Expression Pattern of Antioxidant Enzymes Genes in the Ventral Prostates of Rats Exposed to Procymidone and/or Testosterone after Castration

  • Procymidone is a fungicide with anti-androgenic properties widely used to protect fruits from fungal infection, which induces an excessive reactive oxygen species production in male reproductive organs. In this study, to clarify whether procymidone affect the cellular antioxidant system of prostate at onset of puberty, gene expression patterns of the representative antioxidant enzymes such as cytoplasmic glutathione peroxidase (GPx1), phospholipid hydroperoxide GPx (PHGPx), selenoprotein P (SePP), cytoplasmic copper/zinc superoxide dismutase (SOD1), and manganese SOD (SOD2) were investigated in the rat ventral prostates exposed to procymidone using real-time RT-PCR analyses. Seven-week-old Sprague-Dawley rats castrated at 6 weeks old were treated with procymidone (25, 50, or 100 mg/kg per day) orally for 7 consecutive days after testosterone propionate (0.4 mg/kg per day) administration by subcutaneous injection. As compared to normal control animals, GPx1 mRNA expression in prostates significantly increased by the administration with TP and/or procymidone. However, PHGPx and SOD1 mRNA levels significanatly decreased by over 25 mg/kg of procymidone treatment and SePP and SOD2 mRNA levels was significanatly reduced by over 50 mg/kg of procymidone treatment. These findings indicate that procymidone may affect the antioxidant system of prostatic cells in up-regulation mode of GPx1, but in down-regulation modes of PHGPx, SePP, SOD1, and SOD2, suggesting that procymidone may affect differently the cellular antioxidant system of prostate according to the exposure doses.

The Change of Free Sugar, Hesperidine, Naringin, Flavonoid Contents and Antihypertensive Activities of Yuza Variety according to Harvest Date (유자의 종류별 수확시기에 따른 유리당, 헤스페리딘, 나린진, 플라보노이드 함량과 항고혈압 활성 변화)

  • Yuza (Citrus junos) had 3 varieties and they had variable physiochemical properties. This study analyzed free sugar, hesperidine, naringin, flavonoid contents and antihypertensive activities according to variety and harvest time of Yuza. Three Yuza varieties, native (C. junos I), improved I (C. junos I + Poncirus trifoliata), improved II (C. junos I + C. junos II) were used. Harvest time could influence biochemical properties of Yuza such as acidity and $^{\circ}brix$. When the Yuza harvested at behind of season, the sugar/acid ratio went up because the acidity was decreased but sugar content was increased. Fructose content was dramatically high at November, Yuza of the season. Total flavonoids was high in pulp parts of improved II and in juice of native one harvested at October. The content of hesperidin and naringin in peels and juices were high in immature fruits fresh-picked at July since then they were decreased steadily and there were no difference according to varieties. The angiotensin converting enzyme (ACE) inhibition was powerful at pulp of Yuza in particular native one picked at November.

Effects of Quercetin and Genistein on Boar Sperm Characteristics and Porcine IVF Embyo Developments

  • Quercetin and genistein, plentifully present in fruits and vegetables, are flavonoid family members that have antioxidative function and plant-derived phytoestrogen activity. The antioxidative effects of quercetin and genistein on boar sperm characteristics and in vitro development of IVF embryo were investigated. The sperm motility was increased by addition of genistein $50{\mu}M$ for 6 hr incubation compared to control (p<0.05). The sperm viability was increased by addition of quercetin 1 and $50{\mu}M$ and genestein 1 and $50{\mu}M$ for 3 hr incubation. In addition, the sperm viability seemed to be increased dose-dependantly by addition of quercetin or genistein 1 and $50{\mu}M$, respectively (p<0.05). The membrane integrities were not increased by quercetin or genistein treatments for 3 hr or 6 hr incubation period except for quercetin $1{\mu}M$ for 3 hr incubation. In mitochondrial activities, addition of quercetin $50{\mu}M$ for 6 hr incubation increased mitochondrial activity but decreased at $100{\mu}M$ concentration compared with control (p<0.05). When porcine IVF embryos were cultured in PZM-3 medium supplemented with low concentrations of quercetin ($1{\sim}10{\mu}M$), the developmental rates to morula and blastocyst increased but significantly decreased at high concentrations of quercetin ($25{\sim}50{\mu}M$). The highest developmental rate to blastocysts among all concentrations of quercetin was shown at quercetin $10{\mu}M$ (p<0.05). The developmental rates to morula or blastocysts at low ($0.01{\sim}1{\mu}M$) and high ($5{\sim}10{\mu}M$) concentrations of genistein were not significantly different among all treatment group and genistein did not affect on IVF embryo development. These results suggest that quercetin and genistein seem to have positive effects at certain concentrations on sperm characteristics such as motility, viability and mitochondrial activity. In addition, low concentrations of quercetin (1, 5 and $10{\mu}M$) in this experiment, seem to have beneficial effect on porcine IVF embryo development but genistein did not affect on it at all given concentrations ($0.01{\sim}10{\mu}M$).

  • Gas chromatogram of standardized samples for 10 different kinds of polyphenol components, which contained universally in common vegetables and fruits, and those of polyphenol extracts from various ginseng products and Acanthopanax, were revealed, respectively. The consequent results are as follows; 1. There were practically no obvious difference in the polyphenol pattern among white ginseng with skin of either Korean, American, Canadian products, or Korean red ginseng. There was, however, no coincidence in $t_R$ as indicated by peaks of polyphenol pattern for these ginseng products with those expressed by the standard samples. 2. A great similarity existed in the polyphenol pattern between white ginseng and red ginseng extracts, but the number of peaks for the ginseng extracts was far less than dried ginseng. 3. The polyphenol patterns of Russian and Korean Acanthopanax showed a similarity. However, the polyphenol pattern as represented by Acanthopanax was considerably different from that of ginseng products, especially chlorogenic acid which was not present in the ginseng products was identified in Acanthopanax.

Inhibitory Effects of Quercetin on Muscle-type of Nicotinic Acetylcholine Receptor-Mediated Ion Currents Expressed in Xenopus Oocytes

  • The flavonoid quercetin is a low molecular weight compound generally found in apple, gingko, tomato, onion and other red-colored fruits and vegetables. Like other flavonoids, quercetin has diverse pharmacological actions. However, relatively little is known about the influence of quercetin effects in the regulation of ligand-gated ion channels. Previously, we reported that quercetin regulates subsets of nicotinic acetylcholine receptors such as ${\alpha}3{\beta}4$, ${\alpha}7$ and ${\alpha}9{\alpha}10$. Presently, we investigated the effects of quercetin on muscle-type of nicotinic acetylcholine receptor channel activity expressed in Xenopus oocytes after injection of cRNA encoding human fetal or adult muscle-type of nicotinic acetylcholine receptor subunits. Acetylcholine treatment elicited an inward peak current ($I_{ACh}$) in oocytes expressing both muscle-type of nicotinic acetylcholine receptors and co-treatment of quercetin with acetylcholine inhibited $I_{ACh}$. Pre-treatment of quercetin further inhibited $I_{ACh}$ in oocytes expressing adult and fetal muscle-type nicotinic acetylcholine receptors. The inhibition of $I_{ACh}$ by quercetin was reversible and concentration-dependent. The $IC_{50}$ of quercetin was $18.9{\pm}1.2{\mu}M$ in oocytes expressing adult muscle-type nicotinic acetylcholine receptor. The inhibition of $I_{ACh}$ by quercetin was voltage-independent and non-competitive. These results indicate that quercetin might regulate human muscle-type nicotinic acetylcholine receptor channel activity and that quercetin-mediated regulation of muscle-type nicotinic acetylcholine receptor might be coupled to regulation of neuromuscular junction activity.

Temperature and length of cold storage affect the Quality Maintenance of fresh kiwifruit (Actinidia chinensis Planch) (저온저장 온도 및 저장기간이 키위 "골드"의 품질 유지에 미치는 효과)

  • The effects of temperature and length of cold storage on the quality maintenance of fresh "Gold" kiwifruit were investigated. Physio-chemical properties were analyzed in kiwifruit held at $2^{\circ}C$ and $6^{\circ}C$ temperatures compared to fruit at room temperature ($20{\sim}28^{\circ}C$) during 8 weeks of storage. Low temperatures ($2^{\circ}C$ and $6^{\circ}C$) significantly delayed softening and soluble solids content (SSC) accumulation compared to higher temperature ($20{\sim}28^{\circ}C$). Physico-chemical properties of fruits, including weight losses, firmness, SSC, titratable acidity (TA), SSC/TA ratio, and flesh color properties were monitored during storage. Fast firmness loss was detected in fruit stored at higher temperatures compared to low temperature ($2^{\circ}C$). Similar results were observed for acidity according to storage temperature and length of cold storage, whereas SSC increased to the limited values (%Brix) during storage. The soluble solids content (SSC) increased markedly during the first 60 days of storage and remained almost constant thereafter for all treatments. SSC accumulation rates decreased from 5 weeks after storage probably due to differences between initial and ripe kiwifruits, and SSC decreased with each passing week due to natural starch conversion over time. The SSC/acid ratio increased from 18 to 27 until 5 weeks after storage and then slowly declined in all kiwifruit stored at different low temperatures. Sensory evaluation results showed no differences in kiwifruit flesh color stored at two storage temperatures of $2^{\circ}C$ and $6^{\circ}C$.

Breeding of New Cultivar 'Hongsil' in Forcing Culture of Strawberry for Export (수출용 촉성재배 딸기 신품종 '홍실' 육성)

  • The 'Hongsil' strawberry ($Fragaria{\times}ananassa$ Duch.) is a new cultivar released by Gyeongsangnam-do Agricultural Research & Extension Services in 2015. This cultivar originated from a 2011 cross between '802a312' and 'Okmae' and exhibited suitable fruit characteristics for export due to the high soluble solids content and firmness of the fruit. The breeding line '1116-74-15' was selected as an elite line after examining its characteristics and productivity from 2013 to 2014. A field trial of this line was conducted on a farm in 2015, and it was registered as 'Hongsil' thereafter. The 'Hongsil' strawberry demonstrates intermediate plant type and moderate vigor in growth. It is suitable for forcing culture due to its early flower bud differentiation and low chilling requirement. The soluble solids content of 'Hongsil' is $10.0^{\circ}Brix$, which is approximately $0.4^{\circ}Brix$ higher than that of 'Seolhyang' The Fruit firmness of 'Hongsil' is $13.1g{\cdot}mm^{-2}$, similar to that of 'Maehyang'. Fruits of 'Hongsil' are long conical having a red skin color and marketable yield is $4,269kg{\cdot}10a^{-1}$ which is 16% higher than that of 'Maehyang'. The 'Hongsil' cultivar is sensitive to powdery mildew, and it therefore warrants further investigation regarding disease control.

Impact of Healthy Eating Practices and Physical Activity on Quality of Life among Breast Cancer survivors

  • Following breast cancer diagnosis, women often attempt to modify their lifestyles to improve their health and prevent recurrence. These behavioral changes typically involve diet and physical activity modification. The aim of this study was to determine association between healthy eating habits and physical activity with quality of life among Iranian breast cancer survivors. A total of 100 Iranian women, aged between 32 to 61 years were recruited to participate in this cross-sectional study. Eating practices were evaluated by a validated questionnaire modified from the Women's Healthy Eating and Living (WHEL) study. Physical activity was assessed using the International Physical Activity Questionnaire (IPAQ). A standardized questionnaire by the European Organization of Research and Treatment of Cancer Quality of Life and its breast cancer module (EORTC QLQ-C30/+BR-23) were applied to determine quality of life. Approximately 29% of the cancer survivors were categorized as having healthy eating practices, 34% had moderate eating practices and 37% had poor eating practices based on nutrition guidelines. The study found positive changes in the decreased intake of fast foods (90%), red meat (70%) and increased intake of fruits (85%) and vegetables (78%). Generally, breast cancer survivors with healthy eating practices had better global quality of life, social, emotional, cognitive and role functions. Results showed that only 12 women (12%) met the criteria for regular vigorous exercise, 22% had regular moderate-intensity exercise while the majority (65%) had low-intensity physical activity. Breast cancer survivors with higher level of physical activity had better emotional and cognitive functions. Healthy eating practices and physical activity can improve quality of life of cancer survivors. Health care professionals should promote good dietary habits and physical activity to improve survivor's health and quality of life.

Present Status of Smart Greenhouses Growing Fruit Vegetables in Korea: Focusing Management of Environmental Conditions and Pests in Greenhouses (한국의 과채류 재배 스마트 온실 실태: 온실 환경 및 병해충 관리)

  • Smart greenhouses are primarily used for growing fruits and vegetables, such as sweet peppers, tomatoes, strawberries. Although the number of smart greenhouses has been increasing exponentially, no studies have been performed to evaluate the state of smart greenhouses in Korea. Therefore, this study was conducted to determine current state of smart greenhouses with regard to greenhouse specifications, crop growing methods, pests, and user satisfaction in Korea. Contact information for smart greenhouses was provided by the officials of local agricultural research and extension services. This survey was conducted by visiting each greenhouse. Results showed that approximately 50% of surveyed smart greenhouses were between 3,300 ㎡ and 6,600 ㎡. The most frequently chosen method for pest control was chemical pesticides (97.1%). Powdery mildew and gray mold comprised 54.4% and 33.8% of the crop diseases, respectively. All tomato greenhouse farmers considered whiteflies the most problematic pest. In contrast, 76.5% and 70.6% of sweet pepper farmers believed thrips and aphids posed significant threats, respectively. The mean satisfaction score was 7.5 out of 10 points, with 10 being "extremely satisfied". These results will aid in decision making with respect to the management of current smart greenhouses and the design of future smart farms in Korea.

Effect of Lateral Shoot Pinching and Shift of Drip Irrigation Site on the Number of Lateral Shoot and the Yield in White Spined Cucumber (백침계 오이의 적심방법과 관수부위 확대가 측지발생수와 수량에 미치는 영향)

  • An experiment was conducted to evaluate the efficacy of continuous lateral shoot pinching and gradual adjustment of drip irrigation lines in improving plant health as well as fruit yield and quality in white-spined cucumber grown in the greenhouse. Grafted white-spined cucumber seedlings were established at a density of 18,000 plants per ha in a greenhouse. Main stems were pinched at 21st node. Each of the lateral shoots was pinched at the 3rd node and 2-3 fruits per lateral shoot were harvested. Drip irrigation lines were gradually adjusted to provide water to the area of maximum root concentration. Plants grown with these combined treatments produced $55\%$ more lateral shoots and 38% more marketable yield, as compared to the control.
