Zinc (Zn) is an essential element in biological process, however inadequate Zn status in general population have been recognized. To update the knowledge for Zn-cadmium (Cd) interaction, we studied the intestinal uptake and transport, and the expression of metal transporter proteins (divalent metal transporter 1, DMT1 ; metal transporter protein 1, MTP1 ; zinc transporter 1, ZnTl ; metallothionein 1 , MT1) in duodenum after Cd exposure using Zn deficient animal model. Rats were led Zn deficient (ZnD, 0.5-1.0 mgZn/kg) or Zn supplemented (ZnS, 50mg Zn/kg) diet for 4 weeks, and followed single administration of $^{109}$ CdCl$_2$orally. The body Zn flatus and tissue Cd concentration were determined at 24 hrs after Cd administration. Total body burden of Cd and Cd absorption index (AI, %) were estimated based on the tissue Cd analyzed. DMT1, MTP1, ZnTl and MT1 mRNA were analyzed by using RT-PCR method. Feeding of Zn deficient diet for 4 weeks produced a reduced body weight gain and a depletion of body Zn. Tissue Cd concentration, body burden of Cd and Cd absorption index were higher in the ZnD diet fed rats than the ZnS diet red rats. Especially, Cd concentration in the small intestine (duodenum, jejunum and ileum) and the colon of FeD diet fed rats were higher markedly than in the FeS diet group. The expression levels of DMT1, MTP1 and ZnT1 mRNA in FeD diet fed rats were similar to the FeS diet. The level of MT1 mRNA expression was significantly lower in the FeD than the FeS diet fed rats. Taken together, theses results indicate that Zn deficiency in diet induce an increased intestinal absorption and tissue retention of Cd, and down -regulate the MT1 expression in the intestine which might be play a part of role in Cd absorption and transport in mammalian. These findings suggest that deficiency of essential metal could be enhanced the toxicity of toxic, non-esstial metals through the metal-metal interaction.
Thirty five male Sprague-Dawley rats were treated with cadmium chloride solution ranging from 0.2 to 3.2mg $CdCl_2/kg$ by intravenous single injection. At 48 hours after administration of cadmium, total cadmium, MT bound cadmium and histopathologic finding in liver, kidney, lung, heart, testis, metallothionein in liver, kidney and total cadmium in bleed were examined. Tissue cadmium concentration was highest in liver, followed by in kidney, heart, lung and testis. Cadmium bound to rnetallothionein (MT-Cd) and ratio of MT-Cd to total cadmium were increased in liver and kidney dependently of cadmium exposure dose, but not significantly changed in other organs. On histopathologic finding, the most susceptible organ was heart in considering cadmium exposed dose, but testis in considering cadmium concentration. Blood cadmium concentration was increased with dose-dependent pattern, and significantly correlated with tissue cadmium concentration, so that we may estimate tissue cadmium concentration by measurement of blood cadmium concentration. Metallothionein in liver and kidney was increased with dose-dependent pattern, higher in liver than in kidney, and was significantly correlated with tissue cadmium concentration. However, metallothionein induction efficiency of tissue cadmium(${\mu}g\;MT/{\mu}g\;Cd$) was eater in liver than in kidney, and reverse to tissue concentration or exposed dose of cadmium.
This study was performed to investigate the effects of Korean native plant diets on lipid and cadmium (Cd) metabolisms and also antioxidative enzyme activities in rats. Seventy male Sprague-Dawley rats weighing 210.3$\pm$2.7g were blocked into ten groups according to body weight. Rats were raised for four weeks with diets containing 0 or 0.04%(w/w) cadmium chloride and 5%(w/w) plant powder-Ssook(Artemisia princeps var. orientalis HARA), Chamchwi(Aster scaber THUNB), Gomchwi(Lingularia fischeri TUBCZ). or Solbeerum (Portulaca oleracea LINNE). Food intake, weight gain, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significantly lower in Cd-exposed groups, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significanlty lower in Cd-exposed groups. Plasma total lipid level, liver total lipid, cholesterol and triglycerde concentrations, and fecal total lipid, cholesterol and triglyceride excretions were decreased by Cd administration. Total lipid and triglyceride concentration in plasma and liver were lower in Chamchwi groups together with increased fecal total lipid and triglyceride excretions. Cholesterol concentration in plasma and liver were lower in Gomchwi groups with increased fecal cholesterol excretions. Activities of red-blood-cell superoxide dismutase(SOD), and liver catalase, SOD and glutathione peroxidase (GSH-px) were decreased significantly by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentrations of plasma and liver were increased by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentration of plasma and liver were increased by Cd administration. Administration of plant diets decreased plasma and liver catalase, SOD and GSH-px activities in Cd-free diet groups and increased them in Cd-administered groups. Plasma and liver TBARS concentrations were decreased in animals fed plant diets, with Chamchwi showing the most effective antioxidative capacity. The concentration of Cd accumulated in blood and tissue decreased in Cd-exposed groups fed plant diets. Liver Cd concentration were lower in the Chamchwi and Gomchwi groups, and kidney Cd levels were lower in the Ssook and Chamchwi groups. Among them, Chamchwi was the most effective in Cd detoxification. In conclusion, Chamchwi and Gomchwi were effective in lowering tissue lipid levels, with Chamchwi having the greatest antioxidative and detoxifying effects.
Effects of Zn, P and Fe on Cd uptake and accumulations by tomato (Lycopersicum esculentum Mill) and also their interactions on the uptake of Zn, Fe, Mn, P and Cd were investigated using batch type solution culture technique. Experiment 1 was a factorial scheme with 3 levels of Zn (0, 0.5, 2.5 ppm) and 3 levels of Cd (0, 0.2, 1.0 ppm). At 1.0 ppm Cd, significant yield reduction of dry matter and visual toxicity symptoms (yellowing and necrosis) of Cd was observed for all zinc levels. At this Cd level, increasing Zn treatment from 0 to 2.5 ppm increased Cd concentration from 199 to 235 ppm in leaves and from 124 to 145 ppm in stems. Similarly, Cd treatment did not suppress Zn uptake in leaves, and rather significantly increased in stems. Fe concentrations in leaves and stems were significantly reduced due to Cd treatment while Mn were increased by both Zn and Cd treatment. The results of experiment 2 with 3 levels of P (0.5, 2.0, 4.0m Mol) and 3 levels of Cd (0, 1.0, 2.0 ppm) in a factorial scheme also showed a growth reduction and visual toxic symptons from 1.0 ppm Cd level. Increasing P treatment tend to increase Cd concentrations in leaves and stems although it was not statistically significant. Increasing P concentration due to Cd treatment could be the 'concentration' effect as a result of reduced growth, while there was significant decrease in Fe concentration due to Cd treatment in spite of possible 'concentration' effect. Mn concentration was increased at 1.0 ppm Cd level and then dropped at 2.0 ppm Cd level. Zu concentration in leaves and stems showed significant increase as Cd treatment increased as observed in experiment 1. Experiment 3 had 3 levels of Fe (0.5, 1.0, 2.0 ppm) and 3 levels of Cd (0, 0.8, 1.6 ppm) treatments in a factorial design. Significant growth reduction and visual toxic symptoms as observed in experiment 1 and 2 were also observed from 0.8 ppm Cd level. Increasing Fe treatment obviously alleviated toxic symptoms, improved growth and significantly increased dry matter yield. At 0.8 ppm Cd treatment level, increasing Fe treatment from 0.5 to 2.0 ppm significantly decreased Cd concentration from 141 to 92 ppm in leaves and from 101 to 46 ppm in stems. At 1.6 ppm Cd treatment level the decrease was from 224 to 167 ppm in leaves and from 124 to 109 ppm in stems. As in the case of experiment 1 and 2, Fe concentration in leaves and stems were reduced as Cd treatment increased to 1.6 ppm at 0.5 and 1. 0 Fe treatment levels, whereas at 2.0 ppm Fe level, Cd treatment increased Fe concentration in leaves and stems showing significant interactions of Fe and Cd on Fe uptake. Cd effect on Zn and Mn showed similar results to experiment 1 and 2 and Fe treatments reduced Zn and Mn concentrations in plant tissue. The results of 3 experiments show that P and Zn did not manifest suppressive effect on Cd uptake, Fe significantly demonstrated it. Fe also alleviated Cd toxicity symptoms significantly in terms of visual symptoms and dry matter yield. Visual toxicity symptoms were definitely related to Fe status in plant tissue as well as possible physiological effect of Cd itself, and the results suggest that Fe requirement for normal growth increase as Cd element is present in plant tissue. Zn accumulated more in stems than in leaves whereas Cd, Fe and Mn showed the opposite trend in all experiments.

To study the effects between Cd inhalation toxicity and methanol extract of Radix Achyranthis Bidentatae, 4 rat groups were exposed to Cd aerosol by whole-body inhalation exposure for 6 hours/day, 5 days/week, and 4 weeks. Cd concentration in air was 0.98㎎/㎥ and mass median diameter(MMD) was 1.78㎛. 3 different dose intraperitoneal injections of methanol extract of Radix Achyranthis Bidentatae to 3 inhalation exposure groups applied for 4 weeks and the results were as follows: The highest body weight gain for 4 weeks and food intake per day were from inhalation exposure group Ⅲ(p<0.05). The highest lung weight was from inhalation exposure group Ⅲ and the highest liver and kidney weight were from inhalation exposure group Ⅱ(p<0.05). The lowest Cd content in lung was 22.77㎍/g from inhalation exposure group Ⅲ(p<0.05). The highest Cd concentration in blood was 11.71㎍/㎗ from inhalation exposure group Ⅰ(p<0.05). Cd concentrations of 14.87㎍/g in liver and 17.91㎍/g in kidney were the highest from inhalation exposure group Ⅰ(p<0.05). The lowest Cd concentration in liver and kidney were 5.71㎍/g and 3.17㎍/g from the control(p<0.05). For weekly Cd concentration in urine, the highest value was 0.48㎍/㎖ from inhalation exposure group Ⅲ of the 3rd week and inhalation exposure group Ⅰ, Ⅱ of the 4th week. For weekly Cd concentration in feces, the highest value was 0.32㎍/g from inhalation exposure group Ⅰ, Ⅱ, Ⅲ. The highest metallothionein concentration in lung was 89.02㎍/g from inhalation exposure group Ⅲ(p<0.05). The highest metallothionein concentrations in liver and kidney were 265.47㎍/g and 214.21㎍/g from inhalation exposure group Ⅲ, respectively(p<0.05). The highest Hct, Hb, and WBC values were from inhalation exposure group Ⅱ and the highest RBC value was from inhalation exposure group Ⅲ(p<0.05). Mostly damaged part in liver tissue was hepatic lobule and the degrees of damage were lessened by the intraperitoneal injection of methanol extract of Radix Achyranthis Bidentatae. Proximal, distal convoluted tubules and glomerulus in kidney tissue were mostly damaged part. Degeneration and swelling were partially observed but the degrees of kidney tissue damage were lessened more or less by the intraperitoneal injection of methanol extract of Radix Achyranthis Bidentatae.
Using Megabalanus rosa a sessile filterfeeder, its scope for growth (SFG) by analysis of energy budget were examined and free amino acids composition of whole body tissue were analyzed under the exposure to different concentrations of Hg Cu and Cd. The $96\;hr-LC_{50}$ of the barnacle after 96 hr exposure to Hg, Cu, and Cd were 0.220, 0.269 and $1.380\;mgL^{-1}$, respectively. Hg and Cu showed stronger toxicity than Cd, while Hg and Cu had similar influence on the survival of the barnacle. SFG of the barnacles exposed to sublethal concentrations of mercury was 18.936 $Jgdrywt.^{-1}hr^{-1}$ in control group and as increase of mercury concentration the SFG remarkably reduced to 0.041 $Jgdrywt.^{-1}hr^{-1}\;at\;0.1\;mgL^{-1}$ concentration of Hg. In the case of Cu, the SFG was 29.841 $Jgdrywt.^{-1}hr^{-1}$ in control group and as increase of concentration, the SFG remarkably reduced to -8.304 $Jgdrywt.^{-1}hr^{-1}\;at\;0.1\;mgL^{-1}$ concentration. In Cd the SFG was 15.852 $Jgdrywt.^{-1}hr^{-1}$ in control group, and as increasing concentration, the SFG remarkably reduced to -19.490 $Jgdrywt.^{-1}hr^{-1}\;at\;0.1\;mgL^{-1}.$ Content of free amino acid (FAA) of whole body tissue of the barnacle was 45084 $mgkg^{-1}$ in control group, but it was reduced remarkably to 28,130, 37,500 and 37,106 $mgkg^{-1}$ at 0.1 $mgL^{-1}$ concentration of Hg and Cu, and 0.4 $mgL^{-1}$of Cd, respectively. Sum of threonine + serine was 1,334 $mgkg^{-1}$ if control but reduced remarkably to 1,223, 849 and 888 $mgkg^{-1}$ at 0.1 $mgL^{-1}$ of Hg and Cu, and 0.4 $mgL^{-1}$ of Cd, respectively.
Cadmium (Cd) is an environmental pollutant and categorized as a human carcinogen, which has a tendency to accumulate in the human body. The level of Cd in renal cortex and liver are good indicators as an index of Cd exposure in general population. Geometric mean concentration of Cd is 27.4 and 3.1 /g wet weight in renal cortex and liver, respectively, in Korean. Cd is toxic to a number of tissues, notably the liver, kidney, testis, lung, lymphoid tissue and lung. (omitted)

A sediment exposure experiment was conducted to investigate the influence of acid volatile sulfides (AVS) and simultaneously extracted metals (SEM) in sediments on the bioavailability and toxicity of Cd, Ni and Zn to a marine polychaetes Neanthes arenaceodentata. The test animals were exposed to contaminated sediments spiked by metal mixtures of Cd, Ni, Zn (0.5~15 $\mu$mol/g of total SEM) in low (~1 $\mu$mol/g), medium (~5 $\mu$mol/g) and high AVS series (~10 $\mu$mol/g) to determine bioaccumulation, mortality and individual growth rate in each treatment after 20 days. Cd and Zn bioaccumulation in test animals increased with increasing of overlying water (OW) concentration controlled by AVS. In contrast, Ni bioaccumulation increased with increase of SEM concentration. Mortalities and growth inhibitions of N. arenaceodentata observed in only treatments with [SEM-AVS]>0, due to a high level of OW-Zn. With regard to the mortality, the 20-d LC5O value fur OW-Zn was 9.3(8.0$\pm$11.0) $\mu$M. The LOEC (Lowest Observed Effect Concentration) for Tissue-Zn was 7.8 $\mu$mol/g and the NOEC (No Observed Effect Concentration) was 6.2 $\mu$mol/g. Regarding the inhibition of individual growth rate, the LOEC fer Tissue-Zn was 5.9 $\mu$mol/g, and NOEC was 5.1 $\mu$mol/g. In this study, the toxicity of dissolved metals, especially for Zn, was overemphasized due to the reduced distribution coefficients (K$\_$d/s) of metals in the experimental sediments.
This study was initiated to investigate the effects of organic germanium on cadmium and mercury intoxication. The effect was determined by the metallothionein induction in liver and kidney. Male rats (Sprague-Dawley) were treated with CdCI$_{2}$ (2mg/kg), HgCI$_{2}$ (1 mg/kg) and organic germanium (GE-132) (100 mg/kg) in single and in combination via intraperitoneal injection or intragastric administration every other days for 17 days. Experimental animals were sacrificed after 7, 12 and 17 days treatment. The serum transaminase activities (SGOT, SGPT), concentration of metal and metallothionein, metal-binding capacity of metallothionein in liver and kidney were determined and pathomorphological observations were undertaken. The combined treatment of GE-132 and CdCI$_{2}$ significantly decreased the increment of serum transaminase activities in rats treated with CdCI$_{2}$ only, but the combined treatment of GE-132 and HgCI$_{2}$ did not affect to activities of transaminases induced by mercury only. The concentration of metals (Cd and Hg) except Ge in the liver and kidney of rats increased with the time of treatment. Mercury concentration in kidney of rat treated with HgCI$_{2}$ only was significantly higher than the combined treatment of GE-132 and HgCI$_{2}$. The combined treatment of GE-132 and CdCI$_{2}$ significantly increased the concentration of metallothionein in liver compared to the CdCI$_{2}$ only, although the concentration of cadmium in liver were not significantly different between two groups. This indicates that GE-132 decreased toxicity of cadmium in liver by promoting metallothionein induction. There were no significant differences in metallothionein concentration in liver and kidney of rats between the combined treatment of GE-132 and HgCI$_{2}$ and HgCI$_{2}$ only. Metal-binding capacity of metallothionein varied with each time intervals in liver and kidney of metals treated rats except the liver of the combined treatment of GE-132 and CdCI$_{2}$. This finding explains the concentration of metallothionein in liver keeps abreast with the concention of metal. Furthermore, the combined treatment of GE-132and CdCl$_{2}$ revealed pathologically less changes in liver tissue than CdCl$_{2}$ only; the damages of liver cell, such as lobular necrosis and portal inflammation, were relieved and appeared more later. From the above results, organic germanium is considered to have some beneficial effect on the protection of liver from the cadmium intoxication.
Experiments were carried out to investigate the elimination of cadmium in tissues (gill, intestine, liver, kidney and muscle) of flounder, Paralichthys olivaceus, after sub-lethal Cd exposure (5, 50, 100 ${\mu}g/L$). During the depuration phase, Cd concentration in the gill decreased immediately following the end of the exposure period. The elimination rates at the end of depuration periods were $18\%$ for 50 ${\mu}g/L$ exposure and $70\%$ for 100 ${\mu}g/L$ exposure. Intestine showed fastest elimination rates of Cd at all concentration. At the end of the depuration period, the Cd concentration was similar to that in the control, Cd elimination in liver significantly decreased after 10 days of depuration period. After 20 days of depuration, the elimination rate was $66.20\%$ in the fish exposed to 50 ${\mu}g/L$ and $86.22\%$ in the fish exposed to 100${\mu}g/L$. The order of cadmium elimination in tissues were decreased intestine>liver${\geq}$gill>>kidney during depuration periods. In this study the gill, intestine and liver showed faster elimination rate of Cd in 50, 100 ${\mu}g/L$-Cd exposure concentration. After the end of the Cd exposure, the Cd concentration in the kidney slowly decreased or remained constant and the Cd concentration in the muscle slowly increased or remained constant.
