Growth Characteristics and Economic Efficiency of Nursery Plants Production According to Transplanting Container for Acclimatization of Mass Propagated Plantlets via Bioreactor Culture of Ever-bearing Strawberry 'Goha' (생물반응기를 통해 대량증식된 사계성 딸기 '고하' 소식물체의 순화용기에 따른 생육특성과 묘생산 경제성)

  • This study was conducted to find out the optimum container for increasing acclimatization rate of in vitro mass propagated plantlets of Ever-bearing strawberry (Fragaria ${\times}$ ananassa Duch.) via bioreactor. Four types of containers were used such as transparent plastic container (TPC), plug tray (PT), I-pot (IP), and black vinyl pot (BVP). Number of date maintaining soil water content above 10% was five days in TPC, three to four days in BVP, two days in PT, and one day in IP. Survival rate of plantlets was 80% in BVP, 70% in TPC, 55% in IP, and 15% in PT. In TPC, growth increment of plantlets was the greatest among all the tested containers and the lowest in IP. Numbers of runner per plant were 3.3 in BVP, 2.9 in TPC, 1.6 in PT, and 1.2 in IP. Total cost was 44,405,300 won/10 a in BVP, resulting in reducing more 6,659,400 won/10 a than IP's (51,064,700 won/10 a). Around 102,718 plants/10 a were produced by using BVP, suggesting that 30,265.1 plants/10 a more could be produced than IP (72,452.9 plants/10 a). Production cost per plant was 432.3 won in BVP, resulting in reducing 272.5 won than IP's (704.8 won). As a result, BVP was appropriate for acclimatization of in vitro plantlets through bioreactor system.

Microbiological Hazard Analysis on Perilla Leaf Farms at the Harvesting Stage for the Application of the Good Agricultural Practices(GAP) (깻잎의 농산물우수관리제도(GAP) 적용을 위한 수확단계에서 미생물학적 위해요소 분석)

  • The purpose of this study was to analyze microbiological hazards for plants, cultivation environments and personal hygiene of perilla leaf farms at the harvesting stage. Samples were collected from three perilla leaf farms(A, B, C) located in Gyeongnam, Korea and tested for sanitary indications, fungi and pathogenic bacteria(Escherichia coli O157:H7, Listeria monocytogens, Salmonella spp., Staphylococcus aureus and Bacillus cereus). As a result, total bacteria and coliform in perilla leaf were detected at the levels of 4.4~5.2 and 3.4~4.3 log CFU/g, respectively, but E. coli was not detected in all samples. Among the pathogenic bacteria, B. cereus(perilla leaf: 2.0~2.4 log CFU/g, stem: 1.4~2.1 log CFU/g, water: 0.7 log CFU/ml, soil: 4.2~5.0 log CFU/g, hands: 3.0 log CFU/ hand, gloves: 2.1~2.4 log CFU/100 $cm^2$, glothes: 1.5~2.8 log CFU/100 $cm^2$) and S. aureus(3.4 log CFU/hand) were detected in all samples and worker's hand from farm A, respectively. However, other pathogenic bacteria were not detected. This study demonstrates that perilla leaf at the harvesting stage was significantly contaminated with microbial hazards.

Evaluation of Sanitary Safety for Shellfish in Hansan·Geojeman, Korea (경남 한산·거제만해역에서 생산된 패류의 위생학적 안전성 평가)

  • To evaluate bacteriological and toxicological safety hygienic indicator bacterium and paralytic and diarrhetic shellfish toxins in the shellfish produced in Hansan Geojeman 2013-2017 were investigated. Fecal coliforms were < 18~330 MPN/100 g in 404 oyster samples. But all samples tested, did not exceed 230 E. coli MPN/100 g. Geometric mean of E. coli for oyster samples collected during major shellfish production period was 24.3 MPN/100 g, considerde stable results. Bacteriological quality of oysters collected from Hansan Geojeman meets the standard value based on shellfish hygiene of the Food Sanitation Act of Korea and also meets Grade A, according to classification of shellfish harvesting areas of the European Union. For toxicological evaluation of Hansan Geojeman, 532 oyster samples and 268 mussel samples as an indicator, were analyzed. Paralytic shellfish toxins were detected in the range of 0.42~2.29 mg/kg in eight mussel samples, and exceeded criteria in three samples from early to late April 2013. Diarrhetic shellfish toxin was detected in three of 120 samples, but it was revealed to be under regulation value (0.16 mg Okadaic Acid equ./kg). As a result of toxicological evaluation, paralytic and diarrhetic shellfish toxins were not detected in oyster samples, but it was found that mussel as an indicator species, exceeded the threshold value of paralytic shellfish toxin. Accordingly, sanitary surveys were continuously requested for food safety management of shellfish.

Studies on the Quality and Utilization of Pumpkin Silages (호박 Silage의 품질(品質) 및 이용성(利用性)에 관(關)한 연구(硏究))

  • The preservation efficiency, quality and utilization of silage from 3 species of pumpkins (Mammoth pumpkin, Queensland blue pumpkin, Korean pumpkin) without and with 10, and 20% wheat bran additive were studied in this experiment. Silages were analysised and tested the chemical composition, pH and quality of silages between at 40-60 days and egg performance were carried out with mammoth pumpkin silage without additive. The results were summaried as follows. 1. The losses of all silage ware lower and similar as about 15% at 6 monthes following after silage-making but all raw pumpkins were spoilaged during the winter storaging. 2. The moisture content of silages were higher as about 97% in mammoth pumpkin silage, 94% in Queensland pumpkin silage and 91% in Korean pumpkin silage without additive and all nutrient content of silage without and with additive were depended on its content of raw silage material of pumpkins and wheat bran. The contents of moisture and N-free extract were slightly decrease but not significantly difference during the silaging and other contents were not so much changed. 3. Good quality of silage were made from all pumpkins with and without additive. Organic acid contents were 2.09-2.93% of lactic acid, 0.68-1.71% of acetic acid and 0% of butyric acid and it was pH 3.8-4.0 in silages. 4. Feed intakes, egg production and quality of egg were showed good result in 5.0 and 7.5% silage feeding group as D.M. base for egg performance. (P<0.01) 5. It was concluded that good quality of silage were made from pumpkins with and without wheat bran additive and it was suggested that poor quality feedstuff may be improved it feeding value by extended palatability with pumpkin additive silage.

Influence of Early- and Late-feathering Phenotype on Productive Performance in the Feather-sexing Strains of Korean Native Chicken (한국재래닭 깃털 성감별 계통에 있어 조우성과 만우성이 개체의 생산능력에 미치는 영향)

  • The vent sexing and the auto-sexing by using sex-linked traits are general sexing methods of day-old chicks. Currently, the feather sexing which is based on the differences in the feather characteristics at hatching is the representative sexing method of chicken, because the late-feathering is sex-linked trait. The feather sexing can be used if the breed has dominant feathering gene (K) in maternal and recessive gene ($k^+$) in paternal. Therefore it is necessary to identify the association of feathering genes and quantitative traits in chickens. In this study, we investigated the influence of the rate of feathering on productive traits in Korean Native Chicken. In results, there was no significant difference between early-feathering chickens and late-feathering chickens in reproductive performance such as fertility and hatchability. Livability, body weights, egg production, egg weight and egg quality also did not significantly differ between early- and late-feathering chickens. Age at first egg was the only trait of those tested in which significant difference was observed. The early-feathering chickens laid eggs 3 days earlier than late-feathering chicken. As a result, there is no influence of feathering phenotypes on productive performance in Korean Native Chickens. Consequentially, establishing the feather sexing strain is available using the Korean Native Chicken breed without considering of the effect of feathering genes on productive traits.

Flexural Properties according to Change of Polymerization Temperature of Autopolymerized Resin for Orthodontic (치과 교정용 자가중합형 Resin의 중합 온도 변화에 따른 굽힘 특성)

  • For this experiment, specimen was manufactured by injecting polymer and monomer into silicon mold with volume ratio of 2.5:1 based on ISO 20795-2 so that average thickness, width and length of specimen would be maintained as 3.3 mm, 10.0 mm and 65.0 mm, respectively depending on spray on technique. Specimen was divided into 3 groups ($25^{\circ}C$, $40^{\circ}C$, $70^{\circ}C$) depending on polymerization temperature and 10 specimen was manufactured for each group and it was polymerized in water tank of ${\pm}1^{\circ}C$ under the setting condition of polymerization time of 15 minutes and pressure of 3 bar. After keeping specimen in distilled water of $37^{\circ}C$ for over 48 hours before experiment, flexural strength (FS) and elasticity modulus (EM) of specimen being tested by using Intron (3344; Instron; Instron). SPSS ver. 16.0 was used for analysis and post-hoc test of Scheffe was performed after using one-way ANOVA. When comparing mean value of FS of resin for orthodontics, it was represented in the range of 71.500 MPa for $25^{\circ}C$ group, 74.920 MPa for $40^{\circ}C$ group and 76.880 MPa for $70^{\circ}C$ group and difference was shown in the order of $25^{\circ}C$ group <$40^{\circ}C$ group <$70^{\circ}C$ group but such difference was not significant statistically (p=0.052). Result of EM mean value of resin for orthodontics was more polymerization temperature was high, the more was significant difference represented in the order of $25^{\circ}C$ group <$40^{\circ}C$ group <$70^{\circ}C$ group (p<0.039).

Cold-Sensitive Growth of Bacillus subtilis Mutants Deleted for Putative DEAD-Box RNA Helicase Genes (Bacillus subtilis DEAD-Box RNA Helicase 유전자 결손 균주들의 저온 민감성 생장)

  • Four genes (yqfR, yfmL, ydbR, deaD) were identified as putative DEAD-box RNA helicase genes in the genomic sequence of Bacillus subtilis by homology search. To understand the function of these genes, each of the genes was deleted and the constructed strains were tested for their growth charateristics at different temperatures. The growth rate of ydbR deletion mutant ($T_d$=53 min) was a little bit reduced at $37^{\circ}C$ as compared to that of wild type strain (CU1065). But the growth rate of other three (yqfR, yfmL, deaD) deletion mutants ($T_d$=30-40 min) is nearly equal to the growth rate of wild type ($T_d$=32 min). On the other hands, the growth rate of deletion mutants were reduced at $22^{\circ}C$ in order of yqfR ($T_d$=151 min), yfmL ($T_d$=214 min), ydbR ($T_d$=343 min), which showed cold-sensitive phenotype. The deletion mutant of deaD ($T_d$=109 min) grew equally as compared to the growth rate ($T_d$=102 min) of the wild type at $22^{\circ}C$ and did not show cold-sensitive growth. Double, triple and quadruple deletion mutants of these genes were constructed, and growth rate of these mutants were measured at various temperature conditions ($22^{\circ}C$, $37^{\circ}C$, $42^{\circ}C$) using LB broth. Multiple deletion mutations showed more severe cold-sensitive growth than single deletion mutations, and double deletion of ydbR and yfmL ($T_d$=984 min) showed most cold-sensitive growth than any other double mutants. Such a cold-sensitive growth of these mutations is quite similar to the result of csdA or srmB deletion in E. coli and suggested that physiological role of ydbR and yfmL is related with ribosome assembly.

Novel Method for Urinary 1-Hydroxypyrene Measurement Using Molecular Imprinting (분자주형을 이용한 요중 1-hydroxypyrene의 측정 방법 개발)

  • This study was performed to determine whether or not urinary 1-hydroxypyrene (1-OHP) levels can be accurately detected by our 1-OHP-detecting $TiO_2$-Bead-HPLC assay that we developed based on the molecular imprinting method. Our method showed a variation coefficient of 4.97% and a between-day variation coefficient of 4.43%, suggesting that this may be a very stable method. In addition, the recovery rate of 1-OHP from a mixture of 1-OHP and similar substances using our $TiO_2$-Bead-HPLC method was estimated to be 105.6%. The correlation coefficient between the conventional enzyme-HPLC method and this new method was 0.74 (p<0.01) when the urine samples were tested. Based on this result, it is conceivable that our method could be a useful technique for measuring urinary 1-OHP levels. Moreover, our method has some advantages of being easier and less expensive than the conventional method. The results of this study suggest that our method can facilitate the development of a urine 1-OHP sensor using $TiO_2$-coating beads and that development of beads by molecular imprinting can be applied to analysis of chemicals other than 1-OHP.

Verification of ET and AI Derived Offspring Using on the Genetic Polymorphisms of Microsatellite and Coat Color Related Genes in Jeju Black Cattle (제주흑우 집단에서 모색 관련 유전자와 microsatellite marker의 다형현상을 이용한 수정란이식 및 인공수정 유래 후대우 검증)

  • To find offspring of Jeju Black cattle (JBC) produced by embryo transfer (ET) and artificial insemination (AI), a molecular genetic study was carried out in candidate cattle populations collected from cattle farms in Jeju Island, Korea. The genetic marker set was composed of 11 ISAG microsatellite (MS) markers, 11 SAES MS markers selected by our preliminary analysis for population diversity of JBC and two major coat color related genes: MC1R and ASIP. The results showed a combined non-exclusion probability for first parent (NE-P1) that was higher than that recommended by ISAG (above 0.9995), and a combined non-exclusion probability for sib identity of $5.3{\times}10^{-10}$. Parentage analysis showed that the cases identified the candidate's father only (77.0%), mother only (54.0%), and both parents (40.5%) in the candidate offspring population. The ET and AI calves were identified as 14.7% in the in vitro fertilized eggs provided and 32.4% in total population, respectively. However, the result from ISAG marker analysis showed 3 identical allele-combinations in 7 calves, and that from ISAG/SAES MS marker combination also showed 1 identical allele-combination in 2 calves. Data from MS and coat-color gene analyses provided information for complete identification of all animals tested. Because the present JBC population was mostly bred using small nuclear founders through bioengineering techniques such as AI and ET, the genetic diversity levels obtained from MS analysis in the JBC population were relatively lower than those of other cattle populations, including Hanwoo. The results suggested that the more efficient marker combinations, including coat color related genotypes, should be studied and used for constructing a system for identification and molecular breeding of JBC as well.

The Characteristics of Sediment and Organic Content in the Dalpo Wetland (달포늪의 퇴적물과 유기물함량 특성 연구)

  • In this study, the correlation of organic content with particle size and type of sediment was found out.Particle size, stratigraphic section and organic content of sediments sampled from Dalpo wetland was analyzed. Dalpo wetland consists of three wetlands, and the area of Dalpo wetland is about $31,295m^2$. The particle size analyses for sampled sediments of 7 points (3 points in wetland A, 3 points in wetland B and 1 point in wetland C) were tested. As results of the particle size analyses, the sediment particle size becomes larger as to the edge of the wetland. It is revealed in order of wetland A > wetland C > wetland B. Borehole surveys with horizontal distance in the major and minor axes of wetland A, the major and minor axes of wetland B and the major axis of wetland C were accomplished. Clayey peat deposit is distributed at 10~90 cm depth below ground surface in the major axis of wetland A. The clayey peat deposit was the most thick at the center of wetland A that horizontal distance is 100 m. As the depth below ground surface of clayey peat deposit is less than 27 cm in the wetland B, we can infer that the life for the wetland B is being finished. Sediment composition of wetland C is simple because wetland C is small scale, and clayey peat deposit is distributed at 10~34 cm depth below ground surface. Sediment sampled by borehole survey in the Dalpo wetland was cut at interval of 10 cm, then organic content was analyzed. Organic content of wetland A sediment showed more than 40% until 70 cm depth below ground surface, also sediment of wetland B is similar to wetland A until 10 cm depth below ground surface, but is showed within 20 % above 30 cm depth below ground surface. Organic content of wetland B is showed the lowest as organic content near the ground surface is about 40%. All of the three wetlands, organic content is showed higher at clayey peat deposit near to ground surface. This is caused by finer particles of the clayey peat deposit, also organic materials were supplied from dead vegetation. Organic content of the Dalpo wetland showed in order of wetland A > wetland C > wetland B. This result is caused by thickness of clayey peat deposit in sediment. Through this study, it was verified that organic content of the Dalpo wetland sediment was dominated by particle size of sediment and vegetation of the upper part.
