효모와 고오지 곰팡이의 혼합배양에 의한 주정생산 (Ethanol Production by the Mixed Culture of Some Aspergilli and Saccharomyces cerevisiae)

  • 쌀을 기질로 하여 고오지 곰팡이와 Saccharomyces cerevisiae를 함께 동시 접종하여 혼합 배양에 의한 동시 당화발효를 시도하였다. Aspergillus awamori, A. kawachii, A. niger, A. oryzae 및 A. shirousamii를 각각 S. cerevisiae와 혼합배양하였을 때 A. shirousamii와의 혼합배양에서 가장 높은 주정생산량을 보였다. 이 때의 가수량은 쌀 50g에 대하여 150ml 이었다. 곰팡이와 효모는 각각 $5{\times}10^2\;conidia/ml$$5{\times}10^6\;cells/ml$로 접종한 경우에, 용적에 대한 표면적의 비는 0.1에서 그리고 초기 pH6.5 및 $30^{\circ}C$의 배양조건에서 10일 동안 발효시 최고 12.9%의 주정이 생산되었다.

  • The aim of this study was to develop a new fermentation method in order to improve the digestion of soybean protein, and to promote normal fermentation of soybean. A proximate composition, such as moisture, pH, and reducing sugar, of fermented soybeans by the new fermentation was similar to those of controls. Neutral protease activity, the most important factor for fermented soybean products, was the highest, having about 636 Dig at 54 h fermentation. The content of total free amino acid was almost 3-18 times higher than controls. The three-step fermented soybeans can be used as a functional food ingredient for human consumption, with higher protein digestibility.

  • For the probiotic feed production, aerobic liquid fermentation of pulverized food wastes was attempted with a yeast Kluyveromyces marxianus. After grinding finely, optimal fermentation conditions of the substrate was investigated by shaking culture. The most active growth of the yeast was shown at solid content of 10%. The proper addition of urea(0.5g/l), o-phosphate(0.4g/l), molasses(4g/l), and yeast extract (1g/1) increased cell growth rate and viable cell count. For optimizing, the nutrients were all added to substrate and fermentation was carried in 2 litre jar fermenter. For the stimulation of hydrolyzing enzyme excretion, mixed culture with Aspersillus oryzae was also conducted. In 12 hours of fermentation, viable cell count of the yeast Kluyveromyces marxianus amounted to the number of 1.4 $\times$10$^{10}$ /1 in the culture medium.

Alcohol Fermentation of Opuntia ficus Fruit Juice

  • Prickly pear juice(PPJ) extracted from prickly pear fruit (Opuntia ficus-indica)was used as raw material for the production of alcoholic beverages. Prickly pear juice (PPJ) had 0.88 oBrix of soluble solid, pH 3.96 and 0.14% of total acidity. Alcohol fermentation of 25% PPJ including 22 oBrix of sugar and 1$\times$106 of inoculum was suitable for alcohol fermentation indicating the rapid decrease of sugar content. The 22 oBrix of sugar in PPJ (25%)GJ (50%) mixture was changed to 6.5 oBrix after fermentation at 3$0^{\circ}C$ for 7 days. The alcohol content was 9.2% (w/v). PPJ (70%)/GJ (30%) mixture produced alcoholic beverage with 6.9% alcohol content resulting in the gradual decrease of soluble for 7 days. On the other hand, PPJ (50%)/GJ(50%) mixture carried out completely the alcohol fermentation at 22$^{\circ}C$ for 6 days and enhanced the red color of alcoholic beverages.

Physico-chemical Properties of Lactobacillus casei 00692 during Fermenting for liquid-type Yogurt

  • This study was carried out to find the physico-chemical attributes of yogurt base with Lactobacillus case; 00692 during 72 hr fermentation at $37^{\circ}C$. The pH decreased up to 44 hr and plateaued thereafter, and the titratable acidity increased up to 40 hr. The number of lactic acid bacteria sharply increased with $1.0\times10^7cfu/mL$ up to 48 hr of fermentation and slowly increased thereafter. The free amino acids produced during the fermentation reached the maximum value at 40 hr and gradually decreased thereafter. In the result of electrophoresis, the band was the thickest at 44 hr and mostly disappeared at 72 hr fermentation. The present data showed that the range of optimum fermentation time for yogurt base using Lactobacillus casei 00692 was from 40 to 44 hr.

Production of Microbial Cellulose and Acids in Kombucha

  • Factors affecting the production of bacterial cellulose and organic acids in Kombucha fermentation were investigated. Kombucha was obtained by the fermentation (for 12 days at 3$0^{\circ}C$) of the green/black tea extract, supplemented with 10% white sugar, using an Oriental tea fungus as starter. Hitgher initial pH increased acid production with decreased cellulose production. With a cellulose pellicle or tea fungus broth as a starter, a 1~3 mm thick cellulose layer developed as a top layer every four days, and was removed subsequently while continuing fermentation. Addition of 30 mL tea fungus broth (13%, v/v) in Kombucha fermentation resulted in maximum production of a cellulose pellicle, indicating weak acid production. Yield of cellulose production at an early stage of fermentation was also higher when Kombucha was inoculated with a cellulose pellicle. In fact, addition of 1% (v/v) alcoholic beverage in the Kombucha fermentation activated the cellulose production, coupled with four times higher acid production.

고정화 효모를 이용한 Ethanol 발효 (A Study on Ethanol fermentation by Immobilized yeast.)

  • A Study for ethanol fermentation with immobilized yeast that is entrapped to Ca-alginate beads and batch system was carried out using molasses as substrate. The results are as follows. 1. The yield of alcohol fermentation is more efficacious then that of conventional fermentation process. The beads were used 15times and got a result of fermentation yield rate 89. 57%∼92.35%f, which is comparable with the rate of 86.3% gained from the conventional fermentation process. 2. The concentration of Ca-alginate was 1∼5% For long run use (2520 hours) it is necessary 2% or more concentration of Ca-alginate. 3. The amount of the yeast cells for entrap to Ca-alginate beads was required 1.0g (indried material) or more for 200g Ca-alginate beads.

Effects of Candida norvegensis Live Cells on In vitro Oat Straw Rumen Fermentation

  • This study evaluated the effect of Candida norvegensis (C. norvegensis) viable yeast culture on in vitro ruminal fermentation of oat straw. Ruminal fluid was mixed with buffer solution (1:2) and anaerobically incubated with or without yeast at $39^{\circ}C$ for 0, 4, 8, 16, and 24 h. A fully randomized design was used. There was a decrease in lactic acid (quadratic, p = 0.01), pH, (quadratic, p = 0.02), and yeasts counts (linear, p<0.01) across fermentation times. However, in vitro dry matter disappearance (IVDMD) and ammonia-N increased across fermentation times (quadratic; p<0.01 and p<0.02, respectively). Addition of yeast cells caused a decrease in pH values compared over all fermentation times (p<0.01), and lactic acid decreased at 12 h (p = 0.05). Meanwhile, yeast counts increased (p = 0.01) at 12 h. C. norvegensis increased ammonia-N at 4, 8, 12, and 24 h (p<0.01), and IVDMD of oat straw increased at 8, 12, and 24 h (p<0.01) of fermentation. Yeast cells increased acetate (p<0.01), propionate (p<0.03), and butyrate (p<0.03) at 8 h, while valeriate and isovaleriate increased at 8, 12, and 24 h (p<0.01). The yeast did not affect cellulolytic bacteria (p = 0.05), but cellulolytic fungi increased at 4 and 8 h (p<0.01), whereas production of methane decreased (p<0.01) at 8 h. It is concluded that addition of C. norvegensis to in vitro oat straw fermentation increased ruminal fermentation parameters as well as microbial growth with reduction of methane production. Additionally, yeast inoculum also improved IVDMD.

한외여과공정을 이용한 포도주의 품질개선 (Ultrafiltration for Quality Improvement of Wine)

  • 포도 착즙액의 당도를 $24^{\circ}Brix$로 조절하여 $25^{\circ}C$에서 2주간 발효하여 제조한 후 $15^{\circ}C$에서 14주간 숙성과정을 거친 포도주의 이화학적 성분 및 미생물의 변화를 살펴보았으며 한외여과 후의 포도주의 이화학적 특성의 변화를 관찰하였다. 총 세균수는 발효 초기 $3.3{\times}10^2\;CFU/mL$에서 2주간의 발효 후에는 $2.3{\times}10^6\;CFU/mL$로 증가한 후 숙성과정을 거친 후에는 $1.9{\times}10^3\;CFU/mL$로 다시 감소하였다. 효모의 경우에는 발효 초기 $2.8{\times}10^2\;CFU/mL$에서 발효 후에는 $2.2{\times}10^7\;CFU/mL$로 증가한 후, 숙성과정을 거친 후에는 $1.6{\times}10^4\;CFU/mL$로 역시 감소하는 경향을 보였다. 탁도, pH, 총당, 환원당, 고형물 함량은 발효가 진행됨에 따라 감소하였고, 산도, 알코올 함량은 증가하는 경향을 나타내어 발효가 완료된 후의 산도는 0.64%, 알코올 함량은 8.4%의 값을 보였다. 숙성과정 중에는 대부분의 이화학적 성질은 크게 변화하지 않았다. 포도주를 $0.45{\mu}m$ nitrocellulose 미세여과막을 이용하여 여과한 후 재질과 공경이 서로 다른 한외여과막을 사용하여 한외여과한 결과 Biomax 100K 막이 초기 flux가 $100\;liter/m^2/h$ (LMH)로 가장 높아 한외여과공정의 최적 한외여과막으로 선정하였다. 한외여과에 의해 포도주 내에 존재하는 미생물은 완벽하게 제거되었으며 환원당 함량과 고형물 함량은 약간 감소하였으나 그 이외의 이화학적 특성은 크게 변화하지 않았다. 포도주를 $30^{\circ}C$에서 12주간 저장하였을 경우 저장기간동안 미생물이 전혀 검출되지 않았으며 이화학적 특성도 변화하지 않았다.

경상도 전통 마른 오징어 식해로부터 젖산균의 분리, 동정 및 숙성과정 중 유기산의 변화 (Identification of Lactic Acid Bacteria and Changes of Organic Acid during Aging of Traditional Kyungsando Squid sikhe)

  • 경상도 동해안 연안에서 향토식품으로만 명맥을 유지하고 있는 마른오징어를 주원료로 한 식해를 제조하여 젖산균을 분리 동정하고 숙성 과정 중 성분 변화를 조사하였다. 경상도 전통마른오징어 식해로부터 성장이 우수하고 산생성력이 강한 3 균주는 Lactobacillus brevis SH-1, Lactobacillus plantarum SH-2 및 Leuconostoc mensenteroides SH-3로 잠정적으로 동정되었다. 식해의 발효기간별 총균수 및 젖산균수는 시간이 경과함에 따라 차차 증가하다가 10일을 전후하여 차차 감소하였고 숙성 6일째 총균수 및 젖산균수는 각각 $3.7{\times}10^9$$8.2{\times}10^8\;CFU/g$이였다. 식해의 비휘발성 유기산은 숙성과정 중 젖산을 포함하여 7 종류를 동정하였으며 구연산,주석산 및 젖산의 함량은 숙성과정 중 차차 증가하였고 특히 젖산의 증가는 다른 유기산보다 현저하게 증가하였다. 초산 및 주석산의 함량 변화는 숙성과정 7일 이후에 차차 증가하였다.
