For the production of Korea traditional cereal wine makgeolli, a rice fermentation starter nuruk was Rhizopus oryaze CCS01 commercial nuruk. The carbohydrate content of rice nuruk was higher, the levels of moisture, ash, crude protein, and crude fat were lower. In particular, the saccharifying activity of rice nuruk was 2.2 times higher than commercial nuruk. pH, alcohol, and viable yeast cells of makgeolli made rice nuruk were higher than those of makgeolli made commercial nuruk. In contrast, the levels of acidity, $^{\circ}Brix$, viable lactic acid bacteria (LAB) cells, and browning of makgeolli made rice nuruk were lower than those of makgeolli made commercial nuruk, the alcohol content of glutinous rice (GUR) makgeolli made rice nuruk higher other samples. These results suggest that rice nuruk R. oryzae CCS01 make new type cereal makgeolli.

This study aimed to investigate the effects of temperature and retention time of the pressurized steam chamber on the ruminal fermentation characteristics and nutrient degradability of corn flakes in three Korean native Hanwoo cows and three Holstein cows implanted with a ruminal fistula. Corn kernels were categorized into 13 groups based on the chamber temperature (range, $100^{\circ}C-116^{\circ}C$) and retention time (range, 700-950 s). The pH value was lowest in T1 regardless of breed. Propionate concentration was the highest in T2 (p < 0.05). Total-volatile fatty acid (VFA) concentration was slightly but not significantly greater in T2 than in other conditions. Dry matter (p < 0.05), starch, and crude protein (p < 0.05) degradability were the highest in T1. At different incubation times and with different breeds, dry matter, starch, and crude protein degradability of corn flakes were the highest in T1. Thus, the present results indicate that the optimal temperature and retention time of the pressurized steam chamber should be $100^{\circ}C-105^{\circ}C$ and 700-720 s.

The objective of this study was to evaluate the effect of protein and energy on goats fed oil palm fronds (OPF) as roughages. Twenty-four male Saanen goats aged between 7 and 8 months and weighing $23.4{\pm}1.6kg$ were used in a $2{\times}3$ factorial design. Factors were three levels of urea (3%, 4% or 5%) and two levels of energy (low energy (LE) or high energy (HE)). On average, all parameters measured, including dry matter intake (DMI), nutrient digestibility, digestible nutrient intakes, ruminal ammonia-N ($NH_3$-N), ruminal total volatile fatty acid (total VFA) and individual VFA concentrations (mM/L), microbial N supply, P/E ratio and N retention were higher for HE compared to LE diets. Significant (p<0.05) interactions were found between levels of urea and energy for nonstructural carbohydrate (NSC) and energy (DE) digestibilities, ruminal $NH_3$-N and total VFA concentrations. HE diets had higher N absorption and retention than LE diets. Interactions between urea and energy for plasma urea nitrogen (PUN), heat production (HP), and urine and faeces N excretion were significantly lower (p<0.05) for the HE diets than those recorded for the LE diets. The results indicated that supplementation of energy enhanced utilization of urea and resulted in higher animal performance as a consequence of improved ruminal fermentation, microbial yield and N balance. However, the optimal level of urea supplementation remained at 3% in the HE diet.
Objective: As the climate changes, it influences ruminant's feed intake, nutrient digestibility, rumen methane production and emission. This experiment aimed to evaluate the effect of feeding Sweet grass (Pennisetum purpureum cv. Mahasarakham; SG) as a new source of good quality forage to improve feed utilization efficiency and to mitigate rumen methane production and emission. Methods: Four, growing crossbred of Holstein Friesian heifers, 14 months old, were arranged in a $4{\times}4$ Latin square design to receive four dietary treatments. Treatment 1 (T1) was rice straw (RS) fed on ad libitum with 1.0% body weight (BW) of concentrate (C) supplementation (RS/1.0C). Treatment 2 (T2) and treatment 3 (T3) were SG, fed on ad libitum with 1.0% and 0.5% BW of concentrate supplementation, respectively (SG/1.0C and SG/0.5C, respectively). Treatment 4 (T4) was total Sweet grass fed on ad libitum basis with non-concentrate supplementation (TSG). Results: The results revealed that roughage and total feed intake were increased with SG when compared to RS (p<0.01) while TSG was like RS/1.0C treatment. Digestibility of nutrients, nutrients intake, total volatile fatty acids (VFAs), rumen microorganisms were the highest and CH4 was the lowest in the heifers that received SG/1.0C (p<0.01). Total dry matter (DM) feed intake, digestibility and intake of nutrients, total VFAs, $NH_3-N$, bacterial and fungal population of animals receiving SG/0.5C were higher than those fed on RS/1.0C. Reducing of concentrate supplementation with SG as a roughage source increased $NH_3-N$, acetic acid, and fungal populations, but it decreased propionic acid and protozoal populations (p<0.05). However, ruminal pH and blood urea nitrogen were not affected by the dietary treatments (p>0.05). Conclusion: As the results, SG could be a good forage to improve rumen fermentation, decrease methane production and reduced the level of concentrate supplementation for growing ruminants in the tropics especially under global climate change.

The study was carried out to investigate the ratio of ethanol to n-propanol in blood and urine specimens, and developed a method for distinguishing ingested ethanol from artifactual ethanol in urine samples. In case of no urinary ethanol was detected, the ratio of ethanol to n-propanol concentration was about 12~20 times higher than those of blood. Therefore, it might be a good method to determine whether the detected ethanol is from drinking or from microbial fermentation. During the metabolism of ethanol, the levels of the metabolite of serotonin (5-hydroxytryptamine, 5-HT) and 5-hydroxyindole-3-acetic acid (5-HIAA) were decreased, while 5-hydroxytryptophol (5-HTOL) was increased. The levels of 5-HTOL/5-HIAA in urine samples of drinking suspects were greater than 1, in that of no drinking suspects were less than 1.

Leuconostoc mesenteroides is used as a starter to produce high-quality kimchi products. In this study, an efficient and economical cabbage juice medium (CJM) was developed by process optimization of cabbage extraction and pasteurization and by compositional supplementation of various lacking nutrients. The pasteurized cabbage juice was determined to be a good medium candidate to cultivate L. mesenteroides, showing maximal cell numbers ($9.85{\times}10^8CFU/ml$) after 24 h. Addition of sucrose and yeast extract with soy peptone resulted in increment of bacterial cell counts in CJM, showing the supplementing effect of the lacking nutrients. Furthermore, addition of shell powder gave a protective effect on bacterial cells by preventing pH decline and organic acid accumulation in CJM, resulting in a 2-fold increase of bacterial counts. The optimized composition of CJM was 70% cabbage juice diluted with water, 0.5% (w/v) sucrose, 1% (w/v) yeast extract, 1% (w/v) soy peptone, and 1.5% (w/v) ark shell powder. The CJM developed in this study was able to yield a comparable level of bacterial counts with MRS medium and reduced the cost by almost 10-fold.
This study was performed to examine the cultivation of Pleurotus florida that forms pinhead between 5 and $25^{\circ}C$, on rice straw substrates in Korea. The highest yield of P. florida sporophores was obtained from the fermented rice straw substrates added 5% rice bran to the rice straw by dry weight basis at the starting of fermentation. The pile turning of materials for uniform fermenting was made three times when the temperature of the pile reached $60^{\circ}C$ during out door composting. The $60^{\circ}C$ row steam was treated 6 hrs for pasterization of the substrates in the cultivating house. The quantity of spawn affected the mycelial growth and the yield of P. florida. The optimum amount of spawn was 10% to the rice straw by dry weight basis mixed with the substrates. P. florida produced normal sphorophores from September to middle of July of the following year.

Anaerobic reductive dehalogenation of perchloroethene (PCE) was studied with lactate as the electron donor in a continuously stirred tank reactor (CSTR) inoculated with a mixed culture previously shown to dehalogenate vinyl chloride (VC). cis-1,2- dichloroethene (cDCE) was the dominant intermediate at relatively long cell retention times (>56 days) and the electron acceptor to electron donor molar ratio (PCE:lactate) of 1:2. cDCE was transformed to VC completely at the PCE to lactate molar ratio of 1:4, and the final products of PCE dehalogenation were VC (80%) and ethene (20%). VC dehalogenation was inhibited by cDCE dehalogenation. Propionate produced from the fermentation of lactate might be used as electron donor for the dehalogenation. Batch experiments were performed to evaluate the effects of increased hydrogen, VC, and trichloroethene (TCE) on VC dehalogenation which is the rate-limiting step in PCE dehalogenation The addition of TCE increased the VC dehalogenaiton rate more than an increase in the $H_2$ concentration, which suggests that the introduction of TCE induces the production of an enzyme that can comtabolize VC.

Indigo (Polygonum tinctorium L.) is a typical blue dye which had been used from ancient times. This study was going to shade the complicated traditional methods extracting indigo dye by the fermentation and producing as adsorbate on calcium hydroxide, which says so called as the 'Indigo lime'. Accordingly we were going to make indigo through the hydrolysis of the hot water extractives of indigo leaves simply. During hot-water extraction, ${\beta}$-glucosidase which required hydrolysis of the linkage between indigo and glucose was not activated. To achieve this goal, indican was acid-hydrolyzed to glucose and indigo. The acetic acid, citric acid, hydrochloric acid, and sulfuric acid were used for the hydrolysis of hot water extractives. The hydrolysis conditions of extractives performed in water bath at $80^{\circ}C$ for 120 minutes and in an autoclave for 120 minutes. In the acid hydrolysis of extracted indican by hot water, the indican yields of acetic acid and hydrochloric acid hydrolysis were higher than sulfuric acid in water bath. Also, the indican yield of hydrochloric acid hydrolysis was better than sulfuric acid in autoclave. The hot water extracted indican was confirmed by HPLC analysis and its structure was confirmed by UV-Vis and FT-IR spectroscopy, compared with isolated indigo and commercial synthesized indigo. This improved extraction and hydrolysis methods can be replace the traditional indigo making method.
Pretreatment of cellulosic biomass is necessary before enzymatic saccharification and fermentation. Extrusion is a well established process in food industries and it can be used as a physicochemical treatment method for cellulosic biomass. Aqueous ammonia soaking treatment at mild temperatures ranging from 60 to $80^{\circ}C$ for longer reaction times has been used to preserve most of the cellulose and hemicellulose in the biomass. The objective of this study was to evaluate the effect of extrusion treatment on aqueous ammonia soaking method. Extrusion was performed with miscanthus sample conditioned to 2mm of particle size and 20% of moisture content at $200^{\circ}C$ of barrel temperature and 175rpm of screw speed. And then aqueous ammonia soaking was performed with 15%(w/w) ammonia solution at $60^{\circ}C$ for 1, 2, 4, 8, 12 hours on the extruded and raw miscanthus samples respectively. In the combined extrusion-soaking treatment, most compositions removal occurred within 1~2 hours and on a basis of 1 hour soaking treatment values, cellulose was recovered about 85% and other compositions, including hemicellulose, are removed about 50% from extruded miscanthus sample. The combined extrusion-soaking treated and soaking only treated samples were subjected to enzymatic hydrolysis using cellulase and ${\beta}$-glucosidase. The enzymatic digestibility value of combined extrusion-2 hours soaking treated sample was comparable to 12 hours soaking only treated sample. It means that extrusion treatment can shorten the conventional long reaction time of aqueous ammonia soaking. The findings suggest that the combination of extrusion and soaking is a promising pretreatment method to solve both problems for no lignin removal of extrusion and long reaction time of aqueous ammonia soaking.
