Effect of Younnyeniksoobulrodan(延年益壽不老丹) on Antioxidant Capacity in D-galactose induced Aging Rats (연년익수불로단(延年益壽不老丹)이 노화유발 흰쥐의 항산화능에 미치는 영향)

  • Objectives: Younnyeniksoobulrodan(延年益壽不老丹) composed of Polygonum multiflorum THUNB. and some medical herbs is known as formula of senescence delay effect. The purpose of this study is to investigate the effect of Younnyeniksoobulrodan(延年益壽不老丹) on antioxidant enzyme activity such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase (GSH-px) in rat erythrocytes and liver. Methods: Sprague-Dawley rats divided into 4 gorups, Young group(8 weeks old, N-8), Aging group(18 weeks old, N-18), pathologically induced aging gorup(injected D-galatose 50mg/kg, 1time/day for 6 weeks, CON) and Younnyeniksoobulrodan(延年益壽不老丹) administered group(D-galactose 50mg/kg and Younnyeniksoobulrodan extracts 840.0mg/kg 1time/day for 6 weeks, YIB). Rats were sacrificed and TBARS, SOD, CAT, and GSH-px were measured in rat erythrocytes and liver. Results: Plasma and liver TBARS concentrations of YIB group were significantly lower than those of control. Red blood cell(RBC) SOD activities of YIB group was increased(F=3.445, p=0.033, ANOVA test), and RBC catalase activities of all experimental group were not significantly different. RBC GSH-px activities of YIB group was increased(F=9.365,p=0.0001, ANOVA test). Liver SOD activities of YIB group was higher than those of control(F=4.967, p=0.008, ANOVA test). Liver catalase activities of all experimental group were not significantly different, and liver GSH-px activity of YIB group was significantly higher than that of control(F=3.846, p=0.022,ANOVA test). Conclusions: According to the above results, it is considered that Younnyeniksoobulrodan is effective in inhibiting lipid peroxidation and increasing antioxidative enzyme activities in D-galactose induced aging rat.

Effect of Olive-Oil Prepared Tomato Powder (OPTP) and Refining Lycopene on the Physicochemical and Sensory Characteristics of Seasoned Raw Pork During Storage (올리브유처리 토마토 분말과 정제 Lycopene이 저장 중 양념육의 물리화학적 및 관능적 특성에 미치는 영향)

  • This study was carried out to determine the effects of olive-oil prepared tomato powder (OPTP) and lycopene on seasoned raw pork. 1.5% OPTP was added in T1 and 3.0% in T2; and 0.03% refining lycopene was added in T3. While in storage, pH values of the treated samples were lower (p<0.05) than those of the control samples. Upon increasing storage, salinity values of OPTP samples increased as well, however, the values of T3 did not change. There were significantly higher TBARS values observed in the OPTP samples compared to control, but no significant difference between T3 and the control samples in TBARS values during storage. VBN values of T2 samples were lower than that of other samples at Day 3 and Day 6 of storage. At Day 9 however, the VBN values of T1 and T2 were significantly higher than those of the control and T3. There was no significant difference between the control and the treated samples in the total plate counts. There were relatively higher redness, yellowness and chromatic values observed in the treated samples than in control. In sensory evaluation, there was no significant difference between the control and the treated samples in color, tenderness, juiciness and overall acceptability.

  • This study was conducted to investigate the changes in haem pigments, peroxide value, TBARS, free fatty acid contents and fatty acid composition of five muscles from low fat pork cuts during storage at $4^{\circ}C$ for 14 d. The myoglobin contents (Oxy, Met and Total) did not change significantly (p>0.05) as storage time increased. In addition, the peroxide value did not change significantly (p>0.05), but the thiobarbituric acid reactive substances were significantly (p<0.05) upregulated during chilled storage. The total free fatty acid contents of the longissimus dorsi muscle were significantly (p<0.05) upregulated, but the saturated, unsaturated, mono-unsaturated and poly-unsaturated fatty acid composition of the muscles did not change significantly (p>0.05) during chilled storage.

Effects of Aralia canescens and Phellodendron amurense Extracts on Streptozotocin Induced Diabetic ICR Mice (Streptozotocin으로 당뇨를 유도한 생쥐의 간과 체장에서 황백피와 두릅나무 추출물이 지질과 산화물 생성과 글루타티온 의존성 효소의 활성에 미치는 효과)

  • The effects of Aralia canescens and Phellodendron amurense(AP) extracts on the experimental diabetes in ICR mice were investigated. 96male ICR mice were induced diabetes mellitus by intrape-ritoneal streptozotocin injection(75mg/kg B.W.) and divided into two injection groups which are 5 day injection and 10 day injection group. Then, each injection group was subdivided into 8 groups of 6 animals repspectively. CIC served as control and CI1, CI2 and CI3 were treated with 50, 150, 250mg/kg B.W. of AP extracts powder in 0.9% NaCl solution. Animals of groups DIC, DI1, DI2 and DI3 were strepto-zotocin-induced diabetes. DIC served as diabetic control and the rest groups received 50, 150, 250mg/kg B.W of AP extracts powder in saline solution respectively. The body weight, liver and kiney weight changes and blood levels of glucose, cholesterol and triglyceride were measured. Thiobarbituric acid reactive substance(TBARS), and glutathione reductase(GR) and glutathione peroxidase(GPx) activities were also measured for determining antioxidant effects. AP extracts increased the body weight in diabetic groups. The liver and kidney weight/100g B.W. in DIC group were greater than those of normal ICR group but after AP extracts injection, liver and kidney weight were decreased significantly. These effects were more efficient at 10 days injection group. The total, LDL, VLDL cholesterol and triglyceride levels were significantly higher in DIC group and the extent of decrement responded to AP injection dose. The contents of TBARS and antioxidant enzyme activities were relatively decreased after AP extracts injection. These results suggest that the intraperitoneally administered AP extracts may have not only hypoglycemic effect but act as antioxidants by reducing lipid peroxidation.

Effects of Dietary Cadmium on the Respiratory Burst of Phagocytes and the Antioxidant Defense in Cultured Red Seabream (Pagrus major)

  • To examine effects of cadmium on the respiratory burst of kidney phagocytes and antioxidant defense in liver, juvenile red seabream Pagrus major were fed a cadmium-incorporated diet $(1g\;CdC1_2/kg\;diet)$. The respiratory burst activity measured by chemiluminescence (CL) was significantly reduced by oral intake of cadmium. Lipid peroxidation in liver expressed as thiobarbituric acid reactive substances (TBARS) was significantly higher in the fish fed a cadmium-incorporated diet than that of the fish fed a control diet both on Day 3 and Day 9. Liver Glutathione S-transferase (GST) activitiy was significantly increased both on Day 3 and Day 9 by feeding a cadmium-incorporated diet, when compared with the controls. From the present results, it can be concluded that oral intake of cadmium in red seabream is associated with marked reduction of respiratory burst capacity of kidney phagocytes which can elevate susceptibility of fish against infecting pathogens. Cadmium administration also elicits significant increment of lipid peroxidation in liver, and fish try to detoxify cadmium by increasing GST activity.

  • Effects of cimetidine, a cytochrome P450 inhibitor, on the benzo[a]pyrene (BaP)-mediated cytochrome P450 induction and lipid peroxidation of liver in olive flounder, Paralichthys olivaceus, were investigated. Fish were fed either a cimetidine-supplemented diet or a cimetidine-free control diet once daily to satiation for 3 days. After 6 hrs of last feeding, the fish received intraperitoneal (i.p.) injection of BaP (20 mg/kg of body weight) dissolved in sterile corn oil $(100 \mu L)$ or received only a corn oil i.p. injection. At 1, 2, 3, and 7 days after the injection, hepatic cytochrome P450 and thiobarbituric acid reactive substances (TBARS), an indicator of lipid peroxidation, were analyzed. BaP injection resulted in an increase of hepatic cytochrome P450, and the fish fed the cimetidine-supplemented diet before injection of BaP showed delayed increase of hepatic cytochrome P450 compared to the fish fed a cimetidine-free diet and BaP injected. Injection of BaP clearly induced hepatic lipid peroxidation, and consistently higher TBAR values were shown in the fish fed a cimetidine­supplemented diet before injection of BaP than the fish injected with BaP alone.

Effect of Fresh Garlic on Lipid Oxidation and Microbiological Changes of Pork Patties during Refrigerated Storage

  • The effects of two levels (1.4 vs 2.8%) of fresh garlic on lipid oxidation and microbial growth in pork patties were evaluated. Hunter color (L, a, b), pH, thiobarbituric acid reactive substances (TBARS), oxidative volatile compounds, total bacteria and Enterobacteriaceae in the pork patties with or without fresh garlic were measured during storage at $4^{\circ}C$. Addition of fresh garlic decreased redness (a), while increased pH and yellowness (b) values of the fresh pork patties were observed, regardless of the levels added. The TBARS values of the pork patties were increased with the addition of fresh garlic (p<0.05). Similar results were observed in oxidative volatile compounds. A total of 13 volatile compounds were detected in the patties (5 sulfur-containing compounds, including allyl mercaptan, allyl methyl sulfide, diallyl sulfide, methyl-(E)-propenyl-disulfide, and diallyl disulfide, and the 8 other oxidative compounds, including 1-pentanol, hexanal, 1-hexanol, heptanal, (E)-2-heptenal, 1-octen-3-ol, (E)-2-octenal and nonanal). Fresh garlic accelerated development of oxidative products in the pork patties, especially hexanal and the total oxidative volatile compounds. However, the addition of 1.4 and 2.8% of fresh garlic inhibited the growth of total bacteria and Enterobacteriaceae, indicating low total bacterial counts and Enterobacteriaceae than the controls.

Effects of Fucoxanthin Addition to Ground Chicken Breast Meat on Lipid and Colour Stability during Chilled Storage, before and after Cooking

  • Effects of fucoxanthin (FX), a major carotenoid in algae, on lipid peroxidation and meat colour in ground chicken breast meat were investigated. FX and/or ${\alpha}$-tocopherol (Toc) were added to ground samples at a content level of 200 mg/kg. The samples were subjected to a chilling storage test before and after cooking. In the storage test before cooking, neither FX nor Toc affected the thiobarbituric acid reactive substances (TBARS) on days 1 and 6, and FX decreased the L* value and increased the a* and b* values. In the storage test after cooking, both FX and Toc decreased TBARS values on days 1 and 6. FX decreased the L* value and increased the a* and b* values, similar to what occurred in the storage test before cooking. Based on these results, we concluded that FX is a potent ingredient for improvement of the appearance and shelf life of chicken meat and its products.

Effect of Stem Bark Extract from Cudrania tricuspidata on the Concentrations of Lipid and Lipid Peroxidation in Rats Fed a Cholesterol Diet. (꾸지뽕나무 수피 추출물의 콜레스테롤 급여 흰쥐의 지질 농도 및 과산화지질 농도에 미치는 영향)

  • Male Sprageu-Dawely rats fed a cholesterol diet (Control group) or cholesterol diet supplemented with the water-soluble extract of stem bark from Cudrania tricuspidata(CTSB group) at the level of 0.5% for 2 weeks. Concentration of triglyceride in serum of CTSB group was significantly lower than that of control group. However, the other lipids concentrations of liver and serum, body weight gain, and food intake did not show significant difference between CTSB group and control group. Antioxidative activities of water-soluble extract from stem bark of Cudrania tricuspidata on the lipid peroxidation in rat were also studied in vivo by measuring the formation of thiobarbituric acid reactive substance(TBARS). Concentration of TBATS in the liver homogenates of CTSB group was significantly higher than that in control group. Nonheme iron concentration was significantly increased in the liver of CTSB group compared to control group. suggested that enhanced nonheme irom was associated with enhanced peroxidation of liver in CTSB group. These results suggested that water soluble extract from stem bark of Cudrania tricuspidata exert the hypotriglycerolemic effect and might amplify the lipid peroxidation of tissues in cholesterol fed rats.

Development of Resources for Functional Food and Biological Activity of Lentinus edoes mycelium (표고버섯 균사체의 생리활성과 기능성식품 소재 개발)

  • Objectives : This study was carried out to investigate an antioxidant ability, the change of antioxidant ability, antimicrobial activity and anticancer in functional meat ball and sausage prepared from Lentinus edoes mycelium and texturized soy protein. Methods : This experiments was carried out to investigate antioxidant ability(TBARS, DPPH, SOD-like ability), antibacterial activity and anticancer ability using sarcoma 180 extracts from Lentinus edoes mycelium. Result : Anticancer ability of Lentinus edoes mycelium showed a 28% survival rate and 63% of inhibition rate of tumor, which showed $1.30{\pm}0.4g$ of tumor weight. These results revealed an effective Lentinus edoes mycelium resources as anticancer sources. After heating peoducts prepared from Lentinus edoes mycelium, these products doesn't showed difference between after heating and before heating in measuring of SOD-like activity and DPPH. In DPPH experiment, ethanol extracts showed a high DPPH value as 90.85%, but hot water extracts showed 82.14% in DPPH value. Conclusion : In conclusion, it is very useful resources for preparing functional food on the basis of results from antioxidant(TBARS, DPPH, SOD-like ability), antibacterial activity and anticancer ability using sarcoma 180.
