Three-dimensional analysis of the arrangement of microtubules of the outer segment in the ciliary-type photoreceptor cell in the Onchidium dorsal eye

  • The inverted retina of the Onchidium dorsal eye (DE) is composed only of ciliary-type photoreceptor cells (CC's). The outer segment (OS) of the CC is a concentric lamellar structure consisting of many modified ciliary membranes and stains positively with anti-$\beta$-tubulin antibody. Near the base of the OS there are about 30 basal bodies each connecting individually to a cilium. The cilia are rod-shaped at the base, progressing upwards to a flattened sheet-like shape with increasing surface area. Three-dimensional analysis on serial sections demonstrates the ladle-shape of a modified cilium. Many modified cilia wrap around each other like the leaves of a cabbage. Nine pairs of microtubules (MT's) are located regularly in a ring at the base of the cilium, gradually losing their regular arrangement towards the periphery, where they separate into two subgroups that are contained within two swollen portions of a modified cilium. Within the CC of the Onchidium DE, MT's in the modified cilium exist as two poles extending longitudinally in a thin expanded ciliary membrane. This arrangement may support the photoreceptive OS and serve to maintain its structural integrity.

Surfactant enhanced filtration performances of monochlorophenol isomers through low-pressure membrane

  • Membrane processes are major breakthrough for the removal of organic pollutants in water remediation. The separations of solutes depend on nature of the membranes and solutes. The separation performance depends on the nature of the solutes (i.e., molecular volume, polarity, and hydrophobicity) for the same membrane. As 4-chlorophenol is of more dipolemoment compared to 2-chlorophenol, the orientation of the molecule enables it pass through the pores of the membrane, which is of negatively charged and thus separation order follows: 2-chlorophenol > 4-chlorophenol. Hydrophobicity factor also supports the order. Addition of sodium dodecyl sulfate (SDS) to chlorophenol solution shows remarkable increase in separation performance of the membrane. The improvement in separation is 1.8 and 1.5 times for 4- and 2- chlorophenol consecutively in case of 0.0082 M SDS (1cmc = 0.0082 M) in the solution. 4-chlorophenol has better attachment tendency with SDS because of its relatively more hydrophobic nature and thus reflects in performance i.e. the separation performance of 4-chlorophenol with SDS through the membrane is better compared to 2-chlorophenol.

Effect of Cholesterol on the Phase Change of Lipid Membranes by Antimicrobial Peptides

  • Membrane disruption by an antimicrobial peptide (AMP) was investigated by measuring the $^2H$ solid-state nuclear magnetic resonance spectra of 1-palmitoyl-$d_{31}$-2-oleoyl-sn-glycero-3-phosphatidylcholine (POPC_$d_{31}$) in mixtures of POPC_$d_{31}$/cholesterol and either magainin 2 or aurein 3.3 deposited on thin cover-glass plates. The line shapes of the experimental $^2H$ solid-state nuclear magnetic resonance (SSNMR) spectra were best simulated by assuming the coexistence of a mosaic spread of bilayers containing pore structures and a fasttumbling isotropic phase or a hexagonal phase. Within a few days of incubation in a hydration chamber, an isotropic phase and a pore structure were induced by magainin 2, while in case of aurein 3.3 only an isotopic phase was induced in the presence of a bilayer phase. After an incubation period of over 100 days, alignment of the bilayers increased and the amount of the pore structure decreased in case of magainin 2. In contrast with magainin 2, aurein 3.3 induced a hexagonal phase at the peptide-to-lipid ratio of 1/20 and, interestingly, cholesterol was not found in the hexagonal phase induced by aurein 3.3. The experimental results indicate that magainin 2 is more effective in disrupting lipid bilayers containing cholesterol than aurein 3.3.

항균성 펩타이드인 magainin 2의 활성에 대한 고체 핵자기 공명 분광 연구 (A solid-state NMR study on the activity of an antimicrobial peptide, magainin 2)

  • 얇은 유리판 위에 정렬된 지질 이중막에 작용하는 항균성 펩타이드 magainin 2의 작용활성을 고체 핵자기 공명 분광법을 이용하여 조사하였다. 수화에 따른 지질 이중막의 파괴정도를 파악하기 위해 두 가지 수화 상태에서 실험이 이루어졌다. 95%의 상대 습도에서 수화된 지질 이중막에서는 약 15% 정도의 지질 이중막이 파괴되었으며, 완전히 수화된 지질 이중막에서는 약 20% 정도의 지질 이중막이 파괴되었다. 두 가지 시료 제조방법에 따른 지질 이중막 파괴 정도의 비교를 통해, 항균성 펩타이드의 지질 이중막 결합과정이 지질 이중막 파괴에 중요한 변수임을 알 수 있었다.

대두 사포닌의 활성에 관한 기전 연구

  • Saponins are glycosidic compounds present in many plant foods. They are characterized by their ability to lyse cell membranes due to their surface-active properties. Saponins are believed to interact primarily with cholesterol in the cell membrane. In this study, the interaction of soybean(SS) with cell membrane was investigated using erythrocytes as a model. Mechanisms of interaction was also investigated by measuring their binding capacity with different membrane lipid fractions. Throughout the study, gypsophilla saponin(GS) and quillaja saponin(QS) were used to evaluate the membranolytic activity of soybean saponins. All saponins released hemoglobin in a concentration-dependent manner. SS induced 40% hemolysis at the concentration of 400 ppm, however there was no increase in hemoglobin release above 400ppm concentration. 5ppm of GS and 8 ppm of QS hemolyzed 100% of erythrocytes. Isolation of SS fractions by thin layer chromatography revealed that only one non-polar saponin possesses strong hemolytic activity. When saponins were incubated decreased the release of cholesterol. When the hemolytic activity of saponins was measured in the presence of other major membrane lipid components, sphingomyelin significantly reduced the hemolytic activity of SS, while cholesterol reduced the activity of QS. GS showed high affinity to other component(s) in the incubation media as well as lipids. These results suggest that the membranolytic activity of saponins are related to their specific chemical structure, which determines the interaction behavior between saponins and different membrane components, and thereby influence the biological activity.

셀룰로오스 아세테이트 기반의 햅틱 액추에이터 (A New Haptic Actuator based on Cellulose Acetate)

  • This paper suggests a new film-type haptic actuator based on cellulose acetate electro-active paper. Conventional tiny haptic actuators in mobile devices can create vibrotactile sensation at only near resonant frequency. The strategy of operating near the resonant frequency, however, brought a new issue for creating vibrotactile sensation which can be strong enough to feel in arbitrary frequency. Another problem is that the size of the conventional actuator is not small enough to be embedded into slim mobile devices. In order to achieve these issues, we propose a thin and tiny actuator based on a cellulose acetate material charged with an electric potential. The motion of the actuator can be a concave or a convex by controlling a polarity of both charged membranes and the actuator performance can be modulated by increasing level of biased electric potential.

Functional Nannomaterials Based on Nanoporous Template

  • Nanoporous templates have been widely used for the development of new functional nanostructured materials suitable for electronics, optics, magnetism, and energy storage materials. We have prepared nanoporous templates by using thin films of mixtures of polystyrene-block-poly (methyl methacrylate) (PS-b-PMMA) and PMMA homopolymers. These templates have cylindrical nanoholes spanning the entire thickness of the film. Some applications of nanoporous templates are introduced: a) anti-reflective coating, b) the preparation of conducting polymer nanowires of poly (pyrrole), poly (3,4-ethylenedioxy-thiopene) onto a glass coated with indium-tin-oxide, and c) the separation membranes for biomaterials. We found that when the pore fraction of nanoholes in the film was ~0.68, almost zero reflectance at a specific wavelength, which can be changed with film thickness, was achieved at visible wavelengths Furthermore, ultra high density array of conducting nanowires was successfully prepared onto various substrates including flexible polymer. Due to highly alignment of polymer chain along the nanowire direction, the conductivity was much increased. Furthermore, these nanoporous films were found to be very effective for the separation of human Rhinovirus type 14 (HRV 14), major pathogen of a common cold in humans, from the buffer solution. We also found that when the pore size was effectively controlled down to 6 nm, a single file diffusion was observed.

Preparation and Characterization of New Immunoprotecting Membrane Coated with Amphiphilic Multiblock Copolymer

  • New immunoprotecting membranes were prepared by spin coating the amphiphilic random multiblock copolymers of poly(ethylene glycol) (PEG) and poly(tetramethylene ether glycol) (PTMEG) or poly(dimethyl siloxane) (PDMS) on porous Durapore(R) membrane. The copolymer coating was intended to make a biocompatible, immunoprotecting diffusional barrier and the supporting porous substrate was for mechanical stability and processability. By filling Durapore(R) membrane pores with water, the penetration of coating solution into the pores was minimized during the spin coating process. A single coating process produced a completely covered thin surface layer (~1 ${\mu}{\textrm}{m}$ in thickness) on the porous substrate membrane. The permselectivity of the coated layer was influenced by PEG block length, polymer composition, and thickness of the coating layer. A composite membrane with the coating layer prepared with PEG 2 K/PTMEG 2 K block copolymer showed that its molecular weight cut-of fat any 40 based on dextran was close to the molecular size of IgG (Mw = 150 kDa). However, IgG permeation was detected from protein permeation test, while glucose oxidase (Mw = 186 kDa) was not permeable through the coated membrane.

  • There has been many attempts to improve the membrane performance using pervaporation processes[l-3]. They are 1) blending polymer with the high flux and one with high selectivity, 2) an incorporation of functional groups interacting with permeants into a membrane through copolymerization or modification, 3) composite membrane or asymmetric membrane structure with a thin skin layer which acts as a selective layer. Among them, a polymeric membrane containing ion complex group receives an extensive attention recently because ionic complex is known to activate the water transport through ion-dipole interaction. It is especially advantageous in the separation of organic-water system. We applied the ideas of the activation of water transport through ion-dipole. We have reported on the in-sire complex membrane to separate water from aqueous aceiic acid and pyridme solution[4-5] based on the simple acid-base theory. Water transport was enhanced through in-situ complex between pyridine moiety in the membrane and the incoming acetic acid in the feed. In this case, catalytic transport mechanism was proposed. In the present study we used pyridine solution as a feed and the sulfonic acid group in the membrane.

  • Monolayers of lipids on a water surface have attracted much interest as models of biological membranes, but also as precursors of multilayer systems promising many technical applications. Until now, many methodologies have been developed in order to gain a better understand. Photoisomerization in monolayers of a novel azobenzene compound, azobenzene dendrimer, was investigated for the first time by means of the absorption spectrum and Maxwell displacement current(MDC) technique. Dendrimers are well-defined macromolecules exhibiting a tree-like structure, first derived by the cascade molecule approach According to the absorption spectrum, trans-to-cis conversion ratio was estimated to the third generation of azobenzene dendrimer deposited onto a glass substrate. Temperature-dependent induced charge with trans-cis isomerization was also measured by means of MDC technique.
