Effects of Dietary Fat Level on the Aging Process of the Fibroblast Cells and Immune Function in Rats of Different Ages (식이지방수준이 나이에 따른 흰쥐 섬유아세포의 노화과정 및 면역반응에 미치는 영향)

  • To investigate the influence of age and dietary fat level on the immune function and the growing potential of the fibroblast cells, male rats of 2 month, 6 month and 30 month of age were fed either 6% or 30% fat diet for 16 weeks. The weight of thymus decreased linearly with increasing age. And this age-dependent degeneration of thymus was delayed in rats fed low fat diets. The blastogenesis of spleen lymphocytes to PHA, ConA, and PWM was decreased with increasing age, however, no effect of dietary fat level was observed. The age-related decline in ratios of PHA/ConA response may suggest that T suppressor cell activity increases with age. In cell culture system, lung fibroblast cells from 30M rats showed lower plating efficiency. longer doubling time. and shorter cumulative doubling potential than those from 2M or 6M animals. Also. the morphology of fibroblasts from 30M rats was tended to be rouned rather than flattened and more variable in size and being generally larger. wherease those from 2M and 6M rats were uniform in size and adhered tightly to the culture vessel in ordered arrays. Therefore fibroblast cell culture system tried in this study reflects the changes of cellular aging.

Preventive Effects of Daekumeumja on Fatty Degeneration of Liver and Immunosuppression Induced by Alcohol (대금음자(對金飮子)가 알코올 투여로 유발된 흰쥐의 간 지방 병증과 면역억제의 예방에 미치는 영향)

  • Objective : The present study has been undertaken to investigate the preventive effects of Daekumeumja on fatty degeneration of liver and immunosuppression induced by alcohol in rats. Method : Except for the normal group, we fed rat on 25% alcohol for 55 days. And Daekumeumja(DK) extract was administrated for the same period. We measured the serum component in rat's blood, weight of internal organs, liver triglyceride contents, histomorphometry and histopathological observation of internal organs. Results : 1. In the change measurement of serum components, DK group(50 mg/kg, 200 mg/kg) showed significant decrease of AST, ALT, albumin, ALP and triglyceride in comparison with those of the alcohol control group. 2. In the change measurement of internal organ's weight, DK group(50 mg/kg, 200 mg/kg) showed significant increase of relative body weights of liver, thymus and spleen in comparison with those of the alcohol control group. 3. DK group(50 mg/kg, 200 mg/kg) showed significant decrease of hepatic triglyceride contents in comparison with those of the alcohol control group. 4. In histomorphometrical changes of liver, DK group(50 mg/kg, 200 mg/kg) showed significant decrease of numbers of hepatocytes occupied by over 10% lipid droplets, percentages of regions occupied by lipid droplets and mean diameters of hepatocytes in comparison with those of the alcohol control group. In histomorphometrical changes of thymus, DK group(50 mg/kg, 200 mg/kg) showed significant increase of lobular thickness and cortex thickness in comparison with those of the alcohol control group. In histomorphometrical changes of spleen, DK group(50 mg/kg, 200 mg/kg) showed significant increase of splenic thicknesses, numbers of white pulps and mean diameters of white pulps in comparison with those of the alcohol control group. 5. In histopathological changes of liver, DK group(50 mg/kg, 200 mg/kg) showed effective inhibition of severe fatty changes in comparison with those of the alcohol control group. In histopathological changes of thymus and spleen, DK group(50 mg/kg, 200 mg/kg) showed effective inhibition of atrophic changes in comparison with those of the alcohol control group. Conclusions : Reviewing these experimental results, it appears that Daekumeumja have pharmaceutical preventive efficacy on fatty degeneration of liver and immunosuppression induced by alcohol in rats.

Effects of Akebia quinata extract on alcohol-induced damage of liver, spleen and thymus in rats (목통(木通)이 알코올 투여로 유발된 흰쥐의 간, 비장 및 가슴샘의 손상에 미치는 영향)

  • Objective : This study was conducted to investigate the effects of Effects of Akebia quinata (AQ) extract on alcohol-induced damage of liver, spleen and thymus in rats. Method : Experimental animals were divided in to 4 groups; Normal group, Alcohol group, AQ50 group and AQ200 group. All rats, except for Normal group, were fed 25 % ethanol for 55 days. During experimental period, Normal group and Alcohol group were administrated saline, and AQ50 group and AQ200 group were administrated AQ extract at dose of 50 and 200 mg/kg/day, respectively. We measured organ weight, liver triglyceride contents, and alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and triglycerid levels in serum. Also, we conducted histomorphometry and histopathological observation of liver, spleen and thymus. Results : AQ significantly decreased the level of AST, ALT and triglyceride in serum and the liver triglyceride contents induced by ethanol. Also, AQ significantly inhibited lipid droplets accumulation in hepatocytes. The decreased relative organ weight of spleen and thymus by ethanol were increased by AQ administration. In histopathological analysis of spleen, the rats administrated AQ 200 mg/kg presented significantly increased mean diameters of white pulps, numbers of white pulps and splenic thicknesses. The administration of 200 mg/kg AQ improved decreased lobular thickness and cortex thickness of thymus, which were decreased by ethanol. Conclusions : The results of present study indicated that AQ has an ameliorating effect for fatty degeneration of liver and damage of spleen and thymus.

Subacute Toxicity of DA-125, A New Anthracycline Anticancer Agent in Rats (새로운 Anthracycline 항암제 DA-125의 랫드에서의 아급성독성연구)

  • DA-125, a new anthracycline antitumor antibiotic, was administered to Sprague-Dawley rats intravenously for 4 weeks to investigate the repeated dose toxicity Focal alopecia was noted in three female rats receiving 1.0mg/kg/day. In rats receiving 1.0 mg/kg/day, weight gain decreased in both sexes after first or second week. Hematological examination revealed lower counts of total leukocyte and increased numbers of platelet after second week. At terminal necropsy, atrophy of thymus and spleen was observed. Lymphocytic depletion of thymus and atrophy of white pulp in spleen were observed microscopically. A decrease in the number of hematopoietic cells in the bone marrow and degeneration of germinal epithelia in testes were also observed. These treatment-related effects were mainly confined to rats receiving 1.0 mg/kg/day. And toxic effects with microscopic changes were not observed in rats receiving 0.2 mg/kg/day or 0.04 mg/kg/day.

  • The subacute oral toxicity of 1-(4-methylpiperazinyl)-3-phenylisoquinoline (CWJ- a-5) was investigated in Sprague-Dawley (SD) rats. Five groups of 5 males and 5 females were orally administered at doses of 0, 37.5, 75, 150 and 200 mg/kg with CWJ-a-5 for 2 weeks. In clinical signs, Salivation was observed in the 75, 150 and 500 mg/kg male and female groups. Loss of fur was observed in the 500 mg/kg male and female group. Body weight were significantly decreased in the 150 and 500 mg/kg male groups and in the 500 mg/kg female group. Food consumption was significantly decreased in the 300 mg/kg male group. In serum biochemistry, total cholesterol and phospholipid were significantly increased in 500 mg/kg male and female group. Aspartate aminotransferase was significantly increased in the 500 mg/kg female group. In histopathological examination, vacuolar degeneration of renal tubules in the kidney, vacuolar degeneration of hepatocytes in the liver vacuolar degeneration of myocytes in the heart, vacuolar degeneration of histiocytes in the spleen and thymus, atrophy of seminiferous tubule and degeneration of germinal epithelium in the testis, vacuolar degeneration of corpus luteum, granulosa cell and theca cell in the ovary were observed in the 150 and 500 mg/kg male and female groups. Based on these results, the no observed adverse effect level (NOAEL) with CWJ-a-5 was considered to be 75 mg/kg and the absolute toxic dose was considered to be 150 mg/kg in this study

Single Dose Intravenous Toxicity Study of A New Anthracycline Anticancer Agent (DA-125) in Rats and Mice (새로운 안트라사이클린계 항암제 DA-125의 랫드 및 마우스에서의 정맥투여 급성 독성시험)

  • This Study was conducted to assess the single dose toxicity of DA-125, a new anthracycline anti-cancer agent, in rats and mice. The Drug was administered once intravenously to both sexes of rats and mice. Then followed a 14-day period of observation. The $LD_{50}$ Values (95% confidence limit) were estimated to be 60.9 mg/kg (57.5~64.3 mg/kg) for male rats and 60.2 mg/kg (56.2~64.5 mg/kg) for female rats, and 85.8 mg/kg (81.0~90.9 mg/kg) for male mice and 84.5 mg/kg (78.2~91.9 mg/kg) for female mice. Both sexes of rats and mice given the drug revealed the clinical sign of decreased locomotor activity, emaciation, hair loss, red-dish brown urine, salivation, and watery diarrhea. In addition, body weight from the next day to the 7th day tended to be decreased slightly in rats and mice treated with DA-125. Death occurred from the next day after administration to the 12th day. Macroscopically, congestion of gastrointestinal organ, lung, and adrenal glands were found in both sexes on the dead rats and mice. Histopathological examination of dead rats manifested atrophy of spleen, hypoplasia of bone marrow, hypcplasia and necrosis of lymphocyte in thymus, atrophy of villi in small intestine (duodenum, jejunum, and ileum), hyperplasia of granular epithelium in small intestine, degeneration of germinal epithelium in testis, defer oration of tubular epithelium in kidney, and vacuolation and myolysis of myocardium in heart. Histopathological examination of dead mice revealed hypoplasia of spleen and mesenteric lymph node, local necrosis of liver, atrophy of villi in small intestine, hyperplasia of glandular epithelium in small and large intestine, degeneration of tubular in kidney, degeneration of germinal cells in testis, and slight vacuolar degeneration of myocardium in heart.

Four-Week Repeated-Dose Toxicity Studies of Hyrubicin ID6105, a Novel Anthracycline Anticancer Agent, in Rats (랫드에서 새로운 Anthracycline계 항암제 Hyrubicin ID6105에 대한 4주 반복투여 독성연구)

  • Repeated-dose toxicity of hyrubicin ID6105, a novel anthrarycline anticancer agent, was investigated in Sprague-Dawley rats. ID6105 was injected intravenously to rats at dose levels of 0.04, 0.2 or 1.0 mg/kg/day for 4 week. As a result, there were no dose-related mortality and specific clinical signs of all animals treated with the drug. However body weight gain of both male and female rats treated with a high dose (l.0 mg/kg/day) of ID6105 significantly decreased compared to control. Interestingly, the numbers of RBC and platelets, and concentration of hemoglobin remarkably increased, while protein synthesis was suppressed, which may be related to the atrophy of spleen, thymus and liver. Moreover there were severe lymphocytic depletion in spleen and thymus as well as decrease in the number of hematopoietic cells in bone marrow. Also, degeneration of cardiac muscles and testicular germinal epithelia were observed. Taken together, it is suggested that Long-term administration of ID6105 at high doses over 0.2 mg/kg/day might cause hematopoietic and male reproductive system injuries, in addition to hepatic dysfunction.

Immunohistochemistry and RT-PCR for pathogenesis of Newcastle disease in chickens (닭 뉴캣슬병의 발병기전 규명을 위한 RT-PCR 및 면역조직화학적 연구)

  • The present experiment was carried out to study the pathogenesis of Newcastle disease(ND), ND virus (NDV) antigens and genes in various organs from NDV inoculated chickens were detected by immunohistochemistry and RT-PCR. Immunohistochemically, NDV antigens were detected in the spleen, thymus, cecal tonsil, proventriculus, trachea and lungs at 12 hour post-inoculation (hpi). Viral antigens were localized mainly in the cytoplasm of lymphocytes and macrophages. After 48 hpi, clinical findings of the affected chickens were open-mouth breathing, conjunctivitis, watery diarrhea and edema around the eye and neck. After 72 hpi, chickens showed muscular tremor, paralysis of the legs and wings, and coma. Histopathological results consist of multi-focal necrosis with hemorrhages in lymphoid aggregates of the intestinal tracts, necrosis of the lymphoid tissues, neuronal degeneration and necrosis, and perivascular cuffing. Using RT-PCR, virus genes were detected in the spleen and proventriculus at 48 hpi, and in the brain at 60 hpi.

Effects of Omahwan(OMH) on CD4+, CD8+ T cell and Immune Organ Index in Rat (오마환(烏麻丸)이 백서(白鼠)의 혈액내(血液內) CD4+, CD8+ T cell 및 면역기관(免疫器官) 장기지수(臟器指數)에 미치는 영향(影響))

  • OMH which is known for its properties of recruiting vitality, is composed of Polygonum multiflorum Thunb. and Sesamum indicum DC.. This formula is known to possess the properties of recruiting vitality, blackening white hair and expanding life span. 16-week-old SD-Rats were treated with OMH for 16 days. After 24 hours, the rats were treated with MTX(Mthotrexate is oral administrated for 4 days(1mg/kg/day), in order to lower immunity. Then, These rats were classified in to groups, the N-16 group(not specially tested), the Control group(MTX), the OMH-L group(2.5% OMH+MTX)&the OMH-H group(10% OMH+MTX) and 6 rats were assigned to each group. After 18 hours from MTX treatment the organ index of the rats from each group Thymus and Spleen were measured. The percentage of CD+4, CD8+ T cell were measured and compared by flow cytometer. 1) Rats from the OHM-L&OMH-H group showed higher organ indexes of the Thymus and Spleen compared to the rats from the Control Group. This proves that OMH possesses the properties to mitigate degenerations of immunity($F_{thymus}=20.162,\;F_{spleen}=5.882$, ANDVA, p<0.05). 2) The rats from the two OMH groups showed higher rates of CD4+ T cell counts compared to the control group(F=26.906, ANOVA, p<0.05). CD8+ T cell showed lower rates compared to the Control group, but showed no differences within the two OMH groups(F=1.254, ANOYA, p>0.05). CD4+/CD8+ showed higher rates in the two OMH groups compared to the control group which can be thought as a proof that OMH prevents depression of immune response(F=10.554, ANOVA, p<0.05). In this test 16 week-old rats were used, which can be considered as the middle and prime age of the human being. These rats were treated with OMH which ended up showing properties of mitigating degeneration of immune responses and maintaining T-cell rates within the blood. It was possible to study that OMH possesses the properties to increase immune responses.

Pathological study on rabbit haemorrhagic disease in young rabbits

  • We investigated the pathological changes in young rabbits which were experimentally infected with rabbit hemorrhagic disease virus (RHDV). Experimental infection of RHDV was carried out in both thymectomized and non-thymectomized young immature rabbits and adult rabbits. None of young rabbits infected with RHDV died during the experiment. Histologically, single or focal hepatocellular degeneration and necrosis with mild lymphocyte infiltration were observed in the rabbits killed at 30 hours and 5 days PI. Lymphocyte infiltration was more severe at 5 days PI than at 30 hours PI. RHDV antigens were mainly detected in the degenerating hepatocytes adjacent to the infiltrated lymphocytes at 30 hours PI and 5 days PI. In electron microscopical observation, infiltrated lymphocytes in the lesions had large nuclei without cytoplasmic granules and interdigitated with adjacent hepatocytes. It is assumed that infiltrated lymphocytes in hepatic lesions in RHDV infected young rabbits are T-lymphocytes and originate from peripheral lymphoid organs or tissues rather than from thymus.
