Effects of Taurine Supplementation on Mitochondrial Function in Chronic Ethanol Administered Rats

  • The present investigation was undertaken in vivo to determine whether the functional alterations of hepatic mitochondria induced by ethanol might be prevented by taurine. We examined the effects of supplementation of taurine on hepatic mitochondrial oxidative phosphorylation in the chronic ethanol-administered rats. Isolated hepatic mitochondria from three groups of rats were functionally tested by an analysis of $\beta-hydroxbutyrate-supported$ respiration and the coupling of this process to ATP synthesis in the presence of ADP. The three groups were control group(CO), ethanol(60g/L) administered group (AL), and ethanol (60g/L) + taurine (5g/L) supplemented group (AT). Ethanol and/or taurine were given in drinking water for 10 weeks. The mitochondria from AL group had lower state 4 respiratory rate, respiratory control (RC) ratio and ADP : O(P/O) ratio than those from CO and AT group. It showed that the ethanol administered rats were less coupled and thus less efficient with respect to mitochondrial ATP synthesis than both control rats and ethanol + taurine supplemented rats. It suggests that taurine supplementation might improve the impaired oxidative phosphorylation efficiency in mitochondrial dysfunction that is recognized as a cause of liver diseases in chronic ethanol consumption.

  • The experiments was conducted to investigate the effects of dietary supplementation of feather meal and pyridoxine on the taurine content of egg yolk and performance of laying hens. Feeding trial was conducted with 900 31-wk-old Hy-Line Brown layers for 4wks. The experiment consisted of six dietary treatments: control(basal diet), feather meal(FM) 3 % diet(FM 3 %), FM 3 % + pyridoxine supplemented diet(FM 3 % + Pyridox), FM 6 % diet, FM 6 % + pyridoxine supplemented diet(FM 6% + Pyridox), Synthetic taurine 0.25 % supplemented diet(Taurine). Egg production of birds fed FM 3% was highest and those of the FM diets were also higher than those of Taurine and the control. Egg weight of Taurine was significantly lower than those of FM 3 %, FM 6 % and the control but were not significantly different from those of FM 3 % + Pyridox or FM 6 % + Pyridox. Feed intake of the control was greater than those of FM 6 %, FM 6% + Pyridox or Taurine treatment but was not significantly different from those of FM 3 % and FM 3 % + Pyridox. Feed conversion of the control was significantly higher than other treatments in which that of FM 6 % was lowest. Broken and soft egg production of Taurine was highest while that of the control was lowest among treatments. Taurine content of egg yolk significantly increased by supplementation of taurine(64.7 %). FM 6 % + pyridoxine(57 %), FM 3% + pyridoxine (32.1%) and FM 6% (16.6 %). Sensory evaluation data of Taurine has shown the highest score in most of sensory attributes. It is concluded that taurine can be enriched in egg yolk by supplementation of 6 % FM diet and pyridoxine as well as 0.25 % synthetic taurine.

Taurine Enrichment of Eggs with Feather Meal and Pyridoxine

  • The effects of dietary supplementation of feather meal (FM) and pyridoxine ($B_6$) on the taurine content of egg yolk and performance of laying hens were investigated. A feeding trial was conducted in nine hundred 31-wk-old $Hy-Line^{\circledR}$ Brown layers over 4 wk. The hens received 6 dietary treatments: Control, FM 3% supplemented diet (FM 3%), FM 3%+$B_6$ supplemented diet (FM 3%+$B_6$), FM 6% supplemented diet (FM 6%), FM 6%+$B_6$ supplemented diet (FM 6%+$B_6$), and synthetic taurine 0.25% supplemented diet (Taurine). Parameters of production were significantly (p<0.05) affected by treatments. The egg production of hens fed FM 3% was the highest and hens fed FM diets were more productive than the Taurine and Control groups. The egg weights of the Taurine group were significantly lower than those of the FM 3% and FM 6% groups, but not significantly different from those of other treatments. The feed intake of the Control group was highest among all groups. The feed conversion ratio of the Control group was higher than in groups receiving other treatments of which FM 6% was the lowest. The broken egg production of the Taurine group was highest, while that of the Control group was lowest among treatments. The taurine content of egg yolk was significantly (p<0.01) increased by supplementation of taurine (64.7%), FM 6%+$B_6$ (57%), FM 3%+$B_6$ (32.1%), and FM 6% (16.6%) over a 4 wk average. Sensory evaluation data of the Taurine group showed the highest score in all of the sensory attributes and those of other treatments were not significantly (p<0.05) different. In conclusion, taurine can be enriched in egg yolk by supplementation of 6% FM and $B_6$, as well as 0.25% synthetic taurine.

육계 사료내 우모 Digest 첨가시 생산성과 계육내 Taurine 함량에 미치는 영향 (The Effects of Dietary Supplementation of Feather Meal Digests on the Performances and Muscular Taurine Contents in Broiler Chickens)

  • 우모분(FM)과 우모분 digests가 육계의 성장률과 계육(가슴살, 다릿살) 및 심장의 taurine함량에 미치는 영향을 조사하기 위해 갓 부화한 Ross${\circledR}$ 병아리 100수를 5처리 2반복 반복당 10수씩(암 수 5수씩) 배치하여 6주간 사양시험을 실시하였다. 시험사료는 기초사료(control) 에 FM 5% 첨가사료(FM), NaOH 처리 FM 5% 첨가사료(NaOH-FM), $HNO_3$ 처리 FM5% 첨가사료$(HNO_3$-FM) 그리고 합성 타우린0.5% 첨가사료(Taurine)를 시험사료로 공시하여 42일간 실시하였으며 21일령까지는 대조구사료를 모두에게 급여하였고, 처리 사료는 22일령부터 42일령까지 급여하였다. 증체량에 있어 처리구들은 control에 비해 낮았지만 유의차가 없었다. 사료 섭취량은 Taurine과 FM 급여구가 대조구에 비해 낮았고 NaOH-FM과 $HNO_3$-FM구들이 FM구 보다는 높은 경향이 있었다. 사료요구율은 NaOH-FM구가 FM과 $HNO_3$-FM구 보다 유의하게 높았고 대조구와 Taurine구와는 유의한 차이가 없었다. 대조구의 taurine 함량을 보면 심장근육이 1482$\mu$g/g 으로 가장 높았고 다리근육이 778$\mu$g/g이었고 가슴살이 79$\mu$g/g으로 낮았다. 다리살과 가슴살의 경우 처리간에 유의차(P<0.01)가 있었으나 심장에서는 유의차가 없었다. 다리살의 경우 control에 비해 처리구들의 taurine 함량이 유의하게 높았는데 Taurine 첨가구에서는 170%, FM구는 123%, NaOH-FM구는 122% 그리고$HNO_3$-FM구는 63% 높았다. 가슴 근육의 타우린 함량은 Taurine구가 타처리구들에 비해 유의하게 높았고 대조구에 비해서는 246% 높았다 성장의 경우에는 처리구들간에 차이가 없었다. 결론적으로 육계후기 사료에 우모분을 5% 첨가했을 때, 다리근육의 타우린 함량은 효과적으로 강화시킬수 있었으며 우모분을 NaOH 나 $HNO_3$로 처리하는 것은 taurine 강화에 도움이 되지 않았다.

The Effect of Dietary Supplementation of Feather Meal on the Performance and Muscular Taurine Contents in Growing-finishing Pigs

  • An experiment was conducted to investigate the effects of dietary supplementation of feather meal (FM) on the performance of pigs and taurine content in pork. A total of 135 cross-bred (L${\times}$Y${\times}$D) pigs, weighing an average 46 kg, were assigned to five dietary treatments; Control, 3% FM supplemented diet (3% FM), 3% FM and 10 ppm pyridoxin supplemented diet (3% FMpyridoxin), 6% FM supplemented diet (6% FM) and 6% FM and 10 ppm pyridoxine supplemented diet (6% FM-pyridoxin). Each treatment had 3 replications of 9 pigs (4 gilts and 5 barrows) each. Pigs were fed for 11 weeks under a phase feeding program which consisted of grower (0-3 week), grow-finisher (4-8 week) and finisher (9-11 week) periods. During the whole feeding period, ADG and ADFI of pigs fed FM treatments tended to increase in general compared to the control. Feed conversion ratio (feed/gain) of the control was significantly (p<0.05) lower than FM treatments. Taurine content of the control was highest in heart muscle (1,393.8 ppm) followed by liver (647.3 ppm), tenderloin (601.2 ppm), ham (462.4 ppm) and loin (375.8 ppm). Taurine contents of heart, tenderloin, ham and loin were significantly (p<0.05) increased by FM treatments. Taurine content was highest in the 6% FM-pyridoxin treatment. Taurine content of heart, tenderloin, ham and loin on 6% FM-pyridoxin supplementation were increased by 91%, 23%, 27% and 29%, respectively, compared with the control. Sensory tests of the pork showed that there was no consistent response among the treatments. In conclusion, supplementation of FM and pyridoxin to a pig diet can increase taurine content of the pork.

Taurine supplementation in diet for olive flounder at low water temperature

  • The objective of this study was to examine the effect of dietary supplementation of taurine for juvenile olive flounder (Paralichthys olivaceus) at low water temperature ($16.4{\pm}0.36^{\circ}C$). Fish meal (FM)-based diet was used as the control diet. Four other experimental diets were prepared by adding taurine to FM-based diet at 0.25, 0.50, 1.00, and 1. 50% (T1, T2, T3, and T4, respectively). Each experimental diet was fed to triplicate groups of fish (initial mean body weight, 19.5 g) for 10 weeks. At the end of the feeding trial, growth performance and feed utilization, hematological parameters, non-specific immune responses, whole-body proximate composition, and liver mRNA expression of insulin-like growth factor-1 (IGF-1) were investigated. Feed conversion ratio was significantly reduced while protein efficiency ratio was significantly increased in taurine-supplemented groups. Hematocrit and hemoglobin were also significantly increased while plasma cholesterol levels were decreased in taurine-supplemented groups than those in the control group. Nitroblue-tetrazolium, myeloperoxidase and lysozyme activities, and plasma immunoglobulin level were significantly increased by taurine supplementation. These results suggest that dietary taurine supplementation is effective in improving growth performances, feed utilization, and innate immunity of olive flounder in low water temperature season.

  • The effects of dietary supplementation of feather meal digests(FM) and its digests on the growth of broiler chicks and taurine content in the broiler meat were examined. Total of 100 broiler chickens were assigned to five dietary treatments: T1; Control, T2; feather meal(FM) 5 % diet, T3; NaOH treated FM 5% diet, T4; HNO$_3$treated FM 5 % diet and T5; synthetic taurine 0.5 % supplemented diet. Taurine content of leg muscle was significantly(P<0.01) increased by treatments. The highest increase over the control was shown by 0.5 % taurine diet(170 %), followed by FM diet(123 %), NaOH treated FM diet(122 %) and HNO$_3$treated FM diet(63 %). Taurine content of breast muscle was increased by 246 % in 0.5 % taurine diet but FM diets were not significantly different from the control. Taurine content of heart muscle was not significantly affected by dietary treatments. There were big differences in the average taurine content of the parts or organ of the control birds; 778 $\mu\textrm{g}$/g leg muscle, 79 $\mu\textrm{g}$/g breast muscle and 1482 $\mu\textrm{g}$/g heart muscle. It was concluded that taurine content of leg muscle of broiler can be increased by supplementation of feather meal. Alkaline or acid treatment FM was not effective in improving taurine enrichment of the broiler meat.

우모분 및 우모분 Digest 첨가가 우유 내 Taurine 함량에 미치는 영향 (Effects of Dietary Supplementation of Feather Meal and It's Digest on Taurine Content of Cow Milk)

  • This study, consisting of three experiments, was conducted to determine the effects of feeding feather meal (FM), feather meal digest (FMD), L-cystine and methionine hydroxyl analogue (MHA) on taurine content of milk and milk production of Holstein dairy cows. In experiment 1, FM or FMD was supplemented at 0, 1, 3 and 5% of dry matter intake (DMI), respectively. Taurine concentration of 3% FM and 5% FMD treatment were increased by 14% and 220/0, respectively. The 5% FM treatment had a negative effect on milk yield and FM and FMD treatments had no significant or consistent effects on milk fat, protein, lactose, milk urea nitrogen (MUN) and somatic cell count (SCC). In experiment 2, Lcystine or MHA was supplemented at 0, 1, 3, and 5g or ml/d along with 5% FMD, respectively. Milk yield decreased at 3 and 5g or ml Lcystine or MHA supplementation along with 5% FMD. Fat and lactose in milk were not significantly affected by treatments. However, milk protein level increased significantly in the 5 ml HMA with 5% FMD treatment. SCC decreased significantly in 1ml MHA with 5% FMD supplemented treatment but increased in 5g Lcystine with 5% FMD and 5 ml MHA with 5% FMD treatments. Increase of milk taurine concentration of L'cystine with 5% FMD treatments was not significant but those of MHA with 5% FMD treatments were significantly higher than the control. The highest increase of milk taurine concentration was 65% shown in 1 ml MHA with 5% FMD treatment. In experiment 3, 5% FM, 5% FM+3% molasses or 5% FM+3% molasses+l ml MHA was supplemented to the based TMR diet. The molasses treatments (5% FM+3% molasses and 5% FM+3% molasses+l ml MHA) showed significantly higher milk taurine content than the 5% FM treatment. The molasses treatments significantly reduced MUN but increased SCC. It was concluded that FMD is more effective than FM in enriching taurine in milk. Maximum taurine enrichment (65%) in the milk was obtained by supplementation of 5% FMD/DM1+1 ml MHA/d/cow. Molasses supplementation to 5% FM diet increased milk taurine content. However, MHA supplementation in dairy cows increased ruminal escape, gastrointestinal absorption and response of serum methionine.

Effects of dietary taurine supplementation on plasma and liver lipids in OVX rats fed calcium-deficient diet

  • Taurine supplementation has been shown to have an effect on lowering blood lipids in ovariectomized (OVX) rats. It therefore seemed desirable to find out whether the beneficial effect of taurine on OVX rats fed calcium-deficient diet could also be reproduced. Forty female Sprague-Dawley rats were divided into two groups. One group was OVX and the other group received a sham operation (Sham). Each rat group was further divided into the control diet and the taurine supplemented (2.0g/100g diet) diet group. All rats were fed on calcium-deficient diet and deionized water ad libitum for 6 weeks. Plasma and liver lipids were determined by using commercial kits. LDL-cholesterol concentrations were estimated with the equation of Friedewald et al. (1972). There were no significant differences in body weight gain and food intake between the control and taurine group within Sham and OVX groups, but body weight gain, food intake, and food efficiency ratio was higher in the OVX group. Concentrations of plasma total cholesterol, triglyceride, LDL-cholesterol were significantly lower in the taurine fed group of OVX rats fed Ca deficient diet, while HDL-cholesterol concentration was increased in the taurine fed group. Therefore, in this study, we examined whether taurine also prevented hypercholesterolemia induced by ovarian hormone deficiency in ovariectomized rats when they were fed a calcium-deficient diet. These results indicate that taurine may have some beneficial effects on hypercholesterolemia and hypertriglyceridemia in OVX rats fed calcium-deficient diet.

Effects of taurine on plasma and liver lipids, erythrocyte ouabain sensitive Na efflux and platelet aggregation in Sprague Dawley rats

  • The effects of taurine on plasma and liver cholesterol, erythrocyte ouabain sensitive Na efflux and platelet aggregation were examined in Sprague Dawley rats fed control or 0.5% cholesterol with 0.2% cholate diet. Plasma and liver levels of total cholesterol were increased significantly (p<0.05) in rats fed cholesterol diet compared to the control, and taurine significantly decreased the elevated plasma level of cholesterol in rats fed cholesterol diet (p<0.05). HDL-cholesterol was decreased in groups fed the cholesterol diet regardless of taurine supplementation and the difference between groups with and without cholesterol was significant (p<0.01). Plasma triglyceride was decreased and liver triglyceride was increased both significantly (p<0.05) in rats fed cholesterol compared to the control. Plasma and liver triglyceride in rats fed taurine was decreased significantly compared to the control (p<0.05). Intracellular Na tended to be lower in rats fed cholesterol or taurine and higher in rats fed cholesterol plus taurine compared to the control. Na efflux through Na-K ATPase and the passive leak of Na was somewhat reduced in rats fed cholesterol or taurine and was augmented in rats fed cholesterol plus taurine compared to the control, which showed a similar trend to the intracellular Na. Taurine supplementation caused a suppression of Na efflux in groups fed control diet and restored the suppressed Na efflux in groups fed cholesterol. Platelet aggregation was significantly decreased in the group fed taurine compared to the control (p<0.05) and the group fed cholesterol plus taurine was also a little lower in aggregation than the group fed cholesterol. Microscopic examination showed that taurine prevented fatty liver in rats fed cholesterol diet. Taurine known for stimulating Na-K ATPase in some cell types rather decreased erythrocyte ouabain sensitive Na-K ATPase in the present study. Taurine had hypolipidemic and hypocholesterolemic effects and inhibited platelet aggregation which may be favorable for prevention of cardiovascular diseases.
