Studies on the Species Crossabilities in the Genus Pinus and Principal Characteristics of F1 Hybrids (일대잡종송(一代雜種松)의 교배친화력(交配親和力)과 특성(特性)에 관(關)한 연구(硏究))

  • By means of the interspecific hybridization in the Sub-genus Diploxylon of the Genus Pinus, $F_1$ hybrids of Pinus rigida${\times}$elliottii, Pinus rigida${\times}$radiata, P. rigida${\times}$serotina and P. densiflora${\times}$thunbergii had been produced. And on the basis of the crossabilities of these hybrids the taxonomic affinities of these pines were examined. And the needle characteristics of these hybrid and the occurence of phenolic substances in these $F_1$ hybrid were also investigated to see the potential usefulness of these characteristics for the diagnosis of the taxonomic affinity. And, the growth performances of the $F_1$ hybrids have also been compared with those of parental species. In order to contribute to the establishment of the hybrid seed orchard the introgression phenomena between P. densiflora and P. thunbergii in the eastern coastal area have also been investigated along with the investigation of the heterozygosity of plus trees of P. densiflora growing in the clone bank in Suwon. And the results were summarized as follows. 1. On the basis of crossabilities as well as on the taxonomic affinities according to the systems of Shaw, Pilger and Duffield, it has been proven that the parental species of those hybrids are of close affinities and range of the fertile hybrid seed production rate was as high as 28-58% in the best hybrid combination (Table 13). 2. Among those hybrids, the ${\times}$ Pinus, rigiserotina hybrid seemed to be most promising in the growth performance exhibiting 109-155% more volume growth compared to the seed parent with the statistic significance of 1% level (Tables 16 and 17). 3. Notwithstanding the fact that the all of the pollen parents are cold tender, all hybrids exhibit cold hardiness as much as their seed parent and it seems to suggest that the characteristics of cold hardiness were transmitted from the seed parent. 4. Though a striking difference in needle length was observed between the parental species of each hybrid, it was difficult to distinguish each hybrid from their seed parent by the needle length except ${\times}$P. rigiserotina which is characterized by long needle which is 65% more longer than the needle of the seed parent (Table 21). 5. With regard to the anatomical characteristics of needle, the hypoderm is apparently thicker in most of the $F_1$ hybrid pines and the characteristics of resin canals are dominated by medial in most $F_1$ hybrid. And, the fibrovascular bundles were apart as were in their seed parent. Therefore it was found to be possible to distinguish the hybrids pines from their parents by the needle characteristics. And, it is to be noticed that the ${\times}$P. densithunbergii was more close to the pollen parent having RDI value of 0.73 (Fig.l, Table 22). 6. It has been demonstrated that ${\times}$P. rigielliottii, ${\times}$P. rigiradiata and ${\times}$P. rigitaeda have a phenolic substance (No.7) of light yellow at Rf-0.46, same as their seed parent, but no trace of phenolic substance was observed in their pollen parent. This fact will serve as an important criteria for early identification of hybridity in progeny testing. However, the fact that both of ${\times}$P. rigiserotina and ${\times}$P. densithunbergii exhibit the same reactions of phenolic substances as well their parental species seems to indicate the close affinities between the parental species of the respective hybrid (Fig.2, Table 23). 7. The separation and the reaction of phenolic substance developed on TLC were found to be same in the same species showing no variations between the individuals, and no variations due to tree part of sampling, tree age or pollen sources. And the reaction was also observed regardless of the not varied by the kind of developing solvent whether it is Aceton-Chloroform (3:7 v/v) or Benzene-Methanol-Acetic acid (90:16:8 v/v). 8. The introgression phenomena of natural Pinus densifiora stand in both east and west coastal area indicates that the major part of the red pines investigated are all heterozygous and the heterozygosity of pines are higher in the west coast than in the east coast(Tables 24 and 25). 9. Based on the RDI, among the plus trees of Pinus densiflora selected in Korea and Japan as well, no pure P. densiflora has been found. Since all of the sample trees of Pinus densiflora were found to be as heterozygous bearing part of the characteristics of P. thunbergii, those red pines were considered to be natural heterotic hybrid pines(Figs. 3 and 4. Tables 26 and 27).

Phytoplankton Diversity and Community Structure Driven by the Dynamics of the Changjiang Diluted Water Plume Extension around the Ieodo Ocean Research Station in the Summer of 2020 (2020년 하계 장강 저염수가 이어도 해양과학기지 주변 해역의 식물플랑크톤 다양성 및 개체수 변화에 미치는 영향)

  • The expansion of the Changjiang Diluted Water (CDW) plume during summer is known to be a major factor influencing phytoplankton diversity, community structure, and the regional marine environment of the northern East China Sea (ECS). The discharge of the CDW plume was very high in the summer of 2020, and cruise surveys and stationary monitoring were conducted to understand the dynamics of changes in environmental characteristics and the impact on phytoplankton diversity and community structure. A cruise survey was conducted from August 16 to 17, 2020, using R/V Eardo, and a stay survey at the Ieodo Ocean Research Station (IORS) from August 15 to 21, 2020, to analyze phytoplankton diversity and community structure. The southwestern part of the survey area exhibited low salinity and high chlorophyll a fluorescence under the influence of the CDW plume, whereas the southeastern part of the survey area presented high salinity and low chlorophyll a fluorescence under the influence of the Tsushima Warm Current (TWC). The total chlorophyll a concentrations of surface water samples from 12 sampling stations indicated that nano-phytoplankton (20-3 ㎛) and micro-phytoplankton (> 20 ㎛) were the dominant groups during the survey period. Only stations strongly influenced by the TWC presented approximately 50% of the biomass contributed by pico-phytoplankton (< 3 ㎛). The size distribution of phytoplankton in the surface water samples is related to nutrient supplies, and areas where high nutrient (nitrate) supplies were provided by the CDW plume displayed higher biomass contribution by micro-phytoplankton groups. A total of 45 genera of nano- and micro-phytoplankton groups were classified using morphological analysis. Among them, the dominant taxa were the diatoms Guinardia flaccida and Nitzschia spp. and the dinoflagellates Gonyaulax monacantha, Noctiluca scintillans, Gymnodinium spirale, Heterocapsa spp., Prorocentrum micans, and Tripos furca. The sampling stations affected by the TWC and low in nitrate concentrations presented high concentrations of photosynthetic pico-eukaryotes (PPE) and photosynthetic pico-prokaryotes (PPP). Most sampling stations had phosphate-limited conditions. Higher Synechococcus concentrations were enumerated for the sampling stations influenced by low-nutrient water of the TWC using flow cytometry. The NGS analysis revealed 29 clades of Synechococcus among PPP, and 11 clades displayed a dominance rate of 1% or more at least once in one sample. Clade II was the dominant group in the surface water, whereas various clades (Clades I, IV, etc.) were found to be the next dominant groups in the SCM layers. The Prochlorococcus group, belonging to the PPP, observed in the warm water region, presented a high-light-adapted ecotype and did not appear in the northern part of the survey region. PPE analysis resulted in 163 operational taxonomic units (OTUs), indicating very high diversity. Among them, 11 major taxa showed dominant OTUs with more than 5% in at least one sample, while Amphidinium testudo was the dominant taxon in the surface water in the low-salinity region affected by the CDW plume, and the chlorophyta was dominant in the SCM layer. In the warm water region affected by the TWC, various groups of haptophytes were dominant. Observations from the IORS also presented similar results to the cruise survey results for biomass, size distribution, and diversity of phytoplankton. The results revealed the various dynamic responses of phytoplankton influenced by the CDW plume. By comparing the results from the IORS and research cruise studies, the study confirmed that the IORS is an important observational station to monitor the dynamic impact of the CDW plume. In future research, it is necessary to establish an effective use of IORS in preparation for changes in the ECS summer environment and ecosystem due to climate change.

  • It has intended to identify the members of the Genus Lespedeza in Korea by a chemical colour reaction, and the following five species of the Genus Lespedeza grown in the garden have been used in this experiment. 1. Lespedeza bicolor Turcz 2. Lespedeza bicolor var. melanantha (Nak.) T. Lee 3. Lespedeza cyrtobotrya Miq. 4. Lespedeza japonica var. intermedia Nakai 5. Lespedeza maritima Nakai 6. Lespedeza maximowiczii Schneider 7. Lespedeza maximowiczii var. tomentella Nakai A few drops of each solution of $K_2Cr_2O_7$. $FeSO_4{\cdot}7H_2O$, $FeCl_3$, $KH_2PO_4$, $KMnO_4$, $NH_4OH$, and HCl was added to the methanol extracts of wood dust to get the specific colour reaction. HCl-infused wood was also used for the identification of L. bicolor var. melanantha and L. bicolor. The results can be summarized as the following key; 1. Discrimination of African Yams Containing High Functional Compounds Using FT-IR Fingerprinting Combined by Multivariate Analysis and Quantitative Prediction of Functional Compounds by PLS Regression Modeling (FT-IR 스펙트럼 데이터의 다변량 통계분석을 이용한 고기능성 아프리칸 얌 식별 및 기능성 성분 함량 예측 모델링)

  • We established a high throughput screening system of African yam tuber lines which contain high contents of total carotenoids, flavonoids, and phenolic compounds using ultraviolet-visible (UV-VIS) spectroscopy and Fourier transform infrared (FT-IR) spectroscopy in combination with multivariate analysis. The total carotenoids contents from 62 African yam tubers varied from 0.01 to $0.91{\mu}g{\cdot}g^{-1}$ dry weight (wt). The total flavonoids and phenolic compounds also varied from 12.9 to $229{\mu}g{\cdot}g^{-1}$ and from 0.29 to $5.2mg{\cdot}g^{-1}$dry wt. FT-IR spectra confirmed typical spectral differences between the frequency regions of 1,700-1,500, 1,500-1,300 and $1,100-950cm^{-1}$, respectively. These spectral regions were reflecting the quantitative and qualitative variations of amide I, II from amino acids and proteins ($1,700-1,500cm^{-1}$), phosphodiester groups from nucleic acid and phospholipid ($1,500-1,300cm^{-1}$) and carbohydrate compounds ($1,100-950cm^{-1}$). Principal component analysis (PCA) and subsequent partial least square-discriminant analysis (PLS-DA) were able to discriminate the 62 African yam tuber lines into three separate clusters corresponding to their taxonomic relationship. The quantitative prediction modeling of total carotenoids, flavonoids, and phenolic compounds from African yam tuber lines were established using partial least square regression algorithm from FT-IR spectra. The regression coefficients ($R^2$) between predicted values and estimated values of total carotenoids, flavonoids and phenolic compounds were 0.83, 0.86, and 0.72, respectively. These results showed that quantitative predictions of total carotenoids, flavonoids, and phenolic compounds were possible from FT-IR spectra of African yam tuber lines with higher accuracy. Therefore we suggested that quantitative prediction system established in this study could be applied as a rapid selection tool for high yielding African yam lines.

A Taxonomic Reconsideration of the Genus Lemna L. (Lemnaceae) in Korea (한국산 좀개구리밥속(개구리밥과)의 분류학적 실체에 대한 재고)

  • Duckweed family (Lemnaceae Martinov), including the genus Lemna L., is a typical floating aquatic perennial plant, and about five genera and 40 species in the family are in wide distribution around the world except the polar regions. The genus Lemna is the smallest and the simplest plant among the angiosperms. It has a characteristic of doubling every three days with fast vegetative propagation, which helps the organisms to increase in rapid growth. As such, the plant is ideal for environmental pollution assessment and toxicity test. Although taxonomists and scholars have used different scientific names for the species, many of them have agreed that there is only one member of species of the genus Lemna in Korea. Paying attention to the external morphological variation observed in the Korean genus Lemna, we conducted a molecular phylogenetic analysis to identify the entity of the Korean Lemna species and to investigate the possibility of two or more members of the species existing in Korea. We determined and aligned the DNA sequences of the atpF-H region of the chloroplast DNA in 37 populations of the nationally distributed Lemna species. The results showed that the sequence length of the cp DNA atpF-H region was 463-483 bp, the length of the aligned sequences was 488 bp, and the number of variation site in nucleotide sequences was 47. There were two types of aligned sequences of the cp DNA atpF-H region from 37 populations of Lemna species in Korea. The maximum parsimony analysis revealed that the Korean Lemna consists of two clades, and one of them had two subclades. The results suggest that, contrary to the general understanding, at least two taxa (L.aequinoctialis, L.minor) exist in Korea.

The aplication of fuzzy classification methods to spatial analysis (공간분석을 위한 퍼지분류의 이론적 배경과 적용에 관한 연구 - 경상남도 邑級以上 도시의 기능분류를 중심으로 -)

  • Classification of spatial units into meaningful sets is an important procedure in spatial analysis. It is crucial in characterizing and identifying spatial structures. But traditional classification methods such as cluster analysis require an exact database and impose a clear-cut boundary between classes. Scrutiny of realistic classification problems, however, reveals that available infermation may be vague and that the boundary may be ambiguous. The weakness of conventional methods is that they fail to capture the fuzzy data and the transition between classes. Fuzzy subsets theory is useful for solving these problems. This paper aims to come to the understanding of theoretical foundations of fuzzy spatial analysis, and to find the characteristics of fuzzy classification methods. It attempts to do so through the literature review and the case study of urban classification of the Cities and Eups of Kyung-Nam Province. The main findings are summarized as follows: 1. Following Dubois and Prade, fuzzy information has an imprecise and/or uncertain evaluation. In geography, fuzzy informations about spatial organization, geographical space perception and human behavior are frequent. But the researcher limits his work to numerical data processing and he does not consider spatial fringe. Fuzzy spatial analysis makes it possible to include the interface of groups in classification. 2. Fuzzy numerical taxonomic method is settled by Deloche, Tranquis, Ponsard and Leung. Depending on the data and the method employed, groups derived may be mutually exclusive or they may overlap to a certain degree. Classification pattern can be derived for each degree of similarity/distance $\alpha$. By takina the values of $\alpha$ in ascending or descending order, the hierarchical classification is obtained. 3. Kyung-Nam Cities and Eups were classified by fuzzy discrete classification, fuzzy conjoint classification and cluster analysis according to the ratio of number of persons employed in industries. As a result, they were divided into several groups which had homogeneous characteristies. Fuzzy discrete classification and cluste-analysis give clear-cut boundary, but fuzzy conjoint classification delimit the edges and cores of urban classification. 4. The results of different methods are varied. But each method contributes to the revealing the transparence of spatial structure. Through the result of three kinds of classification, Chung-mu city which has special characteristics and the group of Industrial cities composed by Changwon, Ulsan, Masan, Chinhai, Kimhai, Yangsan, Ungsang, Changsungpo and Shinhyun are evident in common. Even though the appraisal of the fuzzy classification methods, this framework appears to be more realistic and flexible in preserving information pertinent to urban classification.

Effect of Soil Water Potential on Pysico-Chemical Properties of Soil and Cucumber(Cucumis sativus L.) Growth (토양(土壤) 수분(水分)포텐셜이 오이(Cucumis sativus L.)생육(生育)과 토양(土壤) 이화학적(理化學的 ) 특성(特性)에 미치는 영향(影響))

  • A field experiments was carried out to investigate the physicochemical properties of soil and cucumber growth in vinyl house when irrigation point was made at 0.2, 1/3, 0.5 and 1.0 bar. The obtained results was summarized as follow: The taxonomic class of the soil used was loam and each content of the required water was 4.4, 7.3, 9.6 and 13.4 mm per each irrigation time at 0.2, 1/3, 0.5 and 1.0 bar treatments in spring culture, respectively. At 0.2 bar and 1.0 bar treatments, interval of irrigation was 2.3 and 14.8 day, the times of irrigation was 37 and 6, and total irrigation volume was 163.5 and 80.3 mm, respectively. After cucumber culture, pH, EC concentration and exchangeable K content of soil at 0.2 bar treatment was distributed near to the level of improvement target while EC, available $P_2O_5$ and exchangeable base content in other treatments were higher compared to improvement target. At 1.0 bar treatment, ratios of the solid and liquid phase were 44.9 and 27.1%, respectively, and bulk density was $1.26g\;cm^{-3}$ which was the highest among the treatments. At 0.2 bar treatment, the ratio of the solid and liquid phase was 41.7 and 22.8%, respectively, and bulk density was $1.09g\;cm^{-3}$ which was the lowest. The root length and radius at 0.2 bar treatment were best, while those at 1/3 bar were worst. At 0.2 bar treatment, the total yield was 7,269 kg and the weight of good products was 5,677 kg which was the highest among treatments. At 0.33 bar treatment, the yield was the lowest with the high ratio of deformity.

Regrowth Ability and Species Composition of Phytoplankton in International Commercial Ship's Ballast Water Berthed at Pusan and Daesan Ports (부산과 대산항에서 선박평형수에 유입된 식물플랑크톤의 종조성과 재성장능력)

  • The aim of this study is to assess the importance of ballast water discharge as a vector for the introduction of exotic species into Pusan and Daesan Ports, Korea. We also examined to understand the impacts of environmental factors on the survival success of introduced species by ship's ballast water in laboratory experiments. Seven ship's ballast water originated from the coastal water of China (Taicang, Ningbo and Jinshan), Japan (Tokuyama, Moji and Akita), and Singapore. According to PCA (principal components analysis) analysis, environmental factor in the each ballast and shipside waters were different by bioregion. Based on cluster analysis, the phytoplankton community structures were distinguished for ballast water origin. Most of the major taxonomic groups were diatoms and, the others were dinoflagellate, silcoflagellate and several fresh-waters species. In particular, species number and standing crops of phytoplankton in the ballast tanks decreased with the increasing age ofballast water(r = -0.35 for standing crop; r = -0.63 for species number). In the laboratory study, although phytoplankton in ballast water treatment did not survive even in optimal temperature, the in vivo fluorescence of phytoplankton viability increased under the nutrient typical of shipside water and F/2 medium at $15^{\circ}C$ and $20^{\circ}C$. The diatoms species such as Skeletonema costatum and Thalassiosira pseudonana in ballast water were successfully regrown. On the salinity gradient experiments for Shui Shan (2) vessel, several freshwater species, brackish and marine species were successfully adapted. Of these, S.costatum was able to tolerate a wide range of salinities (10 to 30 psu) and its species-specific viability was suitable for colonization.

Isolation and Identification of Oceanisphaera sp. JJM57 from Marine Red Algae Laurencia sp. (Ceramiales: Rhodomelaceae) (해양 홍조류 Laurencia sp. (Ceramiales: Rhodomelaceae)에서 분리한 Oceanisphaera sp. JJM57의 분리 및 동정)

  • A taxonomic study was carried out to assess the phylogenetic characteristics of isolate JJM57 from marine red algae Laurencia sp. collected from intertidal zone in Jeju Island, South Korea. Comparative analysis of 16S rRNA gene sequence shows that this isolate belongs to the genus Oceanisphaera. It shows 98.02% and 97.7% sequence similarity with Oceanisphera litoralis DSM $15406^T$ and Oceanisphera donghaensis KCTC $12522^T$, respectively. Strain JJM57 is a Gram-negative, aerobic, moderately halophilic bacterium able to grow in different NaCl concentration ranges from 0.5 to 8.0% and at varying temperatures from 4 to $37^{\circ}C$. Sharing some of the physiological and biochemical properties with O. litoralis and O. donghaensis, JJM57 strain differs in the utilization of ethanol, proline, and alanine. The G+C contents of the strain JJM57 is 61.94 mol% and it is rich in $C_{16:1}$ ${\omega}7c$ and/or iso-$C_{15:0}$ 2-OH, $C_{16:0}$, and $C_{18:1}$ ${\omega}7c$ fatty acids. The DNA-DNA relatedness data separates the strain JJM57 from other species such as O. litoralis and O. donghaensis. On the basis of these polyphasic evidences, present study proposed that strain JJM57 (=KCTC 22371 =AM983543 =CCUG 60764) represents a novel bacterial species of Oceanisphaera.

  • From 1985 to 1993, we collected 20 isolates throughout Kangwon and obtained 6 isolates from other sources. A. mellea formed rhizomorph actively, and some of A. osroyae were poor in the formation of rhizomorph and some without formation of rhizomorph. A. tabescens was active in the growth of aerial mycelium and poor in the development of rhizomorph. In A. gallica, the mycelium development among the isolates were variable greatly, and especially in isolate A8(KNU-250), the mycelial development was similar to that of A. osroyae, but A8(KNU-250) showed the feature of A. gallica to change medium into brown color. In PCR-RFLP analysis of the IGS region in rDNA, the homology between each isolate in the A. mellea and A. ostoyae showed 100% homology. A. tabescens showed $0.919{\sim}0.974$ homology, and A. gallica showed $0.619{\sim}1.000$ homology. A19 and A12 showed 100% homology as the same group, but compared with other subgroups they showed less than 10% homology as $0.051{\sim}0.108$ value. In RAPD analyses, the isolates of A. mellea showed high homology among themselves as $0.983{\sim}1.000$, and A. ostoyae also showed high similarity. The homology between isolates of A. tabescens showed $0.594{\sim}0.953$ value because A. gallica showed $0.280{\sim}0.733$ value, and the variations between isolates were greater than other species. Especially, A19 and A22 were identified as new novel group which were remoted from other groups, and the homology between these two isolates showed 0.921 value, and the genetic similarity between these groups and other 4 groups showed less than 7% as $0.012{\sim}0.069$ value. Of 5 species identified in this study, 4 species were identified as A. mellea, A. ostoyae, A. tabescens, and A. gallica that were already reported ones and 1 species was suggested as a new specie in Korea.
