단백질 함량 및 철분 농도의 변화에 따라 홍차 추출물이 철분의 용해도에 미치는 영향 (Effects of Protein and Iron Concentrations on Iron Solubility in Black Tea Infusion)

  • Tannins in plant foods and beverages may produce antinutritional or toxic effects although some proteins with high affinity for tannins seem to function as defense mechanism to tannin toxicity. Our objectives were to investigate of tea tannins, iron and protein and to evaluate the role of proteins in tannin effects on iron solubility. Iron solubility in vitro was measured using tea with and without proteins. Mixtures of tea, protein in varying concentrations(either gelatin or bovine serum albumin), and iron(eithe 10 or 50ug/mL) were prepared. Controls contained water in place of tea. Iron bioavailability was assessed by measuring iron solubility in the simulated gastric condition with pepsin digestion. Bound iron was removed by centrifugation and soluble in tea alone. When iron concentratin was 10ug/mL, addition of small amounts of protein to tea dramatically reduced iron solubility, but solubility of iron increased in the tea mixturea as the concentration of protein was increased. The percnetage of iron that precipitated was much greater at 10ug Fe/mL than the values at 50ug Fe/mL suggesting that the iron binding sites on the tea-protein complex was saturated. These results suggest that interactions of iron with tea tannins are influenced by the concentratins of protein and iron.

Effects of Green Tea Infusion on the Preneoplastic Lesions and Peroxidation in Rat Hepatocarcinogenesis

  • The effect of green tea drinking on the hepatocellular chemical cacinogenesis have been studied. Placental glutathione S-transferase(GST-P) positive foci area in a liver tissue, contents of thiobarbituric acid reactive substances(TBARS), total cytochrome P450 and glucose 6-phospphatase(G6P) activity in hepatic microsomes were investigated. Weaning Sprague-Dawley male rats were fed AIN-76A diet with deionized water or green tea infusion, Rats of CTR and CTR+ groups were provided deionized water while GTI and GTI+ groups were provided green tea instead of deionized water for the entire experimental period of 13weeks. Rats of GTP and GTP + groups had deionized water for the first 6 weeks and switched to green tea for the last 7weeks of the experimental period. CTR+, GTI +, and GTP + groups were carcinogen treated groups, Diethylnitrosamine(DEN) was injected as a single dose of 200mg/kg body weight intraperitoneally after 4 weeks of feeding. 2-Acetyla-minofluorene(AAF) was used as a carcinogen proliferater and suppled in the diets of carcinogen treated rats as 0.02% content for the last 6weeks starting from 2weeks after DEN injection. Rats were sacrificed after 13week weeks of feeding. The area and number of GST-P positive foci detected in carcinogen treated rats were decreased by green tea ingestion but when timing and duration of green tea ingestion was delayed after promotion period as in GTP + group, GST-P positive foci were not decreased as much as in GTI+ group. TBARS contents of carcinogen treated rats decreased by 13weeks of green tea ingestion but GTP groups did not show statiscally significant differences. G6P activities tended to decrease by carcinogen treatment but changes were not statiscally significant by green tea ingestion. Total cytochrome P450 contents were increased by carcinogen treatment. Thirteen weeks of green tea ingestion (GTI) also increased to total cytochrome P450 contents while 7weeks of green tea ingestion(GTP) did show any effects. These results suggest that green tea has suppressive effects on hepatocellular chemical carcinogenesis probably through the activities of antioxidant compounds. Enteral Infusion of Green Tea Extract Selectively Enhances the Biliary Secretion of 14C-Benzo[a]pyrene in Rats without Affecting Other Biliary Lipids

  • Recently, we have demonstrated that green tea extract (GTE) decreases the intestinal absorption of benzo[a]pyrene (BAP), which is an extremely lipophilic food contaminant. The present study was conducted to examine if an enteral infusion of GTE would influence the biliary secretion of BAP and lipids in rats. Female rats were fed an AIN-93G diet with or without (control) GTE at 5 g/kg diet for 4 week. Following the 4-week dietary treatment, rats with bile duct cannula were infused continuously for 8 hr at 3.0 mL/hr via a duodenal catheter with a lipid emulsion containing $4.0\;{\mu}mol$ BAP labeled with $^{14}C$ ($^{14}C$-BAP), $20.7\;{\mu}mol$ cholesterol, $452\;{\mu}mol$ triolein, and $3.1\;{\mu}mol$ ${\alpha}$-tocopherol, and $396.0\;{\mu}mol$ Na-taurocholate with or without 76.1 mg GTE powder in PBS buffer (pH, 6.4). Bile was collected hourly via bile cannula for an 8 hr period. Our results showed that bile flow did not differ between groups. However, the biliary secretion of $^{14}C$-BAP was significantly enhanced by GTE infusion, compared with those infused with the lipid emulsion alone. However, GTE did not affect the biliary outputs of cholesterol, fat, phospholipid and ${\alpha}$-tocopherol. These findings indicate that GTE has a profound stimulatory effect on the biliary excretion of BAP in rats, without affecting other biliary lipids. The mechanism(s) by which GTE enhances the biliary secretion of BAP remains to be investigated.

면섬유에 대한 홍차색소의 염색성 (Dyeing Properties of Cotton with Black Tea Colorants)

  • The colorante from black tea infusion were applied to cotton fabric. Dyeing properties were investigated in terms of dye uptakes at various dyeing conditions and the effect of mordants on color change and colorfastness. Cotton showed low affinity to black tea colorants. Effects of mordants on dye uptake was not significant except Sn. Irrespective of mordant type, color of cotton dyed with black tea colorants was brownish and colorfastness was generally good. In order to improve dye uptake, cotton was cationized by treating with Cationon UK(tertiary ammonium salt) and chitosan. Dye uptake of cotton was remarkably increased by cationizing. Cationon UK was more effective than chitosan.

음용 조건으로 추출한 티백 형태 순수 녹차의 카테킨 함량 및 산화방지능 (Effects of commonly used infusion method on catechin content and antioxidant capacities of pure green tea packaged in tea bags)

  • 국내외 티백 녹차 10종을 $70^{\circ}C$의 생수 150 mL에 2분간 음용 조건으로 추출하여, 총페놀 함량, 총플라보노이드 함량, 산화방지능, 카페인 및 카테킨 4종을 정량 분석하였다. 10종의 티백 녹차의 총페놀 함량은 46.3-93.9 mg GAE/g, 총플라보노이드 함량은 10.3-21.7 mg CE/g, DPPH법과 ABTS법을 이용한 산화방지능은 각각 88.8-175.6 mg VCE/g과 87.9-183.5 mg VCE/g이었다. HPLC를 이용한 정량 분석에서 카페인 함량은 9.3-24.0 mg/g, 총카테킨 함량은 33.8-74.7 mg/g이었다. 총카테킨 함량을 녹차 추출 용액 한잔(150 mL)으로 환산했을 때, 건강기능식품 고시형 기능성 원료인 녹차추출물의 기준 및 규격을 고려치 않는 가정 하에 녹차 티백 3.8-12.5잔을 마시면 카테킨 일일섭취량(300-1,000 mg)을 충족시킬 수 있을 것이다.

홍차색소의 성분과 특성 (Components and Characteristics of Black Tea Colorants)

  • Colored components in black tea were extracted, freeze-dried, and analysed to investigate the possibility using as a natural dye. Fractionation of the colored components was carried out by gel permeation chromatography. The colored components in black tea were elected into seven fractions. Each fraction was analyzed by UV spectrophotometer. The early fluted fractions 1-4 did not show any absorption peaks in 320-700 nm and showed the increase in absorption as it approaches to short wavelength and are considered as highly polymerized colored substances. Fractions 5-6 showed tar at 350 m and are considered as thearubigins. Fraction 7 showed absorption peaks at 376 nm and 456 nm and is considered as theaflavin. IR spectra of each fraction show: Strong C=0 stretching band at 1650 cm-1 appears in fractions 1-4, but not in fractions 5-7. Strong C=0 stretching band at 1700 cm-L appears in fraction 3-7. C=0 stretching band at 1610 cm-1 appears as a shoulder in fraction 4 and progressively changes into strong peak in fraction 5-7. From these results, it is assumed that colored components in black tea consist of polyphenolic substances having different molecular weight which were formed during tea manufacturing process. The colorants from black tea infusion were applied to silk, wool, cotton and nylon fabrics. Black tea colorants showed high affinity to wool, silk and nylon, but very low affinity to cotton fabrics.

Chemical Composition of Green Teas According to Processing Methods and Extraction Conditions

  • This study examined the influence of manufacturing processes and extraction conditions on the chemical compositions of green tea. Green tea samples grown in various areas (Korea, China, and Japan) and processed by 4 different methods (steaming, pan-firing, steaming and pan-firing, and heavy roasting after steaming and pan-firing) were collected for study. The chemical compositions of the green tea extracts and infusions were different according to their processing methods and extraction conditions, including catechins, caffeine, and free amino acids contents. In all samples analyzed, (-)-epigallocatechin gallate (EGCG), (-)-epigallocatechin (EGC), and theanine were determined as the major catechins and free amino acid, respectively. Studies of samples grown in the same area (Jeju; Korea) showed that there were significant differences in the concentrations of catechins and caffeine in extract and infusion according to the processing methods. These results indicate that processing methods influenced the chemical compositions of the green tea extracts and infusions.

아로니아잎차의 덖음조건에 따른 항산화 성분과 항산화 활성 및 카페인 함량 (Antioxidant Compounds and Activities as well as Caffeine Content of Aronia melanocarpa Leaf Tea according to Pan-Roasting Conditions)

  • 본 연구는 아로니아 생잎과 여러 가지 덖음조건에 따라 아로니아잎차를 제조하고, 이들의 항산화 유용성분과 항산화 활성도를 비교하였다. 생잎에 비해 아로니아잎을 6회 덖은 경우 총폴리페놀 및 플라보노이드 함량이 각각 37.96 CE mg/g, 19.96 QE mg/g으로 가장 높게 나타났다. 또한, 6회 덖은 아로니아잎 추출물은 DPPH 및 ABTS 라디칼 소거능도 각각 $IC_{50}$ 0.43 mg/mL 및 0.27 mg/mL로 가장 높은 항산화 활성을 나타내었다. 그러나 덖음조건에 따른 폴리페놀 함량 변화에 영향을 주는 인자에 대해서는 추후 연구가 필요하다. 아로니아 생잎과 잎차 모두 카페인은 함유하지 않는 것으로 확인되어 아로니아 잎차는 무카페인 차로써의 가치가 있음을 확인하였다. 결론적으로 아로니아잎에 함유된 총폴리페놀, 총플라보노이드와 같은 항산화 유용성분은 물론 항산화 활성을 극대화할 수 있는 최적화된 덖음조건에 대한 기초자료를 확보할 수 있었으며, 아로니아 잎은 카페인을 함유하지 않는 항산화 기능성 잎차 또는 바이오식품 소재로서 그 활용가치가 기대된다고 할 수 있다.

  • Colorants in green tea were extracted freeze-dried and analyzed to investigate the possibility of using as a natural dye. Fractionation of the colorants was carried out by column chromatograpy. Colorants in green tea were eluted into five fractions. All the fractions except fraction F2 showed absorption peakat 280nm. Fraction f2 showed absorption peak at 270nm and broad peak at 350nm, From the IR analysis it is speculated that fractions F2-F5 having similar stucture but different molecular weight are catechis. Silk fabrics dyes with fractions F1-F4 showed yellowish red color while sample dyed with fraction F5 showed red color. The colorants from green tea infusion was applied to silk wool nylon cotton and rayon fabrices. It showed relatively good affinity to protein and polyamide fibers bur low affinity to cellulose and regenerated cellulose fibers.

산뽕잎 및 은행잎 차의 항산화 활성과 플라보노이드 함량 (Antioxidant Capacities and Flavonoid Contents of Wild Mulberry and Ginko Leaves Teas)

  • 산뽕잎, 은행잎 및 혼합차의 침출시간에 따른 항산화 활성을 DPPH 라디칼 소거 활성과 FRAP 방법으로 조사하였으며, 총 페놀 화합물의 함량은 Folin-Ciocalteu법, 플라보노이드 함량은 HPLC로 분석하였다. 침출시간에 따라서 DPPH 라디칼 소거활성과 FRAP 활성은 은행잎차, 혼합차, 산뽕잎차 순으로 높게 나타났다. 총 페놀 성분 함량은 은행잎차, 혼합차, 산뽕잎차 순으로 높았으며 산뽕잎에 은행잎을 2:1로 혼합하였을 때 항산화 활성이 증가하였다. 침출시간 3분 정도에서 산뽕잎차 및 혼합차는 비교적 항산화 활성이 높았으며 은행잎차는 6분 까지 활성이 증가하였다. HPLC로 분석한 플라보노이드 성분 함량은 quercetin이 모든 차에서 가장 함량이 높았으며, 은행잎차, 혼합차, 산뽕잎차 순으로 플라보노이드의 총 함량이 높게 나타났다. 제조한 침출차 들의 항산화 성분 함량과 항산화 활성의 상관계수는 0.87 이상으로 상관성이 상당히 높았다.
