Structural stability of CD1 domain of human mitotic checkpoint serine/threonine-protein kinase, Bub1

  • Bub1 is one of the spindle checkpoint proteins and plays a role in recruitment of the related proteins to kinetochore. Here, we studied the structural characteristic of the evolutionarily conserved 160 amino acid region in the N-terminus (hBub1 CD1), using Circular Dichroism (CD) and NMR. Our CD results showed that hBub1 CD1 is a highly helical protein and its structure was affected by pH: as pH was elevated to basic pH, the helical propensity increased. This could be related to the surface charge of the hBub1 CD1. However, the structural change did not largely depend on the salt concentration, though the thermal stability a little increased. The previous NMR analysis revealed that the hBub1 CD1 adopts eight helices, which is consistent with the CD result. Our result would be helpful for evaluating the molecular mechanism of the hBub1 CD1 and protein-protein interactions.

Synthesis and Characterization of New Liquid Crystalline Fumarate and Maleate Monomers with Two Symmetrical Mesogens

  • 4-Hydroxy-4'-methoxyazobenzene and 4-hydroxy-4'-cyanoazobenzene were synthesized from phenol with p-anisidine and p-aminobenzonitrile through a diazotization reaction, respectively. They were reacted with 2-chloroethanol, 2-(2-chloroethoxy)ethanol, or 2-[2-(2-chloroethoxy)ethoxy]ethanol to produce six kinds of new mesogenic alcohols having an azobenzene group that is sensitive to the ultraviolet. Twelve kinds of new photoresponsive monomers with two symmetrical mesogens were prepared by the reaction of the mesogenic alcohols with fumaric acid or maleic acid through a Mitsunobu reaction. The resulting monomers have different length of flexible ethyleneoxy spacer tethered to azobenzene group. The length of the spacer affected their thermal stability, solubility, and phase transition temperature. Structures of the monomers were identified by FT-IR and ¹H-NMR spectra. Their phase transition temperatures and thermal stability were also investigated by a differential scanning calorimetry (DSC) and a thermogravimetric analysis (TGA). From an optical polarizing microscopy, all the prepared monomers except fumarate-1 and maleate-1 were found to show enantiotropic liquid crystallinity with a smectic texture like focal-conic, fan-shaped, and batonnet textures.

Biochemical and Thermal Stabilization Parameters of Polygalacturonase from Erwinia carotovora subsp. carotovora BR1

  • With an emphasis on its thermal behavior with different pHs and salts, the kinetic and thermodynamic parameters of the purified polygalacturonase (PG) from E. carotovora subsp. carotovora (Ecc) BR1 were studied, as the characterization of an enzyme is significant in the context of burgeoning biotechnological applications. The thermodynamic parameters for polygalacturonic acid hydrolysis by the purified PG were ${\Delta}H^*$=7.98 kJ/mol, ${\Delta}G^*$=68.86 kJ/mol, ${\Delta}S^*$=-194.48 J/mol/K, ${\Delta}G_{E-S}$=-1.04 kJ/mol, and ${\Delta}G_{E-T}$=-8.96 kJ/mol. In addition, its turnover number ($k_{cat}$) was 21/sec. The purified PG was stable within a temperature range of $20-50^{\circ}C$ and was deactivated at $60^{\circ}C$ and $70^{\circ}C$. The thermodynamic parameters (${\Delta}H^*$, ${\Delta}G^*$, ${\Delta}S^*$) for the irreversible inactivation of the PG at different temperatures ($30-60^{\circ}C$) were determined, where the effectiveness of various salts and different pHs (4-8) for the thermal stability of the PG were also characterized. The efficacy of various salts for the thermal stability of the PG was in the following order: $MgCl_2$ > $BaCl_2$ > KCl > $CaCl_2$ >NaCl. Therefore, the present work presents the biochemical, substrate hydrolysis thermodynamics and the thermal stabilization parameters of the PG from Ecc.

실리카 베이스 무기 바인더 기반의 TiO2 코팅액의 제조 및 특성 평가 (Fabrication and characteristics of TiO2 coating solution with silica-based inorganic binder)

  • 자동화 시스템이 일반화되면서 제품 관리를 위한 라벨지(label)의 수요는 증가하고 있으며, 다양한 환경에서 사용할 수 있는 기능성 라벨지 개발이 빠르게 진행되고 있다. 인쇄회로기판의 경우 제작 과정에서 $300^{\circ}C$ 이상의 리플로우 솔더링 공정과 여러 차례의 세정 공정을 거치기 때문에 열적 화학적 안정성을 갖는 바코드 라벨지(barcode label)가 사용되고 있으나 황변(yellowing) 현상 발생으로 인한 인식률 저하의 문제가 발생하고 있다. 본 연구에서는 실리카 무기 바인더와 이산화티탄 백색안료를 사용한 복합 코팅층을 개발하고, 열적 화학적 안정성을 확보한 기능성 라벨지 연구를 진행하였다. 졸-겔 공정으로 제조한 실리카 무기 바인더는 기재(substrate)로 사용하는 폴리이미드 필름과 우수한 밀착성과 내마모성 특성을 갖는 것으로 확인하였다. 또한 이산화티탄 백색안료와 혼합하여 폴리이미드 필름에 백색의 코팅층을 제조할 수 있었으며, 복합 코팅층은 $400^{\circ}C$ 이상의 고온에서도 우수한 백색도와 광택도를 특성을 유지하는 것을 알 수 있었다. 또한 산성(pH 1.6)과 염기성(pH 13.6) 세정제를 통한 화학 처리 후에도 백색도와 광택도 변화가 일어나지 않는 우수한 화학적 안정성을 확인하였다.

Improved Thermal Stability of a Novel Acidophilic Phytase

  • Phytase increases the availability of phosphate and trace elements by hydrolyzing the phosphomonoester bond in phytate present in animal feed. It is also an important enzyme from an environmental perspective because it not only promotes the growth of livestocks but also prevents phosphorus contamination released into the environment. Here we present a novel phytase derived from Turicimonas muris, TmPhy, which has distinctive structure and properties compared to other previously known phytases. TmPhy gene expressed in the Pichia system was confirmed to be 41 kDa in size and was used in purified form to evaluate optimal conditions for maximum activity. TmPhy has a dual optimum pH at pH3 and pH6.8 and exhibited the highest activity at 70℃. However, the heat tolerance of the wildtype was not satisfactory for feed application. Therefore, random mutation, disulfide bond introduction, and N-terminal mutation were performed to improve the thermostability of the TmPhy. Random mutation resulted in TmPhyM with about 45% improvement in stability at 60℃. Through further improvements, a total of three mutants were screened and their heat tolerance was evaluated. As a result, we obtained TmPhyMD1 with 46.5% residual activity, TmPhyMD2 with 74.1%, and TmPhyMD3 with 66.8% at 80℃ heat treatment without significant loss of or with increased activity.

  • The effects of pH, types of applied current, agitation method and time, additive on the amount of co-deposited $TiO_2$ particles in the matrix were investigated. The deposition rates increased with increasing pH values, while the volume fraction of $TiO_2$ particles and the size of agglomerated $TiO_2$ particles in the composite decreased. The volume fraction of $TiO_2$ particles in the composite decreased when pulsed current of 50% duty cycle was used. And the size of agglomerated $TiO_2$ particles in the nickel matrix of pulsed current was smaller than that of DC current specimen. The volume fraction of $TiO_2$ particles in the matrix decreased with longer time by air agitation, but in case of using magnetic bar, volume fraction in the same range of time was relatively constant. The volume fraction of the electrodeposited Ni-$TiO_2$ composite in the solution containing 0.01 M Dimethylamine borane (DMAB) increased slightly with increasing agitation time regardless of agitation methods. Thermal stability of the electrodeposited Ni-$TiO_2$ composite increased with lower pH at the temperature range of $200{\sim}800^{\circ}C$, and the results showed that the amount of co-deposited $TiO_2$ relies more on the deposition rate than zetapotential of $TiO_2$ particles.

Structural Characterization of Growth-Related Translationally Controlled Tumor Protein P23

  • P23, a translationally controlled turner protein is involved in the interleukin-4 secretion from human basophils and is also known to be an IgE-dependent histamine-releasing factor. However, the precise physiological function and structure of P23 have not been elucidated. In the current study, we constructed the optimal expression and purification protocol of P23 and investigated the secondary structure and structural stability in various conditions. Circular dichroism (CD) investigation showed that the secondary structure of P23 adopts mainly a P-sheet conformation. CD spectroscopy and differential scanning calorimetry revealed that P23 is fairly stable in the pH range of neutral and mild-basic conditions and in the temperature range of 10 - 50$\^{C}$. Since the thermal stability and the P-sheet content of P23 were decreased by the addition of Ca$\^$2+/ ion, it could be suggested that Ca$\^$2+/ion induces structural change by partially destabilizing the structure of P23. In addition various H experiments were monitored to solve the aggregation of P23. Den results will provide the preliminary structural information about P23.

Investigation of Antimicrobial Activity of Brown Algae Extracts and the Thermal and pH Effects on Their Activity

  • The antimicrobial activity of water and ethanol extracts from 30 species of algae was measured using the agar diffusion method and minimum inhibitory concentration (MIC) test. In agar diffusion method, the 95% ethanol extracts from 12 of the algae showed growth inhibition against the tested microorganisms. In particular, Ishige okamurai, Ecklonia stolonifera, Sargassum siliquastrum, Sargassum thunbergii, Colpomenia bullosa, and Ecklonia cava had strong antibacterial activities against Gram-positive bacteria at 4 mg/mL. In the results of the MIC test, S. siliquastrum showed the most antimicrobial activity, where its MIC values ranged from 0.005 to 0.0075% against Listeria monocytogenes, Clostridium perfringens, and Basillus subtilis. In the thermal stability test, for the ethanol extracts of I. okamurai, E. cava, S. siliquastrum, S. thunbergii, and C. bullosa, the extracts proved to maintain high antimicrobial activities when they were treated at $121^{\circ}C$ for 15 min. In the pH stability test, the antimicrobial activity of the S. siliquastrum ethanol extract was stable from pH 2 to 10, whereas the activity of the other species ethanol extracts were weakened under pH 10 against several microbes.

천연색소로서 한국산 유색미 안토시아닌의 안정성 연구 (Physicochemical Stability of Anthocyanins from a Korean Pigmented Rice Variety as Natural Food Colorants)

  • 유용한 천연색소원으로서, 한국산 유색미에서 안토시아닌을 분리하여 pH, 온도, 금속이온, 당 및 유기산에 대한 안정성을 조사하였다. 몇가지 pH 조건에서 21일간 저장하였을 때, pH 2.0와 pH 3.0에서는 안토시아닌의 반감기가 약 36일과 17일로 나타났으며 중성과 염기성 pH에서는 하루 이내에 거의 분해되는 것으로 나타나 산성조건의 식품에서는 안정하게 이용될 수 있음을 시사하였다. 온도의 영향에 대한 실험 결과, 반감기가 $95^{\circ}C$에서 7.4시간(pH 3.0)과 0.17시간(pH 9.0), $75^{\circ}C$에서 23.6시간(pH 3.0), 0.57시간(pH 9.0), $50^{\circ}C$에서는 96.3시간(pH 3.0), 1.7시간(pH 9.0)으로 산성조건에서는 열에 대해서도 비교적 안정하였다. 금속이온 첨가에 따른 농색화 효과는 1가, 2가, 3가로 갈수록 높아졌으며 농도가 10 mM보다 100 mM 첨가시 더 높은 흡광도를 보였고, 2가이상의 금속이온들은 대부분 높은 안정화 효과를 보였다. 당 농도의 경우 단당류는 0.1 M, 이당류는 0.15 M 일때가 가장 높은 농색화 효과를 보여주었으나, 저장하는 동안 fructose가 안토시아닌 분해를 가장 촉진하는 것으로 나타났고, 당 첨가군은 모두 대조군보다 낮은 색소 잔존량을 보여주었다. 유기산 용액의 pH를 3.0으로 고정하여 몇가지 유기산의 영향을 조사한 결과, citric acid가 가장 농색화 및 안정화 효과가 있었으며, tartaric acid 첨가군은 대조군보다 불안정하였다. 20,000 lux의 빛에 노출시켜 빛이 미치는 영향을 조사한 결과, 유색미 안토시아닌의 반감기가 약 14시간으로 나타나 빛에 매우 민감함을 알 수 있었다. 함량이 많은 유리아미노산으로는 glutamic Acid, lysine, arginine, aspartic acid, histidine, leucine, alanine 등으로서 일반토하젓에서의 조성과 상이하였다. 양념토하젓은 glutamic acid를 제외한 대부분의 유리아미노산이 일반토하젓에서보다 낮았다. 일반토하젓과 양념토하젓의 핵산관련물질함량은 각각 $2.64{\sim}4.82\;{\mu}mol$$1.08{\sim}1.93\;{\mu}mol$로서 제품에 따른 차이가 많았다. 일반토하젓에서는 homarine이 $18{\sim}86\;mg$ 검출되었으나 바다새우젓의 30.7%였고 trigonelline은 비슷한 수준이었다. 양념토하젓은 homarine이 흔적${\sim}39\;mg$, trigonelline $0{\sim}5\;mg$으로서 일반토하젓보다 낮았다. 일반토하젓과 양념토하젓의 TMAO와 TMA 함량은 미량으로서 바다새우젓의 20.9%와 27.7% 수준이었다. 일반토하젓과 양념토하젓에서 분석된 각성분들에 대한 질소분포는 모두 유리아미노산이 가장 중요한 함질소성분으로서 평균 65.7%와 60.8%였고, 다음은 oligopeptide류로서 17.8%와 16.6%였다. 핵산관련물질은 3.9%와 2.2%, betaine류는 1.1%와 0.4%, TMAO와 TMA는 1.2%와 1.8%, 그리고 creatine은 0.4%와 9.6%로서 엑스분질소의 회수율은 90.3%와 91.2%였다. 이상에서 분석된 토하젓의 엑스분질소와 함질소 엑스성분들을 상관분석한 결과 엑스분 질소, betaine 총량, 유리아미노산 총량 및 유리아미노산의 일종인 phenylalanine 이 토하젓의 품질지표로 활용가능성이

  • Fluoroplastics have been used widely for chemical or electrical facility materials and lubricable engineering structural materials because they have the superior characteristics such as thermal stability, chemical stability, solid lubricity, arc resistance, wearable durability and good sealing property. In this study, PTFE composite materials for the insolation parts of high voltage and current breaker were investigated and estimated.
