Physicochemical Characteristics of Fermented Phragmites communis Extract and Its Biological Activity (갈대 발효추출물의 이화학적 특성 및 생리활성 연구)

  • This study evaluates the tyrosinase, elastase inhibitory and antioxidant activities of isolated Lactobacillus rhamnosus fermented extracts of Phragmites communis Trinius. After culture for 4 days at $30^{\circ}C$ using 1% P. communis extract, the cell mass of L. rhamnosus reached $1.4{\times}10^{10}CFU/mL$. The number of cells on P. communis extract and MRS medium was similar. This results indicated that P. communis extract can be used as an economical medium for industrial lactic acid bacteria production. The fermented P. communis extract exhibited 4 fold higher tyrosinase inhibitory effect than non fermented P. communis extract. The non fermented P. communis extract has no inhibitory effect on elastase. However the fermented P. communis extract show high inhibitory effect on elastase ($IC_{50}$; $249{\mu}g/mL$). These results indicated that the fermented P. communis extract can potentially be used for developing new cosmetic or health food ingredients.

  • Rheological properties of Shinsunchalbyeo(Japonica) and $Hangangchalbyeo(J{\times}Indica)$ waxy-rice starches gelatinized with thermal or alkali solutions were investigated with rotational viscometer(Brabender Viscotron). The two starches showed Bingham pseudoplastics behavior in $4{\sim}8%$ thermal or alkali gelatinized starch solutions. The shear stress of Hangangchalbyeo starch solution gelatinized with thermal or alkali showed higher values than that of Shinsunchalbyeo starch and the difference of the two varieties gelatinized with alkali showed higher values than that gelatinized with thermal. Consistency index and yield stress values of Hangangchalbyeo starch showed higher than that of Shinsunchalbyeo starch in the two gelatinized methods and the difference of the two varieties gelatinized with alkali showed higher values. And it was higher that the deffendence of consistency index on starch concentration and initial starch concentration of yield stress in the alkali gelatinized samples than those of in the thermal gelatinized ones.

  • B. subtilis JM3 pretense from naturally fermented anchovy sauce was purified in $40{\sim}60%$ ammonium sulfate fraction. In order to accelerate the fermentation of anchovy sauce,2% and 4% of crude B. subtilis JM3 protease were added to 6 month-fermented anchovy sauces, respectively and then the various quality characteristics such as pH, lactic acid, amino-nitrogen, VBN, browning and hydrolysis degree, VBN, and sensory evaluation were analyzed at different storage times. pH was constant during storage time in all samples, whereas lactic acid contents of anchovy sauces hydrolyzed by 2% and 4% proteases were higher than that of control. The amino-nitrogen and volatile basic nitrogen contents of anchovy sauce with 2% and 4% proteases were twice higher than those of control. Anchovy sauces with 2% and 4% pretense increased the hydrolysis rate by 27% and 32%, respectively. Browning degree of anchovy sauce with 4% was higher than those of 2% and control. Anchovy sauce with 2% and 4% proteases was good in sensory evaluation of color, aroma, and taste attributes.

  • It is a recent observation that about 80 per cent of the body's immune system is localized in the gastrointestinal tract. This explains to a large extent why eating right is important for the modulation the immune response and prevention of disease. I addition it is increasingly recognized that the body has an important digestive system also in the lower gastrointestinal tract where numerous important substances are released by microbial enzymes and absorbed. Among these substances are short chain fatty acids, amino acids, various carbohydrates, polyamines, growth factors, coagulation factors, and many thousands of antioxidants, not only traditional vitamins but numerous flavonoids, carotenoids and similar plant- and vegetable produced antioxidants. Also consumption of health-promoting bacteria (probiotics) and vegetable fibres (prebiotics) from numerous sources are known to have strong health-promoting influence. It has been calculated that the intestine harbours about 300 000 genes, which is much more than the calculated about 60000 for the rest of the human body, indicating a till today totally unexpected metabolic activity in this part of the GI tract. There are seemingly several times more active enzymes in the intestine than in the rest of the body, ready to release hundred thousand or more of substances important for our health and well-being. In addition do the microbial cells produce signal molecules similar to cytokines but called bacteriokines and nitric oxide, with provide modulatory effects both on the mucosal cells, the mucosa-associated lymphoid system (MALT) and the rest of the immune system. Identification of various fermentation products, and often referred to as synbiotics, studies of their role in maintaining health and well-being should be a priority issue during the years to come.

Quantitative Analysis of Marker Substances in Solid Fermented Angelicae Gigantis Radix by HPLC (HPLC를 이용한 고체발효 당귀의 지표성분 분석)

  • The purpose of this study was investigation of quantitative analysis of marker substances in solid fermented Angelicae Gigantis Radix by High performance liquid chromatography(HPLC). HPLC was performed for determination of nodakenin and decursin in solid fermented Angelicae Gigantis Radix extract, the separation method was performed on C18 column ($250\;mm\;{\times}\;4.6\;mm$, $5\;{\mu}m$, RS tech) using gradient solvent mixtures of water-acetonitrile with photodiode array detector (330 nm). The flow rate was 1.0 ml/min. Retention time of nodakenin and decursin was about 11.47, 46.79 min and linearity of calibration was showed good result(r2=0.9999, 0.9999), respectively. Content of nodakenin was $0.76\;{\pm}\;0.02%$ in control, $0.31\;{\pm}\;0.00%$ in Angelicae Gigantis Radix extract fermented with Paecilomyces japonica(SDT)(p<0.01), $0.51\;{\pm}\;0.02%$ in Angelicae Gigantis Radix extract fermented with Ganoderma lucidum(SYT)(p<0.01), $0.82\;{\pm}\;0.03%$ in Angelicae Gigantis Radix extract fermented with honey(SST)(p<0.05) and $0.88\;{\pm}\;0.01%$ in Angelicae Gigantis Radix extract fermented with Nuruk(SNT)(p<0.01). Content of decursin was $4.50\;{\pm}\;0.08%$ in control, $2.90\;{\pm}\;0.05%$ in Angelicae Gigantis Radix extract fermented with Paecilomyces japonica(SDT)(p<0.01), $2.65\;{\pm}\;0.08%$ in Angelicae Gigantis Radix extract fermented with Ganoderma lucidum(SYT)(p<0.01), $4.46\;{\pm}\;0.11%$ in Angelicae Gigantis Radix extract fermented with honey(SST) and $4.73\;{\pm}\;0.04%$ in Angelicae Gigantis Radix extract fermented with Nuruk(SNT)(p<0.05), respectively.

  • This study aimed to provide consumer perception and evaluation of natural Cynanchi Atrium Fermented Original Cosmetics for Skin Whitening that can improve skin by using oriental medicinal ingredients. The product was applied three times a day for two weeks by a total of 60 users who agreed to participate in a clinical trial assessing the usability and skin sensitivity to extracts that had already completed component safety testing. This study subjects reported which images were most reminiscent of Korea. They preferred products that bore Korean images because they felt trust and recognized that the quality of the cosmetics was excellent. The responses also showed that many consumers were willing to buy a product, even though it was luxurious, elegant, and expensive. The cognitive evaluation of Original Cosmetics was high and showed that the quality was likely to be good. The usability and CMF(Color, Material, Finishing) evaluation of Original Cosmetics showed good product the product is long-lasting and tightened and moisturized skin naturally. They are think about that this product it has moisturizing effects and good application for use, so it is light and soft to use. And had a whitening effect. It was intended to assist in the development of products that meet consumers' desires and expectations for safe, healthy, and beautiful skin by developing a skin whitening Original Cosmetics prototype that can inhibit the production of melanin and tyrosinase activity safely and effectively.

Improvement of Ethanol Production by Electrochemical Redox Combination of Zymomonas mobilis and Saccharomyces cerevisiae

  • Zymomonas mobilis was immobilized in a modified graphite felt cathode with neutral red (NR-cathode) and Saccharomyces cerevisiae was cultivated on a platinum plate anode. An electrochemical redox reaction was induced by 3 volts of electric potential charged to the cathode and anode. The Z. mobilis produced 1.3-1.5 M of ethanol in the cathode compartment, whereas the S. cerevisiae produced 1.7-1.9 M in the anode compartment after 96 h. The ethanol produced by the Z. mobilis immobilized in the NR-cathode and S. cerevisiae cultivated on the platinum plate was 1.5-1.6 times higher than that produced under conventional conditions. The electrochemical oxidation potential inhibited Z. mobilis, but activated S. cerevisiae. The SDS-PAGE pattern of the total soluble proteins extracted from the Z. mobilis cultivated under the electrochemical oxidation conditions was gradually simplified in proportion to the potential intensity. Z. mobilis and S. cerevisiae were cultivated in the cathode and anode compartments, respectively, of an electrochemical redox combination system. The Z. mobilis culture cultivated in the cathode compartment for 24 h was continuously transferred to the S. cerevisiae culture in the anode compartment at a rate of 300 ml/day. Approx. 1.0-1.2 M of ethanol was produced by the Z. mobilis in the cathode compartment within 24 h, and an additional 0.8-0.9 M produced by the S. cerevisiae in the anode compartment within another 24 h. Thus, a total of 2.0-2.1 M of ethanol was produced by the electrochemical redox combination of Z. mobilis and S. cerevisiae within 48 h.

Antihypertensive effect of Korean Red Ginseng by enrichment of ginsenoside Rg3 and arginine-fructose

  • Background: Ginsenoside Rg3 and arginine-fructose (Arg-Fru) are known as the hypotensive compounds of Panax ginseng; however, their efficacy on antihypertension has not been reported yet to our best knowledge. Thus, hypotensive components-enriched fraction of red ginseng (HCEF-RG) was prepared from fine root concentrate (FR) and their antihypertensive effects were investigated in spontaneously hypertensive rats (SHR). Methods: Male SHRs were divided into six groups: control (Wistar Kyoto, SHR); FR 500; FR 1,000; HCEF-RG 500; and HCEF-RG 1,000; samples (mg/kg body weight) were orally administered every day for 8 wk. Blood pressure was monitored at 1 wk, 2 wk, 3 wk, 4 wk, 6 wk, and 8 wk by tail cuff method. At 8 wk after samples administration, mice were killed for the measurement of renin activity (RA), angiotensin-I converting enzyme inhibition, angiotensin II, and nitric oxide (NO) levels in plasma. Results: HCEF-RG with four-fold more Rg3 and 24-fold more Arg-Fru contents was successfully prepared from reacted mixtures of FR and persimmon vinegar (12 times against FR, v/v) at $80^{\circ}C$ for 18 h. Both FR 1,000 and HCEF-RG 1,000 showed lowered systolic blood pressure than SHR control group and HCEF-RG 1,000 group exhibited a significant decrease in diastolic blood pressure. RA was significantly lowered in all treated groups, while angiotensin II did not affect by FR and HCEF-RG treatment. However, angiotensin-I converting enzyme inhibition and NO in FR 1,000 and HCEF-RG 1,000 were significantly increased compared with SHR control group. Conclusion: HCEF-RG is more effective and useful for alleviating hypertension than FR, implying the health benefit of Rg3 and Arg-Fru.

Effect of Vital Wheat Gluten on the Quality Characteristics of the Dough Frozen after 1st Fermentation (활성글루텐이 1차발효 후 냉동한 생지의 품질특성에 미치는 영향)

  • Frozen dough made by sponge and dough method using sweet dough formula was quickly frozen at $-40^{\circ}C$ and stored for 8 weeks at $-20^{\circ}C$. Effects of vital wheat gluten on number of yeast cells, bread volume, specific loaf volume, hardness, and sensory properties of bread were investigated. Dough added with 4% vital wheat gluten showed higher yeast cell survival rate during freeze storage and larger specific loaf volume than other doughs. Hardness value increased with increasing amount of vital wheat gluten added, whereas, in frozen dough stored more than 4 weeks, dough added with 2% vital wheat gluten showed lower hardness value than others. Bread made with 4% vital wheat gluten showed highest sensory score.

Growth Characteristics of Lactic Acid Bacteria in Whey-Soy Milk Mixtures (유청(乳淸)과 두유(豆乳) 혼합액(混合液)에서의 유산균(乳酸菌) 생육특성(生育特性))

  • Growth characteristics of six lactic acid bacteria in whey-soy milk mixtures were investigated to obtain basic informations for processing cheese-like product by coprecipitation of whey and soy proteins. Streptococcus cremoris and Lactobacillus acidophilus produced more aicd than other lactic acid bacteria both in whey-soy milk mixture and in soy milk. Lactic acid fermentation was accelerated in whey-soy milk mixture than in soy milk with all the lactic aicd bacteria, and specially with S. lactis and S. cremoris in great extent. The number of viable cell of 1:1 mixed culture of S. lactis and S. cremoris in whey soy milk mixture was about 10 times than in soymilk. It was mainly the effect of lactose in the whey that increased the acid production by lactic aicd bacteria in whey-soy milk mixture although the degree of acceleration depended on the ability of microorganism to use carbohydrates. The optimum amount of lactose added to soy milk to accelerate the acid production was 0.8g/100ml soy milk.
