The Frequency of Detecting Prevotella intermedia and Prevotella nigrescens in Korean Adult Periodontitis Patients (한국인 치주 감염 환자에서의 Prevotella intermedia와 Prevotella nigrescens의 발현빈도)

  • Prevotella intermedia has been implicated as a potent pathogen in many kinds of periodontal, pulpal and periapical diseases. However, it has been isolated from periodontally healthy adults and from edentulous children as well. The intraspecies heterogeneity of Prevotella intermedia has been demonstrated in early studies and finally Shah & Gharbia confirmed the existence of 2 DNA homology groups and proposed dividing Prevotella intermedia into 2 species, Prevotella intermedia and Prevotella nigrescens. This study was designed to examine the frequency of Prevotella intermedia and Prevotella nigrescens in diseased periodontal pockets and healthy gingival sulcus of Korean people by PCR based on 16s ribosomal DNA sequence. One hundred adults who had adult periodontitis but not taken any periodontal treatment or antibiotics during previous 6 months and 50 adults who had healthy periodontal tissue were selected for this study. The sulcular fluid was collected into VMGA by sterilized paper point and diluted to 1,000 times in anaerobic chamber. $100{\mu}{\ell}$ of sample was cultured in $37^{\circ}C$ for 10 days. Among the bacterial colonies, BPB were selected and cultured in BHI broth and then Prevotella intermedia was identified through Gram staining and biochemical test. Identified Prevotella intermedia was cultured again and centrifuged. DNA was extracted from the pellet using several reagents. PCR was performed by previously designed primer. The results were followed. 1. BPB were isolated from 39 of 100 samples of diseased periodontal pockets(39%). 2. Prevotella intermedia was identified from 24 of 39 BPB samples. 3. Among 24 Prevotella intermedia, 21 were confirmed as Prevotella inter - media(87.5) and 2 were confirmed as Prevotella nigrescens(8.33%). 4. BPB were isolated from 9 of 50 samples of periodontally healthy patients. Among them only two were identified as Prevotella intermedia, that is, one was confirmed as Prevotella intermedia and the other was Prevotella nigrescens.

Development of On-line Quality Sorting System for Dried Oak Mushroom - 3rd Prototype-

  • In Korea, quality evaluation of dried oak mushrooms are done first by classifying them into more than 10 different categories based on the state of opening of the cap, surface pattern, and colors. And mushrooms of each category are further classified into 3 or 4 groups based on its shape and size, resulting into total 30 to 40 different grades. Quality evaluation and sorting based on the external visual features are usually done manually. Since visual features of mushroom affecting quality grades are distributed over the entire surface of the mushroom, both front (cap) and back (stem and gill) surfaces should be inspected thoroughly. In fact, it is almost impossible for human to inspect every mushroom, especially when they are fed continuously via conveyor. In this paper, considering real time on-line system implementation, image processing algorithms utilizing artificial neural network have been developed for the quality grading of a mushroom. The neural network based image processing utilized the raw gray value image of fed mushrooms captured by the camera without any complex image processing such as feature enhancement and extraction to identify the feeding state and to grade the quality of a mushroom. Developed algorithms were implemented to the prototype on-line grading and sorting system. The prototype was developed to simplify the system requirement and the overall mechanism. The system was composed of automatic devices for mushroom feeding and handling, a set of computer vision system with lighting chamber, one chip microprocessor based controller, and pneumatic actuators. The proposed grading scheme was tested using the prototype. Network training for the feeding state recognition and grading was done using static images. 200 samples (20 grade levels and 10 per each grade) were used for training. 300 samples (20 grade levels and 15 per each grade) were used to validate the trained network. By changing orientation of each sample, 600 data sets were made for the test and the trained network showed around 91 % of the grading accuracy. Though image processing itself required approximately less than 0.3 second depending on a mushroom, because of the actuating device and control response, average 0.6 to 0.7 second was required for grading and sorting of a mushroom resulting into the processing capability of 5,000/hr to 6,000/hr.

Screening for Resistance to Downy Mildew among Major Commercial Cucumber Varieties (주요 오이 품종의 노균병에 대한 저항성 검정)

  • This study was carried out for the downy mildew resistant test between 2010 and 2012. A set of 22 accessions belonging to 2 wild species and 20 varieties of the genus Cucumis, originating mainly from the Asian Vegetable Research and Development Center (AVRDC) Gene Centre, was evaluated for resistance to Pseudoperonospora cubensis, causal agent of cucumber downy mildew. The youngest fully expanded true leaves were found suitable for in vitro screening. Both leaf discs and full leaves could be kept fresh longer when applying 0.2 ${\mu}g/ml$ of gibberellin acid (GA). The incubation temperature of $20^{\circ}C$ was found to be the most suitable temperature for symptom development comparing with 15 and $25^{\circ}C$. Symptom development was faster when contact diseased leaf discs (2 weeks after inoculation) on to fresh leaf samples comparing with using conidia suspension ($10^5$ spores/ml). The numbers of spots in 'C-19' were lower than other varieties. 'C-19' variety was also showed the highest level of downy mildew resistant at $20^{\circ}C$ chamber in 6 days after inoculating with pathogen and displayed 0.90 (under 10%) of the infected rate. However, other varieties displayed susceptible in the pathogen sprayed plots. 'C-19' was the most resistant variety and no lesion was observed. Based on all data, 'C-19' can be a useful variety for the prevention of downy mildew.

Occurrence of Target Spot on Rosemary Caused by Corynespora cassiicola in Korea (Corynespora cassiicola에 의한 로즈마리 점무늬병)

  • The purpose of this experiment was to investigate the development of new spot disease on the leaf and stem of rosemary (Rosmarinus officinalis) in commercial greenhouses at Jeonju and Namwon in Korea. Incidence of target spot on rosemary was higher at the end of the rainy season with high humidity. Those symptoms were black ring spots (3-5 mm in diameter) and withering on green leaves and stems. Conidiophores and conidia were formed on the infected tissue in moist chamber and conidia were shown as the cylindrical and oval types in chain, ranged from 55 to $275{\mu}m$ in length, and 7 to $14{\mu}m$ in width. Conidia with eight to ten pseudosepta were formed on the conidiapore. The optimum growth temperature of isolates was $30^{\circ}C$ on the PDA medium under the dark condition. In the pathogenesis test, the target spot and withering symptoms were appeared on the leaves and stems 3 days after inoculation showing similar symptoms compared to those of in nature. The same fungus was re-isolated from infected lesion, indicating that Corynespora cassiicola caused leaf target spot and twig blight on rosemary. The rDNA ITS nucleotide sequences of the pure cultured isolate from the diseased area on rosemary showed 100% similarity to the sequences of C. cassiicola available in the GenBank database (JQ595296, JQ595297, FJ852715 and AY238606). Therefore, we report that the target spot of leaves and stems in rosemary was caused by C. cassiicola.

Comparison of Airborne Asbestos Concentrations from Soils in Naturally Occurring Asbestos(NOA) Areas - Activity Based Sampling(ABS) vs. Real-time Asbestos Fiber Monitor(F-1 fiber monitor) - (자연발생석면지역의 토양 내 석면함유율에 따른 비산석면 농도평가 - 활동근거시료채취방법(ABS)과 실시간 섬유 측정 장치(F-1 fiber monitor) 결과 비교 -)

  • Objectives: The present study is aimed at performing real-time measurement of fibrous materials using an F-1 fiber monitor, investigating the correlations between the measurements and environmental conditions, and assessing the feasibility of the use of the monitor in actual exposure assessments based on the accuracy and reliability of the device. Methods: Asbestos specimens with a fixed asbestos content were dispersed in a chamber and collected with a particle measuring test device. Measurements obtained by the existing PCM method, and with the F-1 fiber monitor were compared. In addition, concentrations of asbestos fibers obtained by the PCM method, the TEM method, and the F-1 fiber monitor were compared with that of specific ABS scenarios in NOA regions. Correlations of asbestos contents in soil and weather conditions with each method of measurement were analyzed. Results: Laboratory results showed that levels of asbestos fibers measured with each method increased as fiber contents in soil increased. In the accuracy and reproducibility assessment, no significant differences were found between the different methods of measurement. On-site assessment results showed positive correlations among the methods, and these correlations were less significant compared with what was shown by the laboratory results. Levels of asbestos fibers increased as asbestos contents in soil increased, and as temperature increased. Levels of asbestos fibers decreased as humidity increased, and wind speed did not significantly affect the extent to which asbestos fibers were scattered. Conclusions: While it would be premature to replace existing methods with the use of F-1 fiber monitors in real sites based on the results of this study, the monitor may be useful in the screening of the sites, which assesses hazard levels in different regions. Replacement of existing methods with the use of F-1 fiber monitors may be possible after the limitations identified in this study are overcome, and additional assessment data are obtained and reviewed under different conditions to confirm the reliability of the monitor in future research. Obtained assessment results may be used as basic data for the assessment of asbestos hazard in NOA regions.

Assessment of Emitted Volatile Organic Compounds, Metals and Characteristic of Particle in Commercial 3D Printing Service Workplace (실제 3D 프린팅 작업장에서 발생하는 공기 중 유기화합물, 금속 및 입자특성 평가)

  • Objectives: 3D printing technologies have become widely developed and are increasingly being used for a variety of purposes. Recently, the evaluation of 3D printing operations has been conducted through chamber test studies, and actual workplace studies have yet to be completed. Therefore, the objective of this study was to determine the emission of volatile organic compounds(VOCs), metals, and particles from printing operations at a workplace. This included monitoring conducted at a commercial 3D printing service workplace where the processes involved material extrusion, material jetting, binder jetting, vat photo polymerization, and powder bed fusion. Methods: Area samples were collected with using a Tenax TA tube for VOC emission and MCE filter for metals in the workplace. For particle monitoring, Mini Particle Samplers(MPS) were also placed in the printer, indoor work area, and outdoor area. The objective was to analyze and identify particles' size, morphology, and chemical composition using transmission electron microscopy with energy dispersive spectroscopy(TEM-EDS) in the workplace. Results: The monitoring revealed that the concentration of VOCs and metals generated during the 3D printing process was low. However, it also revealed that within the 3D printing area, the highest concentration of total volatile organic compounds(TVOC) was 4,164 ppb at the vat photopolymerization 3D printing workplace, and the lowest was 148 ppb at the material extrusion 3D printing workplace. For the metals monitoring, chromium, which, is carcinogenic for humans, was detected in the workplace. As a characteristic of the particles, nano-sized particles were also found during the monitoring, but most of them were agglomerated with large and small particles. Conclusions: Based on the monitoring conducted at the commercial 3D printing operation, the results revealed that the concentration of VOCs and metals in the workplace were within Korea's occupational exposure limits. However, due to the emission of nano-sized particles during 3D printing operations, it was recommended that the exposure to VOCs and metals in the workplace should be minimized out of concern for workers' health. It was also shown that the characteristics of particles emitted from 3D printing operations may spread widely within an indoor workplace.

Fabrication and packaging of the vacuum magnetic field sensor (자장 세기 측정용 진공 센서의 제작 및 패키징)

  • This work reports the tunneling effects of the lateral field emitters. Tunneling effect is applicable to the VMFS(vacuum magnetic field sensors). VMFS uses the fact that the trajectory of the emitted electrons are curved by the magnetic field due to Lorentz force. Polysilicon was used as field emitters and anode materials. Thickness of the emitter and the anode were $2\;{\mu}m$, respectively. PSG(phospho-silicate-glass) was used as a sacrificial layer and it was etched by HF at a releasing step. Cantilevers were doped with $POCl_3(10^{20}cm^{-3})$. $2{\mu}m$-thick cantilevers were fabricated onto PSG($2{\mu}m$-thick). Sublimation drying method was used at releasing step to avoid stiction. Then, device was vacuum sealed. Device was fixed to a sodalime-glass #1 with silver paste and it was wire bonded. Glass #1 has a predefined hole and a sputtered silicon-film at backside. The front-side of the device was sealed with sodalime-glass #2 using the glass frit. After getter insertion via the hole, backside of the glass #1 was bonded electrostatically with the sodalime-glass #3 at $10^{-6}\;torr$. After sealing, getter was activated. Sealing was successful to operate the tunneling device. The packaged VMFS showed very small reduced emission current compared with the chamber test prior to sealing. The emission currents were changed when the magnetic field was induced. The sensitivity of the device was about 3%/T at about 1 Tesla magnetic field.

MICROLEAKAGE OF RESILON BY METHACRYLATE-BASED SEALER AND SELF-ADHESIVE RESIN CEMENT (Resilon을 이용한 근관충전 시 레진계열의 근관실러와 자가-접착 레진시멘트에 따른 미세누출)

  • The purpose of this study was to compare the apical microleakage in root canal filled with Resilon by methacrylate-based root canal sealer or 2 different self-adhesive resin cements. Seventy single-rooted extracted human teeth were sectioned at the CEJ perpendicular to the long axis of the roots with diamond disk. Canal preparation was performed with crown-down technique using Profile NiTi rotary instruments and GG drill. Each canal was prepared to ISO size 40, .04 taper and 1 mm short from the apex. The prepared roots were randomly divided into 4 experimental groups of 15 roots each and 5 roots each for positive and negative control group. The root canals were filled by lateral condensation as follows. Group 1: Gutta-percha with AH-26, Group 2: Resilon with RealSeal primer & sealer, Group 3: Resilon with Rely-X Unicem, Group 4: Resilon with BisCem. After stored in $37^{\circ}C$, 100% humidity chamber for 7 days, the roots were coated with 2 layers of nail varnish except apical 3 mm. The roots were then immersed in 1% methylene blue dye for 7 days. Apical microleakage was measured by a maximum length of linear dye penetration after roots were separated longitudinally. One way ANOVA and Scheffe's post-hoc test were performed for statistical analysis. Group 1 showed the least apical leakage and there was no statistical significance between Group 2, 3, 4. According to the results, the self adhesive resin cement is possible to use as sealer instead of primer & sealant when root canal filled by Resilon.

Family Involvement and The Entry Mode into Entrepreneurship: The Contingent Role of National Culture (가족관여와 창업방식간의 관계에 대한 연구: 국가문화의 조절효과를 중심으로)

  • This study examines the role of family involvement and national culture in entrepreneurs' choice of mode of entry into entrepreneurship. Mode of entry consists of two choices: starting a new venture or taking over an existing firm. Specifically, we investigate how cultural values moderate the relationship between family involvement and their choice of business takeover. Using a cross-national sample of 3,831 respondents from 33 countries in Flash Eurobarometer data, we develop key variables and test our hypotheses employing hierarchical linear modelling. Our results show that the relationship between family involvement of entrepreneurs and their choice of business takeover is moderated by individualism, short term orientation, and power distance. This research contributes to understanding comparative international entrepreneurship based on national culture. Implications for policy makers and educators are discussed.

Study on the Physical Characteristics of Water Supply Steel Pipe according to Temperature Change (수도용 강관의 온도변화에 따른 물리적 특성에 대한 연구)

  • 'The facilities standards of water supply' issued by the Ministry of Environment in 2004 indicates that expansion joints cannot be used in welding water supply steel pipes. However, their reason is not clear and it is difficult to confirm the stability of the steel pipe for a water supply pipeline. The purpose of this study is to determine whether or not an expansion joint is necessary to improve the stability of water supply in steel pipe through a displacement analysis of the pipework. The test results are as follows. Firstly, it was found that expansion and contraction of the water supply steel pipe (D 2,400 mm) occur repeatedly in 4 cycles per year, and the maximum expansion and contraction amount of the pipe is 13.03 mm in 1.24 km pipelines. Secondly, the thermal stress caused by expansion and contraction of the steel pipe is $13.7{\sim}36.1kgf/cm^2$ according to the burial depth (0~4 m). The main comparison factors to determine the stability of the steel pipe (STWW 400) were the allowable tensile strength and the fatigue limit, which were computed to be $4,100kgf/cm^2$ and $1,840kgf/cm^2$, respectively. Finally, the thermal stress of the steel pipe is very small compared to the allowable tensile stress and fatigue stress. Therefore, thermal stress does not affect the stability of the steel pipe, although the expansion and contraction of the steel pipe occurs by temperature changes. In conclusion, the study demonstrated that expansion joints are not required in water supply steel pipelines.
