LuxR-Type SCO6993 Negatively Regulates Antibiotic Production at the Transcriptional Stage by Binding to Promoters of Pathway-Specific Regulatory Genes in Streptomyces coelicolor

  • SCO6993 (606 amino acids) in Streptomyces coelicolor belongs to the large ATP-binding regulators of the LuxR family regulators having one DNA-binding motif. Our previous findings predicted that SCO6993 may suppress the production of pigmented antibiotics, actinorhodin, and undecylprodigiosin, in S. coelicolor, resulting in the characterization of its properties at the molecular level. SCO6993-disruptant, S. coelicolor ΔSCO6993 produced excess pigments in R2YE plates as early as the third day of culture and showed 9.0-fold and 1.8-fold increased production of actinorhodin and undecylprodigiosin in R2YE broth, respectively, compared with that by the wild strain and S. coelicolor ΔSCO6993/SCO6993+. Real-time polymerase chain reaction analysis showed that the transcription of actA and actII-ORF4 in the actinorhodin biosynthetic gene cluster and that of redD and redQ in the undecylprodigiosin biosynthetic gene cluster were significantly increased by SCO6993-disruptant. Electrophoretic mobility shift assay and DNase footprinting analysis confirmed that SCO6993 protein could bind only to the promoters of pathway-specific transcriptional activator genes, actII-ORF4 and redD, and a specific palindromic sequence is essential for SCO6993 binding. Moreover, SCO6993 bound to two palindromic sequences on its promoter region. These results indicate that SCO6993 suppresses the expression of other biosynthetic genes in the cluster by repressing the transcription of actII-ORF4 and redD and consequently negatively regulating antibiotic production.

Research on Residential Ecology Conversion Education in Home Economics Education for Carbon Neutrality Practice (탄소중립 실천을 위한 가정과교육에서의 주생활 생태전환교육 방향)

  • The advent of the carbon neutrality era has led to a paradigm shift in education toward ecological transition education. It is time to understand the concept and practice of ecological transition education pursued in the 2022 revised curriculum and prepare for changes to the main life education covered in home economics education. Therefore, the purpose of this study is to explore the role of home economics education for carbon neutrality for future generations who will have to live in the era of rapidly changing climate crisis, and to propose a practice plan for ecological transition education in the main living area based on the educational contents of the 2022 revised curriculum. To achieve these objectives, this study reviewed previous research on carbon neutrality and ecological transition education, analyzed the curriculum for main life from the first curriculum to the 2022 revised curriculum, and proposed the direction of main life education for ecological transition education. Through this study, the importance of carbon neutrality and ecological transition can be emphasized in home economics education, and it is expected that it can be used as a basis for realizing ecological transition education in actual education sites.

A Cartographic Study on the Earthly Paradise Represented in the Medieval Mappaemundi (서양 중세 세계지도에 표현된 지상낙원의 지도학적 연구)

  • The achievements of the medieval mappaemundi should be evaluated on their own terms and in the context of their purpose. Medieval mappaemundi expressed christian world view which reflected Augustinian theology. These Christian world view and Greek and Roman classical geography were combined to form the cultural barkground of the medieval mappaemundi. These maps had a function to organize physical space according to religious principles. The Christian concept of the world as a temporal phenomenon, derived from the simultaneous creation of time and space as described in St. Augustin's theology was represented in these maps. The purpose of this paper is to consider geographic characteristics of earthly paradise expressed in medieval mappaemundi and their cartographic characteristics. For this, firstly, we reviewed medieval Christian scholars' opinions on earthly paradise. Secondly, centered on geographic location and representation method, we examined cartographic characteristics of medieval paradise mapping, Thirdly, we considered the shift of paradise according to chronological change. Fourthly, we examined the reason why earthly paradise disappeared from world map after fourteenth century.

0.11-2.5 GHz All-digital DLL for Mobile Memory Interface with Phase Sampling Window Adaptation to Reduce Jitter Accumulation

  • An all-digital delay-locked loop (DLL) for a mobile memory interface, which runs at 0.11-2.5 GHz with a phase-shift capability of $180^{\circ}$, has two internal DLLs: a global DLL which uses a time-to-digital converter to assist fast locking, and shuts down after locking to save power; and a local DLL which uses a phase detector with an adaptive phase sampling window (WPD) to reduce jitter accumulation. The WPD in the local DLL adjusts the width of its sampling window adaptively to control the loop bandwidth, thus reducing jitter induced by UP/DN dithering, input clock jitter, and supply/ground noise. Implemented in a 65 nm CMOS process, the DLL operates over 0.11-2.5 GHz. It locks within 6 clock cycles at 0.11 GHz, and within 17 clock cycles at 2.5 GHz. At 2.5 GHz, the integrated jitter is $954fs_{rms}$, and the long-term jitter is $2.33ps_{rms}/23.10ps_{pp}$. The ratio of the RMS jitter at the output to that at the input is about 1.17 at 2.5 GHz, when the sampling window of the WPD is being adjusted adaptively. The DLL consumes 1.77 mW/GHz and occupies $0.075mm^2$.

Synthesis and Luminescent Characterization of Eu2+/Dy3+-Doped Sr2MgSi2O7 Powders (Eu2+/Dy3+ 이온이 도핑된 Sr2MgSi2O7 분말 합성 및 발광 특성)

  • $Eu^{2+}/Dy^{3+}$-doped $Sr_2MgSi_2O_7$ powders were synthesized using a solid-state reaction method with flux ($NH_4Cl$). The broad photoluminescence (PL) excitation spectra of $Sr_2MgSi_2O_7:Eu^{2+}$ were assigned to the $4f^7-4f^65d$ transition of the $Eu^{2+}$ ions, showing strong intensities in the range of 375 to 425 nm. A single emission band was observed at 470 nm, which was the result of two overlapping subbands at 468 and 507 nm owing to Eu(I) and Eu(II) sites. The strongest emission intensity of $Sr_2MgSi_2O_7:Eu^{2+}$ was obtained at the Eu concentration of 3 mol%. This concentration quenching mechanism was attributable to dipole-dipole interaction. The $Ba^{2+}$ substitution for $Sr^{2+}$ caused a blue-shift of the emission band; this behavior was discussed by considering the differences in ionic size and covalence between $Ba^{2+}$ and $Sr^{2+}$. The effects of the Eu/Dy ratios on the phosphorescence of $Sr_2MgSi_2O_7:Eu^{2+}/Dy^{3+}$ were investigated by measuring the decay time; the longest afterglow was obtained for $0.01Eu^{2+}/0.03Dy^{3+}$.

A new approach to quantify paraquat intoxication from postmortem blood sample by using 1H qNMR method

  • For a case study of suspected paraquat intoxication, we developed a simple and rapid method of $^1H$ qNMR to determine the mili-molar amount of paraquat in postmortem blood samples. There were no interfering signals from endogenous compounds in the chemical shift of paraquat and diquat (internal standard). The amount of sample used ranged from 0.25 mM to 10.0 mM. Diquat, which has similar physicochemical properties with paraquat, was chosen as an internal standard. The NMR experimental conditions, relaxation delay time and CPMG spin-echo pulse sequence were optimized. The developed method was validated in terms of specificity, accuracy, precision, matrix effect, recovery, limit of detection (LOD), and low limit of quantification (LLOQ). The proposed qNMR method provided a simple and rapid assay for the identification and quantification of the quaternary ammonium herbicide, "paraquat" in postmortem blood samples. This method was tested by using the blood from the heart of a man who was intoxicated with paraquat. In this particular case, the level of paraquat was 1.07 mM in the blood. For the determination of quaternary ammonium herbicides, qNMR could also be used to provide a better understanding of the currently available techniques.

Earnings Management Associated with Types of CEO Turnover in Public Institutions (공공기관의 경영자 교체유형과 이익조정 행태와의 관련성)

  • This study examines the influence of CEO(Chief Executive Officer) turnover on earnings management in public institutions. We classified sample into four portfolios based on the joint types of departing CEO(Forced/Voluntary) and incoming CEO(Expert/Nonexpert), and we explore whether earnings management are distinguishable depending on the types of CEO turnover. Empirical findings are summarized as following. The different patterns of earnings management are observed for each portfolio in the turnover year. In particular, when departing CEO leaves the company forcibly and incoming CEO is an expert in that business, there is an evidence of downward earnings management in the turnover year. This means that the incoming CEO takes a bath in his first year by recording big charges to shift blame for failed management on to the predecessor. The results documented in this paper provide an important implication for diverse interest groups participating in the CEO turnover decisions at the present time when normalization of management of public institutions is viewed more important than ever.

  • In this paper, we proposed a new algorithm of pulse generation and detection for the UWB multimedia communication system. The existing UWB systems using Gaussian pulse have some difficulties to cope with bandwidth limitation and frequency transition. In this paper, we introduce a new pulse generation method, which is able to control the bandwidth and center frequency that applies to the frequency modulation method, thus the proposed algorithm could improve the detection performance of receiving. And we proposed the multi channel transmission algorithm which transmits 1 channel synthesized at same time using the wavelet synthesis filter. Because of wavelet filter's perfect reconstruction property, the BER(Bit Error Rate) of transmission data is not changed by the number of accessed user. BER is changed only the property of channel transmission. By the results of simulation, when shift SNR from 2dB to 8dB on AWGN channel, we confirmed that the proposed algerian has $3.9063{\times}10^{-4}$ BER at 4dB SNR(AWGN channel).

Feasibility Study on Introduction of Piggy-back System by Applying Transport Database

  • In this study, The goal is to analyze the feasibility of introducing a Piggyback system that can reduce the time and cost incurred by transshipment work and improve the transportation speed when transporting complex cargo by rail. To this end, the feasibility analysis methodology is reviewed through domestic and international literature review. In order to quantitatively derive the feasibility analysis values, a transportation database was applied to develop a freight transport simulation model and a freight demand prediction model for major freight transport O-D routes with a transportation distance of 200 km or more. As a result of analyzing economic feasibility by setting the analysis period to 15 years on the premise that the Piggyback System will be introduced on major cargo transport O-D routes in 2025, the NPV value was positive and the B/C value was 1.18, indicating that the Piggyback system was economical. The proposed research method can be meaningful data for establishing transportation policies that can improve the competitiveness of railroad transportation.

Position Estimation Technique of High Speed Vehicle Using TLM Timing Synchronization Signal (TLM 시각 동기 신호를 이용한 고속 이동체의 위치 추정)

  • If radio interference occurs or there is no navigation device, radio navigation of high-speed moving object becomes impossible. Nevertheless, if there are multiple ground stations and precise range measurement between the high-speed moving object and the ground station can be secured, it is possible to estimate the position of moving object. This paper proposes a position estimation method using high-precision TDOA measurement generated using TLM signal. In the proposed method, a common error of moving object is removed using the TDOA measurements. The measurements is generated based on TLM signal including SOQPSK PN symbol capable of precise timing synchronization. Therefore, since precise timing synchronization of the system has been performed, the timing error between ground stations has a very small value. This improved the position estimation performance by increasing the accuracy of the measured values. The proposed method is verified through software-based simulation, and the performance of estimated position satisfies the target performance.
