Resistance Characteristics of Chinese Cabbage Cultivars to Black Rot (배추 품종들의 검은썩음병에 대한 저항성 특성)

  • Black rot of Chinese cabbage caused by Xanthomonas campestris pv. campestris (Xcc) is one of the most severe diseases in crop cultivation. To define the resistance characteristics of Chinese cabbage to Xcc, we tested the virulence of eight Xcc isolates in four susceptible cultivars of Chinese cabbage. The isolates of Xcc showed different the virulence on the cultivars. On the other hand, we selected 22 resistant or moderately resistant cultivars of Chinese cabbage to Xcc and tested the occurrence of black rot on the cultivars caused by the isolates of Xcc. Mean disease severity of black rot on the Chinese cabbage caused by each isolate was also positively correlated with the virulence of Xcc isolates. Furthermore, the development of black rot in each cultivar increased according to virulence of Xcc isolates. The number of resistant cultivars of Chinese cabbage to eight isolates of Xcc decreased according to the virulence increase of bacteria. Taken together, these results suggest that resistance of Chinese cabbage cultivars to Xcc is likely affected by the virulence of Xcc isolates, but not result from race differentiation.

Antioxidant, Anti-inflammation, and Anti-aging Effects of Ethyl Acetate Fraction from Cynanchum paniculatum Extracts (산해박 분획물의 항산화, 항염증 및 항노화 활성 효과 연구)

  • In this study, we prepared fractions from Cynanchum paniculatum (C. paniculatum) with ethyl acetate and identified the structure and content of paeonol, an indicator substance. Fractions were obtained by sequential solvent fractions using n-hexane, methylene chloride, ethyl acetate, n-butanol, and distilled water, and showed the highest paeonol content in ethyl acetate (EtOAc). The structure of the ethyl acetate fraction (EA) was confirmed by nuclear magnetic resonance (NMR) and the content was determined by high performance liquid chromatography (HPLC) analysis. The antioxidant activity of EA was tested through its ability to scavenge 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and its ability to inhibit nitric oxide (NO) production in RAW 264.7 cells induced by lipopolysaccharide (LPS) was tested for its ability to inhibit NO production in a concentration-dependent manner. Furthermore, MMP-1 production induced by ultraviolet B (UVB) was decreased in a concentration-dependent manner by the EA, and the ability to produce procollagen type-I was increased in a concentration-dependent manner. In addition, as a result of conducting clinical trials using cosmetic formulations containing EA, the effect of improving wrinkles around the eyes was confirmed. Thus, it is expected that Cynanchum paniculatum can be used as a natural functional cosmetic material with antioxidant and anti-aging effects.

Utility Evaluation of Two-point Calibration Curve applied for TSH, FT4 Tests (TSH, FT4 검사의 Two-point Calibration Curve 적용의 유용성 평가)

  • Purpose The ASAN Medical Center, Nuclear Medicine performs TSH (Thyroid stimulating hormone) and FT4 (Free Thyroxine) tests 8 times per day. Accordingly, 70 ~ 80 kit tubes are consumed every day for the measurements and the time consumed for reagent dispensing averages over 170 seconds, where the TAT (turnaround time) may be effected when the number of test samples is larger than expected. Therefore, the following test was conducted with the purpose to reduce the number of kit tubes consumed, and reduce the time for reagent dispensing. Materials and Methods The test is based on applying the same reagent for tests where the number of samples is 30 or less. The test for TSH was conducted 9 times from July $1^{st}$ 2015 to July $10^{th}$ 2015. The test for FT4 was conducted 4 times from June $18^{th}$ 2015 to June $22^{nd}$, 2015. Standard Solution No.2 (0.153 uU/mL) and No.5 (4.96 uU/mL) was selected as the two-point standards for the TSH test, and Standard Solution No.3 (0.777 ng/dL) and No.4 (2.044 ng/dL) was selected as the two-point standards for the FT4 test. 38 test samples were subject to correlation analysis. Results For TSH, the result of the normal test shows ranges of 0.20 ~ 0.37 uU/mL for Control1, 0.53 ~ 0.71 uU/mL for Control2, and 6.77 ~ 7.94uU/mL for Control3, while the result of two-point calibration curve test shows ranges of 0.18 ~ 0.27 uU/mL for Control1, 0.53 ~ 0.71 uU/mL for Control2, and 7.30 ~ 8.52 uU/mL for Control3. For FT4, the result of the normal test shows ranges of 0.85 ~ 0.94 ng/dL for Control1 and 4.23 ~ 4.57 ng/dL for Control2, while the result of two-point calibration curve test shows ranges of 0.61 ~ 0.75 ng/dL for Control1 and 3.88 ~ 5.71 ng/dL for Control2. For TSH, the CV% of the normal test for Control1, Control2 and Control3 are 10.5, 3.3 and 3.6 respectively, while the CV% of the two-point calibration curve test for Control1 and Control1 are 12.4, 8.2 and 5.1 respectively. The result shows an outstanding correlation of TSH: y = 0.9985x - 0.0459 $R^2=0.9986$. For FT4, the CV% of the normal test for Control1 and Control2 are 0.70 and 0.71 respectively, while the CV% of the two-point calibration curve test for Control1 and Control1 are 8.7 and 16.2 respectively. The result shows an outstanding correlation of FT4: y = 1.2674x - 0.1133 $R^2=0.9824$. Conclusion The two-point calibration curve can be efficiently applied for TSH in cases where the number of test samples is not large, since the number of samples to be re-tested increases when the result is abnormal from the calibration curve. The two-point calibration curve test should not be applied for FT4 where the results do not consistently comply with the quality assessment range. Depending on how the two-point calibration curve is applied, up to 5 test tubes can be conserved per test, and the reduced time for reagent dispensing is anticipated to have a positive effect on the TAT (turnaround time).

Effect of Cooking Methods with Various Heating Apparatus on the Quality Characteristics of Pork (가열기구에 따른 조리방법이 돼지고기의 품질특성에 미치는 영향)

  • Pork belly(PB) and pork shoulder(PS) parts were tested to find out chemical and physical characteristics and sensory evaluation with various cooking methods such as pan heating, boiling, grilling, steaming, charcoal heating, pan heating and double-layer pan filled with phase change material (PCM) cooking. The moisture contents of steamed PB and PS had higher results of 60.2% and 67.2% than other treatments. The highest results of crude fat contents in PB treatments was charcoal roasting as 33.2% (p<0.05) while grilling resulted the highest at 16.0% in the PS (p<0.05). In the crude protein contents, boiling treatment resulted the lowest at 15.4% while steaming was the highest at 18.9% in PB. Also, crude protein content of grilling treatment was 25.2%, a result significantly higher than in other cooking methods in PS. Heating loss, which has a close relationship with water holding capacity, showed the highest result in the charcoal treatment at 40.18% and 39.68% each in the both of PS and PS. In the result of shear force, the lowest result was oven treatment at $2.76kg/cm^2$ in PB (p<0.05) and double-layer pan heating at $3.67kg/cm^2$ in PS (p<0.05). L value in the color test of boiling treatment showed the highest result of 65.16 and 59.72 in the PB and PS respectively (p<0.05), however it scored the lowest of 2.32 in b value in PB (p<0.05). In the 9 point-scale sensory evaluation, grilling treatment showed the highest result of 7.56 in the overall palatability of PB (p<0.05). However, PS in the pan heating which scored 7.22 was the best result while having the lowest score of 5.88 in the boiling treatment (p<0.05).

  • I. Introduction : The purpose of this study is to test empirically hypothetical causality among constructs used in previous studies to build the model of relational market behavior on customers' participation and consumer-brand relationship after introducing theories of relationship marketing, loyalty program, consumer-brand relationship, customers' participation in service marketing as previous studies with regard to relational market behavior, which Bagozzi(1995) and Peterson(1995) commented on constructs and definition suggested by Sheth and Parvatiyar (1995). For this purpose, loyalty program by the service provider, customers' participation and consumer-brand relationship as preceding variables explain relational market behavior defined by Sheth and Parvatiyar(1995). This study proposes that loyalty program as a tool of relationship marketing will be effective in that consumers' participation in marketing relationship results in a narrow range of choice(Sheth and Parvatiyar, 1995) because consumers think that their participation motive result in benefits(Peterson, 1995). Also, it is proposed that the quality of consumer-brand relationship explain the performance of relationship as well as the intermediary effect because the loyalty program could be evaluated based on relationship with customers. We reviewed the variables with regard to performance of relationship based on relation maintain in marketing literature, and then tested our hypotheses related to several performance variables including loyalty and intention of relation maintain based on the previous studies and constructs(Bendapudi and Berry, 1997 ; Bettencourt, 1997 ; Palmatier, Dant, Grewal and Evans, 2006 ; You Jae Yi and Soo Jin Lee, 2006). II. Study Model : Analyses about hypothetical causality were proceeded. The marketing effect of loyalty program on relational market behavior was empirically tested in study regarding a service provider. The research model in according to the path hypotheses (loyalty program ${\rightarrow}$ customers' participation ${\rightarrow}$ consumer-brand relationship ${\rightarrow}$ relational market behavior and loyalty program ${\rightarrow}$ consumer-brand relationship, and loyalty program ${\rightarrow}$ relational market behavior and customers' participation ${\rightarrow}$ consumer-brand relationship, and customers' participation ${\rightarrow}$ relational market behavior) proceeded as an activity for customer relation management was suggested. The main purpose of study is to see if relational market behavior could be brought as a result of developing relationship between consumers and a corporate into being stronger and more valuable when a corporate or a service provider try aggressively to build the relationship with customers (Bettencourt, 1997; Palmatier, Dant, Grewal and Evans, 2006; Sheth and Parvatiyar, 1995). III. Conclusion : The results of research into the membership fitness club, one of service areas with high level of customer participation (Bitner, Faranda, Hubbert and Zeithaml, 1997; Chase, 1978; Kelley, Donnelly, Jr. and Skinner, 1990) are as follows: First, causalities in according to path hypotheses were tested, after the preceding variables affecting relational market behavior and conceptual frame were suggested. In study, all hypotheses were supported as expected. This result confirms the proposition suggested by Sheth and Parvatiyar(1995), who claimed that intention of consumer and corporate to participate in marketing relationship brings high level of marketing productivity. Also, as a corporate or a service provider try aggressively to build relationship with customers, the relationship between consumers and a corporate can be developed into stronger and more valuable one (Bettencourt, 1997; Palmatier, Dant, Grewal and Evans, 2006). This finding supports the logic of relationship marketing. Second, because the question regarding the path hypothesis of consumer-brand relationship ${\rightarrow}$ relational market behavior are still at issue, the further analyses were conducted. In particular, there existed the mediating effects of consumer-brand relationship toward relational market behavior. Also, multiple regressions were conducted to see if which one strongly influences relational market behavior among specific question items with regard to consumer-brand relationship. As a result, the influence between items composing consumer-brand relationship and ones composing relational market behavior was different. Among items composing consumer-brand relationship, intimacy was an influence of sustaining relationship, word of mouth, and recommendation, intimacy and interdependence were influences of loyalty, intimacy and self-connection were influences of tolerance and advice. Notably, commitment among items measuring consumer-brand relationship had the negative influence with relational market behavior. This means that bringing relational market behavior is not consumer-brand relationship without personal commitment, but effort to build customer relationship like intimacy, interdependence, and self-connection. This finding confirms the results of Breivik and Thorbjornsen(2008). They reported that six variables composing the quality of consumer-brand relationship have higher explanation in regression model directly affecting performance of consumer-brand relationship. As a result of empirical analysis, among the constructs with regard to consumer-brand relationship, intimacy(B=0.512), interdependence(B=0.196), and quality of partner(B=0.153) had the effects on relation maintain. On the contrary, self-connection, love and passion, and commitment had little effect and did not show the statistical significance(p<0.05). On the other hand, intimacy(B=0.668) and interdependence(B=0.181) had the high regression estimates on word of mouth and recommendation. Regarding the effect on loyalty, explanation level of the model was high(R2=0.515), intimacy(0.538), interdependence(0.223), and quality of partner(0.177) showed the statistical significance(p<0.05). Furthermore, intimacy(0.441) had the strong effect as well as self-connection(0.201) and interdependence (0.163) had the effect on tolerance and forgive. And these three variables showed effects even on advice and suggestion, intimacy(0.373), self-connection(0.270), interdependence (0.155) respectively. Third, in study with regard to the positive effect(loyalty program ${\rightarrow}$ customers' participation, loyalty program ${\rightarrow}$ consumer-brand relationship, loyalty program ${\rightarrow}$ relational market behavior, customers' participation ${\rightarrow}$ consumer-brand relationship, customers' participation ${\rightarrow}$ relational market behavior, consumer-brand relationship ${\rightarrow}$ relational market behavior), the path hypothesis of customers' participation ${\rightarrow}$ consumer-brand relationship, was supported. The fact that path hypothesis of customers' participation ${\rightarrow}$ consumer-brand relationship was supported confirms assertion by Bitner(1995), Fournier(1994), Sheth and Parvatiyar(1995) about consumer relationship to participate in marketing relationship.

The Effects of 'Climbing learning Method' in the Learning of Mathematics in Elementary School (학습구조차트를 활용하는 등산학습법의 초등수학 적용과 효과에 관한 연구)

  • This study discussed the climbing learning method which studied and practiced by Professor Saito Noboru. This is the learning method which is devised to know not only the relationship of the learning factors but the systemic or structural connection of whole studying contents- affects children's math learning ability through practical class to both the lower and the higher grades. To achieve the purpose of this study, these following issues were set; A. Develop the teaching and learning course of mathematics by applying the climbing learning method. B. Execute the mathematics lesson according to the climbing learning method and analyze the learning achievement. C. Analyze the difference between application of the climbing learning method and that of the learning method by student's level in mathematics. D. Analyze what the climbing learning method gives a shift of the recognition of learning mathematics. In order to accomplish these study issues, we analyzed the text book of math not only for children but also for teachers and developed the teaching and learning course applied the climbing learning method with advice of experts. It was chosen two different homogeneous groups each, third year for lower grade group and fifth year for higher grade group. It was done the experimental group lesson applying the climbing learning method and general lesson for the control group. After then, t-test against independent samples was done depending on the result of the student's assessment(T1, T2). These two groups' students were divided into smaller groups based on result of achievement level regardless of gender. These subgroups were confirmed the difference of learning ability between upper and lower level group. As regarding the result making out grades of faith and attitude for math, t-test was used on independent sample. At the same time, experimental groups were tested using learning attitude with the learning structure chart. Through this study the following results are obtained and the conclusion was drawn. Firstly, although applying the climbing learning method to the lesson does not have significant effect to the lower grade of elementary school student's achievement it has significant influence on the higher grade student's achievement. Second, as a result of analyzing the difference between the climbing learning method and the learning method by student's level in mathematics, it is of no beneficial effect to the lower grade both upper level and lower level. However, it has appreciable effect to the higher grade classes both upper level and low level. Especially, upper level students have higher effect than low level students. Third, climbing learning method does not affect to the faith and attitude of the lower grade students positively, but it has affirmative effect to the higher grade students'. As a result of the survey of the experimental groups which were applied to the climbing loaming method, the lesson by using the learning structure chart proved to be helpful to the both the lower and higher grade. The best advantage of using the learning structure chart, children say, is easily understood whole contents of studying and is useful for review. Furthermore, using the learning structure chart is more efficient compared with previous learning method and is given the successful result to self-directed learning. In conclusion, keeping up with the current of the thought of education, we suggest a scheme as a new teaching method from the constructive learning method which emphasize the self-directed learning.

The Comparison of Results Among Hepatitis B Test Reagents Using National Standard Substance (국가 표준물질을 이용한 B형 간염 검사 시약 간의 결과 비교)

  • Purpose: Hepatitis B is infection caused by Hepatitis B virus (HBV). Currently, there are several methods, Kits and equipments for conducting Hepatitis B test. Due to ununiformed methods, it would cause some differences. To manage these differences, it needs process evaluating function of test system and reagent using particular standard substance. The aim of this study is to investigate tendency of RIA method's reagent used in Asan Medical Center through comparing several other test reagents using national standard substance. Materials and Methods: The standard substance in National Institute of Food and Drug Safety Evaluation's biology medicine consists of 5 things, 4 antigens and 1 antibody. We tested reagents using A, B company's Kits according to each test method. All tests are measured repeatedly to obtain accurate results. Results: Test result of "HBs Ag Mixed titer Performance panel" is obtained match rate compared S/CO unit standard with RIA method and EIA 3 reagents, CIA 2 reagents is that company A's reagent is 94.4% (17/18), 83.3% (15/18), B is 88.9% (16/18), 77.8% (14/18). Test result of "HBs Ag Low titer Performance panel" is obtain that EIA 2 reagents is shown 7 posive results, CIA 3 reagents is 11, and RIA method's company A's reagent is 3, B is 2 of 13 in low panel. "HBV surface antigen 86.76 IU/vial" tested dilution. A is obtain positive results to 600 times(0.14 IU/mL), B is 300 times (0.29 IU/mL). Case of "HBV human immunoglobulin 95.45 IU/vial", A is shown positive result to 10,000 times (9.5 mIU/mL) and B is 4,000 times (24 mIU/mL). Test result of "HBs Ag Working Standards 0.02~11.52 IU/mL" is shown that Company A's kit concentration level was 0.38IU/mL, company B was 2.23 IU/mL and higher level of concentration was positive results. Conclusion: When comparing various test reagents and RIA method according to National Standard substances for Hepatitis B test, we recognized that there were no significant trends between reagents. For hepatitis B virus antigen-antibody titers even in parts of the test up to 600 times the antigen, antibodies to 10,000 times the maximum positive results could be obtained. Therefore, we confirmed that results from Asan Medical Center are performed smoothly by reagents and system for hepatitis B virus test.

A Field Survey and Analysis of Ground Water Level and Soil Moisture in A Riparian Vegetation Zone (식생사주 역에서 지하수위와 토양수분의 현장 조사·분석)

  • Phenomenon of vegetation recruitment on the sand bar is drastically rising in the streams and rivers in Korea. In the 1960s prior to industrialization and urbanization, most of the streams were consisted of sands and gravels, what we call, 'White River'. Owing to dam construction, stream maintenance, etc. carried out since the '70s, the characteristic of flow duration and sediment transport have been disturbed resulting in the abundance of vegetation in the waterfront, that is, 'Green River' is under progress. This study purposed to identify the correlation among water level, water temperature, rainfall, soil moisture and soil texture out of the factors which give an effect on the vegetation recruitment on the sand bar of unregulated stream. To this purpose, this study selected the downstream of Naeseong Stream, one of sand rivers in Korea, as the river section for test and conducted the monitoring and analysis for 289 days. In addition, this study analyzed the aerial photos taken from 1970 to 2009 in order to identify the aged change in vegetation from the past to the present. The range of the tested river section was 361 m in transverse length and about 2 km in longitudinal length. According to the survey analysis, the tested river section in Naeseong Stream was a gaining river showing the higher underground-water level by 20~30 m compared to Stream water level. The difference in the underground water temperature was less than $5^{\circ}C$ by day and season and the Stream temperature did not fall to $10^{\circ}C$ and less from May when the vegetation germination begins in earnest. The impact factor on soil moisture was the underground water level in the lower layer and the rainfall in the upper layer and it was found that all the upper and lower layer were influenced by soil particle size. The soil from surface to 1 m-underground out of 6 soil moisture-measured points was sand with the $D_{50}$ size of 0.07~1.37 mm and it's assumed that the capillary height possible in the particle size would reach around 14~43 cm. On the other hand, according to the result of space analysis on the tested river section of unregulated stream for 40 years, it was found that the artificial disturbance and drought promoted the vegetation recruitment and the flooding resulted in the frequency extinction of vegetation communities. Even though the small and large scales of recruitment and extinction in vegetation have been repeated since 1970, the present vegetation area increased clearly compared to the past. It's found that the vegetation area is gradually increasing over time.

Under the request of the Dept. of Navy, U.S.A. this investigation has been done as a part work of the Navy Research Project of Tropical Woods at the Wood Technology Laboratory, School of Forestry, Yale University, New Haven, Conn., U.S.A. 2. In order to determine the equilibrium moisture content and hysteresis loop of three tropical woods (Ocotea, Tablebuia, and Hymenaea) which have not been tested the physical properties, this investigation has made with small thin specimens (1.5"${\times}$1.0${\times}$0.4) under four different controlled relative humidity conditions (that is, 21%, 53%, 60%, and 83%). 3. As the result, the equilibrium moisture content and hysteresis loop of three tropical woods have been shown in the Table and Figures 2, 3 and 4. 4. According to the results, it is concluded that there are the considerable differences in the equilibrium moisture content under the same relative humidity condition and the type of hysteresis loop between different species which have been tested. 5. Desorption of lumber with slightly oscillating humidity of each species tested, has shown on the Table 9 and it has almost the same tendency of results showing the difference between species as the small specimen. 6. Although it is hard to compare the difference of results, E.M.C., and hysteresis between tropical wood and woods from temperate zone, there are, however, still some difference between species. 7.   • Purpose : This study was conducted to assess the current(2003-2005) prevalence of anti-HBs and immunologic memory for Hepatitis B vaccine in children from the central area of Korea. Methods : Subjects were chosen from children and adolescents who received tests for hepatitis B surface antigen(HBsAg) and anti-HBs at Dankook University Hospital from March 2003 to May 2005. Among these, antibodies to hepatitis B core antigen(IgG anti-HBc) were checked. A single booster vaccination was performed on children whose anti-HBs titers were under 10 mIU/mL. One month after booster vaccination we rechecked the anti-HBs titer. Results : A total of 3,277 subjects were tested for HBsAg/anti-HBs, and 1,913(58.4 percent) of them were positive for anti-HBs. Of these, 29 subjects(0.9 percent) were positive for HBsAg. Positive results for anti-HBs by age were 78.6 percent for 6-12 months of age, 62.7 percent for 1-3 years of age, 51.9 percent for 4-6 years of age, 49.5 percent for 7-12 years of age, 63.4 percent for 13-15 years of age and 72.2 percent for 16-18 years of age. The 80 subjects who were tested negative for HBsAg/anti-HBs received a single booster vaccine, 71 subjects were tested positive for antibodies. IgG anti-HBc titer was checked for 169 of the subjects, 5 subjects were positive. Conclusion : In our study, a significant anamnestic response was observed in 88.8 percent of children. This is believed to be a result of the relatively long immunologic memory effect of the hepatitis B vaccination in children from the central area of Korea.
