Induction of Apoptotic Cell Death in Human Jurkat T Cells by a Chlorophyll Derivative (Cp-D) Isolated from Actinidia arguta Planchon

  • The chloroform and methanol (2;1, v/v) extract from an edible plant, Actinidia arguta Planchon, appeared to possess antitumor activity against human leukemias Jurkat T and U937 cells through inducing apoptosis. The substance in the solvent extract was purified by silica gel column chromatography, preparative TLC, and Sephadex LH-20 column chromatography. Characteristics of the substance analyzed by UV scanning analysis, $^1H$ and $^{13}C$ NMR spectra suggested that the substance belongs to the chlorophyll derivatives-like group. The $IC_{50}$ value of the chlorophyll derivative (Cp-D) determined by MTT assay was $15\mu\textrm{g}/ml$ for Jurkat, $10\mu\textrm{g}/ml$ for U937, and $11.4\mu\textrm{g}/ml$ for HL-60m and was more toxic to these leukemias than to solid tumors or normal fibroblast. In order to elucidate cellular mechanisms underlying the cytotoxicity, the effect of the Cp-D on Jurkat T cells was investigated. When cells were treated with the Cp-D at a concentration of $15\mu\textrm{g}/ml$, [3H]thymidine incorporation declined rapidly and wa undetectable in 1h. However, no significant changes were made in the cell cycle distribution of the cells by 24h. The sub-Gl peak representing apoptotic cells began to be detectable in 36h, at which time apoptotic DNA fragmentation was also detected on agarose gel electrophoresis, demonstrating that the cytotoxic effect of the Cp-D is attributable to the induced apoptosis. Under the same conditions, although the protein level of cyclin-dependent kinases such as cdc4, csk6, cdk2, and cdc2 was not significantly changed until 24h, the kinase activity of all c안 rapidly declined and reached a minimum level within 1-6h and then recovered to the initial level by 12h and sustained until 24h. These results suggest that inactivation of cdks at an inappropriate time during the cell cycle progression in jurkat T cells following a treatment with the Cp-D leads to induction of apoptotic cell death.

Inhibitory Effect of Perilla Sprouts Extracts on Oxidation of Perilla Oil (들깨유의 산화에 대한 들깨 발아 싹 추출물의 억제효과)

  • During 10 days germination of perilla seeds for sprouts preparation, the changes of proximate composition and antioxidant activities were monitored, and the inhibitory effect of sprouts extracts on perilla oil oxidation was also studied. The moisture content in seeds(2.9%) was increased to 9.2% in sprouts at 10 days while crude ash content wasn't significantly. The crude fat and protein contents were reduced from 46.8% and 20.7% in seeds to 18.2% and 18.3% in sprouts, respectively, but reducing sugar and fiber contents increased from 2.2% and 14.8% to 12.8% and 22.4%, respectively. Compared with perilla leaf, sprouts at 10 days contained more fat, carbohydrate, reducing sugar, and fiber while less moisture, ash, and protein. Antioxidant activities during germination were increased and reached to maximum at 8 days in which Trolox equivalent antioxidant capacity(TEAC) based on DPPH and ABTS radical scavenging were 133.1 and 136.8 Trolox eq. mmol/kg, respectively, and ferric ion reducing power(FRAP) was 399.3 Fe(II) eq. mmol/kg. Polyphenol content(19.2 g/kg) was maximum at this stage, too. Perilla leaf showed similar TEAC but higher FRAP than the sprouts. When methanol extract of sprouts at 8 days was added to perilla oil, the oil oxidation was delayed in dose dependent manner. The induction time for oxidation was extended about 2.8 times by adding 2.5%(w/w) extract, that is, from 1.67 hr(control) to 4.62 hr. This induction time corresponded to 38% level of that of perilla oil containing 2.5% BHT.

EFFECTS OF CONDENSED PHOSPHATES ON THE DENATURATION OF ALASKA POLLACK MUSCLE DURING REFREEZING AN COLE STORAGE (재동결 명태육의 냉동변성에 미치는 축합인산염처리의 효과에 대하여)

  • The present study was ,conducted to evaluate the effects of condensed phosphates on the refeezing damage of Alaska pollack muscle. The fillets were dipped in such solution as 5 and $10\%$ sodium polyphosphate, 1 and $5\%$ mixture of sodium polyphosphate and sodium pyrophosphate (1:1, w/w) for 1 and 5 minutes, respectively, before refreezing. And fillets were frozen at $27^{\circ}C\~28^{\circ}C$ and stored for 15 days at $-18^{\circ}\~-20^{\circ}$. The degree of denaturation was estimated by determining amounts of drip relased, content of total solids, nitrogen, and DNA in the drip an cooking-weight-loss. Phosphorus absorbed in the muscle was also determined. Phosphorus absorbed in the fillets treated with loft solution of sodium polyphosphate for 5 minutes amounted to 101 mg/100g muscle as $P_2O_5$. The absorption was dependent on tile concentration of treating solution rather than on the dipping time. The increase of phosphorus absorption seemed to affect to reduction of drip. Among the treating conditions, $10\%$, 5 minutes and $10\%$ 1 minute with sodium polyphosphate appeared most effective ones on drip reduction. The effect of $5\%$, minutes with the mixture of sodium polyphosphate and sodium pyrophosphate did not show so benefitable effect in refrozen fillets. As a tendency total solids, nitrogen, and DNA in tile drip varied proportionally to the amount of drip released. And the content of DNA was lower than the amount. Treatment, at higher the concentration and longer the dipping time, resulted in the lower cooking-weight-loss and the better quality on organoleptic test of thawed fillets.

Anaesthetic Effect of Lidocaine Hydrochloride-Sodium Bicarbonate and MS-222 on the Greenling (Hexagrammos otakii) (쥐노래미 (Hexagrammos otakii)에 대한 염산리도카인-중탄산나트륨과 MS-222의 마취 효과)

  • Anaesthetic effect of lidocaine hydrochloride-sodium bicarbonate mixture $(lidocaine\;HCL/NaHCO_3)$ and tricaine methanesulfonate (MS-222) was tested for the greenling (Hexagyammos otakii) at three different temperature regimes: $12^{\circ}C,\;8^{\circ}C\;and\;24^{\circ}C.$ Based on the exposure and recovery time, effective dose of lidocaine $HCl/NaHCO_3$ was 800 ppm $(18^{\circ}C)\;and\;300\;ppm\;(24^{\circ}C)$ for greenling of $21.0\pm1.4\;cm$ body length. Anaesthetic dose and temperature-dependent relationship in exposure and recovery time were observed. Effective dose of MS-222 at $18^{\circ}C$ was proven to be 125 ppm and 150 ppm. Combination of lidocaine $HCl/NaHCO_3$ and MS-222, considerably reduced the dosage of each anaesthetic required to give rapid, deep anaesthetic condition. In the dry exposure after anaesthetic, the control fish exhibition $22\%$ mortality after dry exposure of 20 min; whereas, the anaesthetic condition with 800 ppm lidocaine $HCl/NaHCO_3$ for 1 min exhibited delayed recoveries from the anaesthetic condition with mortalities of $20\%,\;41\%,\;78\%\;and\;100\%$ after dry exposures of 8 min, 12 min, 16 min and 20 min, respectively. The results indicate that lidocaine $HCl/NaHCO_3$ can be used as suitable anaesthetic for the greenling.

An Experimental Study for Dryer (건조기 고안 제작에 관한 연구)

  • A newly devised dryer with heated air for the farm products, especially suited for high water content materials such as red pepper, Beer ground, each Vegetables, and Low water content materials such as Rough rice was tested for its thermal efficiency and drying mechanism, and the optimum conditions for each sample were established. In order to improve the present rural situation of drying farm products which entirely dependent upon natural solar radiation, a study upon an economic multi-parpose dryer was conducted. A series of drying tests were run first with red pepper which is one of the important cash crop in Korean farm. And successive series of tests were also run with such proaucts as garlic, sweet potatoes, green onion, radish, Beer ground and Rough rice. The results from the above experiment in drying system with heat dryer can be summarized as follows. 1. Drying duration could be shortened by the tempering effect in high water content crop such as red pepper and beer ground. 2. The color changes occured in around 20% water content in red pepper. The degree of color change was heavily affected by high temperature and short drying duration. 3. The drying condition of red pepper was most favourable at the temperature of 85$^{\circ}C$ in early stage and 80$^{\circ}C$ in middle stage and 75$^{\circ}C$ at the final stage, and with the air rate of 0.81㎥/sec and with sample amount of 200kg. 4. The drying condition of Rough rice(I.R.667) was most favourable at the templature of 40$^{\circ}C$ in early stage and 35$^{\circ}C$ in middle stage and final stage and with the air rate of 0.2㎥/sec and with sample amount of 75kg. 5. In order to prevent the color change of red pepper and to assure high efficiency in drying mechanism, it was necessary to lower the temperature as the time passes in drying process. 6. For vege tables, the drying rate were short in early stage and there was also tempering effect. However, for garlics, Constant drying rates through the early and final stages were observed and there were no tempering effects. 7. Evaluation of Corrosion Effects on Permanent Ground Anchors (영구 지반앵커에 대한 부식의 영향 평가)

  • The corrosion rate measurement procedure for the permanent ground anchors using polarization resistance measurements and electrochemical impedance spectroscopy is presented in this paper. The polarization resistance measurements were used to determine the correlation between corrosion rate in the steel and soil characteristics. The electrochemical impedance spectroscopy was used to predict the time dependent corrosion reaction and evaluate the different type of coating systems and the effect of cement grouting on the corrosion attack under various conditions. The results indicate that a low pH soil is a good indicator of a corrosive soil. The low pH soil condition (<5) in both clay and sand has a significant effect on the corrosion reaction of steel members in permanent found anchors. In the case of neutral and alkaline conditions beyond pH 6 in clay and sand, no consistent acceleration of corrosion was measured and the corrosion rate was constant regardless of variations of soil pH levels. Laboratory test data for porcelain clay indicate that the change of soil pH level has a small influence on the corrosion reaction in the steel member. The use of cement footing in the bonded length is sufficient to decrease the corrosion rate to a level close to 0.003∼0.01mm/y at the end of the given period. With epoxy and fusion bonded epoxy coating, the steel specimens remained unaffected and retained the original condition. It is suggested that epoxy and fusion bonded epoxy coating can provide effective protection against corrosion for a long time even in aggressive environment.

The Occupational Structure of the Junior College Students' Transfer and Occupation Status Effect (편입학의 직업지위 효과와 직업구조: 전문대졸자 vs. 일반대 편입학자)

  • This study surveyed and analyzed the labor market result when comparing the case that junior college students found a job after transferring to 4-year university with the case that they found the job directly after graduating the junior college. The difference of the occupational structure following the transfer was surveyed in detail and this study examined what effect the transfer had on the occupation status. The graduate occupation movement survey(GOMS) was used and the samples of 544 persons were extracted to use through propensity score matching(PSM) to raise the choice convenience of the sample. The occupation status index to use as a dependent variable was developed to apply. The study result is as follows. First, as a result of confirming the occupational structure of the transfer, the transfer students were distributed in more various industrial fields evenly compared to the non-transfer students in the horizontal industrial structure, and non-transfer students were intensively distributed in the certain field. In the vertical occupation status structure, transfer students were distributed in the high occupation status more than non-transfer students who were broadly distributed in the low occupation status. Second, it was revealed the college transfer was highly effective for the occupation status, which appeared statistically significantly. The explanation variable that appeared significantly outside of that included the parents' academic background, major affiliation, employment pattern, major job coincidence, and job coincidence. The higher students' academic background was, the higher the occupation status was and in case of the engineering natural science affiliation, the occupation status was higher than the humanities social science. In case of full-time workers, their occupation status was higher than one of part time workers and it was revealed the occupation status was high when the their major coincided with the job and their educational/technical level coincided with their job.

Evaluation of Lipid Accumulation's Inhibitory Activity on 3T3-L1 Cells with Red Yeast Barley Extracts (홍맥 추출물의 3T3-L1세포에 대한 지방 축적 저해 활성평가)

  • Red yeast rice has been extensively used as food and traditional medicine for thousands of years in East Asian countries. It is produced by the fermentation of a particular yeast (in general, Monascus purpureus) as rice and various cereals (barley, soybean, etc.). Monascus sp. produces many secondary metabolites during its growth, including pigments, monacolins, and γ-aminobutyric acid. Some metabolites―specifically, monacolin K, γ-aminobutyric acid, dimerumic acid, and monascus pigments―have been reported to lower cholesterol and blood pressure while showing anti-obesity effects. In this study, we investigated the anti-obesity effect of ethanol extract from red yeast barley (RYB) fermented with Monascus sp. BHN-MK 2 on 3T3-L1 cells. The anti-obesity effects of RYB extract were examined: its lipid accumulation inhibitory effect was tested by Oil Red O staining, and obesity-related mRNA expression levels were tested by real-time RT-PCR in MDI stimulated 3T3-L1 cells. The intracellular lipid content of MDI-stimulated 3T3-L1 cells decreased significantly to 5.04%, 12.24%, and 23.52% in response to 200, 400, and 800 ㎍/ml RYB, respectively. Moreovers, we evaluated that RYB extract significantly downregulated the expression of C/EBPα, SREBP-1, and PPAR-γ gene in a dose-dependent manner. As a result, red yeast barley ethanol extracts exerted the strongest anti-obesity effects. Also, the results indicate that red yeast barley could be used as a functional anti-obesity food material.

Expression of Heat Shock Protein 70 Gene and Body Color Changes in Non-biting Midge Larvae (Glyptotendipes tokunagai) Effected by O3 Treatment (오존(O3) 노출에 의한 조각깔따구(Glyptotendipes tokunagai)의 체색 변화 및 heat shock protein 70 발현 변화)

  • Ozone (O3) is a general disinfectant to remove micro-pollutants in water treatment system. Previous studies have reported effect of ozone to bacteria and pathogens removal, but its effect to the relatively large organisms has little known. In this study, we investigated potential effects of ozone toxicity to the non-bite midge larvae (Glyptotendipes tokunagai) with accumulate mortality, coloration change and expression of heat shock protein 70 (HSP70). The accumulate mortality rate of G. tokunagai increased in a dose-time dependent manner and the highest mortality rate was observed to 75% at 30 minute of exposure duration with 2.0 ppm of ozone concentration. Exposure to ozone was a factor increasing body color of the larvae. The tendency of HSP70 mRNA expression showed up-regulation in ozone exposure at 20 minute. After that time, the expression of HSP70 in exposed group decreased to a similar level of control group. Our results clearly showed that ozone toxicity affects physical and molecular activity of G. tokunagai, implying the potential hazardous of ozone in the aquatic ecosystem including macroinvertebrates.

Ginsenosides Rk1 and Rg5 inhibit transforming growth factor-β1-induced epithelial-mesenchymal transition and suppress migration, invasion, anoikis resistance, and development of stem-like features in lung cancer

  • Background: Lung cancer has a high incidence worldwide, and most lung cancer-associated deaths are attributable to cancer metastasis. Although several medicinal properties of Panax ginseng Meyer have been reported, the effect of ginsenosides Rk1 and Rg5 on epithelial-mesenchymal transition (EMT) stimulated by transforming growth factor beta 1 (TGF-β1) and self-renewal in A549 cells is relatively unknown. Methods: We treated TGF-β1 or alternatively Rk1 and Rg5 in A549 cells. We used western blot analysis, real-time polymerase chain reaction (qPCR), wound healing assay, Matrigel invasion assay, and anoikis assays to determine the effect of Rk1 and Rg5 on TGF-mediated EMT in lung cancer cell. In addition, we performed tumorsphere formation assays and real-time PCR to evaluate the stem-like properties. Results: EMT is induced by TGF-β1 in A549 cells causing the development of cancer stem-like features. Expression of E-cadherin, an epithelial marker, decreased and an increase in vimentin expression was noted. Cell mobility, invasiveness, and anoikis resistance were enhanced with TGF-β1 treatment. In addition, the expression of stem cell markers, CD44, and CD133, was also increased. Treatment with Rk1 and Rg5 suppressed EMT by TGF-β1 and the development of stemness in a dose-dependent manner. Additionally, Rk1 and Rg5 markedly suppressed TGF-β1-induced metalloproteinase-2/9 (MMP2/9) activity, and activation of Smad2/3 and nuclear factor kappa B/extra-cellular signal regulated kinases (NF-kB/ERK) pathways in lung cancer cells. Conclusions: Rk1 and Rg5 regulate the EMT inducing TGF-β1 by suppressing the Smad and NF-κB/ERK pathways (non-Smad pathway).
