Effects of Fermented Milk with Hot Water Extract from Acanthopanax senticosus and Codonopsis lanceolata on the Immune Status of Mouse (가시오가피와 더덕 추출물을 첨가한 발효유가 마우스의 면역기능에 미치는 영향)

  • We investigated the immunomodulatory actions of water extract from Acanthopanax senticosus in male ICR mice. The mice were treated with fermented milk containing three added doses of freeze dried extract: 3 mg/kg (A), 9 mg/kg (B), and 27 mg/kg (C) of body weight with Acanthopanax senticosus: Codonopsis lanceolata (8:2) for 7 and 10 weeks, respectively. Organ weights, plaque-forming cell tests, agglutination tests, IgG tests, differential white cell counts, and histological tests were performed at the 7th and 10th weeks of dietary treatment. There were no significant differences in body weight and organ weight. The spleen indices of group B at 7 weeks and group C at 10 weeks were significantly higher than those of the control group (p<0.05). For the plaque-forming cell test, groups B and C at 7 weeks, and group C at 10 weeks, showed significant increases over the control group (p<0.05). The agglutination test decreased with an extended experimental period. Groups A, B, and C at 7 weeks, and groups B and C at 10 weeks, had greater antibody responses to sheep red blood cells (SRBC) than the control group. The IgG antibody production of group C at 7 weeks and groups B and C at 10 weeks were significantly higher than the control group (p<0.05). In groups B and C, lymphocyte percentage was higher than the control group, and their spleen and thymus tissues showed active immune reactions.

Immunobiological Studies on Doses of Methanol Extract of Astragali Radix (황기의 메탄올 추출물의 용량에 따른 면역생물학적 연구)

  • Effects of methanol extract of Astragali Radix (AR) on the immune responses were studied using ICR mice. Mice were divided into 4 groups (10mice/group), and methanol extracts of AR at doses of 0.05, 0.25 and 1.25g/kg were orally administered to ICR mice once a day for 2 weeks. Mice were immunized and challenged with sheep red blood cells (SRBC). The results of this study were summarized as follows; (1) Methanol extract of AR at 0.05, 0.25 and 1.25g/kg didn't affect the weight ratios of thymus to body, as compared with those in controls, but significantly increased spleen weight ratio. (2)Methanol extract of AR at 0.05 and 0.25g/kg significantly increased hemagglutination titer and splenic plaque forming cells corresponding to humoral immunity, as compared with those in controls, but their enhancements were somewhat lowered at a high dose (1.25g/kg). (3) Methanol extract of AR at 0.05 and 0.25g/kg siginificantly increased delayed-type hypersensitivity reaction resulted from cell-mediated immunity, as compared with those in controls, but not so significant increases were observed at a high dose (1.25g/kg). (4) Methanol extract of AR at 0.05 and 0.25g/kg significantly increased phagocytic activity and the number of circulating leukocyte compared with those in controls, but their enhancements were lowered at a high dose (1.25g/kg). These results suggest that methanol extract of Astragali Radix increased humoral and cell-mediated immune responses, phagocytic activity and the number of circulating leukocyte, dependent upon dose, but inhibited their enhancement effects were decreased at a high dose (1.25g/kg).

A Study on the Subchronic Inhalation Toxicity of 2-Butanethiol Using SD Rats (2-부탄티올의 SD Rats를 이용한 아만성 흡입독성연구)

  • With the 2-Butanethiol, which is an unidentified inhalation toxic material, acute inhalation toxicity was tested with SD rats. The $LC_{50}$ was evaluated to be 2,500 ppm (9.22 mg/L) or higher which falls under the criteria of acute toxicity Category 3 (500<$LC_{50}$<2,500 ppm) in the Industrial Safety and Health Act. In the subchronical inhalation toxicity test by 0, 25, 100, and 400 ppm, 6 hours a day, 5 days a week, for 13 weeks repeated exposure, though no death or particular clinical presentation was observed, in the female 25 and 400 ppm group, including weight change, and in each concentration group including 400 ppm, change of feed rate, eye stimulation, motility change in male group, and lesions in blood and blood biochemical were observed. In the internal organs weight, 25, 100, and 400 ppm groups in male and 400 ppm group in female showed significant (p<0.05) changes in kidney, liver, thymus, and lung. In the pathological tissue test, severe cortical tubular hyaline droplets were observed in the male 400 ppm group, and all male rats of 400 ppm group and 2 female individuals showed tubular degeneration/regeneration accompanied with pigmentation, showing that the target organs of inhalation exposure of 2-Butanethiol are spleen, kidney, nasal cavity, and adrenal. Through the tests, the NOEL of 2-Butanethiol was evaluated to be 25 ppm (0.092 mg/L) or less for both male and female.

Effect of Eucommia ulmoides Extracts on Allergic Contact Dermatitis and Oxidative Damage Induced by Repeat Elicitation of DNCB (두충 추출물이 DNCB로 유발된 알레르기성 접촉피부염과 산화적 손상에 미치는 영향)

  • Inhibitory effects of allergic contact dermatitis of hot water extract of Duchung (Eucommia ulmoides Oliver) leaf, bark and stem growing at Sancheong-gun were investigated for female BALB/c mouse induced by repeat elicitation of DNCB (2,4-dinitro-chlorobenzene). Skin reactions, consisting of increased ear thickness and the presence of ear inflammation, were observed in mice treated with DNCB and Duchung. Weight of lymph node, spleen and thymus in mice treated with Duchung extracts were lower than that of mouse treated with DNCB. Ear weight of mouse treated with Duchung extracts was decreased by increasing the concentration of sample as compared to control group and dropped as low as control level at 1,000 mg/kg. Ear thickness became thinner as test time on Duchung extract progressed. MDA (malondialdehyde) contents in liver tissue were not different in sample group with DNCB group, but were different in ear tissue. NO (nitric oxide) contents was decreased in Duchung extract groups at serum and ear tissues as compared to 1% DNCB group. In the present study, the results suggested that Duchung extract inhibits inflammatory response and oxidative damage induced DNCB allergen.

Effects of Methionine Diets on the Immunotoxicity of Ethanol in ICR Mice (생쥐에 있어서 에탄올의 면역독성에 미치는 메치오닌 식이의 영향)

  • Experiments were performed on mice to investigate the effect of methionine diets (MET) on the immunotoxicity of ethanol. ICR female mice were divided into 5 groups, Met (Basal (B)+0.19% methionine(M), B+1.71% M and B+5.13%W) and ethanol(4%) were administered ad libitum for 21 days. The mice were evaluated for changes in immune status as measured by antibody titer, Arthus reaction, delayed type hypersensitivity (DTH), rosette forming cell(RFC) and plaque forming cell (PFC) to sheep red blood cells (S-RBC). To investigate the change of the non-specific immune response, the number of leukocytes in peripheral blood and phagocyte activity were measured. The results were summarized as follows: (1) The weight ratios of spleen and thymus to body weight were significantly increased by the B+0.19% M, B+0.57% M and B+1.71% M groups in comparison with control group(B), but B+5.13% M group was significantly decreased. (2) Humoral immune responses were significantly increased by the B+0.19% M and B+0.57% M groups in comparison with control group, but B+5.13% M group was significantly decreased. (3) Cellular immune responses were significantly decreased by the B+1.71% M and B+5.13% M groups in comparison with control group. (4) Phagocyte activities were significantly increased by the B+0.19% M, B+0.57% M and B+1.71% M groups in comparison with control groups, but B+5.13% M group was significantly decreased. (5) The number of circulating leukocyte was significantly increased in the B+0.19% M and B+0.57% M groups in comparison with control group, but B+5.13% M group was significantly decreased.

Antitumor and Immunologic Activity of Chitosan Extracted from Shell of Shrimp (새우 껍질에서 추출한 키토산의 항암 및 면역활성)

  • This study was designed to investigate the antitumor effect and immunological activities of chitosan extracted from Solenocera prominetis toward on mire bearing sarcoma-180. The growth inhibition ratio of the chitosan toward sarcoma-180 showed at the highest level of 63.84% when chitosan were administrated at the concentration of 40mg/kg. The direct cytotoxic effect of chitosan was not observed in the mice bearing sar-roma-180 in vitro. In the effect of immunological activities, dose-dependent responses indicated by the increase of leucocyte, peritoneal exudate cell than that of control group when chitosan administered to the mice in the concentation of 30mg/kg and 40mg/kg. Also dose-dependent responses showed also by the increase of immunoorgans weights such as body weight, liver, spleen or thymus in the same concentration of 30mg/kg and 40mg/kg. Food pad swelling having the relationship with arthus reaction of antibody-mediated hypersensitivity and delayed type hypersensitivity was recovered the almost normal level. In the efforts of macrophge on phagocytes, there were not substantial differences in phagorytic and corrected phagocytic index. In the number of plaque forming cell(PFC), PFC on the 10$^{7}$ spleen cells were increased the levels of 18.88% and 31.83% when chitosan were adminstersd at the concentration of 30mg/kg and 40mg/kg.

  • We have investigated the immunomodulatory activity of water extracts of Acanthopanax divaricatus var. alveofructus in male ICR mice. Mice were administrated with fermented milk containing freeze-dried extract 3 mg/Kg (A), 9 mg/kg (B), 27 mg/Kg (C) per body weight with A. divaricatus var. alveofructus (loots : leaves : stem) : Codonopsis lanceolata = (5 : 2 : 1.5) : 1.5 for 7 and 10 weeks, respectively. Body weight, relative organ weight, cellularity of lymphoid organs, plaque- forming cell (PFC) assay, agglutination (AGG) test and lymphoproliferation were examined in various groups of animals. Any significant differences of body weight gain were not recorded in the tested ICR mice. There was significant different (p<0.05) in the spleen index in B group of 10 weeks and C group of 7 weeks fed mouse. The thymus gain weight was increased during administration of the extract, but there was no significant increase on other organs gain. Humoral immunity as measured by PFC showed more decreased PFC level in 10 weeks than in 7 weeks. In the HT, A. divaricatus var. albeofructus extract also showed a significant increase (p<0.05) in C group of 10 weeks. Administration of extracts from A. divaricatus var. albeofructus increased significantly in the production of IgG antibodies on the mice immunized with SRBC in B group of 7 and 10 weeks (p<0.05).

Effect of Selenium-enriched Japanese Radish Sprouts and Rhodobacter capsulatus on the Cholesterol and Immune Response of Laying Hens

  • Immune response and yolk cholesterol are crucial factors for commercial chicken producers. The objectives of this study were to investigate the effect of selenium-enriched Japanese radish sprouts (Se-enriched JRS) and R. capsulatus synergistically on immune response and cholesterol in laying hens. A total of 50 laying hens (20-wk old) were assigned to 5 dietary treatment groups, and fed diets supplemented with 2.5 ${\mu}g/kg$, 5 ${\mu}g/kg$, 10 ${\mu}g/kg$ Se-enriched JRS and 5 ${\mu}g/kg$ Se-enriched JRS+R. capsulatus (0.02%). Egg production and yolk color were significantly improved by the supplementation of Se-enriched JRS+R. capsulatus in the layer diet (p<0.05). Compared to the control, serum cholesterol concentration and triglyceride levels were decreased by all the treatments (p<0.05). After 8-wk of the experiment, supplementation of 5 ${\mu}g/kg$, 10 ${\mu}g/kg$ and Se-enriched JRS+R. capsulatus significantly reduced yolk cholesterol and triglycerides, while the greatest reduction was observed when R. capsulatus was incorporated with Se-enriched JRS. Spleen, bursa and thymus weight were significantly increased by both the 5 ${\mu}g/kg$ and 10 ${\mu}g/kg$ Se-enriched JRS. Compared to the control, supplementation of 5 ${\mu}g/kg$ and 10 ${\mu}g/kg$ Se-enriched JRS significantly increased serum IgG and yolk IgY concentration and foot web index activity by Newcastle Disease Virus (p<0.05). After 4-wk and 8-wk of supplementation, the highest number of leukocytes was observed with Se-enriched JRS+R. capsulatus (p<0.05). The highest concentration of serum and yolk Se was found in Se-enriched JRS plus R. capsulatus treatment. Combined dietary supplementation of Se-enriched JRS and R. capsulatus might be beneficial for better health, disease protection and overall production performance.

  • The subacute oral toxicity of 1-(4-methylpiperazinyl)-3-phenylisoquinoline (CWJ- a-5) was investigated in Sprague-Dawley (SD) rats. Five groups of 5 males and 5 females were orally administered at doses of 0, 37.5, 75, 150 and 200 mg/kg with CWJ-a-5 for 2 weeks. In clinical signs, Salivation was observed in the 75, 150 and 500 mg/kg male and female groups. Loss of fur was observed in the 500 mg/kg male and female group. Body weight were significantly decreased in the 150 and 500 mg/kg male groups and in the 500 mg/kg female group. Food consumption was significantly decreased in the 300 mg/kg male group. In serum biochemistry, total cholesterol and phospholipid were significantly increased in 500 mg/kg male and female group. Aspartate aminotransferase was significantly increased in the 500 mg/kg female group. In histopathological examination, vacuolar degeneration of renal tubules in the kidney, vacuolar degeneration of hepatocytes in the liver vacuolar degeneration of myocytes in the heart, vacuolar degeneration of histiocytes in the spleen and thymus, atrophy of seminiferous tubule and degeneration of germinal epithelium in the testis, vacuolar degeneration of corpus luteum, granulosa cell and theca cell in the ovary were observed in the 150 and 500 mg/kg male and female groups. Based on these results, the no observed adverse effect level (NOAEL) with CWJ-a-5 was considered to be 75 mg/kg and the absolute toxic dose was considered to be 150 mg/kg in this study

Immune Stimulating Efficacy of Soluble β-1,3-glucans (수용성 β-1,3-glucans의 면역 활성 효능에 대한 연구)

  • Background: $\beta$-1,3-glucans are well known to enhance the immune reactions, resulting in antitumor, antibacterial, antiviral, anticoagulatory and wound healing activities. $\beta$-1, 3-glucans have various activities depending on molecular weight, degree of branching, conformation, water-solubility and intermolecular association. However, the $\beta$-1,3-glucan linked backbone structure is essential and $\beta$-D-glucopyranosyl units are required for immuno-potentiating activities. Result: In this study, we tested the immunophamacological activities of soluble $\beta$-1,3-glucans and confirmed the following activities: (1) $IFN-{\gamma}$ production in PBMCs in the presence or the absence of PHA, LPS, or IL-18; (2) induction of various cytokines in the spleen and thymus; (3) adjuvant effect on the antibody production; (4) nitrogen oxide synthesis in macrophages; (5) the cytotoxic and antitumor effects on cell lines and ICR mice. Conclusion: These results strongly suggested that $\beta$-1,3-glucans possessed various immuno-pharmacological activities.
