The aim of this study was to investigate the effects of induced diabetes by streptozotocin (STZ) administration on gene expression of proinflammatory cytokines in adipose tissue of C57/BL6 mice fed either a normal diet (ND) or a high-fat diet (HFD). Four diabetic mice groups (16- or 26-week-old mice fed either ND or HFD) and four control groups of age and diet matched non-diabetic mice were used. By real-time PCR, gene expression levels of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and monocyte chemoattractant protein-1 (MCP-1) were examined in adipose tissue. The results demonstrated that gene expression of TNF-${\alpha}$ was significantly or marginally increased in STZ induced diabetic mice groups compared with non-diabetic groups. On the other hand, MCP-1 gene expression tended to be decreased in diabetic mice compared with non-diabetic controls. Especially, MCP-1 expression level in 16w diabetic mice on HFD was about 26% of that in age and diet matched non-diabetic controls (p<0.001). In addition, MCP-1 gene expression in adipose tissue was correlated with plasma insulin levels (p=0.0002). These results suggest that gene expression of proinflammatory cytokines in adipose tissue is differentially regulated in mouse models of diabetes. The basic data in this study will be useful for elucidating basic mechanisms of inflammatory state and increased expression of proinflammatory cytokines in adipose tissue in obesity, insulin resistance, and diabetes.
Tomato phenylalanine ammonia-lyase 5 (tPAL5) was identified that alternate initiation sites were utilized differentially in response to environmental stimuli (Lee et al, 1992b). In this study, we tried to look into tissue -or cell- specific expression pattern of tPAL5 gene by fusing with ${\beta}-glucuronidase$ (GUS) gene in transgenic tobacco plants. In transgenic plants, root and stem extracts contained 8~12 fold higher levels of GUS activity than petiole or leaf tissue while the highest levels of induction was observed from leaf tissue by mechanical wounding (5~11 fold). In trans-sections of stems and petioles, GUS activity was restricted to phloem cells(outer region) of developing vascular bundle and mainly at apical tip region in the root tissues. The levels of GUS activity was drastically reduced (10~12 fold reduction) when the 5'-upstream region of tPAL5 gene (-1151bp from ATG codon) was deleted up to -665. The levels of GUS expression, however, raised up by 6~8 fold when deleted up to -455. Therefore, we conclude that there are positive cis-elements at the region -1151 to -1008 and at -455 to -195 while the negative cis-element is at -1008 to -455.
Background: The relationship between the degree of expression of matrix metalloproteinases or tissue inhibitor of metalloproteinases and venous reflux remains to be investigated. Materials and Methods: Primary varicose vein tissues were obtained from 23 patients, 18 females and 5 males, aged from 19 to 73. Cephalic or basilic veins were obtained for the control group from 10 patients who underwent vascular access for maintenance hemodialysis. Two operative techniques (high ligation with stripping or endovenous laser coagulation) were used. The expression of matrix metalloproteinase-2 and 13 and tissue inhibitor of metalloproteinase-4 in the varicose vein group and control group was assessed semi-quantitatively by immunohistochemical slides stained with primary antibodies. Results: Twenty (87%) of the varicose vein group patients had greater or lesser saphenous vein diseases with reflux. The focal weak (+) stain for matrix metalloproteinases-2, and 13, and tissue inhibitor of matrix metalloproteinase-4 was dominant in the varicose vein group; the focal or diffuse strong stain (++ or +++) was prevalent in the control group. The differences were statistically significant (p<0.01). The degree of reflux and the duration of symptoms were not significantly related to the expression of MMP-13 (p=0.317 and p=0.654, respectively). Conclusion: Further study should be performed to investigate the relationship between the clinical characteristics related to venous hypertension or reflux and expression of MMPs and TIMP in varicose veins.
Transglutaminase2 (TGM2) is a multi-functional calcium dependent enzyme that affects angiogenesis, apoptosis, differentiation, attachment, and changes in the extracellular matrix. However, its function in periodontal tissue has not yet been studied. The aim of this study was to investigate the association of the TGM2 expression and the modulation of inflammatory mediators in inflamed periodontal ligament (PDL) cells induced by pro-inflammatory cytokines such as Interleukin-$1{\beta}$ and the Tumor necrosis $factor-{\alpha}$. The expression of TGM2 was increased in the inflamed periodontal tissue and PDL cells. Over-expressed TGM2 in the PDL cells increased expression of MMP1, MMP3, IL-6, CXCL8, and PTGS2. Conversely, inhibition of TGM2 activity using LDN27219, a TGM2 inhibitor, resulted in decreased expression of MMP1, MMP3, IL-6, and CXCL8. The mRNA expression was confirmed by RT-PCR and quantified by qRT-PCR. Protein levels were also confirmed by immunofluoroscence staining. These results suggest that TGM2 plays an important role in the regulation of inflammatory mediators which exacerbate tissue damage in inflamed periodontal tissue.
The effect of SV40 and murine cytomegalovirus (MCMV) enhancers on the general and tissue-specific gene expression was investigated. Recombinant plasmids containing these transcriptional engancers downstream of a structural gene for chloramphenicol acetyl transferase (CAT) were constructed. The transient expression of CAT gene from these plasmids was measured in monkey (CV1PD) and HeLa cells. Both SV40 and MCMV engancers activated the expression of CAT gene by more than 20 and 150 folds, respectively, compared with engancerless plasmids. When the SV40 promoter, upstream of CAT gene, was replaced with 2.2 kbp of promoter regulatory region of bovine growth hormone (bGH) gene, there was no expression of CAT even in the presence of enhancers, reflecting the tissue-specific expression of bGH genes. However, when the bGH regulatory region was shortened to 230 bp, the expression level increased dramatically in the presence of SV40 enhancers. In contrast, the expression from the shortened promoter was only marginally activated by the stronger MCMV enhancer.
Human Tissue factor is an essential enzyme activator that forms a catalytic complex with factor VII/ VIIa, and catalyzes both the extrinsic and intrinsic blood coagulation cascades. The extracellular domain of human tissue factor is responsible for association with the biological partner. The efficient procedures for preparing biologically active human tissue factor are essential for the preclinical and clinical studies with coaguligands. An expression vector in Escherichia coli has been constructed to direct the production of extracellular human tissue factor without a fusion protein or a $His_6$ at the N-terminus. The recombinant human tissue factor was expressed in large amounts as a non-native state in E. coli. The recombinant protein was simply renatured during the DEAE-sephacel chromatographic purification procedure. Our expression and purification system does not require a protease treatment or an additional chromatographic step to remove a fusion contaminant, which provides a very useful alternative to conventional expression systems for the production of human tissue factor.

This study investigated whether swimming exercises improves obesity through regulation of angiogenesis in white adipose tissue. Female mice with high-fat diets were divided into sham-operated group (Sham), ovariectomized group (OVX), and swim-trained ovariectomized group (OVX + Swim). Compared to the Sham, OVX increased body weight, adipose tissue mass and size of adipocyte. However, these factors (: such as body weight, adipose tissue mass and size of adipocyte) of OVX + Swim decreased compared with OVX. Compared with the Sham, OVX increased the mRNA expression of angiogenic activator and MMPs and decreased the mRNA expression of angiogenic inhibitors in white adipose tissue. But OVX + Swim decreased the mRNA expression of angiogenic activator and MMPs and increased the mRNA expression of angiogenic inhibitors in white adipose tissue, compared with the OVX. Theses results suggested that swimming exercises the angiogenesis in white adipose tissue, resulting to improve obesity in high-fat diet-fed female OVX mice.
Changes in the expression profiles of specific proteins leads to serious human diseases, including colitis. The proteomic changes related to colitis and the differential expression between tuberculous (TC) and ulcerative colitis (UC) in colon tissue from colitis patients has not been defined. We therefore performed a proteomic analysis of human TC and UC mucosal tissue. Total protein was obtained from the colon mucosal tissue of normal, TC, and UC patients, and resolved by 2-dimensional electrophoresis (2-DE). The results were analyzed with PDQuest using silver staining. We used matrix-assisted laser desorption ionization time-of-flight/time-of-flight spectrometry (MALDI TOF/TOF) to identify proteins differentially expressed in TC and UC. Of the over 1,000 proteins isolated, three in TC tissue and two in UC tissue displayed altered expression when compared to normal tissue. Moreover, two proteins were differentially expressed in a comparative analysis between TC and UC. These were identified as mutant ${\beta}$-actin, ${\alpha}$-enolase and Charcot-Leyden crystal protein. In particular, the expression of ${\alpha}$-enolase was significantly greater in TC compared with normal tissue, but decreased in comparison to UC, implying that ${\alpha}$-enolase may represent a biomarker for differential diagnosis of TC and UC. This study therefore provides a valuable resource for the molecular and diagnostic analysis of human colitis.

The Bombyx mori Microarray Database (BmMDB; http://silkworm.swu.edu.cn/microarray) provides information for tissue-specific gene expression by using the whole-genome oligonucleotide microarray in the silkworm. We analyzed the tissue-specific expression patterns in the silk gland, fat body, and midgut five days of fifth instar larvae during the development of B. mori. To verify the tissue-specific expression, analysis was conducted using quantitative Real-time RT-PCR and the highly expressed endogenous Actin RNA as an intrinsic reference. Finally, we confirmed five genes, (sw15872, sw00692, sw20990, sw05300,and sw2250), out of 18 candidates expressed in two different tissues, which was consistent with the data published by Dr. Xiang's group, thereby supporting the BmMDB. Further studies for promoter regions of candidate genes can be applied in creating transgenic silkworms as biomedical insects for use in producing biomaterials, and to serve as well-characterized models for understanding the mechanism for the genetic regulation of tissue-specific development.
The endometrium undergoes a cyclic growth and tissue remodeling as changes of epithelial cells, and plasminogen activators (PAs) are related to endometrium tissue remodeling. This study was to evulate expression of urokinase-type plasminogen activator (uPA) and tissue-type plasminogen activator (tPA) in porcine uterine epithelial cells. In results, the uPA and tPA were expressed in uterine tissue, epithelium and secretory glands in porcine endometrial cell. In addition, the uPA and tPA were expressed in cultured epithelial cells, and it were mainly expressed in cytoplasm. In porcine uterine tissue and epithelial cells, uPA activity was higher than activity in tPA. In PAs mRNA expression levels, uPA mRNA level was significantly higher than tPA mRNA level (P<0.05). The fluorescence intensity of uPA protein was also higher than fluorescence intensity of tPA protein, and uPA protein expression was significantly higher than in tPA protein expression (P<0.05). Therefore, we suggest that a physiological function in porcine uterine epithelial cells should be more influenced by uPA than in tPA during pre-ovulatory phase.
