A scanning electron microscopic study was performed on the surface ultrastructure of Pygidiopsis summa (Digenea: Heterophyidae) adults. Metacercariae were collected from gills and muscles of mullets (Mugil cephalus) caught in a known endemic area, and adult flukes were harvested from dogs after 8 weeks of experimental infection. The worm was calabash form with its posterior part broader than the anterior part. Tegumental spines were densely distributed over the body surface, except on the suckers and genital apparatus, and around the excretory pore. Well differentiated spines were observed on the anterior half of the body, with 14-16 tips ventrally, and 19-20 tips dorsally. On the oral sucker. three pairs of type I sensory papillae (uni-ciliated knob-like swellings) and one pair of type II sensory papillae (aciliated round-swellings) were observed on the anterior and posterior parts of the lip, respectively. On the lip of the ventral sucker, one pair of type II sensory papillae was distributed only on its posterior part. Sperms were seen emerging from or entering into the genital apparatus. The results showed that the surface ultrastructure of P. summa was unique among the heterophyid trematodes, especially in digitation of tegumental spines and in distribution of sensory papillae on oral and ventral suckers.
An electron microscopic study was performed to know the basic tegumental structure of Cysticercus cellulosae. The scolex and bladder portions of cysticerci (human and porcine strains) were prepared for transmission and scanning electron microscopy by conventional procedures. In general, the tegument of C. cellulosae showed the basic ultrastructure of cestode tegument on electron micrographs. The teguments of both scolex and bladder portions consisted of such components, i.e., an outer vesicular layer with numerous microtriches and inner filroug layer. Below the fibrous layer, there were layers of muscle bundles and tegumental cells. The microtriches which covered the surface of cysticercus revealed two distinctly different shapes. The characteristic bladder-like, elongated pyramid shaped "tetrahedrial form" was observed on the surface of the scolex portion, whereas the elongated cylindrical "filamentous form" was distributed on the stirface of bladder portion. In spite of the difference of isolated host and location, the cysticerci showed tole same result. But dimensional variations of the tegument according to topography of the worm were observed. The possibility of application in making differential diagnosis from other larval cestodes and possible functions of this larval tegument were discussed.
The surface ultrastructure of Acanthotrema felis (Trematoda: Heterophyidae) adults, recovered from a kitten experimentally infected with the metacercariae, was observed using a scanning electron microscope. The worm was leaf-like, ventrally concave and covered with scale-like multi-pointed tegumental spines. The spines on the anterior surface were short but broad, and had 10-12 pointed tips. The cytoplasmic processes protruded around the spines, like pockets for the spines. The ventrogenital opening was crescent, or kidney-shaped, and had protuberances with minute spines on its surrounding tegument. The spines on the posterior surface were long, but narrow, with 6-8 pointed tips. The cytoplasmic processes on this tegument were ridge-like, and elevated along the row of the spines. The surface ultrastructure of A. felis is generally similar to that of other heterophyid flukes, but some features are characteristic, and may be of taxonomic and bio-ecological significance.
The surface ultrastructure of metacercariae and adults of Gymnophallokles seoi, the only known gymnophallid infecting humans, was observed by scanning electron microscopy. Metacercariae were ovoid or pyriform in shape and slightly concave ventrally. The oral sucker had two sizes of type I papillae, small and large, encircling its lip. Type I papillae were arranged in a row on both side oi the body. The ventral pit had several type I papillae on its inner surface. The ventral sucker was covered with cobble-stone like cytoplasmic processes and had 6 type I papillae on its lip. The surface of the body was covered with the tegumental spines except for the ventromedian area between the two suckers. The spines at anterior body were digitated into 3-5 points, and their size decreased at posterior one third or the body. Adult worms were rhomboid or ellipsoid in shape and covered with tegumental spines except for the ventromedian area. The shape and distribution of the tegumental spines and sensory papillae were similar to those of metacercariae. However, sensory papillae arranged in a row on the ventral surface of metacercariae were not observed in adults. The ventral pit became larger and more prominent as the fluke grew. It is suggested that the ventral pit function as an additional adhesive organ to the host tissue.
A scanning electron microscopic study was performed to observe the tegumental ultrastructures of Paragonimus iloktuenensis according to its developmental stages. The metacercariae were obtained from the liver of the brackish water crab, Sesarma dehaani. Juvenile and adult p. iloktsuenensis were recovered from the experimental rats on 2, 4 and 8 weeks after infection. The findings were summarized as follows: 1. The encysted metacercariae were charaeteristically gourd-shape, with their whole body surface beset with numerous spade-shape spines. The Barge, type II sensory papillae (non-ciliated round swellings) were arranged along the rim of the oral and ventral suckers, 11~12 and 6~8 in numbers respectively, 2. Two-week old juvenile worms, recovered chieay from the liver of the experimental rats, were slender in body shape: with their ventral sucker near the anterior one-third level. The distribution of tegumental spines was less dense than in the encysted metacercariae. The spines were with 1~2 pointed tips and 3~.4 longitudinal splits. Numerous ciliated knob-like, type I papillae were observed in both sides of the oral sucker, and 6 large, type If papillae were arranged along the rim of the ventral sucker. 3. Four week old worms, recovered from the thoracic cavity and/or lung parenchyme of the experimental rats, were thicker than wide in body configuration, and their ventral sucker was located near the anterior one.fourth level. The tegumental spines at ventral surface were grouped, each group with 3~5 aggregated ones. The type I and type II papillae (small-sired) were distributed chiefly around the rim of two suckers. 4. Adult (eight-week old) worms, recovered from the capsules in the lung parenchyme, were very stout, and covered densely with bearfoot like spines. At dorsal surface, cobblestone-like cytoplasmic processes were well-developed, with many tegumental spines embedded in them. It was observed in this study that the tegument of p. iloktsuenensis continued to change and differentiate as the worms grew to be adults.
Cercaria caribbea LVIII Cable, 1963 (Digenea: Cyathocotylidae) was detected from a brackish water gastropod species (Cerithideopsilla cingulata) in a coatal area of Shinan-gun, Jeollanam-do (Province), the Republic of Korea, and its surface ultrastructure was studied using a scanning electron microscope. The cercariae were found freely swimming or enveloped within daughter sporocysts when the snail host was mechanically broken. They were morphologically characterized by a linguiform and ventrally concave body, a long and bifurcated tail, and the presence of a holdfast (=tribocytic) organ posterior to the ventral sucker. On the whole ventral and dorsal surfaces, peg-like tegumental spines were densely distributed. Around the oral sucker, several sensory papillae, each with a short cilium, were distributed, and on the tail, sensory papillae, each with an extensively long cilium, were observed. This is the first record describing a cyathocotylid cercaria from a brackish water gastropod in the Republic of Korea.
Antigenic localization in Parofonimn iloktsuenensis worm tissues (tegument, intestine and vitelline gland) in different developmental stages of 2 weeks, 3 weeks, 4 weeks, 5 weeks and 33 weeks from albino rats (Sprague-Dawley) infected with P iloktsuenensis was observed by electron microscopy. These worm tissues of different developmental stage of R iloktsuenensis was observed on electromicrograph by immunogold labeling method using R iLoktsuenensis infected rat serum of 10 weeks. Antigenic localization was demonstrated as labeling of gold particles in tissues on electronmicrograph. In tegument, gold particles were labeled on tegumental tissue, generally more numerous on secretory granules in tegumental syncytium 2 weeks than those on the other elder developmental stages, but there was a little variation in antigenicity according to individual worm tissue. In general, antigenicity in tegumental tissue was not strong (gold particles: 0.1-5/1 Mm2). In intestine, a large number of gold particles (15-18/1 Mm2) were labeled in intestinal epithelium. Gold particles were concentrated especially on secretory granules in cytoplasm, and gold particles were labeled not only in cytoplasmic protrusions, but also in intestinal luminal contents. Intencity of labeling of gold particles was not correlated with developmental stage of worms. In vitelline gland, a large number of gold particles were labeled on vitelline globules. The gold particles in vitelline globules (8- 11/1 Mm2) were concentrated in protoplasm among segmental globules . In order to observe the antigenic localization in the tissues of the young adult Paragenimus westermani, immunogold labeling method was applied using serum immunoglobulins (IgG) of the dog which infected with isolated metacercariae from Cambaroides similis. The sectioned worm tissue was embedded in Lowicryl HM 20 medium and stained with infected serum IgG and protein A gold complex(particle size; 12 nm) It was observed by electron microscopy at each tissues of the worm. The gold particles were not observed on the basal lamina of the tegument, interstitial matrix of the parenchyma, the muscle tissue and mitochondria of the tegument. The gold particles were specifically labeled in the secretory granules in the vitelline cells. They were predominantly labeling on the epithelial lamela and lumen of caecum. The above finding showed that antigenic materials in young adult worm tissue were specifically concentrated on the tegumental syncytium as well as cytoplasm of tegumental cells.
An electron microscopic study was performed to observe the ultrastructure of the tegument of U seoulensis. The outer surface of the tegument was covered with a tnlaminated plasma membrane. The electron-dense cytoplasmic layer was $2.5{\;}\mu\textrm{m}$ wide In the anterior portion and contained numerous vacuoles, mitochondriae and granular materials in its matrix. The basement layer was 330 nm wade or so, and Its numerous extensions protruded into the cytoplasmic layer. The sensory organ was composed of a small vesicle of $1.7{\;}{\times}{\;}1.1{\;}\mu\textrm{m}$ in dimensions, which possessed a cilium of $1.2{\;}{\times}{\;}0.19{\;}\mu\textrm{m}$ in size. The pharynx was composed of the epithelial layer of about $0.5{\;}\mu\textrm{m}$ wide, well developed muscle layer and basement layer. The tegument of the oral sucker was composed of a cytoplasmic layer of $0.4-0.5{\;}\mu\textrm{m}$ width, a narrow basement layer, a well developed muscle layer and tegumental cells. Some kinds of secretory granules that seemed to be originated from the cells of the oral sucker were observed In the parenchymal portions of the adjacent cells. The tribocytic organ consisted of numerous microvilli. The microvilli were 5 nm wide and heptalaminated. Two types of secretory granules originated from the gland cells of tribocytic organ were observed In the tegument and parenchyme. The tegumental cells were irregular in shape, and of which nuclei were multifarious.
The present study was performed to observe the ultrastructure of the integument of adult Paragonimus westermani. Dogs experimentally infected with 60 metacercariae of F. westermani were autopsied 4 months after the infection. Adult p. westermani were extracted from the dogs and the fine structure was studied by means of scanning and transmission electron microscope. The findings are as follows: 1. Scanning electron microscopic findings showed that the spines and the papillae are distributed at whole body surface but the well developed spines or papaillae are shown around the oral sucker and ventral sucker. 2. At the end of the body, excretory pore was found, the shape was irregular. 3. Transmission electron microscopic findings showed that plasma membrane, tegument, basal lamina, connective tissue, circular muscle layer, longitudinal muscle layer. nerve axon and tegumental cell were observed. 4. In higher magnification, plasma membrance and bar-shaped granules were found at the outer surface of the tegument.
