Study on Electric Stimulus Pattern in Cochlear Implant Using a Computer Model (신경모델링을 이용한 인공와우 전기자극 패턴 연구)

  • A cochlear implant system uses charge-balanced biphasic pulses that are known to reduce tissue damage than monophasic pulses. In this study, we investigated effect of pulse pattern on neural responses using a computer model, based on the Hodgkin-Huxley equation. Electric pulse phase, pulse duration, and phase gap have been systematically varied to characterize auditory nerve responses. The results show that neural responses, dynamic range and threshold are represented as a function of stimulus patterns and duration. The results could greatly extend to develop more efficient cochlear implant stimulation.

Improvement of osteogenic potential of biphasic calcium phosphate bone substitute coated with two concentrations of expressed recombinant human bone morphogenetic protein 2

  • Purpose: The aim of this study was to determine whether biphasic calcium phosphate (BCP) bone substitute with two different concentrations of Escherichia coli-expressed recombinant human bone morphogenetic protein 2 (ErhBMP-2) enhances new bone formation in a standardized rabbit sinus model and to evaluate the concentration-dependent effect of ErhBMP-2. Methods: Standardized, 6-mm diameter defects were made bilaterally on the maxillary sinus of 20 male New Zealand white rabbits. Following removal of the circular bony windows and reflection of the sinus membrane, BCP bone substitute without coating (control group) was applied into one defect and BCP bone substitute coated with ErhBMP-2 (experimental group) was applied into the other defect for each rabbit. The experimental group was divided into 2 subgroups according to the concentration of ErhBMP-2 (0.05 and 0.5 mg/mL). The animals were allowed to heal for either 4 or 8 weeks and sections of the augmented sinus and surrounding bone were analyzed by microcomputed tomography and histologically. Results: Histologic analysis revealed signs of new bone formation in both the control and experimental groups with a statistically significant increase in bone formation in experimental group 1 (0.05 mg/mL ErhBMP-2 coating) after a 4-week healing period. However, no statistically significant difference was found between experimental group 1 and experimental group 2 (0.5 mg/mL ErhBMP-2 coating) in osteoinductive potential (P<0.05). Conclusions: ErhBMP-2 administered using a BCP matrix significantly enhanced osteoinductive potential in a standardized rabbit sinus model. A concentration-dependent response was not found in the present study.

  • Experiments for the characterization of inactivation were performed in a series of batch processes with the total coliform used as a general indicator organism based on the chlorine residuals as a disinfectant. The water samples were taken from the outlet of a settling basin in a conventional surface water treat- ment system that is provided with the raw water drawn from the mid-stream of the Han River, The inactivation of total coliform was experimentally analysed for the dose of disinfectants contact time, filtration and mixing intensity. The curves obtained from a series of batch processes were shaped with a general tailing-off and biphasic mode of inactivation, i.e. a sharp loss of bacterial viability within 15 min followed by an extended phase. In order to observe the effect of carry-over suspended solids on chlorine consumption and disinfection efficiency, the water samples were filtered, prior to inoculation with coliforms, with membranes of both 2.5$\mu$m and 11.0 $\mu$m pore size, and with a sand tilter of 1.0 mm in effective size and of 1.4 in uniformity coefficient. As far as the disinfection efficiency is concerned, there were no significant differences. The parameters estimated by the models of Chick-Wat-son, Hom and Selleck from our experimental data obtained within 120 min are: log(N/N$\_$0/)=-0.16CT with n=1, leg(N/N$\_$0/)=-0.71C$\^$0.87/ with n 1 for the Chick-Watson model, log (N/N$\_$0/)=-1.87C$\^$0.47/ T$\^$0.36/ for the Hom model, log (MHo)=-2.13log (1+CT/0.11) for the Selleck model. It is notable that among the models reviewed with regard to the experimental data obtained, the Selleck model appeared to most closely resemble the total coliform survival curve.

  • A general theory of polymer adsorption at a semi-permeable oil-water interface of the biphasic solution is presented. The configurational factor of the solution in the presence of the semi-open boundary at the interface is evaluated by the quasicrystalline lattice model. The present theory gives the feature of the bulk concentration equilibria between oil-water subsystems and the surface excesses of ${\Gamma}^{\alpha}$ and ${\Gamma}^\{beta}$ of the polymer segments as a function of the degree of polymerization $\gamma$, the Flory-Huggins parameter in $\beta$-phase $x_{\rho}^{{\beta}_{\rho}}$, the differential adsorption energy parameter in $\beta$-phase $x_{\sigma}^{{\beta}_{\rho}}$, the differential interaction energy parameter ${\Delta}x_{\rho}$ and the bulk concentration of the polymer in ${\beta}-phase ${\varphi}_2^{{\beta(*)}_2}$. From our numerical results, the characteristics of ${\Gamma}^{\alpha}$ are shown to be significantly different from those of ${\Gamma}^{\beta}$ in the case of high polymers, and this would be the most apparent feature of the adsorption behavior of the polymer at a semi-permeable oil-water interface, which is sensitively dependent on ${\Delta}x_{\rho}$ and r.

  • The development of routine techniques for the isolation and in vitro maintenance of conducting airway epithelial cells in a differentiated state provides an ideal model to study the factors involved in the regulation of the expression of mucocilicary differentiation. Several key factors and conditions have been identified. These factors and conditions include the use of biphasic culture technique to achieve mucociliary differentiation and the use of such stimulators, the thickness of collagen gel substratum, the calcium level, and vitamin A, and such inhibitors, the growth factors EGF and insulin, and steroid hormones, for mucous cell differentiation. Using the defined culture medium, the life cycle of the mucous cell population in vitro was investigated. It was demonstrated that the majority of the mucous cell population in primary cultures is not involved in DNA replication. However, the mucous cell type is capable of self-renewal in culture and this reproduction is vitamin A dependent. furthermore, differentiation from non-mucous cell type to mucous cell type can be demonstrated by adding back a positive regulator such as vitamin A to the “starved” culture. Cell kinetics data suggest that vitamin A-dependent mucous cell differentiation in culture is a DNA replication-independent process and the process is inhibited by TGF-${\beta}$1.

  • Software and hardware for cochlear implant system have been developed to create a speech signal processing system which, in real-time, extracts model parameter including formants, pitch, amplitude information. The system is based on the Texas Instruments TMS320 family. In hardware, computer interface has been desisted and implemented that allows presentation of biphasic pulse stimuli to patients with the hearing handicapped. The host computer sends a stream of bytes to the parallel port. Upon receipt of the data the interface generates the appropriate burst sequence that is delivered to the patient's external transmitter coil. The coded information is interpreted by the Nucleus-22 internal receiver that delivers the pulse to the specified electrodes at the specified amplitude and pulse width.

  • Effect of interfacial electrical conditions such as, the f potential of PET fiber and u-Fe203 particles, the stability parameter and potential energy of interaction on adhesion of a-Fe903 particles to PET fabric and their removal from the fabric, were investigated as functions of pH, electrolyte and ionic strength. The stability parameter, potential energy of interaction between a-Fe2O3 particles and PET fabric were calculated by using the heterocoagulation theory for a sphere-plate model The adhesion of a-Fe2O3 particles to PET fabric and their removal from PET fabric were carried out by using water bath shaker and Terg-O-Tometer under various solution conditions. The adhesion of a-Fe2O3 particles to the PET fabric and the removal of a-Fe2O3 particles from the PET fabric were biphasic and were maximum and minimum at pH 7~8, respectively. With high pH and polyanion electrolytes in solution, the adhesion of a-Fe2O3 particles to the PET fabric was low but effects of electrolytes on the removal of a-Fe2O3 particles from the PET fabric was small. The adhesion of a-Fe2O3 particles to the PET fabric and the removal of a-Fe2O3 Particles from the PET fabric were biphasic, and were lowest and highest at the ionic strength 1$\times$10-3, respectively. The adhesion of a-Fe2O3 particles to PET fabric was well related with the interfacial electrical conditions; it was negatively correlated with the f potentials of a-Fe2O3 Particles of its absolute value, the stability parameter and the maximum of total potential energy, while, the adhesion was not related with the t potentials of PET fiber itself. Therefore, the primary factor determining the adhesion of a-Fe203 particles to PET fabric may be the stability of dispersed particles caused by the electrical repulsion of particles. The removal of a-Fe203 particles from PET fabric was not related to such interfacial electrical conditions as the t potentials of PET fiber, the stability parameter and the maximum of total potential energy but removal was related to t potential of a-Fe203 particles.

Distinctive bone regeneration of calvarial defects using biphasic calcium phosphate supplemented ultraviolet-crosslinked collagen membrane

  • Purpose: To overcome several drawbacks of chemically-crosslinked collagen membranes, modification processes such as ultraviolet (UV) crosslinking and the addition of biphasic calcium phosphate (BCP) to collagen membranes have been introduced. This study evaluated the efficacy and biocompatibility of BCP-supplemented UV-crosslinked collagen membrane for guided bone regeneration (GBR) in a rabbit calvarial model. Methods: Four circular bone defects (diameter, 8 mm) were created in the calvarium of 10 rabbits. Each defect was randomly allocated to one of the following groups: 1) the sham control group (spontaneous healing); 2) the M group (defect coverage with a BCP-supplemented UV-crosslinked collagen membrane and no graft material); 3) the BG (defects filled with BCP particles without membrane coverage); and 4) the BG+M group (defects filled with BCP particles and covered with a BCP-supplemented UV-crosslinked collagen membrane in a conventional GBR procedure). At 2 and 8 weeks, rabbits were sacrificed, and experimental defects were investigated histologically and by micro-computed tomography (micro-CT). Results: In both micro-CT and histometric analyses, the BG and BG+M groups at both 2 and 8 weeks showed significantly higher new bone formation than the control group. On micro-CT, the new bone volume of the BG+M group (48.39±5.47 ㎣) was larger than that of the BG group (38.71±2.24 ㎣, P=0.032) at 8 weeks. Histologically, greater new bone area was observed in the BG+M group than in the BG or M groups. BCP-supplemented UV-crosslinked collagen membrane did not cause an abnormal cellular reaction and was stable until 8 weeks. Conclusions: Enhanced new bone formation in GBR can be achieved by simultaneously using bone graft material and a BCP-supplemented UV-crosslinked collagen membrane, which showed high biocompatibility and resistance to degradation, making it a biocompatible alternative to chemically-crosslinked collagen membranes.

  • To maximize the performance of recombinant cell fermentation process through optimizing environmental conditions, the production of invertase from recombinant Saccharomyces cerebisiae Containing SUC2 gene was studied as a model. The recombinant cells showed biphasic growth on glucose. Since the promoter of the SUC2 is regulated by the concentration of glucose in the medium, expression of invertase by recombinant yeast began when the glucose concentration decreased in a range of 0.25-0.4 g/L during the batch culture. Plasmid segregation occured frequently during glucose fermentation, and infrequently during ethanol oxidation. A rapid appearance of invertase activity with glucose was observed under nonaerated condition, and the maximum specific invertase activity was about 1.5 times as high as under aerobic condition, In fed batch culture, when n low level of glucose was continuously supplied to the tormentor after the time of glucose depletion during growth phase, specific and total invertase activity increased about 1.7 and 2.9 fold, respectively, in a batch culture.

  • 오염 퇴적물내 중금속의 방출과 이에 따른 생이용성(bioavalability)은 기존의 가역 평형관계로써 설명하기에 불충분한 것으로 알려져 있다. 최근 연구결과에 의하면 이러한 탈착 저항성을 설명하기 위한 비가역적 모델에 의해 퇴적물내 중금속의 탈착저항성 부분이 존재함이 밝혀졌다. 탈착저항성에 대해서는 아직 충분한 규명이 이루어지지 않았으나, 오염물질의 노화(aging)에 의해 일단 탈착저항성을 띠게 되면 생이용성(bioavailability)이 감소되는 것으로 알려져 있다. 본 연구에서는 노화(aging)가 연안 퇴적물 내 납과 카드뮴의 흡/탈착 특성에 미치는 영향을 알아보기 위해 연속 탈착실험과 biphasic 탈착모델을 적용함으로써 납과 카드뮴의 탈착저항성을 규명하고자 하였다. 그리고 연속 추출 실험을 통해서 노화(aging)에 따른 퇴적물 내 납과 카드뮴의 흡착 기작을 규명하고자 하였다. 연속탈착 실험 결과 시간이 경과함에 따라 탈착저항성부분의 크기가 증가하였으며, 연속추출 실험 결과 납의 경우 carbonate fraction에서 추출된 납이 가장 많았으며, 노화(aging) 따라 exchangeable fraction에서 추출된 납이 감소하는 반면 reducible, organic material, residual fraction에서 추출된 납의 양이 증가하였다. 카드뮴의 경우 가장 많은 양이 추출된 단계는 exchangeable fraction이였으며, organic material fraction에서는 카드뮴이 추출되지 않았다. 노화(aging) 따라 reducible fraction과 residual fraction에서의 추출량이 증가하는 경향을 보였으나 그 양은 매우 적은 것으로 나타났으며, 노화(aging)에 따른 exchangeable fraction과 carbonate fraction에서의 추출량은 큰 변화가 없었다. 대해서는 북한지역의 분포상황을 밝혔다.것을 알 수 있었으며, 크롬과 비소의 경우는 초기에 많이 용출되고, 구리의 경우는 꾸준히 용출되는 것을 알 수 있었다. 3년 된 통나무집이 8년 된 통나무집보다. 용출양이 더 컸으며, 이는 CCA성분이 초기에 많이 용출된다는 것을 의미한다. 억제 효과를 나타내었고 Hep3B에서는 부탄을 분획물 (1 mg/mL)에서 82%의 비교적 높은 성장억제효과를 나타내었다.as a "front" and "back".
