Enhanced Progression from Lung Adenoma to Adenocarcinoma in Progeny by Prozygotic Testicular X-ray Exposure of Mice

  • Testicles of nine-week-old male CBA/J mice were X-ray irradiated (1 Gy or 2 Gy) and were mated one week later with untreated virgin 12-week-old females of the same strain. The 1-Gy offspring (88 males and 62 females), 2-Gy offspring (100 males and 93 females) and additional offs pring (83 males and 84 females) were treated once subcutaneously with 0.1 mg/g body weight of urethane at 6 weeks of age. These three groups of off offspring showed similar incidences of lung tumors in both sexes. Depending on the doses of paternal X-ray irradiation, increasing incidences of adenocarcinoma were observed in the male 1-Gy and 2-Gy offspring groups. An increased multiplicity of lung carcinomas was observed in the male 2-Gy progeny that was statistically significant when compared with the control group. The results indicate that prezygotic testicular X-ray exposure of patemal animals causes the shift of adenoma-carcinoma sequence towards malignancy in the progeny.

  • Epidermoid cysts can be found anywhere in the body, particularly in areas where embryonic elements fuse together. Most cases have been reported in the ovaries, the testicles, as well as the hand and feet. Epidermoid cysts in mouth are uncommon and account for less than 0.01% of all oral cysts. The treatment of choice is complete surgical excision and prognosis is excellent. The purpose of this article is to describe two cases of an epidermoid cyst in the oral cavity mucosa and a review of the literature.

A Study on the Effect of "ADAPTAGEN"$^{\textregistered}$ Korean Ginseng Components, for the Injured Mouse by X-ray($^{60}Co$) Irradiation (X-방사선($^{60}Co$)에 조사된 새앙쥐의 상해에 대한 "아답태겐"$^{\textregistered}$의 효과에 관한 연구)

  • The results ok feeding experiments to the mice with ginseng extract, ginseng Powder, and ADAPTAGEN, for 30 days before X-ray irradiation and for 40 days after the X-ray irradiation at 750 rads were as follow: 1. The 50% lethals days (LD50, ) by the X-ray irradiation were 9 days at 1, 000 rads. 10 days at 900 rads, 11 days at 800 rads, 14 days at 760 rads, and 19 darts at 750 rads. Therefore, the standard radiation dose was set at 750 radb/8 min. 2. The 80% of the control group mice exposed to the X-ray radiation without ginseng feeding died in periods ranging from 14 to 24 days and the 20~30% of the ginseng extract and ginseng powder feeding groups died. But the 100% of the mice fed with ADAPTAGEN survived. 3. Testicles of the control group became smaller in weight than the nomad group by 26.5 to 29.0% and those of the ginseng extract and ginseng powder feeding group reduced by 44.6 to 60.4%. However, testicles of the ADAPTiIGEN feeding group increased in size by 77.4% to 87.1% and in weight by 61%, showing a recovery phenomenon approarhing to those of the ordinary mice. The ADAPTAGEN feeding group mice were also as active in color as the ordinary ones. 4. An electron micrograph(X8, 000X2.2) of the liver cells of the mice which had been 40 days after X-ray irradiation showed as follows; The control group appeared that is physiological action stopped due to the frequent occurrence of morphological change of the nucleus and diffusion of chromosome, reduction in microspores and expansion of microsomts, and endoplasmic change of mitochondria. The liver cells or the ADAPTAGEN feeding group were in a state similar to those of the ordinary mice restoring to normalcy In contrast, the liver cells of the ginseng extract and ginseng powder feeding groups were still far from being normal. 5. A serological analysis showed that the control group sharply decreased in albumin, Y-g1obu1in, and IgG so far as to cause dystrophy and to weaken antibody resistance but that ginseng extract and ginseng powder feeding groups, though in a little more restoring state than the control group, were still far from the normal group. The ADAPTAGEN feeding group restored to a state as comparable to the normal group in the contents of albumin ${\gamma}$-globulin, IgG and serum protein. In order words, it is noteworthy that ADAPTAGEN feeding was effective in revitalizing the destroyed cells of a living body and that it has the function of normalizing antibody components.

Half-castration is a newly effective method for increasing yield and tenderness of male cattle meat

  • Objective: For improving meat quality especially tenderness, male cattle are usually castrated to removes both the testicles. This study was conducted to evaluate the effect castration method (half- and complete-castration) on meat yield and quality characteristics of Hanwoo male cattle. Methods: Thirty-two similar age (5.9 months) Hanwoo male calves were divided into: half-castration (HC) and complete-castration (CC) groups (n = 16 per group). At 7 months of age, all the animals were castrated in which the HC calves had only one testicle surgically removed while, the CC calves had both testicles surgically removed. The castrated animals were reared under identical conditions until 25 months of age. After slaughter, the carcasses were evaluated for carcass traits and meat yield of primal cuts. For examination of the castration effect on meat quality, L. lumborum and semimembranosus muscles were used. The meat samples were analyzed for chemical composition, color, pH, shear force and water holding capacity, fatty acids, metabolites and volatile aroma compounds. Results: The HC group showed higher meat yields of all primal cuts (p<0.05). As a result, the total meat yield was higher by approximately 44 kg in the HC group (303.32 kg, corresponding to 67.88%) compared to the CC group (259.30 kg, corresponding to 62.11%) (p<0.05). In terms of meat quality, the HC resulted in two times greater fat content in both muscles examined compared to intact males. More importantly, the shear force values did not differ between HC and CC groups for L. lumborum muscles (p>0.05). The meat from HC animals exhibited higher amount of free amino acids associated with sweetness (p<0.05). Furthermore, the castration method only exhibited a negligible effect on metabolites and volatile aroma compounds in the cooked meat. Conclusion: Half-castration emerged as an alternative practice to be used for increasing the yield and tenderness of male cattle meat.

Lipoid Congenital Adrenal Hyperplasia Diagnosed in an Infant with Hyperpigmentation Only by Targeted Exome Sequencing

  • Lipoid congenital adrenal hyperplasia (LCAH) is the severe form of congenital adrenal hyperplasia and is characterized by adrenal insufficiency with hyperpigmentation and female external genitalia irrespective of genetic sex. The steroidogenic acute regulatory protein (StAR) is required for the transport of cholesterol into the mitochondria for steroidogenesis, and defects in the StAR gene account for the majority of LCAH cases. In this report, we present a two-day-old hyperpigmented infant with phenotypical female genitalia. With consideration of the clinical and laboratory findings, the infant was suspected of having adrenal insufficiency due to LCAH and treated with glucocorticoid, mineralocorticoid, and sodium chloride. Karyotyping revealed 46, XY. Upon pelvis ultrasonography, adrenal hyperplasia with abdominal masses (thought to be the testicles) was reported. Molecular analysis with targeted exome sequencing revealed the homozygote mutation of c.772C>T ($p.Q258^*$) in exon 7 of the StAR gene. The early detection and treatment of adrenal insufficiency in infants with hyperpigmentation can prevent clinically apparent adrenal crises. During follow-up, the patient had a good clinical condition and maintained normal electrolyte and adrenocorticotropic hormone levels with medication.

  • A wide ranges of chemicals released into the environment have potential to interfere with physiological and development process by disrupting endocrine pathways. Endocrine system embraces a multitude of mechanisms of action, including effect on growth, behavior, reproduction and immune function. These environmental endocrine disruptors are present in environment and pose potential health consequences to human and wildlife. The best known form in endocrine distruptors involves substances which mimic or block the action of natural hormone in the body. Endocrine disruptor have been variously defined as exogenous agents that interfere with the synthesis, secretion, transport, metabolism, binding action or elimination of the natural hormones in the body which are responsible for the maintenance of homeostasis, reproduction developmental and/or behavior. Many compounds polluted into the environment by human activity are capable of disrupting the endocrine system of animals, including fish, wildlife, and humans. Among these chemicals are pesticides, industrial chemicals, and other anthropogenic products. It has been alleged that several adverse effects on human health are linked with exposure to chemicals which are claimed to be endocrine disrupters, that is, increased incidence of testicular, prostate and female breast cancer, time dependent reductions in sperm quality and quantity, increased incidence of cryptorchidism (undescended testicles) and hypospadias(malformation of the penis), altered physical and mental de velopment in children. This observation is currently the only example of chemically mediated endocrine disruption which has resulted in a clear effect at the population level.

A Study on the Reproduction of the Sunrise Sculpin Pseudoblennius cottoides in Korea (한국산 가시망둑(Pseudoblennius cottoides)의 생식에 관한 연구)

  • The ecological characteristics of Pseudoblennius cottoides were investigated off Dolsan Island, Yeosu, Korea, from June to December, 2001. Their habitats were the reef zone and surrounding embankment which was covered with seaweeds. Their range of total length (TL) was 6.10-8.40 cm (n=15), and the individuals (over 8.10 cm) of them accounted for in June. It was 8.60-15.7 cm (n=67), and the individuals 11.1-12.0 cm in TL accounted for in October, 2001. The female to male sex ratio was 1:0.48 (n=185). The gonads were in the shape of a rod and the ovaries were large in size. In June, the histology of the seminiferous tubules was observed. In August, the testicles appeared. In October, spermatocyte was observed in the testes, and ovaries were observed in relation to circumference popularity and oocyte.

  • Sertoli cell tumor (SCT) of the testicle arises from the supporting cells within the seminiferous tubules. SCT is common in dogs, especially in cryptorchid testicles, but also has been reported in the stallion, ram, cat, and bull. Sertoli cell tumor sample was collected from 7-years male german shepherd. In this study, SCT arose from one testicle. Sample size is approximately 1.7 cm in diameter and it has a round form. In the microscopic, cells within the tumor variably resemble Sertoli cells (SCs) that normally populate the seminiferous tubules and interstitial area. There is abundant stroma of dense, mature fibrous connective tissue in SCT. In the immunohistochemical staining, cytokeratin AE1/AE3 was not expressed in the control and SCT. S-100 protein was expressed by SCs, germ cells and fibrous connective tissue of SCT. Melan A was expressed by leydig cells (LCs) of SCT. A study by using S-100 and melan A in canine SCT was almost never carried out. S-100 and melans A is considered to suggest for diagnosis and pathogenesis of canine SCTs. Inhibin-alpha and Vimentin were well known as the marekers of SCTs. Also, they were expressed by Sertoli cells and LSs of SCT in this study.

Comparison of Spermatozoa Recovery Methods on Cauda Epididymal Sperm of Hanwoo Bulls

  • In this study, two epididymal spermatozoa recovery methods in relation to sperm number, motility, viability and acrosome reaction were examined. Seven bulls were castrated and 7 testicles with epididymides were transferred to the laboratoy. Epididymis in each bull was randomly used for flushing and mincing methods with semen extender (Optixcell, IMV, France). The recovered spermatozoa with adjusted sperm concentration to $40{\times}10^6cells/mL$ was diluted with optixcell and cryopreserved. In experiment 1, the difference in the total number of spermatozoa using flushing and mincing methods was insignificant (2570.0 and $2505.2{\times}10^6cells/mL$, respectively). For experiment 2, the percentage of motile spermatozoa and motility parameters between flushing and mincing methods were studied through the use of sperm class analyzer after frozen-thawing. The percentage of total motile sperm between flushing and mincing methods was almost the same with $89.5{\pm}12.8$ and $91.4{\pm}7.9%$, respectively. The same is the case with experiment 3 wherein the viability and acrosomal integrity of frozen-thawed epididymal spermatozoa by flushing and mincing was insignificantly different. The results from the study showed that both flushing and mincing methods can be used for epididymal spermatozoa recovery in bull.

Effect of Cassia tora L. Powder Added-Diets on the Accumulation of Cadmium in Rat (결명자 첨가식이가 흰쥐의 체내 카드뮴 축적에 미치는 영향)

  • The purpose of this study is to investigate the effect of raw Cassia tora L. powder added-diets on reducing cadmium accumulation in rats. The experimental animals were Sprague-Dawley family(♂, 4 weeks) which was classified into normal group CN, compared group CS, Cd-added group Cl and groups C2, C3, C4 in which 0.5, 1.0 and 1.5% of the Cassia tora L. powder are added, respectively. The growth rate and food efficiency ratio, and the amounts of accumulated cadmium in rats for S weeks were measured and analyzed. The results are as follows; 1. The rates of weight gain decreased in the order of C3>C2>C4>Cn>Cs>Cl groups, and Cl group to which only cadmium water had been fed was the lowest among them. The correlation between groups Cl and C3 was significantly different at the 1% level. 2. Food efficiency ratio(FER) decreased in the order of C3>C2>Cs>Cn>C4>Cl, and the FERs of C3, C2, CS, CN and C4 are greater than that of Cl by 22.87, 19.59, 18.54, 14.20 and 13.17%, respectively. 3. As for the Cassia tora L. powder-added groups, the amounts of cadmium accumulated in organs and tissues, that is, the brain, heart, spleen, liver, lungs, testicles. kidney, femoral muscle and leg bones were 0.45 $\pm$ 0.04 to 0.83$\pm$0.04, 1.68$\pm$0.02 to 2.16$\pm$0.02, 3.26$\pm$0.05 to 4.62$\pm$0.27, 37.52$\pm$0.09 to 47.71$\pm$0.73, 1.07$\pm$0.10 to 1.66$\pm$0.04, 1.04$\pm$0.06 to 1.24$\pm$0.08, 36.79$\pm$0.20 to 39.61 $\pm$0.53, 0.87$\pm$0.02 to 1.00$\pm$0.02 and 0.65$\pm$0.17 to 1.27 $\pm$ 0.06 $\mu\textrm{g}$/g, respectively. 4. The accumulated Cd content for C4 was the lowest among Cassia tora L. powder-added groups. When the results for C4 are compared with those for Cl, it is observed that each cadmium content accumulated in the brain, heart spleen, liver, lungs, testicles, kidney. femoral muscle and leg bones is dropped by 49.03, 22.56, 36.02, 35.75, 41.75, 36.20, 37.00, 22.77 and 56.67 %, respectively.
