies suggest that tissue immune responses are regulated by their microbiome composition. Each tissue has their specific microenvironment, which makes their microbiome composition different, particularly in the context of different types of cancer, such as breast, colorectal, kidney, lung, and skin. Herein, we review the current understanding of the relationship of immune responses and tissue microbiome in cancer in both animal and human studies. Moreover, we discuss the cancer-microbiome-immune axis in the context of cancer development and treatment. Finally, we speculate on strategies to control tissue microbiome alterations that may synergistically affect the immune system and impact cancer treatment outcomes.

Effect of Polysaccharide from Schizophyllum commune on Burn and Wound Healing (치마버섯 유래 다당체의 화상 및 상처 치유효과)

  • Polysaccharides extracted from mushroom have been most commonly used to enhance the immune system. Also polysaccharides maintaining the moisture extent on epidermal tissue have an effect on the removal of necrotic tissue and the restoration of epidermal tissue through enhancing the immune system at skin layers. In this work, polysaccharides were from Schizophyllum commune studied about the burn and wound healing activity in the epidermal tissue on rats through in vivo experiment and hematological values. And antibacterial activities were examined using pathogenic microorganisms causing the secondary inflammation.

Analysis of fungal hyphae, distribution and motility of bacteria in oral cavity according to halitosis (구취에 따른 구강 내 형태별 세균의 분포 및 운동성, 진균 균사 분석)

  • Halitosis is primarily caused by bacterial decay. The bacteria, which originate from biofilms such as dental plaque, show abnormal proliferation due to dental caries, periodontal diseases, soft tissue infections, and tongue diseases. Most studies on halitosis have exclusively focused on gram-negative bacteria in the oral cavity rather than on general oral microorganisms including oral fungi. This study analyzed oral fungal hyphae, as well as distribution and motility of oral microorganisms, and provided basic data on the control of halitosis. Our results revealed that the greater is the number of cocci bacteria, the higher is the halitosis value, or bad breath value (BBV), suggesting that cocci have a strongly positive correlation with halitosis (r=0.379, p=0.030). Moreover, there was no significant difference in the morphology or distribution of motile bacteria and motility score, with respect to BBV. Lastly, we investigated the relationship between halitosis and oral fungal hyphae. We found that a higher BBV corresponded with a greater number of fungal hyphae and that patients with fungal hyphae scored a higher BBV. However, this result was not statistically significant. In conclusion, this study provided the preliminary data on oral microorganisms and halitosis, but further studies are needed to analyze the relationship between oral microorganisms and halitosis.
