Isolation of Defense-Related Genes from Nicotiana glutinosa Infected by Tobacco Mosaic Virus Using a Modified Differential Screening

  • Many of plant defense responses are consequence of transcriptional activation of related genes. We have developed a modified differential screening procedure to isolate tobacco genes that are involved in the defense responses against TMV infection. A cDNA library was constructed from Nicotiana glutinosa leaves infected by TMV under temperature shift conditions. Each of plasmid DNA in the library was hybridized on a set of slot blots to a pool of cDNA probes prepared from either TMV-infected or mock-treated tobacco leaves. Among 900 plasmid DNAs, 81 clones exhibiting significantly enhanced or reduced level of hybridization to either probe were selected for nucleotide sequencing. The clones were listed into 61 genes considering redundancy between the sequences. The genes were identified to be defense-related genes including PR-genes and genes involved in primary or secondary metabolisms. This results supports the implication that plant defense process entails a major shift in total cellular metabolisms rather than activation of a limited number of defense-related genes. Expression patterns of a number of defense-related genes. Expression patterns of a number of selected genes were examined in northern blot analyses. It is notable that the clone 630 of unknown function exhibits expression pattern similar to those of previously known PR-genes. Experiments to elucidate the roles in defense mechanism of a couple of genes newly identified in this study are in progress.

Analysis of Long-term Changes of Days with 25℃ or Higher Air Temperatures in Jeju (제주의 여름철 기온이 25℃ 이상인 날수의 장기변화 분석)

  • In this study, the time series of the number of days with $25^{\circ}C$ or higher temperatures in the Jeju region were analyzed and they showed a strong trend of increase until recently. To determine the existence of a climate regime shift in this time series, the statistical change-point analysis was applied and it was found that the number of days with $25^{\circ}C$ or higher temperatures in the Jeju region increased sharply since 1993. Therefore, in order to examine the cause of the sharp increase of the days with $25^{\circ}C$ or higher temperatures in the Jeju region, the differences between the averages of 1994~2013 and the averages of 1974~1993 were analyzed for the large-scale environment. In the Korean Peninsula including the Jeju region, precipitable water and total cloud cover decreased recently due to the intensification of strong anomalous anticyclones near the Korean Peninsula in the top, middle and bottom layers of the troposphere. As a result of this, the number of days with $25^{\circ}C$ or higher temperatures in the Jeju region could increase sharply in recent years. Furthermore, in the analysis of sensible heat net flux and daily maximum temperatures at 2 m, which is the height that can be felt by people, the Korean Peninsula was included in the positive anomaly region. In addition, the frequency of typhoons affecting the Korean Peninsula decreased recently, which reduced the opportunities for air temperature drops in the Jeju region.

Effects of reversing the coiling direction on the force-deflection characteristics of nickel-titanium closed-coil springs

  • Objective: To investigate the effects of reversing the coiling direction of nickel-titanium closed-coil springs (NiTi-CCSs) on the force-deflection characteristics. Methods: The samples consisted of two commercially available conventional NiTi-CCS groups and two reverse-wound NiTi-CCS groups (Ormco-Conventional vs. Ormco-Reverse; GAC-Conventional vs. GAC-Reverse; n = 20 per group). The reverse-wound NiTi-CCSs were directly made from the corresponding conventional NiTi-CCSs by reversing the coiling direction. Tensile tests were performed for each group in a temperature-controlled acrylic chamber ($37{\pm}1^{\circ}C$). After measuring the force level, the range of the deactivation force plateau (DFP) and the amount of mechanical hysteresis (MH), statistical analyses were performed. Results: The Ormco-Reverse group exhibited a significant shift of the DFP end point toward the origin point (2.3 to 0.6 mm), an increase in the force level (1.2 to 1.3 N) and amount of MH (1.0 to 1.5 N) compared to the Ormco-Conventional group (all p < 0.001), which indicated that force could be constantly maintained until the end of the deactivation curve. In contrast, the GAC-Reverse group exhibited a significant shift of the DFP-end point away from the origin point (0.2 to 3.3 mm), a decrease in the force level (1.1 to 0.9 N) and amount of MH (0.6 to 0.4 N) compared to the GAC-Conventional group (all p < 0.001), which may hinder the maintenance of force until the end of the deactivation curve. Conclusions: The two commercially available NiTi-CCS groups exhibited different patterns of change in the force-deflection characteristics when the coiling direction was reversed.

  • In this paper, the performance of the selective combining according to the channel selection decision method of frequency diversity is evaluated in the underwater frequency selective channel. The underwater acoustic channel in the shallow sea has a complex multipath characteristic by combining various environmental factors such as boundary surface reflection and sound wave refraction according to the water temperature layer. In particular, frequency selectivity due to multipath causes energy fluctuation in a communication channel, which reduces SNR (Signal to Noise Ratio) and deteriorates communication performance. In this paper, we applied the frequency diversity technique using multiple channels to secure the communication performance according to the frequency selectivity by multipath. For each channel, 4-FSK (Frequency Shift Keying) and selective combining were applied, the performance was evaluated by applying the maximum value, average value, and majority decision of the signal in order to decide the demodulation channel selection of the selective combining.

  • Currently, a new energy system is emerging that implements consumption reduction by improving energy efficiency. Accordingly, as smart grids spread, the rate system by timing is expanding. The rate system by timing is a rate system that applies different rates by season/hour to pay according to usage. In this study, external factors such as temperature/day/time/season are considered and the time series prediction model, LSTM, is used to predict energy power usage data. Based on this energy usage prediction model, energy usage charges are reduced by analyzing usage patterns for each device and transferring power energy from the maximum load time to the light load time. In order to analyze the usage pattern for each device, a clustering technique is used to learn and classify the usage pattern of the device by time. In summary, this study predicts usage and usage fees based on the user's power data usage, analyzes usage patterns by device, and provides customized demand transfer services based on analysis, resulting in cost reduction for users.

Water balance change at a transiting subtropical forest in Jeju Island

  • Jeju island has a humid subtropical climate and this climate zone is expected to migrate northward toward the main land, Korea Peninsula, as temperature increases are accelerated. Vegetation type has been inevitably shifted along with the climatic change, having more subtropical species native in southeast Asia or even in Africa. With the forest composition shift, it becomes more important than ever to analyze the water balance of the forest wihth the ongoing as well as upcoming climate change. Here, we implemented the Ecosystem Demography Biosphere Model (ED2) by initializing the key variables using forest inventory data (diameter at breast height in 2012). Out of 10,000 parameter sets randomly generated from prior distribution distributions of each parameter (i.e., Monte-Carlo Method), we selected four behavioral parameter sets using remote-sensing data (LAI-MOD15A2H, GPP-MOD17A2H, and ET-MOD16A2, 8-days at 500-m during 2001-2005), and evaluated the performances using eddy-covariance carbon flux data (2012 Mar.-Sep. 30-min) and remote sensing data between 2006-2020. We simulated each of the four RCP scenarios (2.6, 4.5, 6.0, and 8.5) from four climate forcings (GFDL-ESM2M, HadGEM2-ES, IPSL-CM5A-LR, and MIROC5 from ISIMIP2b). Based on those 64 simulation sets, we estimate the changes in water balance resulting from the forest composition shift, and also uncertainty in the estimates and the sensitivity of the estimates to the parameters, climate forcings, and RCP scenarios.

Changes of Methanogenic Pathway with Incubation Temperatures in the Littoral Sediment of Reservoir Paldang, Korea (팔당호 연안대 저질토에서 배양온도에 따른 메탄발생 경로의 변화)

  • Changes in methanogenic pathway at low temperature were studied by incubation experiments of sediment slurries from the littoral zone of Reservoir Paldang. Methane production rates in sediment slurries increased exponentially between $5^{\circ}C$and $45^{\circ}C$, reached a maximum rate of $7.4\;nmol\;{\cdot}\;g^{-1}\;{\cdot}\;h^{-1}$ at $45^{\circ}C$, and then declined to low rate. The shift of incubation temperature from high temperature ($30^{\circ}C$) to lowtemperature ($15^{\circ}C$) resulted in a decrease of methane production rate and of hydrogen accumulation rate, and the transient accumulation of acetate concentration. Chlorofarm inhibited perfectly methanogenesis and resulted in the accumulation of hydrogen and acetate as immediate precursors for metltane formation at both incubation temperatures of $15^{\circ}C$ and $30^{\circ}C$. In terms of equivalent methane which was calculated from the two intermediary metabolites accumulated in absence of methanogenesis, methane production from acetate was accounted for 14% of total methanogenesis at $30^{\circ}C$ and 75% at $15^{\circ}C$, respectively. When the high acetate concentrations above 19 mM were added to sediment slurries, methane production was inhibited at the low temperature ($15^{\circ}C$) . Our results demonstrate that contribution of acetate on methanogenesis increases at low temperature, but this pathway is inhibited by high concentration of acetate. Therefore acetate-utilizing methanogensis appears to be a key reaction at low temperature, and seems to be one of bottlenecks of the low temperature anaerobic degradation of organic matter in littoral sediments of the reservoir.

Temperature-dependent DNA binding of DicA protein in vivo and in vitro (In vivo와 in vitro에서 DicA 단백질의 온도 의존적 DNA 결합)

  • In Escherichia coli, DicA protein is involved in cell division control. DicA protein is known to bind DNA better at $25^{\circ}C$ than at $37^{\circ}C$. However, the molecular cause of the temperature dependent binding is not clear. In this study, we investigated how DicA binds DNA and why its DNA binding activity depends on temperature. An unique in vivo DNA binding assay developed in this laboratory showed that unlike the homologous proteins such as RovA or SlyA, DicA uses its N-terminal domain for DNA binding. The in vivo DNA binding assay of DicA also demonstrated that the temperature-dependent DNA binding activity does not come from Cnu or H-NS that is known to bind DNA better at $25^{\circ}C$ than at $37^{\circ}C$. Electrophoretic Mobility Shift Assay (EMSA), when performed with purified DicA protein, did not show temperature-dependent DicA binding activity. However when EMSA was performed with crude protein from WT E. coli cells, temperature-dependent DicA binding activity was observed, suggesting that there is a factor(s) that confers temperature DNA binding activity of DicA in vivo.

Identification of High School Students' Understanding on the Reaction Rate Change During Chemical Equilibrium Shift (화학 평형 이동시 반응 속도 변화에 대한 고등학생들의 이해 조사)

  • The purpose of this study is to investigate the students' conceptions about the reaction rate changes during the chemical equilibrium shifts and also whether the questions about basic concepts of the reaction rate are helpful for the students' understanding of reaction rate changes during the chemical equilibrium shifts. The subjects were 100 students in the 12th grade. The questionnaires were composed of A, B, and A' set, which had to be answered sequentially. The A set consisted of questions asking the change of reaction rate when chemical equilibrium was shifted, the B set was to testify the basic concepts of the reaction rate, and the A' set was the same as the A set. The results showed that the students' understanding of the reverse reaction rate change was lower than that of the forward reaction rate change during the equilibrium shift. Also it was found that students' understanding of the reaction rate change caused by adding the reactant was fairly good while their understanding of the reaction rate change caused by temperature increment was very poor. Since the students marked very high scores in the B set questions, their poor understanding for the reaction rate changes during the equilibrium shifts was not seemed to be due to the lacks of the basic knowledge of reaction rate. Instead, it was due to the failure of applying the basic knowledge of reaction rate to the changes of reaction conditions. It was also found that the average scores of A' set were statistically higher than those of A set. It means the B set items were helpful for the students to solve the A' set items. These results evidenced the possibility of set questionnaires could help the students to connect the related concepts in solving the problems.

Analysis of Anesthesia and Recovery Room Nurses's Activities (일 대학 병원 마취${\cdot}$회복실 간호사의 간호활동 분석)

  • Purpose: This study was aimed to specify roles of nurses from the anesthesia and recovery room by analyzing nursing activities as well as anesthesia nursing during surgery. Method: The objects were 12 RNs working in the recovery room of a university hospital located in Incheon. Self-report was performed by measuring stop-watch for five days from May 17th to May 21st in 2004. Research method was designed to record the time and frequency of the nursing activity from anesthesia and recovery room based on nurses' statement and other references for five days. The data were analyzed with descriptive statistics, ANOVA, and t-test via SPSS Win 10.0 program. Result: Nursing activities in anesthesia recovery room were classified into two different characteristics such as direct and indirect nursing activity. The activities consisted of 11 direct nursing areas and 39 nursing activities in anesthesia preparation room. The indirect nursing was classified into 8 nursing areas and 32 nursing activities. The direct nursing was classified into 12 nursing areas and 55 nursing activity. Also, the indirect nursing was classified into 7 nursing areas and 21 nursing activities in recovery room. In terms of prevalence of nursing activities in the anesthesia preparation room, observation and cooperation of anesthesia was the most prevalent activity, drug and eqipment management the second prevalent, and drug administration the third. On the other hand, in the recovery room, the most prevalent activity was vital sign checking and observation, the second most prevalent activity informative activity, and the third body temperature control. Nursing activity time was recorded according to the nursing characteristics. In the anesthesia preparation room, the direct nursing spent for 8092.20 minutes was larger than the indirect nursing spent for 7198.50 minutes. Also, in the recovery room, the direct nursing spent for 2361.16 minutes was larger than the indirect nursing spent for 1134.13 minutes. 4. Nursing activity time was compared to duty shifts. In the anesthesia preparation room, the direct nursing was more prevalent on day shift and the indirect nursing was prevalent on evening shift. However, in the recovery room, both direct and indirect nursing activities were prevalent on day shift. Conclusion: The role of anesthesia and recovery room nurses was analysed according to the time, frequency, and its characteristics.
