  • Effects of double packaging (combinational use of aerobic and vacuum conditions) and antioxidants on physicochemical properties in irradiated restructured chicken rolls were determined. Chicken breast treated with antioxidants (none, sesamol+a-tocopherol) was used to process restructured chicken breast rolls. The sliced rolls were vacuum, aerobic, or double packaged (vacuum for 7 d then aerobic for 3 d) and electron beam irradiated at 2.5 kGy. Color, 2-thiobarbituric acid reactive substances (TBARS), oxidation reduction potentials (ORP), and volatile profiles of the samples were determined at 0 and 10 d. Irradiation made restructured chicken rolls redder (p<0.05), and the increased redness was more distinct in irradiated vacuum-packaged than irradiated aerobic or double packaged meats. TBARS values of antioxidant-treated double packaged rolls were lower than even nonirradiated vacuum-packaged meat, and those were distinct at 10 d (p<0.05). ORP and lipid oxidation values were lower in irradiated vacuum and double packaged samples than those in irradiated aerobic packaged ones at 0 d (p<0.05). Irradiation of restructured chicken rolls increased the amount of total volatiles. Considerable amounts of off-odor volatiles were reduced or not detected by double packaging and antioxidant treatment at 10 d. Therefore, the combined use of antioxidants and double packaging would be useful to reduce redness and control the oxidative quality changes of irradiated restructured chicken rolls

  • Characterization and utilization of the bioactive compounds from natural resources is one of the most concerns to maintain quality properties of foods, especially to prevent the oxidation of lipids in meat products. Phytochemical components and antioxidant activities of Cudrania tricuspidata (CT) leaves extracted using various solvents and their effects on physicochemical properties of pork patties during refrigerated storage were measured. The combined solvents of 80% ethanol, 80% methanol and pure double-distilled (dd)-water obtained the higher total phenolic compounds, flavonoids content, and antioxidant activities as compared to the pure solvent alone. Among the individual antioxidant components, catechin was the predominant polyphenol in CT leaves in all extracts. The addition of CT leaves extracts into pork patties showed high antioxidant activities since thiobarbituric acid-reactive substances (TBARS) values of added CT extracts were lower than those of the control (p<0.05). In conclusion, CT leaf phytochemical components displayed antioxidant activity that varied with the extract solvent used. CT extracts were superior to control in retarding lipid oxidation of pork patties, which was evident as reduced TBARS and peroxide values (POV).

  • The aim of this research was to evaluate the effects of rosemary and kimchi powder on the quality characteristics of pork patties. Cooked pork patties was stored at $3^{\circ}C$ for 15 days. Rosemary powder, either alone or with Kimchi powder, was highly effective in inhibiting lipid oxidation. The AV and TBARS values of R4(additional group of 4% rosemary powder) and RK(additional group of 2% rosemary powder +2% kimchi powder) were slightly lower than those of the control group. Sensory preferences for pork patties decreased with increased of storage time period. In a sensory evaluation, R4 showed the low sensory scores, whereas RK was highly evaluated compared to control. In conclusion, we determined that the pork patties with added 2% rosemary powder and 2% kimchi powder had the most favorable antioxidant effects and sensory qualities.

  • This study was performed to investigate the effects of lipid on cellular senescence, lipid peroxide production, and morphological changes. For this study we used primary skin fibroblasts from neonate rats grown in media various lipid contents. Fibroblasts were cultured until they lost their proliferation potential either in control medium (Dulbecco's modified Eagle's medium supplement with 10% fetal bovine serum) or in media supplemented with various concentrations of lipid-cholesterol rice component from bovine serum. Cumulative population doublings(CPD, as an index of cellular life span), and cellular thiobarbituric acid reactive substances (TBARS, as an index of lipid peroxide) concentrations were measured and morphological changes were observed. CPD were shortened with increasing lipid concentration in media ; 28.12 for cells grown in control medium and 13.42, 11.42, and 6.19 for those grown in 0.1%, 1% and 5% lipid rich components containing media, respectively. Cellular proliferation ratios were those grown in 5% lipid rich components containing media were delayed and they were degenerated soon. TBARS concentrations were increased with increasing concentration of lipid in media. Morphological changes were observed in cells grown in control medium by cellular senescence. Especially lipid droplets were observed in cells grown in 5% lipid rich components containing media. Therefore it seems that lipid contents in media had an effect on cellular proliferation and cellular life span, possibly via lipid peroxide production.

  • This study was performed to see effects of whole grape, grape pomace or grape juice intakes on lipid metabolism during aging in old Sprague-Dawely male rats. One hundred twenty rats of 13 months old Sprague-Dawely were blocked into eight groups according to body weight and raised for 3, 5 or 7 months with diets containing 2% (w/w) dried powders of three different pars of grape and 0.02% (w/w) CdCl₂. Body weights of Cd groups were lower than Cd free groups. Kidney and spleen weights were incre ased with age, and EEP weights of Cd groups were lower than those of Cd free groups. Total lipid, triglyceride and total cholesterol concentration in plasma increased with age. Whole grape, grape pomace or grape juice intake lowered plasma total lipid triglyceride and total cholesterol concentrations, and especially grape pomace lowered them markedly. Whole grape, grape pomace or grape juice intake decreased liver total lipid, triglyceride and total cholesterol concentrations and increased fecal lipid excretion. Grape diets decreased and Cd administration increased TBARS concentration in LDL fraction. In conclusion, grape diets were effective in decreasing lipid levels of liver and plasma, TBARS in LDL, and in increasing HDL cholesterol. The grape pomace was most effective among three grape parts. It is plausible that grape might be recommended for the treatment or prevention of cardiovascular disease and delaying aging. (Korean J Nutrition 35 (7): 713∼728, 2002)

  • We investigated whether the fermented soymilk extract (FSE) has protective effects against high glucose-induced oxidative stress in human umbilical vein endothelial cells (HUVECs). FSE was prepared via fermentation of soymilk with Bacillus subtilis followed by methanol extraction. To determine the protective effect of FSE, oxidative stress was induced by exposing of HUVECs to the high glucose (30 mM) for 48 hr. Exposure of HUVECs to high glucose for 48 hr resulted in a significant (p<0.05) decrease in cell viability, catalase, SOD and GSH-px activity and a significant (p<0.05) increase in intracellular ROS level and thiobarbituric acid reactive substances (TBARS) formation in comparison to the cells treated with 5.5 mM glucose. However, at concentration of 0.1 mg/mL, FSE treatment decreased intracellular ROS level and TBARS formation, and increased cell viability and activities of antioxidant enzymes including catalase, SOD and GSH-px in high glucose pretreated HUVEC. These results suggest that FSE may be able to protect HUVECs from high glucose-induced oxidative stress, partially through the antioxidative defense systems.

  • This study was conducted to evaluate and compare the quality changes of commercial sliced and pizza cheeses processed by gamma and electron beam irradiation. The $L^*$-value of sliced and pizza cheeses decreased and the $a^*$-value decreased only in pizza cheese by both irradiation sources. There was no change in pH. There was no difference in 2-thiobarbituric acid reactive substances (TBARS) value between non-irradiated and irradiated samples at a dose of 3 kGy or less (p<0.05). However, both irradiation sources resulted in increased TBARS value in sliced and pizza cheeses at 5 kGy. Sensory evaluation revealed that irradiation influenced odor, taste and overall acceptability of both cheeses and may cause the limitation of consumers' acceptance for irradiated cheese products. Results indicate that both gamma and electron beam irradiations with less than 3 kGy may not influence significantly the physicochemical quality of sliced and pizza cheeses. However, to meet a market requirement, a method to overcome the sensory deterioration of cheeses should be developed and applied.

  • This study was performed to investigate the effects of dried powder and juice of Aster scaber on lipid metabolism and antioxidative capacity in rats. Twenty one male Sprague-Dawley rats weighing 170.9$\pm$15.1g were blocked into 3 groups according to body weight and raised for 4 weeks with diets containing 5%(w/w) dried powder and juice from the same amount of Aster scaber. Food intake, body weight gain, food efficiency ratio and organ weights were not different among all the experimental groups. On lipid metabolism, both dried powder and juice supplementation decreased plasma total lipid, triglyceride(TG), total cholesterol and liver cholesterol levels by increasing fecal lipid excretions. Plasma thiobarbituric acid reactive substance(TBARS) concentrations were significantly decreased in dried powder and juice groups, whereas liver TBARS concentrations were not affected by experimental diets. Red blood cell(RBC) superoxide dismutase(SOD) activities were significantly increased by dried powder and juice supplementation, while activities of other antioxidative enzymes in RBC and liver were decreased or showed no changes. In conclusion, both Aster scaber dried powder and juice had the effects on lowering lipid levels by increasing fecal lipid excretions, and inhibiting of plasma lipid peroxidation in animals and the effects of juice were higher than that of dried powder. • We investigated the effects of antioxidant vitamins supplementation on antioxidative status and plasma lipid profile in female Korean non-insulin diabetes mellitus(NIDDM) patients. Forty-five patients were groups by types of vitamin to take into three groups-Vitam in E group(400IU/day, n=15), Vitamin C group (1,000mg/day, n=15) and Vitamin E plus C group (400 IU plus 1,000mg/day). Supplementation period was 4 weeks. After vitamins supplementation, plasma vitamin E concentration significantly increased in vitamin E and vitamin E + C group, but plasma retinol concentration were not affected by vitamin E or vitamin C supplementation. And plasma levels of lipid peroxide measured as thiobarbituric acid reactive substances(TBARS), indicator of lipid peroxidation and increased susceptibility of LDL towards lipid peroxidation, were significantly decreased in all three groups after vitamins supplementation. Also catalase activities in erythrocytes were significantly decreased after antioxidant vitamin supplementation in all subjects. And after vitamins supplementation, post prandial 2 hour glucose and total cholesterol levels was decreased in all subjects. And after vitamins supplementation, post prandial 2 hour glucose and total cholesterol levels was decreased in all patients, especially there was a significant difference in vitamin C, Vitamin E + C group. In this study, antioxidant vitamins supplementation might have a protective function against the free radical-mediated lipid peroxidation and decrease the plasma total cholesterol levels in Korean female NIDDM patients.

  • The present study was designed to investigate the efficacy of selective $ET_A$ receptor antagonist, ambrisentan on hyperhomocysteinemia-induced experimental vascular dementia. L-methionine was administered for 8 weeks to induce hyperhomocysteinemia and associated vascular dementia in male rats. Ambrisentan was administered to L-methionine-treated effect rats for 4 weeks (starting from $5^{th}$ to $8^{th}$ week of L-methionine treatment). On $52^{nd}$ day onward, the animals were exposed to the Morris water maze (MWM) for testing their learning and memory abilities. Vascular endothelial function, serum nitrite/nitrate levels, brain thiobarbituric acid reactive species (TBARS), brain reduced glutathione (GSH) levels, and brain acetylcholinesterase (AChE) activity were also measured. L-methionine-treated animals showed significant learning and memory impairment, endothelial dysfunction, decrease in/serum nitrite/nitrate and brain GSH levels along with an increase in brain TBARS levels and AChE activity. Ambrisentan significantly improved hyperhomocysteinemia-induced impairment of learning, memory, endothelial dysfunction, and changes in various biochemical parameters. These effects were comparable to that of donepezil serving as positive control. It is concluded that ambrisentan, a selective $ET_A$ receptor antagonist may be considered as a potential pharmacological agent for the management of hyperhomocysteinemia-induced vascular dementia.
