The metabolic pathway synthesis algorithm was applied to estimate the maximum ethanol yield from xylose in a model recombinant Saccharomyces cerevisiae strain containing the genes involved in xylose metabolism. The stoichiometrically independent pathways were identified by constructing a biochemical reaction network for conversion of xylose to ethanol in the recombinant S. cerevisiae. Two independent pathways were obtained in xylose-assimilating recombinant S. cerevisiae as opposed to six independent pathways for conversion of glucose to ethanol. The maximum ethanol yield from xylose was estimated to be 0.46 g/g, which was lower than the known value of 0.51 g/g for glucose-fermenting and wild-type xylose-fermenting yeasts.
In order to ferment D-xylose directly to ethanol, Yeasts capable of utilizing D-xylose as a sole carbon source and energy source were isolated from soil, sawdust and rotten woods. Among them, the yeast strain, which showed the best ability to produce ethanol, was identified as Candida sp. L-16 isolated from rotten woods. The optimal conditions for production of ethanol were 60rpm of agitation speed, 28j.deg.C of temperature, 4.5 of initial pH and 5% of D-xylose concentration. Ethanol production was reached to maximum state for 4 days culture. Under these optimal conditions, the maximum ethanol concentration and theoretical ethanol yield were 2.4%(v/v) and 74.4% of theoretical value, respectively.
A yeast strain, BT001, which can directly ferment D-xylose to ethanol was isolated from forest soils, and then identified as Candida sp. Cultural conditions for the optimum ethanol production, along with the effects of aeration on cell growth and ethanol production were investigated. Aeration stimulated the cell growth and the volumetric rate of ethanol production, but decreased the ethanol yield. Optimum temperature and initial pH for the ethanol production were $33{\circ}^C$ and 6.0, respectively. In a shake flask culture, this strain produced 52.3 g ethanol per liter from 12%(w/v) D-xylose after incubation for 96 hours. Ethanol yield was 0.436 g per g D-xylose consumed. This corresponds to 85.8% of theoretical yield. Also, this yeast strain produced ethanol from D-galactose, D-glucose and D-mannose, but not from L-arabinose and L-rhamnose. Among these sugars, D-glucose was the fastest in being converted to ethanol sugars.
Brettanomyces custersii CBS 5512 which has reported as a thermotolerant glucose-cellobiose co-fermentable yeast strain was mutated with UV and NTG to improve ethanol yield at higher than 4$0^{\circ}C$ B. custersii H1-23, H1-39, H1-55 and H1062 were finally selected for hyper-fermentable strains at higher than 4$0^{\circ}C$ from thermotolerant 7510 colonies through 5th selection. Among the selected strains, H1-39 mutant had better fermentability at 4$0^{\circ}C$ and 43$^{\circ}C$ from different concentrations of glucose. H1-39 and H1-23 mutants yielded more than 70% of the theoretical ethanol yield in 4 and 8% mixed sugars at above 4$0^{\circ}C$, which was 5-11% higher than those by original strain. Especially, H1-39 mutant had better fermentability in 4% mixed sugar. It showed 78.5% of the theoretical yield at 4$0^{\circ}C$ and 72.2% of the theoretical yield at 43$^{\circ}C$. On the other hand, theoretical yield of ethanol by H1-39 mutant in 8% mixed sugar at 4$0^{\circ}C$ and 43$^{\circ}C$ were 75.2% and 70.2%, respectively. Theses values increased up to 7-11% as compared to those by orginal strain. By the simultaneous saccharification and fermentation, ethanol production by H1-39 mutant increased up to more than 23% as compared to that by original strain.
To enhance the hyperproductive and low energy-consuming ethanol fermentation rate, the thermotolerant yeast S. cerevisiae RA-74-2 cells were immobilized. An efficient immobilization condition was proved to be $1.5{\%}$ (w/v) alginate solution, neutral pH and 20 h activation of beads. The fermentation characteristics and stability at various temperatures were examined as compared with free S. cerevisiae RA-74-2 cells. The immobilized cells had excellent fermentation rate at the range of pH 3-7 at 30-$42^{\circ}C$ in 15-$20{\%}$ glucose media. When the seed volume was adjusted to 0.12 (v/v) (6ml bead/50 ml medium), $11{\%}$ (w/v) ethanol was produced during the first 34 hand $12.15{\%}$ (w/v) ethanol [$95{\%}$ (w/v) of theoretical yield] during the first 60 h in $25{\%}$ glucose medium. In repetitive fermentation using a 2 litre fermentor, 5.79-$7.27{\%}$ (w/v) ethanol [76-$95{\%}$ (w/v) of theoretical yield] was produced during the 40-55 h in $15{\%}$ glucose media. These data suggested the fact that alginate beads of thermotolerant S. cerevisiae RA-74-2 cells would contribute to economic and hyperproductive ethanol fermentation at high temperature.
An ethanol-producing yeast strain, CHFY0201, was isolated from soil in South Korea using an enrichment technique in a yeast peptone dextrose medium supplemented with 5% (w/v) ethanol at $30^{\circ}C$. The phenotypic and physiological characteristics, as well as molecular phylogenetic analysis based on the D1/D2 domains of the large subunit (26S) rDNA gene and the internally transcribed spacer (ITS) 1+2 regions, suggested that the CHFY0201 was a novel strain of Schizosaccharomyces pombe. During shaking flask cultivation, the highest ethanol productivity and theoretical yield of S. pombe CHFY0201 in YPD media containing 9.5% total sugars were $0.59{\pm}0.01$ g/l/h and $88.4{\pm}0.91%$, respectively. Simultaneous saccharification and fermentation for ethanol production was carried out using liquefied cassava (Manihot esculenta) powder in a 5-l lab-scale jar fermenter at $32^{\circ}C$ for 66 h with an agitation speed of 120 rpm. Under these conditions, S. pombe CHFY0201 yielded a final ethanol concentration of $72.1{\pm}0.27$ g/l and a theoretical yield of $82.7{\pm}1.52%$ at a maximum ethanol productivity of $1.16{\pm}0.07$ g/l/h. These results suggest that S. pombe CHFY0201 is a potential producer for industrial bioethanol production.
A cone-type tower fermentor packed with Sacchromyces uvarum was employed to examine the continuous ethanol fermentation process. The maximum yeast concentration in the cone-type tower fermentor was 37.5-39.5g/$\ell$, the maximum ethanol productivity at the dilution rate of $hr^{-1}$ was 16.3g/$\ell$ . hr and the average ethanol yield was 0.48g EtOH/g glucose, which was 94% of the maximum theoretical yield. It was concluded that a cone-type tower fermentor might offer better perspectives for continuous ethanol fermentation.
The intracellular metabolic fluxes can be calculated by metabolic flux analysis, which uses a stoichiometric model for the intracellulal reactions along with mass balances around the intracellular metabolites. In this study, metabolic flux analyses were carried out to estimate flux distributions for the maximum in silico yields of various metabolites in Escherichia coli. The maximum in silico yields of acetic acid and lactic acid were identical to their theoretical yields. On the other hand, the in silico yields of succinic acid and ethanol were only 83% and 6.5% of their theoretical yields, respectively. The lower in silico yield of succinic acid was found to be due to the insufficient reducing power. but this lower yield could be increased to its theoretical yield by supplying more reducing power. The maximum theoretical yield of ethanol could be achieved, when a reaction catalyzed by pyruvate decarboxylase was added in the metabolic network. Futhermore, optimal metabolic pathways for the production of various metabolites could be proposed, based on the results of metabolic flux analyses. In the case of succinic acid production, it was found that the pyruvate carboxylation pathway should be used for its optimal production in E. coli rather than the phosphoenolpyruvate carboxylation pathway.
This study was carried out to optimize the fermentation conditions for direct alcohol fermentation of xylose by Pichia stipitis CBS 5776. The best cell growth and the ethanol production were obtained under 0.05 VVM aeration and 300rpm agitation at $30^{\circ}C$ using 100 g/l xylose medium of the initial pH 5.0. In the above condition, the maximum specific growth rate and maximum cell concentration were 0.14hr-1 and $1.3 \times109$ cells/ml, respectively. Pichia stipitis CBS 5776 also produced 40.2g/l ethanol utilizing about 96% of 100g/l xylose after 72hr fermentation. At this point, the overall volumetric ethanol productivity was 0.56g/1-hr and the ethanol yield was 0.42 g-ethanol/g-xylose consumed, which corresponds to 82% of the theoretical yield.
Microorganisms capable of utilizing D-xylose as a sole carbon and energy source were isolated to ferment D-xylose directly to ethanol. Among them, the strain, which showed the best ability to pro-duce ethanol, was selected and was identified as Fusarium sp. The optimal conditions for the pro-duction of ethanol were 8.0 of initial pH, 33$^{\circ}C$ of temperature, and 2% of substrate concentration. Under this optimal condition, the following results were obtained : maximum ethanol concentration, 7.0g/$\ell$; ethanol yield, 0.35g of ethanol per g of D-xylose (68.6% of theoretical); biomass yield, 0.27g of dry biomass per g of D-xylose.
