Phosphorylation of tyrosine-14 on Caveolin-1 enhances lipopolysaccharide-induced inflammation in human intestinal Caco-2 cells

  • Caveolin-1 (Cav-1) is the main structural component of the caveolae on the plasma membrane, which regulates various cellular processes, including cell growth, differentiation, and endocytosis. Although a recent study demonstrated that Cav-1 might be involved in diabetes-associated inflammation, its exact role in the intestine was unclear. In this study, we examined the intestinal expression of Cav-1 in diabetic conditions. We also investigated its effect on lipopolysaccharide (LPS)-induced inflammation by expressing this protein in human intestinal Caco-2 cells lacking Cav-1. We observed that increased Cav-1 levels and decreased expression of tight junction proteins affected intestinal permeability in high-fat diet-induced diabetic mice. When Caco-2 cells were treated with LPS, Cav-1 enhanced the NF-κB signaling. Moreover, LPS reduced the expression of tight junction proteins while it increased cell-cell permeability and reactive oxygen species generation in Caco-2 cells and this effect was amplified by cav-1 overexpression. LPS treatment promoted phosphorylation of tyrosine-14 (Y14) on Cav-1, and the LPS-induced NF-κB signaling was suppressed in cells expressing non-phosphorylatable Cav-1 (tyrosine-14 to phenylalanine mutant), which reduced intestinal barrier permeability. These results suggest that Cav-1 expression promotes LPS-induced inflammation in Caco-2 cells, and phosphorylation of Y14 on Cav-1 might contribute to the anti-inflammatory response in LPS-induced NF-κB signaling and cell permeability.

대황(大黃)과 황금(黃芩) 추출물 혼합물이 급성 역류성 식도염 흰쥐에 미치는 효과 (Effect of a Mixture of Rhei Rhizoma and Scutellariae Radix Extract on Acute Reflux Esophagitis Rats)

  • Objective : Reflux esophagitis is a disease caused by reflux of stomach contents, stomach acid, and pepsin into the esophagus, and is currently increasing worldwide. This study was conducted to evaluate the effect of a mixture of Rhei Rhizoma and Scutellariae Radix (RS) extract on acute reflux esophagitis in rats. Methods : Rats were divided into five groups for examination: Normal group (Nor, n=8), water-treated acute reflux esophagitis rats (Con, n=8), tocopherol 30 mg/kg body weight/day-treated acute reflux esophagitis rats (Toco, n=8), RS 100 mg/kg body weight/day-treated acute reflux esophagitis rats (RS100, n=8), RS 200 mg/kg body weight/day-treated acute reflux esophagitis rats (RS200, n=8). All rats fasted for 18 h and then were derived by linking the metastatic junction between pylorus and forestomach and corpus. And rats were sacrificed 5 h after surgery. We analyzed the expression of NADPH, MAPK, inflammatory, anti-inflammatory, and tight junction related proteins by western blot in esophageal tissue and observed the level of reactive oxygen species (ROS), alanine aminotransferanse (ALT), and aspartate aminotransferase (AST) in serum. Results : RS administration significantly protected the esophageal mucosal damage of reflux esophagitis, and ROS, AST, and ALT levels were significantly reduced in RS administration compared to Con group. In addition, RS administration effectively suppressed MAPK and NF-κB pathways and upregulated protein expressions of tight junction protein. Conclusions : These results suggest that RS protected the esophageal mucosa by inhibiting the MAPK and NF-κB pathways and upregulating tight junctions.

Differential Expression of the Tight Junction Protein, Occludin, in Brain Tumors

  • Objective : Cerebral edema develops in the brain tumors by loosening of the endothelial tight junction. Tight junction[TJ] proteins, such as occludin and claudin bind adjacent cells tightly. Authors examine the expression rate of occludin in human brain tumors to evaluate the effect of altered expression of occludin on cerebral edema. Methods : Seventy surgical specimens stored at $-70^{\circ}C$ were used. It included 14 astrocytic tumors, 27 meningiomas, 12 scwannomas, 7 pituitary adenomas, 6 hemangioblastomas. and 4 craniopharyngiomas. After protein extraction, expression of occludin was investigated by Western blot analysis. The tumors were classified according to World Health Organization[WHO] classification. Results : The expression rates of occludin in brain tumors were : glioma [8/14=57.1%]. meningioma [16/27=59.3%], schwannoma [10/12=83.3%], pituitary adenoma [6/7=85.7%], hemangioblastoma [6/6=100%], and craniopharyngioma [3/4=75.0%]. The expression rate in glioma and meningioma was lower than other brain tumors. In gliomas, high grade tumor [1/4=25.0%] exhibited lower expression rate of occludin than low grade one [7/10=70.0%]. Conclusion : These results suggest that the expression of occludin is different among the various kinds of brain tumors. In gliomas, its expression is correlated with the histological grade. It may indicate that occludin plays a role in the development of edema in the brain tumors.

홍삼에 의한 Caco-2 단세포층 간극의 안정화 (Effect of Korean Red Ginseng on the Stability of the Tight Junction of Intestinal Epithelial Cells)

  • 본 연구에서는 홍삼성분에 의한 장상피세포 간극(tight junction)의 안정화에 대해서 조사하였다. Transwell에 배양한 Caco-2 단세포층을 HBSS로 치환한 후 단세포층의 상층부에 홍삼추출물을 처리하고 96시간동안 배양하면서 TEER(transepithelial electrical resistance)수치의 변화를 조사하였다. 홍삼 추출물을 $20\;{\mu}g/mL$ 이하의 농도로 상층부에 첨가하였을 때에는 시간이 지남에 따라 점차 감소하는 대조구와는 달리 TEER수치가 덜 감소하였다. 한편 $130\;{\mu}g/mL$ 이상의 농도로 처리하였을 때는 TEER수치가 현저히 감소하였으나 MTT assay 결과 세포독성에 의한 것은 아니었다. 인삼으로부터 분리한 저분자물질, 다당류, 단백질, 사포닌, 폴리페놀 조획분을 각각 상층부에 첨가하였을 때, 장상피세포 간극의 안정화 활성을 나타내는 성분은 폴리페놀 및 사포닌 조획분이었으며, 전자는 높은 활성을 후자는 낮은 활성을 나타내었다. 이처럼 홍삼추출물의 폴리페놀 조획분과 사포닌 조획분은 장상피세포의 간극을 안정화시킴으로서, 식품 알레르겐의 체내유입을 차단하여 알레르기억제에 기여할 수 있을 것으로 추측된다.

Effects of multi-strain probiotic supplementation on intestinal microbiota, tight junctions, and inflammation in young broiler chickens challenged with Salmonella enterica subsp. enterica

  • Objective: This study assessed the effects of probiotics on cecal microbiota, gene expression of intestinal tight junction proteins, and immune response in the cecal tonsil of broiler chickens challenged with Salmonella enterica subsp. enterica. Methods: One-day-old broiler chickens (n = 240) were randomly allocated to four treatments: negative control (Cont), multi-strain probiotic-treated group (Pro), Salmonella-infected group (Sal), and multi-strain probiotic-treated and Salmonella-infected group (ProSal). All chickens except those in the Cont and Pro groups were gavaged with 1×108 cfu/mL of S. enterica subsp. enterica 4 days after hatching. Results: Our results indicated that body weight, weight gain, and feed conversion ratio of birds were significantly reduced (p<0.05) by Salmonella challenge. Chickens challenged with Salmonella decreased cecal microbial diversity. Chickens in the Sal group exhibited abundant Proteobacteria than those in the Cont, Pro, and ProSal groups. Salmonella infection downregulated gene expression of Occludin, zonula occludens-1 (ZO1), and Mucin 2 in the jejunum and Occludin and Claudin in the ileum. Moreover, the Sal group increased gene expression of interferon-γ (IFN-γ), interleukin-6 (IL-6), IL-1β, and lipopolysaccharide-induced tumor necrosis factor-alpha factor (LITAF) and reduced levels of transforming growth factor-β4 and IL-10 compared with the other groups (p<0.05). However, chickens receiving probiotic diets increased Lactobacillaceae abundance and reduced Enterobacteriaceae abundance in the ceca. Moreover, supplementation with probiotics increased the mRNA expression of Occludin, ZO1, and Mucin 2 in the ileum (p<0.05). In addition, probiotic supplementation downregulated the mRNA levels of IFN-γ (p<0.05) and LITAF (p = 0.075) and upregulated IL-10 (p = 0.084) expression in the cecal tonsil. Conclusion: The administration of multi-strain probiotics modulated intestinal microbiota, gene expression of tight junction proteins, and immunomodulatory activity in broiler chickens.

Diet-Induced Gut Dysbiosis and Leaky Gut Syndrome

  • Chronic gut inflammation promotes the development of metabolic diseases such as obesity. There is growing evidence which suggests that dysbiosis in gut microbiota and metabolites disrupt the integrity of the intestinal barrier and significantly impact the level of inflammation in various tissues, including the liver and adipose tissues. Moreover, dietary sources are connected to the development of leaky gut syndrome through their interaction with the gut microbiota. This review examines the effects of these factors on intestinal microorganisms and the communication pathways between the gut-liver and gut-brain axis. The consumption of diets rich in fats and carbohydrates has been found to weaken the adherence of tight junction proteins in the gastrointestinal tract. Consequently, this allows endotoxins, such as lipopolysaccharides produced by detrimental bacteria, to permeate through portal veins, leading to metabolic endotoxemia and alterations in the gut microbiome composition with reduced production of metabolites, such as short-chain fatty acids. However, the precise correlation between gut microbiota and alternative sweeteners remains uncertain, necessitating further investigation. This study highlights the significance of exploring the impact of diet on gut microbiota and the underlying mechanisms in the gut-liver and gut-brain axis. Nevertheless, limited research on the gut-liver axis poses challenges in comprehending the intricate connections between diet and the gut-brain axis. This underscores the need for comprehensive studies to elucidate the intricate gut-brain mechanisms underlying intestinal health and microbiota.

인간 대장상피세포 밀착연접 형성과정에서 NQO1 저해 효과 (Inhibition of NAD(P)H:Quinone Oxidoreductase 1 by Dicumarol Reduces Tight Junction in Human Colonic Epithelial Cells)

  • 본 연구는 최근 연구자 등이 보고한 NQO1 knockout (결핍) 생쥐의 점막기능 감소 및 장염유발 현상을 인간 대장상피세포에서 재확인하는 것이다. 이를 위해, 사람 대장상피세포주인 HT29 세포에 NQO1 억제제인 dicumarol을 처치한 다음 밀착연접 조절인자의 변화를 평가하였다. HT29 세포에 10 μM dicumarol을 처치하여 NQO1을 억제하면 인간 대장상피세포의 밀착연접이 유의하게 줄고 구성인자들(ZO1, occludin)의 단백질 양도 감소함을 확인하였다. 생쥐 대장 내강에 dicumarol (10 μM)을 직접 처치한 결과 점막 투과율이 현저하게 증가함도 확인하였다(장벽기능 감소). 이는 인간 대장상피세포의 밀착연접 형성과정이 NQO1에 의존적임을 보여준다. 그러나 dicumarol 처치는 ZO1과 occludin의 전사량을 억제하지 않았다. NOQ1 결핍 생쥐에서 ZO1과 occludin의 전사량이 크게 감소한다는 이전 보고를 감안하면, NQO1에 의한 밀착연접 단백질의 양적 조절 과정이 전사를 포함하는 다양한 경로와 연관되어 있음을 시사한다. 즉, NQO1에 의한 ZO1과 occludin 조절 과정에 프로테오좀 의존-단백질 변성과 같은 단백질 파괴 경로가 이용될 수 있다고 사료된다.

Differential expression and localization of tight junction proteins in the goat epididymis

  • The blood-epididymis barrier (BEB) forms a unique microenvironment that is crucial for the maturation, protection, transport, and storage of spermatozoa in the epididymis. To characterize the function of tight junctions (TJs), which are constitutive components of the BEB, we determined the expression and localization of TJ proteins such as zonula occludens (ZO)-1, 2, and 3, occludin, and claudin3 (Cldn3) during postnatal development in the goat epididymis. To assess the expression patterns of TJ proteins in immature (3 months of age) and mature (14 months of age) goat epididymides, two different experimental methods were used including immunofluorescence labeling and western blotting. We show that, ZO-1, 2, and 3, and occludin, were strictly expressed and localized to the TJs of the goat epididymis, whereas Cldn3 was present in basolateral membranes as well as TJs. All TJ proteins examined were more highly expressed in the immature epididymis compared to levels in mature tissue. In conclusion, our study indicates that at least five TJ proteins, namely ZO-1, ZO-2, ZO-3, occludin, and Cldn3, are present in TJs, and the expression strength and pattern of TJ proteins tend to be age dependent in the goat epididymis. Together, these data suggest that the distinct expression patterns of TJ proteins are essential for regulating components of the luminal contents in the epididymal epithelium and for forming adequate luminal conditions that are necessary for the maturation, protection, transport, and storage of spermatozoa in the goat epididymis.

Traf4 is required for tight junction complex during mouse blastocyst formation

  • Traf4 (Tumor necrosis factor Receptor Associated Factor 4) is a member of the tumor necrosis factor receptor (TNFR) - associated factors (TRAFs) family. TRAF4 is overexpressed in tumor cells such as breast cancer and associated with cytoskeleton and membrane fraction. Interestingly, TRAF4 was localized with tight junctions (TJs) proteins including OCLN and TJP1 in mammary epithelial cells. However, the expression patterns and biological function of Traf4 were not examined in preimplantation mouse embryos although Traf4-deficient mouse showed embryonic lethality or various dramatic malformation. In this study, we examined the temporal and spatial expression patterns of mouse Traf4 during preimplantation development by qRT-PCR and immunostaining, and its biological function by using siRNA injection. We found upregulation of Traf4 from the 8-cell stage onwards and apical region of cell - cell contact sites at morula and blastocyst embryos. Moreover, Traf4 knockdown led to defective TJs without alteration of genes associated with TJ assembly but elevated p21 expression at the KD morula. Taken together, Traf4 is required for TJs assembly and cell proliferation during morula to blastocyst transition.

트레드밀 운동 및 환경강화가 알츠하이머 질환 동물 모델의 인지기능, 근 기능 및 밀착연접 단백질 수준에 미치는 영향 (The Effect of Treadmill Exercise and Environmental Enrichment on Cognitive Function, Muscle Function, and Levels of tight junction protein in an Alzheimer's Disease Animal Model)

  • 본 연구의 목적은 알츠하이머질환(Alzheimer's disease: AD) 동물 모델을 대상으로 트레드밀 운동(Treadmill exercise: TE)과 환경강화(environmental enrichment: EE) 처치가 인지기능, 근 기능, 및 밀착연접 단백질 발현에 미치는 영향을 확인하는데 있다. AD 동물 모델을 제작하기 위해 aluminum chloride(AlCl3)를 90일간(40mg/kg/하루) 투여 하였으며 동시에 TE(10-12m/min, 40-60min/day) 혹은 EE에 노출시켰다. 그 결과 AlCl3 투여에 의한 인지기능 저하와 근 기능 감소가 TE와 EE에 의해 완화된 것으로 나타났다. 또한, TE와 EE는 AD 질환에서 나타나는 β-amyloid(Aβ), alpha-synuclein 및 tumor necrosis factor-α(TNF-α) 단백질의 발현 증가를 감소시킨 것으로 나타났다. 게다가 TE와 EE는 AlCl3 투여에 의해 감소된 밀착연접 단백질(Occludin, Claudin-5 및 ZO-1)의 발현을 통계적으로 유의하게 증가시킨 것으로 나타났다. 마지막으로 Aβ 단백질과 밀착연접 단백질과의 상관분석을 실시한 결과 부적 상관관계(Occludin: r=-0.853, p=0.001; Claudin-5 : r=-0.352, p=0.915; ZO-1 : r=-0.424, p=0.0390)로 나타났다. 따라서 이를 종합해 보면 TE 혹은 EE 처치는 AD에 나타나는 병리학적 특징들을 일부 완화시켜 인지기능과 근 기능을 일부 개선 시킬 수 있는 효과적인 운동 방법이라고 생각된다.
