Isolation and Identification of 13-Deacetyl-taxchinin I, a New Taxoid from Plant Cell Cultures of Taxus chinensis (식물세포 Taxus chinensis 배양으로부터 신물질 13-Deacetyl-taxchinin I의 분리 및 동정)

  • 13-Deacetyl-taxchinin I, a taxoid having a rearranged 11(15\longrightarrow1)-abeo-taxane skeleton, has been isolated and identified from plant cell cultures of Taxus chinensis. The compound has not previously been encountered in nature. Its structure was elucidated by 1-and 2D NMR techniques including H-H COSY, HMQC, and HMBC experiments. This taxoid also provides information for better understanding of structure-activity relationships and biosynthesis, as well as improving the quality control of paclitaxel production.

A Case of Subungal Abscess and Onycholysis Induced by Docetaxel (Docetaxel 투여 후 발생한 조갑하 농양 및 조갑 박리증 1예)

  • Docetaxel is a taxoid antineoplastic drug, which is widely used to treat locally advanced or metastatic non-small cell lung cancer (NSCLC). Among the adverse dermatological reactions, nail disorders such as bending, onycholysis, hypoor hyperpigmentation are rare. We report a case of a 62-year-old male with advanced NSCLC (cT4N3M1, stage IV), who developed purulent discharge and onycholysis in the nail of all his fingers and the left great toe after five courses of anti-neoplastic chemotherapy, which included docetaxel (cumulative dose: $370mg/m^2$, 590 mg). Seven days after the final session of chemotherapy, the patient had become aware of discoloration and swelling of the nail beds with out pain. Three days later, greenish-yellow purulent discharge oozed out from the involved nails. Microbiologic studies revealed Pseudomonas aeruginosa. Intravenous and topical antibiotics (mupirocin) were applied. After 2 weeks, regrown nails were observed and the onycholysis had improved.

  • Chemical investigation on a sample of the needles of Himalayan Taxus baccata has resulted in the isolation of several taxoids including taxol (1) 10-deacetyl-baccatin III (2) and 2-deacetoxytaxinine J (3) along with different lignans (6 and 7) and simple phenolics (8, 9, 10, 11 and 12). The occurrence of 4-(4'-hydroxyphenyl)-butane-2-one and 4-(4'-hydroxyphenyl)-trans-but 3-ene-2-one (8) in Taxus species is reported for the first time. The $^{13}C-NMR$ spectral data of two rearranged taxiod constituents, brevifoliol (4) and 13-decinnamoyltaxchinin B (5) are presented. The acid-catalyzed decomposition of taxol has been discussed. The synthesis of other two constituents, rhododendrol (10) and hibalactone (7) has been described.

  • In developing a HPLC purification process, it was hoped that a single chromatographic system would be sufficient to abtain pure paclitaxel in high yield. However, no such system was found, due in part to the complex taxoid profile of crude paclitaxel and to the rigorous nature of the product specification. A two step HPLC purification was adopted using reverse-phase separation on C(sub)18 as a first step, and normal-phase separation on silica as the final polishing step. Impurity profiles were established and maintained for paclitaxel, which identified and quantified each impurity observed in purified paclitaxel from these two steps, all impurities at or above 0.1% were identified. Results provide information for improving the quality control of paclitaxel production.
