The Inhibitory Effect of Opioid on the Hyperpolarization-Activated Cation Currents in Rat Substantia Gelatinosa Neurons

  • The action of opioid on the hyperpolarization-activated cation current $(I_h)$ in substantia gelatinosa neurons were investigated by using whole-cell voltage-clamp recording in rat spinal brain slices. Hyperpolarizing voltage steps revealed slowly activating currents in a subgroup of neurons. The half-maximal activation and the reversal potential of the current were compatible to neuronal $I_h.$ DAMGO $(1\;{\mu}M),$ a selective- opioid agonist, reduced the amplitude of $I_h$ reversibly. This reduction was dose-dependent and was blocked by CTOP $(2\;{\mu}M),$ a selective ${\mu}-opioid$ antagonist. DAMGO shifted the voltage dependence of activation to more hyperpolarized potential. Cesium (1 mM) or ZD 7288 $(100\;{\mu}M)$ blocked $I_h$ and the currents inhibited by cesium, ZD 7288 and DAMGO shared a similar time and voltage dependence. These results suggest that activation of ${\mu}-opioid$ receptor by DAMGO can inhibit $I_h$ in a subgroup of rat substantia gelatinosa neurons.

Mercury induced the Accumulation of Amyloid Beta (Aβ) in PC12 Cells: The Role of Production and Degradation of Aβ

  • Extracellular accumulation of amyloid beta protein ($A{\beta}$) plays a central role in Alzheimer's disease (AD). Some metals, such as copper, lead, and aluminum can affect the $A{\beta}$ accumulation in the brain. However, the effect of mercury on $A{\beta}$ accumulation in the brain is not clear. Thus, this study was proposed to estimate whether mercury concentration affects $A{\beta}$ accumulation in PC12 cells. We treated 10, 100, and 1000 nM $HgCl_2$ (Hg) or $CH_3HgCl_2$ (MeHg) for 48 hr in PC12 cells. After treatment, $A{\beta}_{40}$ in culture medium increased in a dose- and time-dependent manner. Hg and MeHg increased amyloid precursor protein (APP), which is related to $A{\beta}$ production. Neprilysin (NEP) levels in PC12 cells were decreased by Hg and MeHg treatment. These results suggested that Hg induced $A{\beta}$ accumulation through APP overproduction and reduction of NEP.

  • When chemotherapy is administered during pregnancy, it is important to consider the fetus chemotherapy exposure, because it may lead to fetal consequences. Paclitaxel has become widely used in the metastatic and adjuvant settings for woman with cancer including breast and ovarian cancer. Therefore, we attempted to clarify the transport mechanisms of paclitaxel through blood-placenta barrier using rat conditionally immortalized syncytiotrophoblast cell lines (TR-TBTs). The uptake of paclitaxel was time- and temperature-dependent. Paclitaxel was eliminated about 50% from the cells within 30 min. The uptake of paclitaxel was saturable with $K_m$ of $168{\mu}M$ and $371{\mu}M$ in TR-TBT 18d-1 and TR-TBT 18d-2, respectively. [$^3H$]Paclitaxel uptake was markedly inhibited by cyclosporine and verapamil, well-known substrates of P-glycoprotein (P-gp) transporter. However, several MRP substrates and organic anions had no effect on [$^3H$]paclitaxel uptake in TR-TBT cells. These results suggest that P-gp may be involved in paclitaxel transport at the placenta. TR-TBT cells expressed mRNA of P-gp. These findings are important for therapy of breast and ovarian cancer of pregnant women, and should be useful data in elucidating teratogenicity of paclitaxel during pregnancy.

Anti-Helicobacter and Anti-inflammatory Effects of Sohamhyungtang in Helicobacter pylori-Infected Human Gastric Epithelial AGS cells

  • This study evaluated the anti-Helicobacter and anti-inflammatory effects of Sohamhyungtang (SHHT). The minimum inhibitory concentration (MIC) of SHHT against Helicobacter pylori (H. pylori) was determined by the agar dilution method. Expression of the H. pylori cagA gene in the presence of SHHT was determined by quantitative real-time polymerase chain reaction (qRT-PCR). Inhibition of H. pylori urease by SHHT was determined by the phenol-hypochlorite assay. Antiadhesion activity of SHHT was measured by urea-phenol red reagent. Inhibition of nitric oxide (NO) production in AGS cells was measured with Griess reagent. Inducible nitric oxide synthase (iNOS) and IL-8 mRNA expression in AGS cells which were infected with H. pylori was determined by qRT-PCR. IL-8 level was measured by enzyme-linked immunosorbent assay (ELISA). The MIC of SHHT was $100{\mu}g/mL$ and the expression of cagA gene was decreased about 25 folds in the presence of SHHT. H. pylori urease was inhibited 90% by SHHT. SHHT inhibited H. pylori adhesion on AGS cell in a concentration dependent manner. mRNA expression of iNOS and IL-8 and the production of NO and IL-8 were significantly decreased in the presence of SHHT. In conclusion, SHHT showed anti-Helicobacter activity and has potent anti-inflammatory effect on H. pylori-induced inflammation in human gastric epithelial AGS cells.

Welding Deformation Analysis of Plates Using the Inherent Strain-based Equivalent Load Method (고유변형률 기반 등가하중법을 이용한 판의 용접변형 해석)

  • IIn this study, used is the equivalent loading method based on the inherent strain to predict the welding deformation of panel members. Equivalent loads are computed from the inherent strain distribution around weld line, and then applied for the linear finite element analysis. Thermal deformation of panel members can be, of course, carried out through the rigorous thermal elasto-plastic analysis procedure but it is not practical in applying to predicting the welding deformation of large structures such as blocks found in a ship structure from view of computing time. The present equivalent load approach has been applied to flat plate model to verify the present approach, and to several curved plate models having the curvature in the welding direction to investigate the effect of the longitudinal curvature upon the weld-induced deformation. The results are compared with those by thermal elasto-plastic analysis. As far as the present results are concerned, it can be said that the present approach shows good agreement with the results by welding experiment and the rigorous thermal elasto-plastic analysis. The present approach has been also applied to predict the welding deformation of panel block as for application illustration to practical model.

Modeling of the Mechanical Drivetrain of an Electric Vehicle for Investigation of Torsional Oscillation Characteristics (전기자동차 기계적 구동계의 모델링 및 비틀림 진동특성 분석)

  • Torsional oscillations of the mechanical drivetrain in electric vehicles are generated under rapid driving conditions. These lead to an uncomfortable jerking of the vehicle and to an increased stress of the mechanical components. To analyze this phenomenon, a drivetrain model is constructed with lumped parameters. The model parameters are identified by geometrical design data and experimental tests. The proposed model is validated by simulation and experimental tests in the time and the frequency domains. As a result, the torsional oscillations are observed at 7Hz of a low damped natural frequency. Also, the analysis of the effect of the parameter variations on the oscillations shows that the oscillation characteristic is mainly dependent on the rotor inertia, and the stiffness of the mounting of the drive aggregate and the driveshaft. The results will be utilized on the basis of the design of an electric drivetrain and an active control of drivetrain oscillations.

The Effect of Static Stretching and Evjenth-Hamberg Stretching for Isokinetic Muscle Strength of Knee Joint (정적인 스트레칭과 Evjenth-Hamberg 스트레칭이 슬관절 등속성 근력에 미치는 효과)

  • Purpose: The purpose of this study was to examine the effects of static stretching and Evjenth-Hamberg stretching on isokinetic muscle strength of knee flexors and extensors. Methods: The subjects were composed of eighty healthy males without weight-training experience. ROM of knee joint measured active maximal extension and isokinetic Peak Torque measured $60^{\circ}/sec,\;120^{\circ}/sec$ using an the En-Knee. Three tests(Baseline, 4 weeks, 8 weeks, respectively) was operated to examine change of each variable. Data were analyzed with a $2{\times}3$ analysis of variance ($group{\times}test$) for repeated measures on last factor by SPSS package 10.0. The data analysis revealed muscle strength were dependent on stretching method. Results: The results were as follows. First, Evjenth-Hamberg stretching(E-HS) was more effective than static stretching(SS) on ROM. Second, Peak Torque of knee flexors and extensors was improved in both methods by each time. but E-HS was more improved than SS. Conclusion: In conclusion, This study indicates that E-HS is more efficient than SS on muscle strength improvement.

Effects of combined therapy with thalidomide and glucantime on leishmaniasis induced by Leishmania major in BALB/c mice

  • For treating Leishmania major infection in BALB/c mice, we used thalidomide in conjunction with glucantime. Groups of mice were challenged with $5{\time}10^3$ metacyclic promastigotes of L. major subcutaneously. A week after the challenge, drug treatment was started and continued for 12 days. Thalidomide was orally administrated 30 mg/kg/day and glucantime was administrated intraperitoneally (200 mg/kg/day). It was shown that the combined therapy is more effective than single therapies with each one of the drugs since the foot pad swelling in the group of mice received thalidomide and glucantime was significantly decreased (0.9 :t 0.2 mm) compared to mice treated with either glucantime, thalidomide, or carrier alone (1.2 :t 0.25, 1.4 :t 0.3, and 1.7 :t 0.27 mm, respectively). Cytokine study showed that the effect of thalidomide was not dependent on IL-12; however, it up-regulated IFN-yand down-regulated IL-10 production. Conclusively, thalidomide seems promising as a conjunctive therapy with antimony in murine model of visceral leishmaniasis.

Ammonia Nitrogen Removal in Wastewater Using Microwave Irradiation (마이크로웨이브를 이용한 폐수 내 고농도 암모니아성질소 제거)

  • Industrial use of microwave heating as an alternative to conventional heating is becoming popular mainly due to dramatic reductions in reaction time. Therefore, this work experimentally determined the effect of microwave irradiation on ammonia nitrogen removal in wastewater. The effects of air flow rate (0.3~0.9 L/min), treatment temperature ($70{\sim}100^{\circ}C$), and initial pH (9~11) were characterized. As the air flow rate increased from 0.3 to 0.9 L/min, the ammonia removal rate constant (k) increased from -0.6642 to $-1.0755min^{-1}$. As the temperature increased from 70 to $100^{\circ}C$, k increased -0.0338 to $-1.0755min^{-1}$. As the pH increased from 9 to 11, k increased -0.2443 to $-1.0755min^{-1}$. Ammonia removal was strongly dependent on temperature and pH rather than air flow rate. The results show that microwave irradiation is effective in ammonia nitrogen removal in wastewater due to advantages of fast and effective processing.

Effects of Material Characteristics on the Dynamic Response of the Reinforced Concrete Slabs (재료 특성이 철근 콘크리트 슬래브의 동적 거동에 미치는 영향)

  • The reinforced concrete slab is one of main structure members in the construction industry sector. However, most of researches regarding to RC slabs have been focused on two-dimensional Mindlin-type plate element on the basis of laminated plate theory since three-dimensional solid element has a lot of difficulties in finite element formulation and costs in CPU time. In reality, the RC slabs are subjected to dynamic loads like a heavy traffic vehicle load, and thus should insure the safety from the static load as well as dynamic load. Once we can estimate the dynamic behaviour of RC slabs exactly, it will be very helpful for design of it. In this study, the 20-node solid element has been used to analyze the dynamic characteristics of RC slabs with clamped edges. The elasto-visco plastic model for material non-linearity and the smeared crack model have been adopted in the finite element formulation. The applicability of the proposed finite element has been tested for dynamic behaviour of RC slabs with respect to characteristics of concrete materials in terms of cracking stress, crushing strain, fracture energy and Poisson's ratio. The effect on dynamic behaviour is dependent on not crushing strain but cracking stress, fracture energy and Poisson's ratio. In addition to this, it is shown the damping phenomenon of RC slabs has been identified from the numerical results by using Rayleigh damping.
