Cell cultures of Taxus wallichiana Zucc. were made to enhance the production of anticancer agent paclitaxel. In suspension cultures, the maximum cell growth rate in exponential growth phase was 0.14 $day^{-1}$ which was correlated to 4.96 days of cell doubling time. The production of paclitaxel was non-growth associated. The paclitaxel production was started after the exponential growth phase and increased to declined phase where the maximum concentration was observed. Various elicitors were tested to enhance the production of paclitaxel. The combination of two elicitors of jasmonic acid and cellulase increased the production of paclitaxel 1.8 and 3.1 times compared to paclitaxel production by individual elicitor respectively.
This study was carried out for the purpose of supplying the basic data for artificial forestation, natural regeneration and ecological conservation etc., and obtaining information on alpine vegetation, by establishing vegetation units on the basis of phytosociological classification of community and studying growth pattern on the basis of species composition, hierarchy structure and population dynamics, for Taxes cuspidata naturally growing at the alpine districts in Korea. The importance value of Taxus cuspidata by districts mostly showed above 100 in the upper story but at Mt. Chiri and Mt. Hanra communities its value was comparatively low. In the middle story it showed lower value than that of upper story and not quite showed at Mt. Chiri. Taxus cuspidata communities in the lower story were only in Mt. Hanra, Mt. Odae and Mt. Sobaek, and their importance values were about 10 which were relatively low values. The communities were classified into five groups as Taxus cuspidata-Males baccata var. mandshurica, Taxus cuspidata-Abies nephrolepis, Taxus cuspidata-Abies koreana, Taxus cuspidata-Acer mono and Taxus cuspidata-Euonymus quelpaertensis by TWINSPAN analysis. Taxus cuspidata-Abies koreana community was distributed at the northern aspect of the mountain ridges and at higher elevation than other communities and distributed. Taxus cuspidata-Acer mono community was relatively low than the others and distributed at the hillsides of mountain. And Taxus cuspidata-Euonymus quelpaertensis communities were distributed at the relatively high elevation and northern and eastern aspect of the mountain tap, and Taxus cuspidata-Malus baccata var. mandshurica communities were distributed at the medium elevation, and southern and eastern aspect of the mountain ridge. In the relation between communities and environmental factors, it was correlated with aspect, elevation and topography at the first axis, and elevation, slope at the second axis.

Chemical investigation on a sample of the needles of Himalayan Taxus baccata has resulted in the isolation of several taxoids including taxol (1) 10-deacetyl-baccatin III (2) and 2-deacetoxytaxinine J (3) along with different lignans (6 and 7) and simple phenolics (8, 9, 10, 11 and 12). The occurrence of 4-(4'-hydroxyphenyl)-butane-2-one and 4-(4'-hydroxyphenyl)-trans-but 3-ene-2-one (8) in Taxus species is reported for the first time. The $^{13}C-NMR$ spectral data of two rearranged taxiod constituents, brevifoliol (4) and 13-decinnamoyltaxchinin B (5) are presented. The acid-catalyzed decomposition of taxol has been discussed. The synthesis of other two constituents, rhododendrol (10) and hibalactone (7) has been described.
13-Deacetyl-taxchinin I, a taxoid having a rearranged 11(15\longrightarrow1)-abeo-taxane skeleton, has been isolated and identified from plant cell cultures of Taxus chinensis. The compound has not previously been encountered in nature. Its structure was elucidated by 1-and 2D NMR techniques including H-H COSY, HMQC, and HMBC experiments. This taxoid also provides information for better understanding of structure-activity relationships and biosynthesis, as well as improving the quality control of paclitaxel production.
To investigate the structure and the conservation strategy of natural forest at Turobong-Sangwangbong area in Odaesan, 19 plots(10*10m) were set up with random sampling method. Several characteristics - needle injury, rate of fallen needle, stem injury, shoot and twig, apical dominance, tree form- of Abies nephrolepis and Tavus cuspidata were investigated with checklist. Two groups(Taxus cuspidata community and Quercus mongolica-Betula ermanii community) were classified by cluster analysis. High positive correlations were prover between Taxus cuspidata and Tripterygium regelii, Sorbus commixta; Quercus mongolica and Euonymus sieboldianus. High negative correlations were proved between Taxus cuspidata and Abies nephrolepis, Quercus mongolica and Betula ermanii; Abies nephrolepis and Trioterygum regelii. Species diversity(H') of investigated area was 0.9608-1.24990. Mean score of Abies nephrolepis was calculated at 6.1 point and ratio of dead individuals was 10.9%. Mean score of Taxus cuspidata was calculated at 17.3 point. Injury of Taxus cuspieata was classified three types, such as branch dieback, wound cavity on the stem and crooked stem.
For the proper tree management of Taxus cuspidata, distributed in the High 1 Ski Slope construction area in Baekwoonsan, Jeungsun-gun, Kangwon-do, the growing states of 323 trees(including transplanted 57trees) are investigated. Tree height, rootcollar diameter, D.B.H., stem condition, apical shoot, dead branch, needle growth condition, 2-year-old needle loss, root condition, tree form are investigated. Taxus cuspidata are mainly growing at the sites, high elevated mountain ridge or slopes facing north. Growing states of Taxus cuspidata, distributed in Baekwoonsan are relatively better than those of Taxus cuspidata and Abies koreana at other subalpine zone in Korea. Damaged trees are mainly due to root-removal through transplanting, root-damages by raising the ground level and digging. Several methods of Taxus cuspidata conservation were suggested.
Time course monitoring of FCW/DCW ratio, production of intra and extracellular protein, and peroxidase activity were performed during suspension culture of Taxus chinensis cells. The observed FCW/DCW ratio was 12 at day 14, which was the lowest value during cultivation, and the specific protein production, based on dry cell weight, was also the lowest at day 14, which showed 4.3 mg/g DCW. The pattern of POD activity was similar to that of protein production. The results in this report were obtained using actively growing cells in flasks, therefore it is possible to use those results to control the process and indicate the stresses imposed on cells during large-scale cultivation.
This study presents the negative pressure cavitation acetone-pentane fractional precipitation to dramatically improve the precipitation efficiency of paclitaxel from Taxus chinensis. When paclitaxel was precipitated under a negative pressure of -200 mmHg, most paclitaxel (>99.9%) could be recovered in a short precipitation time (5 min). The precipitation rate constant increased by 1.512~5.073 times (at -50 mmHg to -200 mmHg) compared to the control. The activation energy decreased by -3,737~-6,536 J/mol due to negative pressure, which increased the precipitation rate. With the introduction of negative pressure, the precipitate size decreased by 5.3 times, and the diffusion coefficient of paclitaxel increased by 7.0 times.
Antifungal substances were isolated and identified from garlic and taxus extracts to develop safe and broad fungicide. The inhibitory effect of seedborn disease of sesame, pepper, radish, chinese cabbage by formulation of antifungal substances was investigated. The antifungal substance isolated through column chromatography from garlic and taxus extracts was confirmed by GC-MS as allicin($C_6H_{10}OS_2$) and taxol($C_{47}H_{51}NO_{14}$) and the quantified content from each extracts by HPLC analysis was 0.62%, 0.29%, respectively. The formulation composed of garlic and taxus extracts controlled effectively the seedborn fungi tested in this study at 10X dilution, but at 100X dilution the inhibitory effect decreased. Phytotoxicity of these formulations did not recognized.
Yew (Taxus cuspidata Sibe) selected cultivation as drug, food and decorative plant in Gyong-gi province in Korea. To extract the water soluble active ingredients, as a extracting method, there was extracted with 20g of dried seeds with each 20g of butylene glycol(BG) and propylene glycol(PG), and 40 mL of water mixing 72 hours at $40{\pm}5^{\circ}C$, and then they were filtrated by 400 mesh. Appearance of extract of seeds was pale brown, $pH=4.5{\pm}0.5$, $gravity=1.013{\pm}0.05$, a reflective $index=1.373{\pm}0.05$, and yield=75%. Also, to extract the high purity oil from seeds, it minutely pulverized the dried seeds and added the hexane, mixing 2 hours at $20{\pm}57^{\circ}C$. And then, this filtrated it with 400-mesh. It got the purified oil through evaporating them at $55^{\circ}C$ during under vacuum. As the results, appearance was slightly brown, gravity=0.922 acid value=0.12, saponification value=192, and it should be obtained the $40{\pm}5%$ of yield. As the efficacy evalution of cosmetic field, the antioxidative activities by NBT method were stronger 86.0% from extract of talus seeds than 52.0% from green tea extract and 35.0% from skullcap extract as well as the antioxidative activities by DPPH method were stronger 93.7% from extract of seed than 60.3% from extract of green tea and 27.1% from extract of skullcap. These are more effective than other plant extracts. The collagen synthesis rate on the activating fibroblast for Taxus cuspidata Sibe extract showed 35.43%. As the activity of the skin elasticity, PPE(porcine pancreatic elastase)-inhibitory activities of talus extract was 50.8%. Anti-inflammatory activity was more effective to be taken 41.1% of taxus seed oil than 24.2% of steady glycyrrhetinate (SG) as a control.
