Antifungal Activity of Crude Extract Compound from Rhus verniciflua Against Anthracnose Fungi (Collectotrichum spp.) of Red-Pepper (고추 탄저병균에 대한 옻나무 추출물의 항진균 효과)

  • BACKGROUND: Anthracnose disease caused by Collectotrichum spp. is one of the most important worldwide devastating diseases in red pepper plants. Fungicides using plant extracts have several advantages, compared to synthetic chemical fungicides, because they are naturally occurring compounds, are usually safe for agricultural environment and are used for producing highly valuable agricultural products. Efforts for seeking an anti-fungal activities using naturally occurring compounds were mostly conducted from medicinal plant extracts. Sap of Rhus verniciflus was known to have healing effects on several human diseases. Recently, the extracts of Rhus verniciflus were actively tested for anti-cancer, anti-oxidative, and anti-fungal effects. In this study, the extract of Rhus verniciflus was tested for anti-fungal activity against Colletotrichum spp., which cause anthracnose in red-pepper. METHODS AND RESULTS: After neutralizing extracts of Rhus verniciflus (urushiol contents 70%) with autoclave, the crude extracts were used to investigate inhibitory effects for mycelial growth and spore germination of Colletotrichum spp. on PDA media. The mycelial growth and spore germination of Colletotrichum spp. were inhibited 18-39% and over 50% in response to crude extract of R. verniciflus (1.0 mg/mL). After spraying the extracts at the same concentrations above and then artificially inoculating Colletotrichum spp. on blue and red-pepper fruits, in vitro inhibition effects were examined. At 1.0 mg/mL, the crude extract of R. verniciflus showed inhibition activity in anthracnose incidence on blue- and red-pepper as 68.1-75.0%, through a artificial inoculation of Colletotrichum spp. in a laboratory. For in vivo inhibitory effects, the extracts (1.0, 0.1, and 0.01 mg/mL) were treated on red-pepper plants grown in green house 3 times at the interval of 1 week. Then inhibitory effects were determined by counting diseased fruits at 1 week after final treatment. The incidence of anthracnose was inhibited over 60% in the greenhouse by treatment of crude extract of R. verniciflus (1.0 mg/mL). CONCLUSION(s): Extracts of Rhus verniciflus were shown to have inhibitory effects on mycelial growth, spore germination of Colletotrichum spp. in vitro and on occurrence of anthracnose on pepper fruit in green house.

Ethanol Extract of Schisandra chinensis (Turcz.) Baill. Reduces AICAR-induced Muscle Atrophy in C2C12 Myotubes (마우스 C2C12 근관세포에서 AICAR로 유도된 근위축에 미치는 오미자 추출물의 영향)

  • Muscle atrophy, known as a sarcopenia, is defined as a loss of muscle mass resulting from a reduction in the muscle fiber area or density due to a decrease in muscle protein synthesis and an increase in protein breakdown. Schisandrae fructus (SF) extract of the fruits of Schisandra chinensis (Turcz) Baillon has been used as a tonic in traditional medicine for thousands of years. Although a great deal of work has been carried out on the therapeutic potential of SF, its pharmacological mechanisms of action in muscle diseases actions remain unclear. In the present study, we investigated the inhibitory effects of SF ethanol extracts on the production of muscle atrophy factors in C2C12 myotubes stimulated with 5-aminoimidazole-4-carboxamide-ribonucleotide (AICAR), an AMP-activated kinase (AMPK) activator, and sought to determine the underlying mechanisms of action. AICAR upregulated atrophy-related ubiquitin ligase muscle RING finger-1 (MuRF-1) and stimulated the levels of the forkhead box O3a (FoxO3a) transcription factor in the C2C12 myotubes. SF supplementation effectively and concentration- dependently counteracted AICAR-induced muscle cell atrophy and reversed the increased expression of MuRF-1 and FoxO3a. Our study demonstrates that SF can reverse the muscle cell atrophy caused by AICAR through regulation of the AMPK and FoxO3a signaling pathways, followed by inhibition of MuRF-1.

Development and Validation of Analytical Method for Determination of Biphenyl Analysis in Foods (식품 중 비페닐 분석법 개발 및 유효성 검증)

  • Biphenyl is used as an intermediate in the production of crop protection products, a solvent in pharmaceutical production, and as a component in the preservation of citrus fruits in many countries. Biphenyl is not authorized for use and also does not have standards or specifications as a food additive in Korea. National and imported food products are likely to contain biphenyl. Therefore, control and management of these products is required. In this study, a simple analytical method was developed and validated using HPLC to determine biphenyl in food. These methods are validated by assessing certain performance parameters: linearity, accuracy, precision, recovery, limit of detection (LOD), and limit of quantitation (LOQ). The calibration curve was obtained from 1.0 to $100.0{\mu}g/mL$ with satisfactory relative standard deviations (RSD) of 0.999 in the representative sample (orange). In the measurement of quality control (QC) samples, accuracy was in the range of 95.8~104.0% within normal values. The inter-day and inter-day precision values were less than 2.4% RSD in the measurement of QC samples. Recoveries of biphenyl from spiked orange samples ranged from 92.7 to 99.4% with RSD between 0.7 and 1.7% at levels of 10, 50, and $100{\mu}g/mL$. The LOD and LOQ were determined to be 0.04 and $0.13{\mu}g/mL$, respectively. These results show that the developed method is appropriate for biphenyl identification and can be used to examine the safety of citrus fruits and surface treatments containing biphenyl residues.

Effect of Flower and Fruit Thinner on Fruit Set and Fruit Quality of 'Gamhong' Apples (적화제 및 적과제가 '감홍' 사과의 착과와 과실 품질에 미치는 영향)

  • This study investigated the effects of flower and fruit thinning agents on fruit set and harvested fruit quality attributes in 'Gamhong' apples. Lime sulfur, MaxCel (1.9% BA), and Fruitone (3.5% NAA) were applied either at post-bloom or fruitlet stages to mature 'Gamhong/M.9' trees. In 2011, the numbers of fruits per cluster in terminal flowers were 1.74, 0.82, and 1.15 for the control, lime sulfur, and Maxcel (applied at 10-mm fruit stage) treatments, respectively. The percentages of single fruit per cluster were 36.0, 47.9, and 48.7% for the control, lime sulfur, and Maxcel (10 mm) treatments, respectively, while the percentages of clusters with three fruits per cluster were 22.9, 1.4, and 5.8%. In lateral flowers, fruit numbers per cluster were 1.20, 0.36, and 0.50 for the control, lime sulfur, and Maxcel (10 mm) treatments, respectively. In 2012, all the thinning treatments showed a positive effect on flower and fruit thinning, compared with the control. Moreover, the treatment with thinning agents did not affect fruit quality. Overall, the results suggest that a single application of flower or fruit thinning agents would be sufficient, rather than the mixed application of thinning agents, based on the observed decrease in fruit setting.

Breeding of New Ever-bearing Strawberry 'Miha' with High Hardness (고경도 사계성 딸기 '미하' 육성)

  • 'Miha' is a new strawberry (Fragaria x ananassa Duch.) cultivar, which was released by the Highland Agriculture Research Institute in 2019. The 'Miha' cultivar originates from a 2014 cross between 'Monterey' and 'Saebong No. 3', both of which exhibited excellent ever-bearing characteristics, including continuous flowering and large fruits under long-day and high temperature conditions. This new cultivar was initially named 'Saebong No. 12' after examining its characteristics and productivity during summer cultivation between 2015 and 2019. After regional adaptability tests, 'Miha' was selected from 'Saebong No. 12' as an elite cultivar. The general characteristics of 'Miha' include intermediate, elliptic leaves, and strong growth. The fruits are conical and of a dark-red color. The number of leaves of 'Miha' was 21.9, which was 6.2 fewer than that of the control cultivar, 'Goha' with 28.1. The number of flower clusters of 'Miha' was similar to that of 'Goha'. The average fruit weight of 'Miha' was 13.4 g, which was 4.3 g heavier than that of 'Goha'. The fruit hardness of 'Miha' was 36.2 g·mm-2, which was 10.1 g·mm-2 harder than that of 'Goha'. The marketable yield of 'Miha' was 37,393 kg·ha-1, 156% more than that of 'Goha' with 23,970 kg·ha-1. Therefore, the new cultivar of ever-bearing strawberry 'Miha' is expected to be very popular in the export or bakery market because it is hard.

The Effect of Renewal Topworking on Early Y Shape Tree Formation and Yields in Peach Trees (고접갱신이 복숭아 Y자 수형 조기 구성 및 생산성에 미치는 영향)

  • The effects of the early tree shapes with renewal by top-working on Y-shaped peach tree, and the influence on the maintaining fruit quantity with gradual renewal of interstock cultivar were determined. In the comparison of the places of top-working tree, top working tree on the inside of the main branch of interstock cultivar had higher graft union rate and branch growth than those of top working tree on the outside. Tree width, basal diameter of shoot, and number of bearing shoots were smaller in top working tree than in replanted tree. Although labor time was not different to control top working tree and replanted tree, labor time was much required to manage top working tree with interstock cultivar. Accumulated fruit production was 2,384 kg/10a in top working tree and 2,025 kg/10a in replanted tree for three years. However, top working tree had no loss of fruits because interstock cultivar of top-working tree had 3,727 kg/10a of fruits. No variation on fruit quality was observed between top working tree and replanting tree. In terms of economic value of top-working tree, labor to manage interstock cultivar, fertilizer price, fruit bagging, and grading and packing price increased. However, fruit production increased, and price of seedling, rental equipment, pulling-out trees, and repairing supporting system decreased. Therefore, gradual renewal of topworking tree has effects on the maintaining fruit quantity, supplementation on fruit loss, and renewal cultivar.

Effects of Fermented Soybean Extracts and Rain-shelter System on Growth and Disease Occurrence of 'Niitaka' Pear (발효 콩 추출물과 비가림 시설이 '신고' 배나무의 생육과 병해충 발생에 미치는 영향)

  • Growth of 6-year old 'Niitaka' pear (Pyrus pyrifolia Nakai) trees and control of insect and disease occurrences were compared between fermented soybean extracts and rain-shelter system for two years. Foliar application of fermented soybean extracts was applied at 6 times as a pre-experiment in the open-field in 2013, with a rain-shelter system in 2014. Fermented soybean extract treatment increased foliar concentrations of approximately 0.46% T-N, 0.17% K, 0.19% Ca, and 0.06% Mg in 2013 compared to the control, with similar macro-nutrients between the control and soybean extract treatment observed in 2014. Rain-shelter system increased foliar concentrations of T-N, Ca, and Mg compared to the open-field. There were no significantly different between the control and soybean extract treatment for number of leaves per fruit, leaf dry weight, phytotoxicity, and completed shoot growth on August during the two years. Rain-shelter system increased leaf dry weight and did not affect phytotoxicity in the leaves. Fruit quality parameters were mostly similar to control and soybean extract treatment for two years, with higher fruit firmness observed for soybean extract treatment. Rain-shelter system advanced 4 days of harvest dates, and increased approximately 7.0 ton fruit yield per ha, 20 g mean fruit weight, and fruit soluble solid contents compared to open-field in 2014. Soybean extract treatment little suppressed occurrence of disease and insect on the leaves and fruits in both years. Rain-shelter system increased occurrence of Venturia nashicola on the leaves and to 63.8% of Gymnosporangium asiaticum on the fruits in 2014. Strong winds and storms in May elevated relative humidity in the rain-shelter system and caused high infection of the disease occurrence, requiring for an additional green control method. Soybean extract treatment little affected tree growth and would have initiated for a long-term study to evaluate tree physiological characteristics. Rain-shelter system improved fruit productivity and advanced harvest dates, which could have been more effective facility at a Thanks Giving Day between middle and end of September.

Effect of Waste Nutrient Solution and Fertigation Nutrient Solution on the Growth and Qualities of Tomato Grown by Fertigation (관비재배시 토마토 생육과 품질에 미치는 폐양액과 기존 비료의 효과)

  • Waste nutrient solution (WNS) that was the drained nutrient solution of Horticultural Research Institute of Japan for culture tomato in perlite hydroponics showed $1.9-2.4dS{\cdot}m^{-1}$ of EC and 5.7-7.1 pH from April to July. Although ${NH_4}^+-N$ concentration of WNS decreased remarkably, the other nutrients did not change significantly, as compared with supplied solution. There were no significant differences in plant height, stem diameter, and the other growth characteristics of tomato plants grown by 2 fertigation nutrient solutions; BHF (Bountiful Harvest Fertilizer, 10% of N, 13% of $PO_4$, 13% of K, 0.05% of B, 0.05% of Zn, and 0.0023% of Cu that made in Korea) and Megasol (11% of N, 8% of $PO_4$, 34% of K, 0.032% of Mn, 0.002% of B, 0.048% of Fe, 0.0122% of Zn, and 0.0023% of Cu that made in Belgium.); however, the chlorophyll content of tomato leaf was highest in WNS. The fresh and dry weight of tomato plants were higher in 3 fertigation treatments than irrigation of tap water, while there were no significant differences in fresh and dry weight among the 3 fertigation treatments. The mineral content of tomato leaf also did not show any differences among the 3 fertigation treatments and any regular tendency in all minerals. Total yield, fruit weight and fruit numbers of tomato were higher in WNS, followed by Megasol, BHF and control, although there were not any difference among the 3 fertigation nutrient solution treatments. BER(blossom-end rot)of tomato fruits decreased in fertigation treatments, especially, fruits grown in WNS and BHF showed lower BER. However, the transpiration rate of leaf was higher in control, followed by BHF, WNS and Megasol, The fruit size and soluble solids content was higher in 3 fertigation nutrient treatments than control. These results suggest that WNS can be used for fertigation solution in tomato because yield and quality of tomato fruit grown in WNS fertigation treatment were similar to those in 2 fertigation nutrient solutions treatments(BHF, Megasol).

Fruit Quality Characteristics of 'Fuji' Apple Fruits in Response to Air Storage after CA Storage (Controlled Atmosphere (CA) 환경설정 해제 후 저온저장이 '후지' 과실의 품질에 미치는 영향)

  • This study aimed to evaluate the fruit quality characteristics and incidence of flesh browning in response to air storage at $0{\pm}1^{\circ}C$ a fter controlled atmosphere s torage (CA condition: $O_2$ $2.5{\pm}0.5$%, $CO_2$ $1.5{\pm}0.5$%) at $0{\pm}1^{\circ}C$ in 'Fuji' apples (Malus domestica Borkh.). The storage system was performed as followed: air storage for one month, CA storage 4 months + air storage 3 months ( CA 4M + A ir 3M), CA storage 5 m onths + air storage 2 months ( CA 5M + Air 2M) a nd C A storage 6 m onths + air storage 1months (CA 6M + Air 1M), while the control fruits were stored at CA storage for 8 months right after harvest. The incidence of flesh browning ranged from 17.1% to 30.2% during CA storage but not detected under the treatments of CA 4M + Air 3M and CA 5M + Air 2M. The respiration rate was not affected by storage treatments for 6M while the respiration rate was lower in the treatments of CA 4M + Air 3M and CA 5M + Air 2M than the other storage treatments after 7 months. Ethylene production and internal ethylene concentration were lowest in rapid CA storage and increased with a decreasing CA storage duration. Therefore, the results indicate that CA 5M + Air 2M storage treatment should be recommended to maintain the fruit quality and reduce the risk development of flesh browning rather than typical CA storage in 'Fuji' apples.

Mutagenicity of residual pesticides using to cultivate the fruits and vegetables in the Salmonella typhimurium (과채류 재배에 사용되는 잔류성 농약의 돌연변이 유발성에 관한 연구)

  • Twelve residual pesticides which is applied mainly on the fruits and vegetables cultivation were tested for mutagenic activity using Salmonella typhimurium. The results were as follows. 1) The pesticides tested - Thalonil, Monopho, Tedion, Danoton, Ometon, Prosing, EPN, Phentoate, Parathion, 8appiran and Captan-, except Dicofol, showed mutagenic activity. Especially, the pesticides which activated with 8-9 mixture gave strong mutagenic activity. 2) With Salmonella typhimurium TA100, the pesticides Monopho, Parathion and Sappiran showed mutagenic activity at $0.5{\mu}g/plate;\;Ometon,\;0.01\; t.tl/plate;Thalonil,\;0.20{\mu}g/plate;\;Prosing,\; 0.20{\mu}l/plate;$ Tedion, Danoton and Phentoate, $1.0\;{\mu}l/plate;\;and\;Captan,\;1.0\;{\mu}g/plate.$ 3) With Salmonella typhimurium TA98, the pesticide Sappiran showed mutagenic activity at $0.02\;{\mu}g/plate;\;Thalonil,\;0.05\;{\mu}g/plate;\;EPN,\;0.05\;{\mu}/plate;\; Phentoate,\;0.10\; {\mu}l/plate;\;Danoton,\;0.50\;{\mu}l/plate;$ Prosing, $ 0.50\;{\mu}g/plate;\;and\;Tedion\;and\;Monopho,\;1.0\;{\mu}l/plate.$ 4) With Salmonella typhimurium TA1535, The pesticides Captan and Danoton showed mutagenic activity at $0.10\;{\mu}g/plate\;and\;0.20\;{\mu}g/plate,\;respectively.$ 5) With Salmonella typhimurium TA1538, The pestides Phentoate showed mutagenic activity at $0.05\;{\mu}l/plate;\;Monopho,\;0.10\;{\mu}l/plate;\;Thalonil,\;0.20 {\mu}g/plate;\;and\;Tedion\;and\;EPN,\;1.0\;{\mu}l/plate.$ 6) The pesticides which commonly showed high mutagenic activity were Thalonil, Phentoate, Parathion and Sappiran. The pesticides Ometon and EPN showed mutagenic activity at only TA100 and TA98, respectively.
