Multi-responsive hydrogel cross-linked synthesized spiropyran-based hydrophilic cross-linker (스피로피란 기반 친수성 가교제를 활용한 다중 자극 감응형 하이드로젤)

  • Stimuli-responsive hydrogels are being extensively studied to alter their physiochemical properties in response to external changes such as temperature, pH, light and mechanical stress. This study reports multi-responsive hydrogel having optical change response to external stress. First, we synthesized a novel spiropyran cross-linker successfully by grafting poly(ethylene glycol) diacrylate (PEGDA) on both side of spiropyran and introduced to hydrogel. In the results, the yellow spriopyran structure was conversed to purple merocyaine structure by internal stress during swelling of the hydrogels cross-linked with the SP-PEGDAs. Also, the hydrogel could be visualized the swelling and deswelling process in response to pH, by converting MC and prontonated MC structure.

  • Hydrogels have gained considerable attention in various fields due to their easily transformative ability by different stimulation. In addition, metal-based conductive additives can enable the hydrogels to be conductive with dimension change. Although the development of the additives offered enhanced electrical properties to the hydrogels, correspondingly enhanced mechanical properties may limit the volume and electrical properties switching after stimulation. Here we prepared poly(N-isopropylacrylamide) (PNIPAM) thermo-responsive hydrogel that has a 32℃ of low critical solution temperature and added liquid metal particles (LMPs) as conductive additives, possessing soft and stretchable benefits. The LMPs enabled PNIPAM (PNIPAM/LMPs) hydrogels to be constricted over 32℃ with a high volume switching ratio of 15.2 when deswelled. Once the LMPs are spontaneously oxidized in hydrogel culture, the LMPs can release gallium ions into the hydrogel nature. The released gallium ions and oxidized LMPs enhanced the modulus of the PNIPAM/LMPs hydrogel, triggering high mechanical stability during repeated swelling/deswelling behavior. Lastly, highly constricted PNIPAM/LMPs hydrogel provided a 5x106 of electrical switching after deswelling, and the switching ratio was closely maintained after repeated swelling/deswelling transformation. This study opens up opportunities for hydrogel use requiring thermo-responsive and high electrical switching fields.

Expression of dehydration responsive element-binding protein-3 (DREB3) under different abiotic stresses in tomato

  • We investigated the expression pattern of dehydration responsive element-binding protein-3 in tomato under different abiotic stresses. Full length LeDREB3 cDNA was isolated from tomato plant, followed by phylogenetic analysis based on deduced amino acid sequences that revealed significant sequence similarity to DREB proteins belonging to diverse families of plant species. Southern blot analysis showed duplicate copies of LeDREB3 in the tomato genome while organ-specific expression profiling indicated constitutive expression of LeDREB3 in all tested organs, which was particularly strong in flower. LeDREB3 expression was significantly induced by Nacl, drought, low temperature and $H_2O_2$. Moreover, LeDREB3 was slightly regulated by treatment with ABA and MV. These observations suggest that the LeDREB3 gene may be involved in the response of the tomato plant to stress.

  • The swelling behavior of hyddrogels has been interested in many applications of drug carriers. These gels show reversible swelling changes in response to pH, electric currcnt, and temperature. Among others, the temperature-responsive behavior of poly(N-isopropylacrylanxide) (p(NIPAAm)) was studied, because a lower critical solution temperature(LCST) is in the vicinity of 32$\circ$C, and remarkable temperature-response can be obtained. We propose a novel composite membrane, which is appropriate for transporting drug ingredients above the transition temperature. Our object was to design a high permeation system above the shrinking temperature of p(NIPAAm). The membrane was composed of a matrix polymer and thermosensitive p(NIPAAm) hydrogel. The flux pattern of 4-acctamidophen through membrane in response of temperature was opposite to that of p(NIPAAm) membrane.

  • The composites of temperature-sensitive hydrogel and activated carbons were prepared in order to improve both the mechanical strength of hydrogel matrix and the loading capacity of drug in a hydrogel drug delivery system. The swelling of composite hydrogel was varied depending on the temperature. Both the swelling and the release behavior of the composite hydrogel were varied depending on the kind of activated carbon. The release behavior showed the high efficiency which is important for practical applications.

  • Thermo-responsive poly(N-isopropylacrylamide) (PIPAAm) was covalently patterned by masked el electron beam irradiation. Introduction of PIPAAm on tissue culture polystyrene dish was confirmed by ATR-FTIR and ESCA measurements. Hepatocytes were cultured at $37^{circ}C$ on these surfaces. Cells adhered on PIPAAm-grafted domains were detached by reducing culture temperature to $20^{circ}C$. Endothelial cells were then seeded and cultured on the same surfaces. Seeded endothelial cells were selectively attached on hepatocytes detached and PIPAAm-grafted domains and could be co-cultured with hepatocytes on the same culture dishes with clear pattern. This co-culture method enabled long-term co-culture of hepatocytes with endothelial cells.

Expression of a rice DREB1 gene, OsDREB1D, enhances cold and high-salt tolerance in transgenic Arabidopsis

  • OsDREB1D, a special DREB (dehydration responsive element binding protein) homologous gene, whose transcripts cannot be detected in rice (Oryza sativa L), either with or without stress treatments, was amplified from the rice genome DNA. The yeast one-hybrid assay revealed that OsDREB1D was able to form a complex with the dehydration responsive element/C-repeat motif. It can also bind with a sequence of LTRE (low temperature responsive element). To analyze the function of OsDREB1D, the gene was transformed and over-expressed in Arabidopsis thaliana cv. Columbia. Results indicated that the over-expression of OsDREB1D conferred cold and high-salt tolerance in transgenic plants, and that transgenic plants were also insensitive to ABA (abscisic acid). From these data, we deduced that this OsDREB1D gene functions similarly as other DREB transcription factors. The expression of OsDREB1D in rice may be controlled by a special mechanism for the redundancy of function.

  • Control of shape/volume, mechanical, optical, electrical, and chemical switching of materials by external stimuli such as light, temperature, pH, electric field, and pressure has attracted great attention. Among these materials, photo-responsive materials containing photochromic compounds such as azobenzene, spiropyran, and cinnamic acid groups have been the subject of intense interest in recent years. In this review, we describe the recent progress in the area of azobenzene containing polymer materials that can convert light energy into mechanical energy directly. Especially we focus our attention on light-driven actuators such as artificial muscle, motor, and valve. We summarize the photomechanical effects in liquid crystal elastomer, amorphous polymer, monolayer, and supramolecules containing azobenzene, respectively.

  • An automatic target sensing and triggering system for small fire arms is proposed. The system consists of an optical collector, an infrared ray sensor responsive to human body temperature, an electric actuator and a trigger mechanism. TRIZ methodologies are used to develop solutions to several contradictory problems. Experimental results on the system performance is compared with predictions.
